Sanchez-Fortún: Writing – original draft, Investigation, Formal analysis., The physical and chemical properties of silver nanoparticles (AgNPs) have led to their increasing use in various fields such as medicine, food, and industry. Evidence has proven that AgNPs cause adverse effects in aquatic ecosystems, especially when the release of Ag is prolonged in time. Several studies have shown short-term adverse effects of AgNPs on freshwater phytoplankton, but few studies have analysed the impact of long-term exposures on these populations. Our studies were carried out to assess the effects of AgNPs on growth rate, photosynthesis activity, and reactive oxygen species (ROS) generation on the freshwater green algae Scenedesmus armatus and the cyanobacteria Microcystis aeruginosa, and additionally on microcystin (MC-LR) generation from these cyanobacteria. The tests were conducted both in single-species cultures and in phytoplanktonic communities exposed to 1 ngL-1 AgNPs for 28 days. The results showed that cell growth rate of both single-species cultures decreased significantly at the beginning and progressively reached control-like values at 28 days post-exposure. This effect was similar for the community-cultured cyanobacteria, but not for the green algae, which maintained a sustained decrease in growth rate. While gross photosynthesis (Pg) increased in both strains exposed in single cultures, dark respiration (R) and net photosynthesis (Pn) decreased in S. armatus and M. aeruginosa, respectively. These effects were mitigated when both strains were exposed under community culture conditions. Similarly, the ROS generation shown by both strains exposed in single-species cultures was mitigated when exposure occurred in community cultures. MC-LR production and release were significantly decreased in both single-species and community exposures. Acetaminophen (APAP)-induced liver injury is one of the most prevalent causes of acute liver failure (ALF). We assessed the role of the bone morphogenetic protein (BMP) type I receptors ALK2 and ALK3 in APAP-induced hepatotoxicity. The molecular mechanisms that regulate the balance between cell death and survival and the response to oxidative stress induced by APAP was assessed in cultured human hepatocyte-derived (Huh7) cells treated with pharmacological inhibitors of ALK receptors and with modulated expression of ALK2 or ALK3 by lentiviral infection, and in a mouse model of APAP-induced hepatotoxicity. Inhibition of ALK3 signalling with the pharmacological inhibitor DMH2, or by silencing of ALK3, showed a decreased cell death both by necrosis and apoptosis after APAP treatment. Also, upon APAP challenge, ROS generation was ameliorated and, thus, ROS-mediated JNK and P38 MAPK phosphorylation was reduced in ALK3-inhibited cells compared to control cells. These results were also observed in an experimental model of APAP-induced ALF in which post-treatment with DMH2 after APAP administration significantly reduced liver tissue damage, apoptosis and oxidative stress. This study shows the protective effect of ALK3 receptor inhibition against APAP-induced hepatotoxicity. Furthermore, findings obtained from the animal model suggest that BMP signalling might be a new pharmacological target for the treatment of ALF.

Inflammation is related to several pathological processes. The aim of this study was to investigate the protein expression of the different subunits of the nuclear factor Kappa b (NFkBp65, p50, p105, p52, p100) and the protein expressions of IkB beta and alpha in the hearts from a murine model of accelerated aging (SAM model) by Western blot. In addition, the translocation of some isoforms of NFkB from cytosol to nuclei (NFkBp65, p50, p52) and ATP level content was studied. In addition we investigated the effect of the chronic administration of growth hormone (GH) on these age-related parameters. SAMP8 and SAMR1 mice of 2 and 10 months of age were used (n = 30). Animals were divided into five experimental groups: 2 old untreated (SAMP8/SAMR1), 2 young control (SAMP8/SAMR1) and one GH treated-old groups (SAMP8). Age-related changes were found in the studied parameters. We were able to see decreases of ATP level contents and the translocation of the nuclear factor kappa B p50, p52 and p65 from cytosol to nuclei in old SAMP8 mice together with a decrease of IKB proteins. However p100 and p105 did not show differences with aging. No significant changes were recorded in SAMR1 animals. GH treatment showed beneficial effects in old SAMP8 mice inducing an increase in ATP levels and inhibiting the translocation of some NFkB subunits such as p52. Objective: The aim of the present study was to investigate the effect of aging on several parameters related to glucose metabolism, proliferation and differentiation in the pancreas and how GH administration to old SAMP8 mice could affect these parameters. Materials and methods: Pancreas samples were obtained from two types of male mice models: senescence-accelerated prone (SAMP8) and senescence-accelerated-resistant (SAMR1) mice SAMP8 and SAMR1 mice and the influence of exogenous administration of GH (2mgs.c./kg/day) on SAMP8 mice. RNA was isolated from pancreas samples of male mice using the kit RNeasy total RNA kit Ref. 50974104 (Qiagen). Insulin was measured in plasma by RIA kit and glucose was measured in plasma by an assay kit. Results: Aging decreases the expression of differentiation in the pancreas of Pdx-1, FoxO 1 and FoxO 3A but not of Sirt 1 or of the expression of the proliferative genes PCNA and Sei1. The expression of glucagon and GLUT2 were increased with aging and no differences were observed in somatostatin and insulin expressions. Insulin levels in plasma were increased with aging in SAMP8 mice. IGF-1 expression was reduced with aging. The treatment with GH was able to increase the expression of Sirt 1, Pdx-1, FoxO 3A and IGF-1. On the other hand, the treatment decreased the expression of glucagon, GLUT2, somatostatin and insulin, furthermore GH was able to decrease the plasma levels of insulin in old male SAMP8 mice (p<0.0004). Conclusion: The present study has shown that aging is associated with significant alterations in the relative expression of pancreatic genes involved in insulin secretion as well as in the differentiation and in the intra islet glucose metabolism. Mulching and organic soil amendment are two agricultural practices that are being increasingly used to preserve soil from degradation, although they may modify the fate of herbicides when applied in soils subjected to these practices. This study has set out to compare the impact of both agricultural practices on the adsorption-desorption behaviour of the herbicides S-metolachlor (SMOC), foramsulfuron (FORAM), and thiencarbazone-methyl (TCM) involving winter wheat mulch residues at different stages of decomposition and particle size, and unamended soils or those amended with mulch. The Freundlich Kf adsorption constants of the three herbicides by mulches, and unamended and amended soils ranged between 1.34 and 65.8 (SMOC), 0-34.3 (FORAM), and 0.01-1.10 (TCM). The adsorption of the three compounds was significantly higher in mulches than in soils (unamended and amended). The adsorption of SMOC and FORAM increased significantly with mulch decomposition, with this positive impact also being observed on the adsorption of FORAM and TCM after mulch milling. Simple and multiple correlations between mulches, soils, and herbicide properties, and adsorption-desorption constants (Kf, Kd, Kfd) reflected the organic carbon (OC) content and/or dissolved organic carbon (DOC) content of the adsorbents as main variables controlling the adsorption and/or desorption of each herbicide. The statistic R2 revealed that >61 % of the variability in the adsorption-desorption constants could be explained by jointly considering the OC of mulches and soils and the hydrophobicity (for Kf) or water solubility of herbicides (for Kd or Kfd). The same trend observed for Kfd desorption constants as for Kf adsorption ones resulted in higher percentages of herbicide remaining adsorbed after desorption in amended soils (33 %-41 % of SMOC, 0 %-15 % of FORAM, and 2 %-17 % of TCM) than in mulches (< 10 %). The changes of physicochemical and biochemical parameters of a silty loam (S1) and sandy loam (S2) vineyard soils added with spent mushroom substrate (SMS) or SMS composted with ophite (OF) as rock dust (SMS + OF) were studied. Two doses of SMS or SMS + OF (25 and 100 Mg ha-1) were applied for two consecutive years (2020-2021) and changes of soil physicochemical parameters, and dehydrogenase activity (DHA), respiration (RES), microbial biomass (BIO), and the phospholipid fatty acids (PLFAs) profile were assayed on a temporal basis. The results showed an increase in soil organic carbon (OC) content, total and mineralised N, P, and K, especially when the highest SMS dose was applied to soils. Repeated application caused OC content over time up to 2.3 times higher than initial content in the silty loam soil. This increase was not observed in sandy soil, possibly due to a higher bioavailability of OC, as indicated by the evolution of extractable humic acid/fulvic acid pools. In both soils, all biochemical parameters increased after amendment, being favoured both by the OC and by the presence of OF. Significant positive correlations were found between DHA, RES and BIO, and OC content especially in the first part and then levelled off after the second dose application. Total bacterial or fungal PLFAs patterns reflected the variation of BIO by SMS application. The higher growth of fungi vs. bacterial community in amended soils was recorded after the first SMS application, although the opposite effect occurred after the second application, with similar results in both soils. The findings indicate that the application of SMS or SMS + OF in vineyard soils could be an appropriate agronomic management practice for maintaining soil sustainability, although doses and application times of these amendments should first be evaluated depending on soil texture.

Prion diseases are characterized by the self-assembly of pathogenic misfolded scrapie isoforms (PrPSc) of the cellular prion protein (PrPC). In an effort to achieve a theranostic profile, symmetrical bifunctional carbazole derivatives were designed as fluorescent rigid analogues of GN8, a pharmacological chaperone that stabilizes the native PrPC conformation and prevents its pathogenic conversion. A focused library was synthesized via a four-step route, and a representative member was confirmed to have native fluorescence, including a band in the near-infrared region. After a cytotoxicity study, compounds were tested on the RML-infected ScGT1 neuronal cell line, by monitoring the levels of protease-resistant PrPSc. Small dialkylamino groups at the ends of the molecule were found to be optimal in terms of therapeutic index, and the bis-(dimethylaminoacetamido)carbazole derivative 2b was selected for further characterization. It showed activity in two cell lines infected with the mouse-adapted RML strain (ScGT1 and ScN2a). Unlike GN8, 2b did not affect PrPC levels, which represents a potential advantage in terms of toxicity. Amyloid Seeding Assay (ASA) experiments showed the capacity of 2b to delay the aggregation of recombinant mouse PrP. Its ability to interfere with the amplification of the scrapie RML strain by Protein Misfolding Cyclic Amplification (PMCA) was shown to be higher than that of GN8, although 2b did not inhibit the amplification of human vCJD prion. Fluorescent staining of PrPSc aggregates by 2b was confirmed in living cells. 2b emerges as an initial hit compound for further medicinal chemistry optimization towards strain-independent anti-prion compounds.

Understanding the impact of land use and land cover change on surface energy and water budgets is increasingly important in the context of climate change research. Eddy covariance (EC) methods are the gold standard for high temporal resolution measurements of water and energy fluxes, but cannot resolve spatial heterogeneity and are limited in scope to the tower footprint (few hundred meter range). Satellite remote sensing methods have excellent coverage, but lack spatial and temporal resolution. Long-range unmanned aerial systems (UAS) can complement these other methods with high spatial resolution over larger areas. Here we use UAS thermography and multispectral data as inputs to two variants of the Two Source Energy Balance Model to accurately map surface energy and water fluxes over a nutrient manipulation experiment in a managed semi-natural oak savanna from peak growing season to senescence. We use energy flux measurements from 6 EC stations to evaluate the performance of our method and achieve good accuracy (RMSD ≈ 60 W m− 2 for latent heat flux). We use the best performing latent heat estimates to produce very high-resolution evapotranspiration (ET) maps, and investigate the drivers of ET change over the transition to the senescence period. We find that nitrogen and nitrogen plus phosphorus treatments lead to significant increases in ET (P < 0.001) for both trees (4 and 6%, respectively) and grass (12 and 9%, respectively) compared to the control. These results highlight that the high sensitivity and spatial and temporal resolution of a UAS system allows the precise estimation of relative water and energy fluxes over heterogeneous vegetation cover.

2023 Acuerdos transformativos CRUE, Zero-valent nano-iron particles (nZVI) are increasingly present in freshwater aquatic environments due to their numerous applications in environmental remediation. However, despite the broad benefits associated with the use and development of nZVI nanoparticles, the potential risks of introducing them into the aquatic environment need to be considered. Special attention should be focused on primary producer organisms, the basal trophic level, whose impact affects the rest of the food web. Although there are numerous acute studies on the acute effects of these nanoparticles on photosynthetic primary producers, few studies focus on long-term exposures. The present study aimed at assessing the effects of nZVI on growth rate, photosynthesis activity, and reactive oxygen activity (ROS) on the freshwater green algae Scenedesmus armatus and the cyanobacteria Microcystis aeruginosa. Moreover, microcystin production was also evaluated. These parameters were assessed on both organisms singly exposed to 72 h-effective nZVI concentration for 10% maximal response for 28 days. The results showed that the cell growth rate of S. armatus was initially significantly altered and progressively reached control-like values at 28 days post-exposure, while M. aeruginosa did not show any significant difference concerning control values at any time. In both strains dark respiration (R) increased, unlike net photosynthesis (Pn), while gross photosynthesis (Pg) only slightly increased at 7 days of exposure and then became equal to control values at 28 days of exposure. The nZVI nanoparticles generated ROS progressively during the 28 days of exposure in both strains, although their formation was significantly higher on green algae than on cyanobacteria. This research presents a case study of the biases and discontinuities that were introduced in observed long-term mean wind-speed and gust data-series due to anemometer changes in a meteorological station in northern Spain, operated by the Spanish State Meteorological Agency: San Sebastian-Igueldo. Field and wind-tunnel experiments with predefined conditions have been presented in the literature, however this research uses a real case study to assess the impact of anemometer changes on wind speed measurements due to three factors being: (i) the 3-cup anemometer model (SEAC vs. THIES companies); (ii) sensor height (∼19.95 m vs. ∼20.45 m) and (iii) sensor age (20-years old vs. new). Our results show (a) substantial biases in the measured wind speed and daily peak wind gusts, with the new THIES anemometer reporting stronger surface winds than the old SEAC anemometer; (b) opposing biases under weak (negative) and moderate-strong (positive) winds; and (c) significant breakpoints in the long-term wind data-series, which highlight the importance of data homogenization. National Weather Services and climate assessment groups will benefit from these findings since errors in wind speed and gust measurements can be minimized by implementing systematic observation protocols. Robust anemometer observations provide a basis for accurate quantification of the magnitude of changes and the variability of surface winds.

Many satellite missions rely on modeling approaches to acquire global or regional evapotranspiration (ET) products. However, a current challenge in ET modeling lies in dealing with sub-pixel heterogeneity, as models often assume homogeneous conditions at the pixel level. This is particularly an issue for heterogeneous landscapes, such as tree-grass ecosystems (TGE). In these areas, while appearing homogeneous at larger spatial scales pertaining to a single land cover type, the separation of the spectral signals of the main landscape features (e.g. trees and grasses) may not be achieved at the conventional satellite sensor resolution (e.g. 10–1000 m). This leads to important heterogeneity within the pixel grid that may not be accounted for in traditional modeling frameworks. This study examined the effect of pixel heterogeneity on ET simulations over a complex TGE in central Spain. High resolution hyperspectral imagery from five airborne campaigns forced the two-source energy balance (TSEB) model at 1.5–1000 m spatial resolutions. Along with this, the sharpened (20 m) and original (1000 m) Sentinels for Evapotranspiration (Sen-ET) products were evaluated over the study site for 2017. Results indicated that TSEB accurately simulated ET (RMSD: ~60 W/m2) when the pixel scale was able to robustly discriminate between grass and tree pixels (<5 m). However, model uncertainty drastically increased at spatial resolution greater than 10 m (RMSD: ~115 W/m2). Model performance remains relatively constant between 30 and 1000 m spatial resolutions, with within pixel heterogeneity being similar at all these scales. For mixed pixels (≥30 m), forcing an effective landscape roughness into TSEB (RMSD: ~80 W/m2) or applying a seasonally changing TSEB (TSEB-2S; RMSD: ~65 W/m2) improved the modeling performance. Wildfire occurrence is expected to increase in future climate and Land Use Land Cover (LULC) change scenarios, especially in vulnerable areas as the European Mediterranean Basin. In this study future probability of wildfire occurrence was estimated for a 20-year time period (2041–2060, centered on 2050) by applying a statistically-based regression model using LULC-derived contact areas with the forest cover (interfaces) as proxy for the human-related factor and a combination of Live Fuel Moisture Content and seasonal climate-related variables as predictors. Future wildfire occurrence was mapped under RCP 8.5 high emissions scenario in four Spanish regions with heterogeneous socioeconomic, LULC and natural fire-related characteristics at 1 km2 target spatial resolution. Results showed increased wildfire probability in ∼19–73% of 1 km2 cells, observing regional differences in the variable effects. This approach could be applied to other spatial scales offering tools for planning and management actions and to obtain different possible future scenarios.

S-metolachlor is a widely used herbicide that may contaminate groundwater when applied to irrigated crops, especially when the soil has a low organic carbon (OC) content. The objective here was to assess the capacity of two organic wastes, namely, green compost (GC) and pelletised organo-mineral manure fertilizer (PM), applied to two soils (S) with different textures at a rate of 10% dry weight to modify the fate of S-metolachlor. The herbicide's Freundlich adsorption coefficient (Kf) increased within a range of 3.2–8.2 times in S + GC and 3.8–6.8 times in S + PM. A positive correlation between adsorption and OC and the coefficient of variation of the OC normalised adsorption coefficients (Kfoc) higher than 20% indicated the evident influence on this process of soil OC content and its nature. The increase in adsorption did not prevent the dissipation of S-metolachlor in the amended soils, although the degradation rate decreased up to ~2 times or was not significantly modified across the different soil types. The S-metolachlor metabolites, metolachlor ethane sulfonic acid and metolachlor oxanilic acid, were detected in the herbicide's dissipation in the unamended soils, but they were not detected in the amended soils. The mobility experiments indicated leached amounts of S-metolachlor higher than 50% in unamended soil. The amounts decreased 1.1–1.7 times and 1.7–1.8 times in the S + GC and S + PM when a saturated flow was applied. Moreover, breakthrough curves indicated a slow leaching kinetics of herbicide in amended soils, with low concentrations continuously detected in the leachates together with a decrease in the maximum peak concentration. The results show the effect of the application of organic wastes especially in sandy soils to promote the immobilisation and/or degradation of S-metolachlor, avoiding its transfer to other environmental compartments.

Remote sensing capabilities to monitor evergreen broadleaved vegetation are limited by the low temporal variability in the greenness signal. With canopy greenness computed from digital repeat photography (PhenoCam), we investigated how canopy greenness related to seasonal changes in leaf age and traits as well as variation of trees’ water fluxes (characterized by sap flow and canopy conductance). The results showed that sprouting leaves are mainly responsible for the rapid increase in canopy green chromatic coordinate (GCC) in spring. We found statistically significantly differences in leaf traits and spectral properties among leaves of different leaf ages. Specifically, mean GCC of young leaves was 0.385 ± 0.010 (mean ± SD), while for mature and old leaves was 0.369 ± 0.003, and 0.376 ± 0.004, respectively. Thus, the temporal dynamics of canopy GCC can be explained by changes in leaf spectral properties and leaf age. Sap flow and canopy conductance are both well explained by a combination of environmental drivers and greenness (96% and 87% of the variance explained, respectively). In particular, air temperature and vapor pressure deficit (VPD) explained most of sap flow and canopy conductance variance, respectively. Besides, GCC is an important explanatory variable for variation of canopy conductance may because GCC can represent the leaf ontogeny information. We conclude that PhenoCam GCC can be used to identify the leaf flushing for evergreen broadleaved trees, which carries important information about leaf ontogeny and traits. Thus, it can be helpful for better estimating canopy conductance which constraints water fluxes.

Cumulative evidence indicates the relevance of cocoa in the modulation of cardiometabolic diseases. Nevertheless, pure cocoa may be rejected by many consumers due to its bitter taste. Carob (Ceratonia siliqua L.) is a Mediterranean legume with pods rich in dietary fibre and polyphenols. In this study, carob flour was combined with cocoa, to obtain a blend which was subjected to nutritional evaluation (proximate composition, carbohydrate and fatty acid profiles, phytochemical composition, dietary fibre properties, antioxidant capacity) and sensory analysis, in order to determine whether it may be a suitable alternative to commercial sugar-rich soluble cocoa powders. The blend showed a high content of dietary fibre (56%), methylxanthines and polyphenols (55% as non-extractable proanthocyanidins). The product had good solubility and was less bitter than cocoa, as evaluated in sensory analysis, thereby suggesting that this blend may be acceptable to consumers, particularly when information about potential biological activity is provided (a significant improvement in taste evaluation was observed). The beneficial environmental impact of carob and the advantages of valorizing the commonly discarded carob pod mean that further studies of applications of this blend and its potential health effects would be of great interest.

Providing accurate information on fire effects is critical to understanding post-fire ecological processes and to design appropriate land management strategies. Multispectral imagery from optical passive sensors is commonly used to estimate fire damage, yet this type of data is only sensitive to the effects in the upper canopy. This paper evaluates the sensitivity of full waveform LiDAR data to estimate the severity of wildfires using a 3D radiative transfer model approach. The approach represents the first attempt to evaluate the effect of different fire impacts, i.e. changes in vegetation structure as well as soil and leaf color, on the LiDAR signal. The FLIGHT 3D radiative transfer model was employed to simulate full waveform data for 10 plots representative of Mediterranean ecosystems along with a wide range of post-fire scenarios characterized by different severity levels, as defined by the composite burn index (CBI). A new metric is proposed, the waveform area relative change (WARC), which provides a comprehensive severity assessment considering all strata and accounting for changes in structure and leaf and soil color. It showed a strong correlation with CBI values (Spearman's Rho = 0.9 ± 0.02), outperforming the relative change of LiDAR metrics commonly applied for vegetation modeling, such as the relative height of energy quantiles (Spearman's Rho = 0.56 ± 0.07, for the relative change of RH60, the second strongest correlation). Logarithmic models fitted for each plot based on the WARC yielded very good performance with R2 (± standard deviation) and RMSE (± standard deviation) of 0.8 (± 0.05) and 0.22 (± 0.03), respectively. LiDAR metrics were evaluated over the King Fire, California, U.S., for which pre- and post-fire discrete return airborne LiDAR data were available. Histamine, one of the most toxic and commonly encountered biogenic amines (BA) in food, is produced by the microbial decarboxylation of histidine. It may accumulate at high concentrations in fish and fermented food. Cheese has some of the highest histamine concentrations, the result of the histidine-decarboxylase activity of certain lactic acid bacteria (LAB). The present work describes the nucleotide sequence and transcriptional organization of the gene cluster responsible for histamine biosynthesis (the HDC cluster) in Lactobacillus vaginalis IPLA 11064 isolated from cheese. The influence of histidine availability and pH on histamine production and the expression of the HDC cluster genes is also examined. As expected, the results suggest that the production of histamine under acidic conditions improves cell survival by maintaining the cytosol at an appropriate pH. However, the transcriptional regulation of the HDC cluster is quite different from that described in other dairy histamine-producing LAB, probably due to the lack of a termination-antitermination system in the histidyl-tRNA synthetase gene (hisS).

This paper reports the mobility and total balance of chlorotoluron (CTL), flufenacet (FNC) and bromide ion (Br−) throughout a sandy soil profile after the application of spent mushroom substrate (SMS) and green compost (GC). Obtaining mobility dataset is crucial to simulate the herbicides' fate under amended soil scenarios by application pesticide leaching models with regulatory application (FOCUS models). The application of organic residues is nowadays increased to improve the crop yields and there is a gap in the simulations of this kind of amended scenarios. A two-year field experiment involving unamended soil (S) and SMS- or GC-amended soil plots was conducted. CTL, FNC, and Br− were annually applied and their residual concentrations were determined in soil profiles (0–100 cm) regularly sampled. In all the treatments the order of mobility is followed as FNC < CTL < Br−. SMS and GC increased herbicide retention in the top 10 cm by the higher organic carbon (OC) content than the unamended soil, and their ability to increase the soil's water-holding capacity and to decrease water percolation. Simultaneously dissolved organic carbon (DOC) content facilitated herbicide transport being it favoured by the initial soil moisture content and the rainfall shortly after the chemicals' initial application. Over the first year, residual amounts (<2.6%) of Br−, CTL and FNC were leached down to 90–100 cm depth in the three treatments. However, over the second year low CTL and FNC amounts (<1.0%) reached the bottom layer only in S + SMS although high Br− concentrations did so in the three treatments (<20%). According to the total balance of Br−, CTL, and FNC in the soil profiles other processes (degradation, mineralisation, bound residues formation, and/or crop uptake) different from leaching below 1 m depth might play a key role in their dissipation especially in the amended soil profiles. This study sets out to evaluate the effect on the leaching of prosulfocarb through packed soil columns of applying green compost (GC) as an organic amendment (20% w/w), herbicide ageing over 28 days in the soil (incubation vs. no incubation), and two different irrigation regimes (saturated or saturated-unsaturated flows). Peak concentrations decreased after herbicide incubation in the columns for both unamended (S) and amended (S + GC) soils under both flow regimes. The leached amounts decreased when the herbicide was incubated for 28 days in S (2.1 and 1.9 times) and S + GC (2.9 and 1.6 times), under saturated or saturated-unsaturated flow, respectively. In the S columns, the total amounts retained (43.3%–60.8%) were lower than the ones obtained for the S + GC columns under saturated flow (77.4%–85.2%), suggesting a stronger interaction between the herbicide and the GC-amended soil. This behaviour was not observed under saturated-unsaturated flow, as the total amounts retained were similar in both the S and S + GC columns. Prosulfocarb was primarily retained in the first segment of the S (>28%) and S + GC (>43%) columns under all conditions. Incubation time did not greatly affect the herbicide retention, but it significantly increased the mineralized amount under saturated flow. The total balances of 14C-prosulfocarb were >73% and >80% in the S and S + GC columns, respectively, indicating that amendment decreased prosulfocarb loss by volatilization. Several factors, such as amendment, herbicide ageing and water flow, proved to be important for controlling the leaching of this herbicide through the soil profile.

Addition of organic residues to soil is a current farming practice but it is not considered in the modelling studies for pesticide risk assessment at regulatory level despite its potential impact on the pesticide dynamics in soil. Thus, the objective of this work was to examine and to compare the ability of PRZM and MACRO pesticide fate models to simulate soil water content, and bromide (Br−, tracer), chlorotoluron and flufenacet concentrations in the soil profiles (0–100 cm) of one agricultural soil, unamended (control soil, S), amended with spent mushroom substrate (S + SMS) or amended with green compost (S + GC). Based on a two-year field-scale dataset, the models were first calibrated against measurements of water and solutes contents in the soil profiles (first year) and then tested without any further model calibration by comparison with the field observations of the second year. In general, the performance of MACRO to simulate the whole dataset in the three soil treatments was higher than that of PRZM. MACRO simulated satisfactorily the water dynamics along the soil profiles whereas it was poorly described by the capacity model PRZM. Both models predicted very well the Br− mobility in control and amended soils after dispersion parameters were fitted to observations. No calibration was necessary to reproduce correctly herbicides vertical distribution in the control soil profile. In the amended soils, MACRO simulations were highly correlated to the observed vertical distribution of flufenacet and chlorotoluron, but calibration of the Kd of chlorotoluron was needed. On the contrary, modelling with PRZM required calibration of Kd and DT50 of both herbicides to obtain an acceptable agreement between observations and predictions in the amended soils. The dissipation and persistence of two cereals herbicides, chlorotoluron and flufenacet, were studied in a field experiment including three replicated plots of unamended soil (S), soil amended with spent mushroom substrate (S + SMS), and soil amended with green compost (S + GC), during the winter wheat cultivation campaign. The SMS and GC organic residues were applied to the soil at rates of 140 or 85 t residue ha−1, and herbicides were sprayed as Erturon® and Herold® formulations for chlorotoluron and flufenacet, respectively. Concentrations of both herbicides and of their metabolites were regularly measured in the three soil treatments (0–10 cm) from 0 to 339 days. The dissipation kinetics fitted well the single first order (SFO) model, except that of chlorotoluron that fitted the first order multi-compartment (FOMC) model better in the unamended soil. The dissipation rates of herbicides were lower in amended than in unamended soils. The results also showed that the DT50 of chlorotoluron (66.2–88.0 days) and flufenacet (117–145 days) under field conditions were higher than those previously obtained at laboratory scale highlighting the importance of the changing environmental conditions on the dissipation process. Similarly, the formation of chlorotoluron and flufenacet metabolites under field conditions was different from that previously observed in the laboratory. The performance of the MACRO pesticide fate model, parameterized with laboratory data, was then tested against field data. There was a very good agreement between measured and simulated chlorotoluron residue levels in the three soil treatments, while the ability of the model to reproduce the dissipation of flufenacet was good in the unamended soil and very good in S + SMS and S + GC soils. MACRO might be used to estimate the remaining amounts of herbicides in amended soils from degradation data previously obtained at laboratory scale. The spread of organic pollutants from soil to other environments is one important source of environmental pollution. The addition of organic amendments to soil is an interesting strategy to control pollutants leaching. However, the contribution of different carbon types of organic amendments to organic pollutants adsorption is not clear. Hence, the objective of this work was to determine the role of carbon types of organic amendments into the adsorption of four herbicides. To this extent, organic amendments were characterized by elemental analysis and 13C-NMR and adsorption–desorption isotherms of herbicides by the organic amendments and two soils amended with them were obtained. Adsorption coefficients were correlated with the organic carbon content of the organic amendments and the adsorption process was enhanced by the hydrophobicity of herbicides and the aliphatic and aromatic carbon of amendments. Organic amendments increased the adsorption of herbicides by soils but it is not possible to extrapolate results from one soil to another because organo-mineral interactions between soils and organic amendments can modify this process. Desorption isotherms of herbicides from organic amendments and/or amended soils presented hysteresis indicating the irreversible adsorption of herbicides. Desorption results indicated, the abundance of O-alkyl and N-alkyl groups in organic amendments enhanced the hysteresis in amended soils

The most recent efforts to provide remote sensing (RS) estimates of plant function rely on the combination of Radiative Transfer Models (RTM) and Soil-Vegetation-Atmosphere Transfer (SVAT) models, such as the SoilCanopy Observation Photosynthesis and Energy fluxes (SCOPE) model. In this work we used ground spectroradiometric and chamber-based CO2 flux measurements in a nutrient manipulated Mediterranean grassland in order to: 1) develop a multiple-constraint inversion approach of SCOPE able to retrieve vegetation biochemical, structural as well as key functional traits, such as chlorophyll concentration (Cab), leaf area index (LAI), maximum carboxylation rate (Vcmax) and the Ball-Berry sensitivity parameter (m); and 2) compare the potential of the of gross primary production (GPP) and sun-induced fluorescence (SIF), together with up-welling Thermal Infrared (TIR) radiance and optical reflectance factors (RF), to estimate such parameters. The performance of the proposed inversion method as well as of the different sets of constraints was assessed with contemporary measurements of water and heat fluxes and leaf nitrogen content, using pattern-oriented model evaluation. The multiple-constraint inversion approach proposed together with the combination of optical RF and diel GPP and TIR data provided reliable estimates of parameters, and improved predicted water and heat fluxes. The addition of SIF to this scheme slightly improved the estimation of m. Parameter estimates were coherent with the variability imposed by the fertilization and the seasonality of the grassland. Results revealed that fertilization had an impact on Vcmax, while no significant differences were found for m. The combination of RF, SIF and diel TIR data weakly constrained functional traits. Approaches not including GPP failed to estimate LAI; however GPP overestimated Cab in the dry period. Updated and harmonized land cover (LC) data is essential for wildfire estimation in fire-prone areas as is the case in southern Europe. CORINE Land cover (CLC) and ESA Climate Change Initiative Land Cover (CCI-LC) maps have been analyzed and compared their performance in the estimation of wildfire occurrence in Europe at regional and local scales for the period 2010–2014. LC maps legends were harmonized and similarities and discrepancies were compared. Overall agreement between the two maps for the whole Europe was ˜75%. Forest and agriculture showed the largest agreement, while shrubland and grassland the lowest. Quantity and allocation disagreements were calculated including exchange and shift components (Pontius and Santacruz, 2014) which provided detailed information about the contribution of each class to the overall disagreement. Spatial discrepancies were found in areas where grassland and shrubland were the dominant classes as in United Kingdom or East Turkey. Land Use and Coverage Area frame Survey (LUCAS) was used as ground truth for validation purposes. The agreement with LUCAS was slightly higher for CCI-LC (59%) than for CLC (56%). Generalized Linear Models (GLM), based on presence-absence of wildfires, were used to estimate wildfire occurrence at 3 × 3 km grid cell resolution from both LC maps at the European scale. LC interfaces and climatic variables (temperature and precipitation) where used as explicative variables while fires from European Forest Fire Information System EFFIS (2010–2014 period) were used as response variable. Wildfire occurrence was also estimated with the two maps at local scale in a test region (Zamora, Spain) using a more precise location of the response variable (x, y fire ignition points). Soil is a non-renewable resource, and its degradation compromises human health, 2 natural ecosystems, and even the climate. The application of organic amendments and 3 herbicides is commonplace in agriculture, and their impact on soil fertility needs to be 4 evaluated. Therefore, the objective was to evaluate the effect on soil microbial activity 5 and structure of amendments, sewage sludge (SS) and green compost (GC), and the rate 6 of herbicide pethoxamid applied (2, 10 and 50 mg kg-1). Herbicide dissipation kinetics, 7 soil dehydrogenase activity (DHA) and the profile of phospholipid-derived fatty acids 8 (PLFAs) extracted from the soil have been determined in unamended (S) and amended 9 (S+SS and S+GC) soils. The dissipation curves of pethoxamid applied at the three rates 10 closely fitted a single first order kinetics model in all the soils. The dissipation rate 11 decreased with the rate applied in the order 2 mg kg-1 > 10 mg kg-1 > 50 mg kg-1 in 12 unamended and amended soils. However, the half-life or time required for 50% 13 dissipation (DT50) of pethoxamid was not significantly different in unamended and 14 amended soils when applied at 2 and 10 mg kg-1, but it was lower in the amended soils 15 than in the unamended one when the herbicide was applied at the highest rate. The 16 highest DHA mean values were obtained in S+GC treated with pethoxamid at 2, 10 and 17 50 mg kg-1; however, DHA was lower in S+SS than in S. Peak DHA values were 18 observed in S and S+GC soils treated with pethoxamid at 2 mg kg-1 at 15 days of 19 incubation, and in S, S+GC and S+SS treated with the herbicide at 10 and 50 mg kg-1 at 20 35 days of incubation. These peak DHA values are close to 50% of herbicide 21 dissipation. A statistical analysis of the PLFA results has revealed significant effects for 22 sampling time in all the soils, for the pethoxamid rate, and for the interaction between 23 time and pethoxamid rate only in S+GC and S+SS. Adding organic amendments to soil could modify the bioavailability of herbicides and lead to changes in the microbial community’s activity and structure. The objective here was to study the dissipation and total mass balance of 14C-labeled prosulfocarb applied at two rates (4 and 10 mg kg-1) in unamended and green compost (GC)-amended soil. Soil dehydrogenase activity (DHA) and phospholipid fatty acid (PLFA) profile analysis were determined to evaluate the effect of herbicide residues on microbial community´s activity and structure over the dissipation period. The dissipation rate of prosulfocarb decreased after soil amendment due to higher herbicide adsorption by the amended soil. The 50% dissipation time (DT50) increased 1.7 times in the unamended soil when the concentration of prosulfocarb increased 2.5 times. The mass balance results indicate that the sum of water and organic extractable fractions represented the highest amounts up to the dissipation of 50% 14C-prosulfocarb. The 14C-herbicide was then mainly mineralized (up to 11%-31%) or formed non-extractable residues (up to 35%-44%). The amount of 14C-prosulfocarb residues extracted with methanol was slightly higher in amended soils than in unamended ones. 14C-prosulfocarb mineralization was higher in unamended soils than in amended ones. The formation of non-extractable residues was continuous, and increased over time. Soil DHA decreased in the unamended soil and was maintained in the GC-amended soil at the end of the assay. The microbial structure was barely disturbed over the prosulfocarb degradation process, although it was clearly influenced by the application of GC. The results obtained reveal the influence organic amendment has on herbicide bioavailability to decrease its biodegradation and buffer its impact on the soil microbial structure.

In the last twenty years, pesticide use in Chile has increased more than 160%, generating a greater risk of water resources pollution. The objective of this study was to assess the presence of 22 pesticides and 12 degradation products in surface water samples from the Cachapoal River basin, Central Chile, an area characterized by intense agricultural activity. Pesticide concentrations in the dissolved phase (DP) and particulate phase (PP) in samples collected in the dry season and after precipitation events was assessed. The solid-phase extraction technique was used to preconcentrate the samples and GC/MS and LC/MS were used to detect pesticides. The results present spatio-temporal variations in the proportion and concentration of pesticides and their degradation products in both the DP and PP for each site and sampling period. The most ubiquitous compounds in the dissolved phase were atrazine, atrazine-2-hydroxy (HA), cyprodinil, pyrimethanil, and tebuconazole, while in the particulate phase HA, imidacloprid, diazinon and pyrimidinol were detected. The results presented in this study make up the first record of pesticides in the dissolved and particulate phases in surface water in Chile. They show that the problem of pesticide contamination undoubtedly affects the quality of bodies of water in agricultural areas in Chile and support the need for a proper assessment of the water quality of the Cachapoal River in the future.

Soil microbiology could be affected by the presence of pesticide residues during intensive farming, potentially threatening the soil environment. The aim here was to assess the dissipation of the herbicides triasulfuron and prosulfocarb, applied as a combined commercial formulation, and the changes in soil microbial communities (through the profile of phospholipid fatty acids (PLFAs) extracted from the soil) during the dissipation time of the herbicides under field conditions. The dissipation of herbicides and the soil microbial structure were assessed under different agricultural practices, such as the repeated application of herbicides (twice), in unamended and amended soils with two organic amendments derived from green compost (GC1 and GC2) and with non-irrigation and irrigation regimes. The results obtained indicate slower dissipation for triasulfuron than for prosulfocarb. The 50% dissipation time (DT50) decreased under all conditions for the second application of triasulfuron, although not for prosulfocarb. The DT50 values for both herbicides increased in the GC2 amended soil with the highest organic carbon (OC) content. The DT50 values decreased for prosulfocarb with irrigation, but not for triasulfuron, despite its higher water solubility. The herbicides did not have any significant effects on the relative population of Gram-negative and Gram-positive bacteria during the assay, but the relative abundance of Actinobacteria increased in all the soils with herbicides. At the end of the assay (215 days), the negative effects of herbicides on fungi abundance were significant (p < 0.05) for all the treatments. These microbiological changes were detected in non-irrigated and irrigated soils, and were more noticeable after the second application of herbicides. Actinobacteria could be responsible for the modification of herbicide degradation rates, which tend to be faster after the second application. [EN] Sea level rise due to climate change, as well as social pressure to decrease the visual impact of coastal structures, have led to reduced crest freeboards, and this increases the overtopping hazard. In previous studies, pedestrian safety during overtopping events was assessed considering the overtopping layer thickness (OLT) and the overtopping flow velocity (OFV). This study analyzed the statistics of OLT and OFV on mound breakwaters without crown walls during severe wave storms. Small-scale 2D physical tests were conducted on mound breakwaters with dimensionless crest freeboards between 0.29 and 1.77, testing three armor layers (single-layer Cubipod (R), and double-layer cubes and rocks) in depth-limited breaking wave conditions and with two bottom slopes. Neural Networks were used to develop new estimators for the OLT and OFV exceeded by 2% of the incoming waves with a high coefficient of determination (0.866 < R-2 < 0.876). The best number of significant figures in the empirical coefficients of the new estimators was determined according to their variability. The 1 parameter Exponential and Rayleigh distribution functions were proposed to estimate the extreme values of OLT and OFV with 0.803 < R-2 < 0.812, respectively.

Highlights • Carbon dioxide + propane mixtures are studied through accurate measurements of speeds of sound and phase equilibria. • Heat capacities and virial coefficients are obtained from speeds of sound. • Retrograde condensation zone is found for the (0.60 CO2 + 0.40 C3H8) mixture. • Results are compared with reference equations of state such as GERG-2008 and AGA8-DC92. This work presents phase envelope and speed of sound data for the (0.60 CO2 + 0.40 C3H8) and (0.80 CO2 + 0.20 C3H8) binary mixtures. Phase equilibria was measured using a cylindrical resonator working in the microwave band whereas an acoustic resonator was used for speed of sound measurements. The experimental results were compared with GERG-2008 equation of state, obtaining average absolute deviations by 0.24% in pressure for phase equilibria data and 0.025% for speed of sound data. Speed of sound values were used to derive perfect-gas heat capacities, acoustic virial coefficients, and second density virial coefficients. In addition, AGA8-DC92 equation of state performance was checked for the results derived from speeds of sound.

The coupling of radiative transfer, energy balance, and photosynthesis models has brought new opportunities to characterize vegetation functional properties from space. However, these models do not accurately represent processes in ecosystems characterized by mixtures of green vegetation and senescent plant material (SPM), in particular grasslands. These inaccuracies limit the retrieval of vegetation biophysical and functional properties. Green and senesced plants feature contrasting spectral properties and carry out different functions that current coupled models do not represent separately. Besides, senescent pigments' absorption features change as SPM decomposes, and neither is this process well parameterized in radiative transfer models. This manuscript aims at overcoming these limitations. On the one hand, we have developed senSCOPE, a version of the Soil-Canopy Observation of Photosynthesis and Energy fluxes (SCOPE) that separately represents light interaction and physiology of green and senesced leaves. On the other, we have characterized new specific absorption coefficients of senescent pigments (Ks) from optical measurements of SPM from a Mediterranean grassland. Sensitivity analyses revealed that compared to SCOPE, senSCOPE 1) predicts variables that respond more linearly to the faction of green leaf area; and 2) keeps high levels of absorbed photosynthetically active radiation in the green leaves, which leads to significant differences in leaf photosynthesis, non-photochemical quenching, and transpiration. Moreover, we compared SCOPE vs. senSCOPE's capability to provide estimates of functional and biophysical parameters of vegetation. A major international effort has been made to monitor sun-induced chlorophyll fluorescence (SIF) from space as a proxy for the photosynthetic activity of terrestrial vegetation. However, the effect of spatial heterogeneity on the SIF retrievals from canopy radiance derived from images with medium and low spatial resolution remains uncharacterised. In images from forest and agricultural landscapes, the background comprises a mixture of soil and understory and can generate confounding effects that limit the interpretation of the SIF at the canopy level. This paper aims to improve the understanding of SIF from coarse spatial resolutions in heterogeneous canopies by considering the separated contribution of tree crowns, understory and background components, using a modified version of the FluorFLIGHT radiative transfer model (RTM). The new model is compared with others through the RAMI model intercomparison framework and is validated with airborne data. The airborne campaign includes high-resolution data collected over a tree-grass ecosystem with the HyPlant imaging spectrometer within the FLuorescence EXplorer (FLEX) preparatory missions. Field data measurements were collected from plots with a varying fraction of tree and understory vegetation cover. The relationship between airborne SIF calculated from pure tree crowns and aggregated pixels shows the effect of the understory at different resolutions. For a pixel size smaller than the mean crown size, the impact of the background was low (R2 > 0.99; NRMSE < 0.01). By contrast, for a pixel size larger than the crown size, the goodness of fit decreased (R2 < 0.6; NRMSE > 0.2). This study demonstrates that using a 3D RTM model improves the calculation of SIF significantly (R2 = 0.83, RMSE = 0.03 mW m−2 sr−1 nm−1) when the specific contribution of the soil and understory layers are accounted for, in comparison with the SIF calculated from mixed pixels that considers only one layer as background (R2 = 0.4, RMSE = 0.28 mW m

Cocoa is a source of flavanols, and these phenolic compounds exert beneficial effects on health and aging, and reduce the risk of suffering chronic diseases (cardiovascular diseases, metabolic disorders, cancer). An increasing body of evidence has emerged to suggest that cocoa flavanols potentially are important chemopreventive natural agents. This review summarizes human studies from the past two decades, providing data related to the effects derived from cocoa intake on health and disease. Most human studies have reported beneficial effects of cocoa consumption on health and chronic diseases; however, outcomes are not unequivocal. Review of human studies enable to identify different mechanisms of action for cocoa, although they are not fully understood at present. In addition, it remains unclear whether cocoa consumption should be recommended to healthy subjects or to patients and what is the appropriate dosage or duration of cocoa consumption. Elucidation of information regarding these crucial issues could lead to cocoa use as an approach for decreasing the risk of certain chronic diseases, as well as improving health and quality of life.

In recent years, human gut microbiota (GM) has emerged as a potential target to modulate health and disease. GM could be shaped by different factors, being diet one of the major influences. It metabolizes ingested nutrients to produce an extensive range of microbial metabolites that may have an important impact on human physiology. In turn, dietary components modulate the composition and metabolic activity of the GM, causing either positive or negative effects on human health. However, nutrients are rarely consumed independently and research on diet–microbiota relationship has progressively moved from the study of individual dietary components toward overall dietary patterns. This review summarized recent advances on the influence of food components and dietary patterns on GM and their subjacent impact on human health.

Colorectal cancer is the third most common cancer in the world and is one of the major causes of cancer-related mortality in the world. Cancer is defined as a multistage process and divided into the following stages: initiation, promotion, and progression. In this disease, oxidative stress is involved in a large array of carcinogenic pathways associated with the proliferation of initiated cells and cellular malignancy. Therefore, the suppression of oxidative stress by natural antioxidant compounds has gained interest as an effective approach in colorectal cancer prevention. Accordingly, cocoa and its phenolic components have demonstrated to exert antioxidant activity and to be able to interfere with multiple carcinogenic signaling pathways. In this chapter, we reviewed the different in vitro and in vivo studies that have identified potential targets and mechanisms whereby cocoa and their flavonoid compounds could interfere with colonic cancer cells. In addition, recent evidence from human studies is also illustrated.

Certain agricultural practices, such as the repeated application of herbicides or organic amendments to soil, can influence herbicide dissipation. This research has studied the effects of two repeated applications of mesotrione, pethoxamid, and triasulfuron on their dissipation rates in unamended soil (S) and soil amended with green compost (S+GC) or sewage sludge (S+SS). The dissipation experiment has also included an evaluation of the adsorption of the three herbicides by soils and of changes in soil dehydrogenase activity (DHA). The adsorption of the three herbicides by amended soils (Kf range 0.83–2.98) was higher than by unamended soil (Kf range 0.20–0.64). The adsorption coefficients (Kd) of mesotrione and triasulfuron were higher for S+SS, while that of pethoxamid was higher for S+GC, but no relationship between values for the time to 50% degradation (DT50) and adsorption coefficients could be determined. The repeated application of mesotrione decreased its dissipation rate in unamended soil (DT50 increased from 4.75 to 8.15 days) and amended soils (DT50 increased from 11.7 to 28.2 days in S+GC and from 17.7 to 37.9 in S+SS), whereas the repeated application of pethoxamid increased its dissipation rate in all the treatments, and the rate for triasulfuron increased only in amended soils. The highest DT50 values for pethoxamid (12.3 days) and triasulfuron (57.1 days) were in S+GC, and the lowest in S+SS (8.35 and 24.7 days). Soil DHA was stimulated by the presence of GC in the soil and by the first application of mesotrione. The second application of mesotrione and pethoxamid positively affected soil DHA, but this did not occur for triasulfuron. The repeated applications of herbicides and soil organic amendments have different effects on herbicide dissipation, adsorption, and soil DHA, and they should be taken into account when assessing soil quality and other potential environmental implications of pesticide use.

Spermine and spermidine are polyamines (PA) naturally present in all organisms, in which they have important physiological functions. However, an excess of PA has been associated with health risks. PA accumulates at quite high concentrations in some foods, but a quantitative assessment of the risk they pose has been lacking. In the present work, the cytotoxicity of spermine and spermidine was evaluated using an in vitro human intestinal cell model, and employing real-time cell analysis. Both spermine and spermidine showed a dose-dependent cytotoxic effect towards the cultured cells, with necrosis the mode of action of spermidine and perhaps also that of spermine. Spermine was more cytotoxic than spermidine, but for both PA the concentrations found to be toxic were above the maximum at which they have been found in food. The present results do not, therefore, support the idea that spermine or spermidine in food is harmful to healthy people.
