Soil pollution from antibiotics has become increasingly severe, posing significant environmental and human health threats. Many soil organisms can survive and sustain their roles in maintaining soil ecosystems, even in polluted conditions. Exploring the life-sustaining mechanisms of these organisms in contaminated environments is scientifically significant. This study used Eisenia fetida as the test organism and antibiotics (oxytetracycline hydrochloride) as exogenous stress substances. Oxidative stress response experiments were conducted using the artificial soil method to examine the response of earthworms to oxidative stress. Additionally, 16S rRNA technology was employed to analyze the succession of microbial community structures inside and outside the earthworms. A screening canonical correlation analysis (SCCA) model was developed to investigate the relationship between microbial communities and earthworm oxidative stress system under oxytetracycline stress, revealing survival strategies in antibiotic-contaminated soil. The results showed that Proteobacteria and Bacteriodetes were the dominant phyla of microbial communities in earthworms under oxytetracycline stress, while Proteobacteria and Firmicutes were dominant bacterial phyla in soil. Bacteriodetes and Firmicutes in earthworms worked synergistically with catalase (CAT) and glutathione peroxidase (GPX) in oxidative stress responses. In soil, Actinobacteria, Verrucomicrobia, and Spirochaeta synergistically resisted oxytetracycline stress alongside peroxidase (POD) and glutathione S-transferase (GST). Earthworm mucus played a crucial role in this synergistic resistance. These findings provide a scientific and experimental basis for assessing the ecological safety risks of antibiotic-contaminated soil.

Diverse microbial community structures (MCS) in wastewater treatment plants (WWTPs) are vital for effectively removing nutrients and chemicals from wastewater. However, the regular monitoring of MCS in WWTP bioreactors remains unattractive owing to the skill and cost required for deploying modern microbial molecular techniques in the routine assessment of engineered systems. In contrast, low-resolution methods for assessing broad changes in the MCS, such as phospholipid fatty acid (PLFA) analysis, have been used effectively in soil studies for decades. Despite using PLFA analysis in soil remediation studies to capture the long-term effects of environmental changes on MCS, its application in WWTPs, where the microbial mass is dynamic and operational conditions are more fluid, remains limited. In this study, microbial communities in a controlled pilot plant and 12 full-scale activated sludge plants (ASPs) were surveyed over a two-year period using PLFA analysis. This study revealed that changes in the MCS in wastewater bioreactors could be detected using PLFA analysis. The MCS comprised 59 % Gram-negative and 9 % Gram-positive bacteria, 31 % fungi, and 1 % actinomycetes. The abundances of Gram-negative bacteria and fungi were strongly inversely correlated, with an R2 = 0.93, while the fatty acids cy17:0 and 16:1ω7c positively correlated (R2 = 0.869). Variations in temperature, solid retention time, and WWTP configuration significantly influenced the MCS in activated sludge reactors. This study showed that WWTP bioreactors can be routinely monitored using PLFA analysis, and changes in the bioreactor profile that may indicate imminent bioreactor failure can be identified.

The fresh carbon (C) induced priming effect (PE) on soil organic C (SOC) decomposition is critical for global C cycling. Climate warming could raise absorptive roots production and turnover, and then increase the input of absorptive roots litter (ABRL). Therefore, it is urgent to understand the PE induced by ABRL under warming. We conducted a 210-day experiment by adding ABRL of Cunninghamia lanceolata into a C4 soil and incubating them at 19 °C and 23 °C. We found that adding ABRL caused positive PE throughout the incubation. At the early stage (ES: first 30 days), labile C compounds dominated the decomposition of ABRL, significantly higher dissolved organic C (DOC), microbial biomass C (MBC), and absolute hydrolase activities were found in the ABRL treatment than in the control. These results supports that labile C inputs stimulating microbial growth, enzyme activities and cause positive PE via co-metabolic. At the later stage (LS: after 180 days), the release of structural C compounds dominated the decomposition of ABRL, a significantly lower available nitrogen (N) and a significantly higher specific potential N-acquisition (Nacq) enzymes were found in the ABRL treatment than in the control. These results suggests that microbes utilizing the C as energy to increase Nacq enzymes to decompose SOC for N mining under N limitation induced positive PE. Warming significantly increased the PEs at both stages. At the ES, warming increased the MBC at the cost of DOC, suggesting that warming intensifies the microbial co-metabolism. At the LS, warming significantly decreased the available N and increases the absolute potential oxidases activities, suggesting an increased N limitation and oxidation for N-rich recalcitrant SOC, i.e., a promoted microbial N mining. Nevertheless, we did not observe a significant effect of ABRL addition on the temperature sensitivity of SOC decomposition compared to the control. This study provides a valuable insight that warming could consistently increase the ABRL induced PE but through different microbial mechanisms along with the decomposition processing.

The soaking and fermentation of the stems and leaves is an important intermediate step in the processing of Indigo Naturalis. However, the relationship between microbiota and Indigo Naturalis yields is still poorly understood. This study aimed to compare microbial communities and metabolite profiles at various stages of soaking fermentation, followed by validation of the results using HPLC. A total of 731 compounds were identified through metabolite analysis, with the levels of indigo and indirubin peaking after 36 h of fermentation. Metagenomes revealed Firmicutes, Proteobacteria, Bacteroidetes and Actinobacteria were identified as the most abundant microbial phyla in soaking fermentation. Correlation analysis indicated that the yields of indigo and indirubin may be affected by Lactococcus, Clostridium, and Enterobacter through the regulation of related synthetic enzymes. The findings offered novel perspectives on the relationship of microorganisms and Indigo Naturalis yields.

Acid rain and invasive plants have quintessential adverse impacts on terrestrial ecosystems. As an environmentally safe method for disposal of invasive plants, we tested the effect of biochar produced from these plants in altering soil deterioration under acid rain as compared with lime. Given the impacts of the feedstock type and soil properties on the response of soil to the added biochar, we hypothesized that the microbial community and functions would respond differently to the charred invasive plants under acid rain. A pot experiment was conducted to examine the response of soil microbiomes and functions to the biochar produced from Blackjack (Biden Pilosa), Wedelia (Wedelia trilobata), and Bitter vine (Mikania micrantha Kunth), or quicklime (CaO) at a rate of 1 % (w/w) under acid rain. Like soil pH, the nutrient contents (nitrogen, phosphorus, and potassium), calcium, and cation exchange capacity (CEC) were important as dominant edaphic factors affecting soil microbial community and functionality. In this respect, lime decreased nutrients availability, driven by 11-fold, 44 %, and 2-fold increments in calcium content, pH, and C/N ratio. Meanwhile, biochar improved nutrients availability under acid rain owing to maintaining a neutral pH (∼6.5), increasing calcium (by only 2-fold), and improving CEC, water repellency, and aggregation while decreasing the C/N ratio and aluminum content. Unlike biochar, lime decreased the relative abundance of Nitrosomonadaceae (the dominant ammonia-oxidizing bacteria) while augmenting the relative abundance of some fungal pathogens such as Spizellomycetaceae and Sporormiaceae. Given the highest nitrogen and dissolved organic carbon content than other biochar types, Wedelia-biochar resulted in the greatest relative abundance of Nitrosomonadaceae; thus, the microbial carbon and nitrogen biomasses were maximized. This study outlined the responses of the soil biogeochemical properties and the related microbial community structure and functionality to the biochar produced from invasive plants under acid rain. This study suggests that biochar can replace lime to ameliorate the effects of acid rain on soil physical, chemical and biological properties.

With the rapid expansion of artificial light in the global nocturnal environment, the ecological impacts of artificial light at night (ALAN) on plant communities have garnered increasing research interest. However, the role of phyllosphere microorganisms as mediators in the regulation between ALAN and plants remains unclear. In this study, we conducted a field experiment in natural grassland plots, employing two treatments: one with no artificial light at night (control) and another with artificial light at night (ALAN). We then investigated the response of phyllosphere microorganisms in three grassland plant species (Leymus chinensis, Vicia sepium, and Aster altaicus) to ALAN exposure. The results revealed that the constructive species L. chinensis demonstrated a markedly higher diversity of phyllosphere bacteria and fungi compared to V. sepium and A. altaicus and thus had the most abundant microbial community. However, ALAN exposure reduced the diversity of phyllosphere bacteria and fungi in all three plant species to varying degrees. ALAN induced certain changes to the phyllosphere bacterial community composition, particularly manifesting in a reduction in the abundance of beneficial bacteria, including Sphingomonas, Hymenobacter, Methylobacterium-Methylorubrum, and Bacillus. Compared with phyllosphere bacterial communities, the alterations in phyllosphere fungal communities were more sensitive, characterized by a significant decrease in the abundance of certain fungi, including Zymoseptoria, Mycosphaerella, Cladosporium, and Epicoccum. The functional predictions of phyllosphere bacteria and fungi further corroborate these conclusions. In summary, ALAN exposure may suppress the functional capacity of phyllosphere microorganisms in grassland plants while decreasing the probability of fungal pathogen occurrence.

This study clarified the functional characteristics of microorganisms during the co-composting of bamboo powder and tea leaves. Three treatments were established for 14 weeks of composting: B100 (600 kg bamboo powder), B40 (240 kg bamboo powder and 360 kg tea leaves), and B20 (120 kg bamboo powder and 480 kg tea leaves), with two replicates each. The functional characteristics of the microorganisms were analyzed using a BIOLOG EcoPlate for carbon substrate utilization patterns using kinetic parameters, microbial diversity analysis by calculating diversity indices, and microbial community structure analysis via principal component analysis of kinetic parameters. When bamboo powder was composted alone, the microbial diversity was high in the early composting stage because of the utilization of the rich and complicated carbon substrates. However, in the late composting stage, the microbial diversity was low because of the utilization of poor and simple carbon substrates. In contrast, when bamboo powder and tea leaves were combined, carbon substrate utilization and microbial diversity were low during the early composting stage. In the middle and late stages of composting, abundant carbon substrates became available, and microbial diversity increased. Differences in carbon substrate utilization patterns can be separated by microbial community structure analysis, especially at the end of composting. The communities were divided according to the proportions of bamboo powder and tea leaves. In the early stage of co-composting bamboo powder and tea leaves, microbial activity decreased because of the lower compost temperature and higher tea polyphenol content. However, in the middle and late stages, microbial activity may increase owing to the supply of tea-derived soluble sugars and amino acids. The co-composting of bamboo powder and tea leaves may enhance microbial activity, diversity, and carbon substrate utilization during the middle and late composting stages.

Soil life revolves around microorganisms that are crucial for soil ecosystems and health. In megacities, the combined exposure of multiple pollutants exerts a significant impact on the structures and functions of soil microorganisms; however, there is a lack of empirical studies on this topic. Hence, we conducted a study including urban parks in Beijing, China. The results indicate that bacteria were abundant in the soils of Beijing parks, showing the same dominant groups but different rare groups. The dominant groups included Actinobacteria and Proteobacteria. Candidate phyla radiation bacteria, a large evolutionary radiation of bacterial lineages whose members remain mostly uncultivated, were the main specialists. Under the combined exposure of multiple pollutants, the structures of soil microbial communities in different parks were similar. Community change due to pollutants (31%) was greater than that due to natural factors (2.4%). Among multipollutants, organophosphate esters, led by dibutyl phosphate, had the highest influence on microbial abundance and distribution. An increase in dibutyl phosphate concentration decreased the abundance of Firmicutes, while the abundance of Synergistota was increased. The interactions among pollutants affecting the bacteria were different. Bis(2-chloroethyl) phosphate, nickel and benzo[g,h,i]perylene influenced microorganisms by working with organophosphate esters. High-molecular-weight polycyclic aromatic hydrocarbons, such as benzo[a]pyrene and benzo[g,h,i]perylene, mainly acted on the functional genes and thus affected multiple biogeochemical cycles. Benzo[a]anthracene, bis(2-chloroethyl) phosphate, and arsenic were the primary pollutants affecting metabolic pathways. Our research helps to better understand the impacts of urban environmental pollution on soil microorganisms.

Nitrous oxide (N2O) emissions from the wastewater treatment sector are a significant contributor to global greenhouse gas levels. This investigation delves into the mechanisms of N2O generation and uptake, correlating microbial processes with variables such as influent characteristics and operational parameters. The nature of carbon substrates in the influent profoundly influences microbial consortia and N2O output. Elevating the carbon-to-nitrogen (C/N) ratio has been shown to curtail N2O emissions by alleviating the competitive dynamics among denitrifying enzymes. Optimal activity of N2O reductase is achieved by maintaining a neutral to mildly alkaline pH and stable ambient temperatures. It is imperative to circumvent extreme aeration rates and prolonged aeration periods to reduce N2O release. The study underscores the importance of an effective carbon feed strategy and advocates for prolonged hydraulic retention times (HRT) and sludge retention times (SRT) in activated sludge suspension systems to inhibit N2O escape. Notably, excessive internal recycling, coupled with heightened dissolved oxygen (DO) levels in aerobic zones, intensifies N2O emission risks. Moreover, the presence of hazardous contaminants, such as heavy metals and antibiotics, interferes with nitrogen elimination processes, warranting a comprehensive assessment of consequent N2O emission hazards. This research provides a scientific basis as well as practical management approaches to diminish N2O emissions.

Breast cancer has become the number one cancer in the world, and intestinal flora may be closely linked to it. Geographic location also has an important impact on human intestinal flora. We conducted the first study on the intestinal flora of breast cancer patients and non-breast cancer patients in a tropical region - Hainan Province in China. At the same time, Pacbio platform based on third-generation sequencing was used for the first time to conduct 16S full-length sequencing of fecal microorganism DNA. We completed the species diversity analysis and differential species analysis of the intestinal flora between the two groups, inferred their functional genetic composition and performed functional difference analysis. There were statistically significant differences in alpha diversity between the two groups in Hainan Province. By species composition difference analysis, at the phylum level, Bacteroidales (P = 0.006) and Firmicutes (P = 0.002) was differed between the two groups, and at the genus level, 17 breast cancer-related differential species such as Bacteroides were screened. According to the five grouping methods including ER level, PR level, HER2 status, Ki67 index and histological grade of breast cancer patients, 4, 1, 9, 6, 5 differential microbiota were screened out respectively, which were in total 25 (P

Cadmium (Cd) removal from soil to reduce Cd accumulation in plants is essential for agroecology, food safety, and human health. Cd enters plants from soil and affects plant growth and development. Hydrogels can easily combine with Cd, thereby altering its bioavailability in soil. However, few studies have evaluated the effects of hydrogel on the complex phytotoxicity caused by Cd uptake in plants and the microbial community structure. Herein, a new poly (acrylic acid)-grafted starch and potassium humate composite (S/K/AA) hydrogel was added to soil to evaluate its impact on tobacco growth and the soil microenvironment. The results indicate that the addition of S/K/AA hydrogel can significantly improve the biomass, chlorophyll (Chl) content, and photosynthetic capacity of tobacco plants during Cd stress conditions, and decrease Cd concentration, probably by affecting Cd absorption through the expression of Cd absorption transporters (e.g., NRAMP5, NRAMP3, and IRT1). Moreover, the application of S/K/AA hydrogel not only reduced the accumulation of reactive oxygen species (ROS), but also reduced the antioxidant activities of peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT), suggesting that S/K/AA hydrogel alleviates Cd toxicity via a non-antioxidant pathway. Notably, we further analyzed the effectiveness of the hydrogel on microbial communities in Cd-contaminated soil and found that it increased the Cd-tolerant microbial community (Arthrobacter, Massilia, Streptomyces), enhancing the remediation ability of Cd-contaminated soil and helping tobacco plants to alleviate Cd toxicity. Overall, our study provides primary insights into how S/K/AA hydrogel affects Cd bioavailability and alleviates Cd toxicity in plants.

Groundwater is an important human resource. Daejeon in South Korea faces severe water quality issues, including radon, uranium, and fluoride pollution, all of which pose health risks to humans. With climate change, threats to potable water, such as heavy rain and typhoons, have become common. Therefore, examining the seasonal effects on groundwater quality and resultant health risks is important for understanding the mechanisms of different hydroclimatological conditions to enable the implementation of sustainable management plans in radon-contaminated groundwater areas. However, this issue has not yet been studied. To bridge this gap, in this study, major ions and microbial community structures were employed and groundwater quality index (GWQI) were calculated with hazard index based on limits set by the World Health Organization (WHO) to investigate the hydrochemical characterization and to assess pollution levels. The results showed that the rainy season had distinct hydrochemical characteristics with high correlations between radon and fluoride, and most groundwater samples collected after the typhoon had characteristics similar to those collected during the dry season, owing to the flow path. Furthermore, the microbial diversity and hazard quotient (HQ) values of fluoride revealed that pollution worsened during the dry season. All of the calculated effective dose values of radon exceeded the threshold limit set by the WHO, despite the low GWQI. Infants and children were particularly susceptible to radon-contaminated groundwater. The statistical results of self-organizing map (SOM) suggested that radon analysis was sufficient for public health intervention in the rainy season; however, in the dry season, combined analyses of radon, fluoride, and microbial diversity played important roles in health risk assessment. Our study presents a comprehensive understanding of radon-contaminated groundwater characteristics under seasonal effects and can serve as a reference for other similar zones to provide significant insights into the effective management of radon contamination.

Relying on the high mobility of water flow, the dissemination of antibiotic resistance genes (ARGs) in the water tends to be exacerbated and enlarged. It caused negative impacts on a wider scope of the environment. The ARGs dissemination monitoring and the methods efficiently reducing their concentration in water became the focus of interest. Green chemicals with antibacterial effects such as tea polyphenols (TPs) and catechins (CA) have been considered as auxiliary disinfectants for ARGs removal in the water environment. However, the antibacterial performance of TPs and CA under the stress of external antibiotics still lacks sufficient research. The results show that more operational taxonomic units can be observed in water samples with TPs and CA than in those without the ingredients under pressure of tetracycline. An unexpected increase along with the increase of ARGs concentrations and the diversity of microbial communities under the low-concentration TPs or CA (1 mg/L). Besides, under the stress of tetracycline, the inhibition of TPs was detected to be strengthened for increase of inti1 and tetC but weakened towards for the increase of tetA. Whilst CA substantially diminished abundances of tetC and tetA under tetracycline pressure. This research demonstrated that TPs and CA are able to assuage development of ARGs under the pressure of antibiotic in water system.

Biochar (BC) amendment has been proposed as a promising strategy for mitigating greenhouse gas (GHG) emissions, specifically carbon dioxide (CO 2 ), methane (CH 4 ), and nitrous oxide (N 2 O). Conducting a meta-analysis to evaluate the impact of biochar on microbial genetic profile, community structure, and phospholipid fatty acid (PLFA) contents can aid in identifying key microbial groups involved in GHG production and consumption, and assessing the overall effectiveness of biochar in reducing GHG emissions. The present meta-analysis revealed that the addition of biochar resulted in a 22 % and 41 % reduction in pmoA and mcrA genes of methanogenic microorganisms, respectively. The mcrA/pmoA ratio significantly increased by 81 %. Gene abundances exhibited a positive response to biochar amendment, with increases observed in nifH, nirK, nirS, nosZ, and nosZ (nirS + nirK) genes by 13 %, 32 %, 37 %, 42 %, and 79 %, respectively. Moreover, biochar amendment influenced the microbial community structure accordingly. The concentration of PLFAs increased in response to BC treatment in the following order: A-bacteria (+49 %) < Fungi (+30 %) < Gram-pb (+21 %) < G-bacteria (+17 %) < Gram-nb (+11 %). Aerobic granular sludge (AGS) exhibits excellent resistance to adverse environment due to its unique layered structure. However, the mechanism about how salinity fluctuations in municipal wastewater impact AGS formation and its physicochemical properties has not been thoroughly revealed. In this study, AGS was cultivated under additional 0 % salinity (R1), additional 1.5 % constant salinity (R2), and additional 0-1.5 % fluctuant salinity (R3), respectively. The results indicate that increased salinity can enhance extracellular polymeric substances (EPS) production and improve sludge settleability, thereby facilitate AGS formation. However, the AGS experienced frequent environmental conversion between dehydration and swell due to salinity fluctuations, resulting in higher content of loosely-bond EPS and low settleability, which delayed the maturation of AGS for over 14 days. Additional salinity significantly inhibited the nitrification process, but the formation of AGS promoted the recovery of ammonia oxidation activity and facilitated the construction of short-range nitrification denitrification processes, resulting in over 16.0 % higher total nitrogen removal efficiency than R1. The microbial community analysis revealed that Thauera played an important role in the granulation process under salinity stress, due to its salt tolerance and EPS secretion abilities. As expected, the formation of AGS enhanced the salt resistance of microorganisms, allowing for the enrichment of functional bacteria, such as Flavobacterium and Candidatus&#95;Competibacter. Generally, microorganisms required extended adaptation periods to cope with salinity fluctuations. Vermicomposting is an efficient bioconversion technology for recycling nutrients from organic waste materials. The biodegradability of raw materials has a significant impact on the earthworm transformation product. However, the management of carbon bioavailability is often overlooked during the vermicomposting process due to the varying degradability of C-rich source in different organic waste. This research aims to investigate the impact of different bioavailable carbon compositions on vermicomposting and to develop a strategy for efficient carbon management. The study involved systematic vermicomposting using four different biodegradable carbon sources (pineapple peels, rice straw, tomato straw, and sawdust) with varying carbon‑nitrogen ratios (ranging from 24 to 42). The earthworm production and vermicompost quality were comprehensively evaluated, along with the influence of carbon components on microbial community structure. The results indicated that the optimal vermicomposting treatments were achieved at PCM24, RCM30, TCM30, and MCM30 treatments. Maintaining an approximate ratio of 1:(0.5-1.3) between available and recalcitrant carbon components based on the optimal carbon‑nitrogen ratio was found to be optimal for regulating vermicomposting products. Increasing the proportion of available carbon enhanced the quality of vermicompost fertilizer, while a higher proportion of recalcitrant carbon could improve earthworm biomass production efficiency. Labile carbon proportion I (LCP1) and available carbon component (ACC) were identified as key indicators in influencing the formation of microbial community structure. Different carbon compositions led to the specific development and formation of microbial communities, further resulting in significant variations in vermicompost quality under the mediation of microbes. The suppression of soil carbon mineralization has been demonstrated to be effectively facilitated by carbon‑iron interactions, yet the specific mechanisms by which artificial humic substances (A-HS) coupled with ferrihydrite influence this process remain insufficiently explored. This study is to investigate how the A-HS, specifically artificial fulvic acid (A-FA) and artificial humic acid (A-HA), coupled with ferrihydrite, affect carbon mineralization under anaerobic system that simulates paddy flooding conditions. The object is to investigate trends in carbon emissions and to delineate microbial community structure and functional pathways. The findings indicate that A-HA and A-FA substantially reduce CO 2 and CH 4 emissions, with A-FA having a particularly pronounced effect on carbon fixation, halving CO 2 concentrations. The low concentration of Fe(II) observed suggest that A-FA and A-HA impede the dissimilatory iron reduction (DIR) process. Detailed 16S rDNA sequencing and gene prediction analyses reveal changes in microbial community structures and functions, highlighting Methanobacterium as the dominant hydrogenotrophic methanogens. The reductive citric acid cycle, predominantly utilized by Clostridium carboxidivorans, was identified as the principal carbon fixation pathway. Numerous reservoirs encounter challenges related to taste and odor issues, often attributed to odorous compounds such as geosmin (GSM) and 2-methylisoborneol (2-MIB). In this study, two large reservoirs located in northern and southern China were investigated. The Jinpen (JP) reservoir had 45.99 % Actinomycetes and 14.82 % Cyanobacteria, while the Xikeng (XK) reservoir contained 37.55 % Actinomycetes and 48.27 % Cyanobacteria. Most of the 2-MIB produced in surface layers of the two reservoirs in summer originated from Cyanobacteria, most of the 2-MIB produced in winter and in the bottom water originated from Actinomycetes. Mic gene abundance in the XK reservoir reached 5.42 × 10 4 copies/L in winter. The abundance of GSM synthase was notably high in the bottom layer and sediment of both reservoirs, while 2-MIB synthase was abundant in the surface layer of the XK reservoir, echoing the patterns observed in mic gene abundance. The abundance of odor-producing enzymes in the two reservoirs was inhibited by total nitrogen, temperature significantly influenced Actinomycetes abundance in the JP reservoir, whereas dissolved oxygen had a greater impact in the XK reservoir. The scarcity of freshwater poses significant challenges to agriculture, often necessitating the use of alternative water sources such as reclaimed water. While reclaimed water offers a viable solution by providing water and nutrients to crops, its potential impacts on soil microbial communities remain a subject of investigation. In this investigation, we conducted a field experiment cultivating Maize (Zea mays) and Lavender (Lavandula angustifolia), employing irrigation with reclaimed water originating from domestic wastewater, while control samples were irrigated using freshwater. Utilizing high-throughput sequencing, we assessed the effect of reclaimed water on soil bacteria and fungi. Plant biomass exhibited a significant response to treated wastewater. Alpha diversity metrics of soil microbial communities did not reveal significant changes in soils irrigated with reclaimed water compared to control samples. Reclaimed water, however, demonstrated a selective influence on microorganisms associated with nutrient cycling. Co-occurrence network analysis unveiled that reclaimed water may alter soil microbial community structure and stability. Abstract Background Soils polluted with animal charcoal from skin and hide cottage industries harbour extremely toxic and carcinogenic hydrocarbon pollutants and thus require a bio-based eco-friendly strategy for their depuration. The effects of carbon-free mineral medium (CFMM) amendment on hydrocarbon degradation and microbial community structure and function in an animal charcoal-polluted soil was monitored for 6 weeks in field moist microcosms consisting of CFMM-treated soil (FN4) and an untreated control (FN1). Hydrocarbon degradation was monitored using gas chromatography-flame ionization detector (GC-FID), and changes in microbial community structure were monitored using Kraken, while functional annotation of putative open reading frames (ORFs) was done using KEGG KofamKOALA and NCBI’s conserved domain database (CDD). Results Gas chromatographic analysis of hydrocarbon fractions revealed the removal of 84.02% and 82.38% aliphatic and 70.09% and 70.14% aromatic fractions in FN4 and FN1 microcosms in 42 days. Shotgun metagenomic analysis of the two metagenomes revealed a remarkable shift in the microbial community structure. In the FN4 metagenome, 92.97% of the population belong to the phylum Firmicutes and its dominant representative genera Anoxybacillus (64.58%), Bacillus (21.47%) and Solibacillus (2.39%). In untreated FN1 metagenome, the phyla Proteobacteria (56.12%), Actinobacteria (23.79%) and Firmicutes (11.20%), and the genera Xanthobacter (9.73%), Rhizobium (7.49%) and Corynebacterium (7.35%), were preponderant. Functional annotation of putative ORFs from the two metagenomes revealed the detection of degradation genes for aromatic hydrocarbons, benzoate, xylene, chlorocyclohexane/chlorobenzene, toluene and several others in FN1 metagenome. In the FN4 metagenome, only seven hydrocarbon degradation genes were detected. Conclusion This study revealed that though CFMM amendment slightly increases the rate of hydrocarbon degradation, it negatively impacts the structural and functional properties of the animal charcoal-polluted soil. It also revealed that intrinsic bioremediation of the polluted soil could be enhanced via addition of water and aeration.

Vegetation restoration in metallic tailing reservoirs is imperative to restore the post-mining degraded ecosystems. Extracellular enzymes determine microbial resource acquisition in soils, yet the mechanisms controlling the enzyme activity and stoichiometry during vegetation restoration in metallic tailing reservoirs remain elusive. Here, we investigated the variations and drivers of C-, N- and P-acquiring enzymes together with microbial community along a 50-year vegetation restoration chronosequence in the China's largest vanadium titano-magnetite tailing reservoir. We found a parabolic pattern in the enzyme activity and efficiency along the chronosequence, peaking at the middle restoration stage (∼30 years) with approximately six-fold increase relative to the initial 1-year site. The enzyme ratios of C:P and N:P decreased by 33 % and 68 % along the chronosequence, respectively, indicating a higher microbial demand of C and N at the early stage and a higher demand of P at the later stage. Soil nutrients directly determined the enzyme activities and stoichiometry, whereas microbial biomass and community structure regulated the temporal pattern of the enzyme efficiency. Surprisingly, increased heavy metal pollution imposed a positive effect on the enzyme efficiency indirectly by altering microbial community structure. This was evidenced by the increased microbial diversity and the conversion of copiotrophic to oligotrophic and stress-tolerant taxa along the chronosequence. Coalbed methane (CBM) presents a promising energy source for addressing global energy shortages. Nonetheless, challenges such as low gas production from individual wells and difficulties in breaking gels at low temperatures during extraction hinder its efficient utilization. Addressing this, we explored native microorganisms within coal seams to degrade guar gum, thereby enhancing CBM production. However, the underlying mechanisms of biogenic methane production by synergistic biodegradation of lignite and guar gum remain unclear. Research results showed that the combined effect of lignite and guar gum enhanced the production, yield rate and concentration of biomethane. When the added guar gum content was 0.8 % (w/w), methane production of lignite and guar gum reached its maximum at 561.9 mL, which was 11.8 times that of single lignite (47.3 mL). Additionally, guar gum addition provided aromatic and tryptophan proteins and promoted the effective utilization of CC/CH and OCO groups on the coal surface. Moreover, the cooperation of lignite and guar gum accelerated the transformation of volatile fatty acids into methane and mitigated volatile fatty acid inhibition. Dominant bacteria such as Sphaerochaeta, Macellibacteroides and Petrimonas improved the efficiency of hydrolysis and acidification. Electroactive microorganisms such as Sphaerochaeta and Methanobacterium have been selectively enriched, enabling the establishment of direct interspecies electron transfer pathways. This study offers valuable insights for increasing the production of biogenic CBM and advancing the engineering application of microbial degradation of guar gum fracturing fluid. Microaerobic sludge bed systems could align with low-energy, reasonable carbon-nitrogen (C/N) ratio, and synchronous removal objectives during wastewater treatment. However, its ability to treat municipal wastewater (MW) with varying low C/N ratio, low NH 4 + concentration, along with managing sludge bulking and loss are still unclear. Against this backdrop, this study investigated the performance of an Upflow Microaerobic Sludge Bed Reactor (UMSR) treating MW characterized by varying low C/N ratios and low NH 4 + concentrations. The study also thoroughly examined associated sludge bulking and loss, pollutant removal efficiencies, sludge settleability, microbial community structures, functional gene variations, and metabolic pathways. Findings revealed that the effluent NH 4 + -N concentration gradually decreased to 0 mg/L with a decrease in the C/N ratio, whereas the effluent COD was unaffected by the influent, maintaining a concentration below 50 mg/L. Notably, TN removal efficiency reached 90% when C/N ratio was 3. The decrease in the C/N ratio (C/N ratio was 1) increased microbial community diversity, with abundances of AOB, AnAOB, aerobic denitrifying bacteria, and anaerobic digestion bacteria reaching 8.34%, 0.96%, 5.07%, and 9.01%, respectively. Microorganisms' metabolic pathways significantly shifted, showing increased carbohydrate and cofactor/vitamin metabolism and decreased amino acid metabolism and xenobiotic biodegradation. Aquatic ecosystems are being increasingly polluted by microplastics (MPs), which calls for an understanding of how MPs affect microbially driven biogenic element cycling in water environments. A 28-day incubation experiment was conducted using freshwater lake water added with three polymer types of MPs (i.e., polyethylene, polypropylene, polystyrene) separately or in combination at a concentration of 1 items/L. The effects of various MPs on microbial communities and functional genes related to carbon, nitrogen, phosphorus, and sulfur cycling were analyzed using metagenomics. Results showed that Sphingomonas and Novosphingobium, which were indicator taxa (genus level) in the polyethylene treatment group, made the largest functional contribution to biogenic element cycling. Following the addition of MPs, the relative abundances of genes related to methane oxidation (e.g., hdrD, frhB, accAB) and denitrification (napABC, nirK, norB) increased. These changes were accompanied by increased relative abundances of genes involved in organic phosphorus mineralization (e.g., phoAD) and sulfate reduction (cysHIJ), as well as decreased relative abundances of genes involved in phosphate transport (phnCDE) and the SOX system. Landfill leachate contributes significantly to the presence of pharmaceuticals and personal care products (PPCPs) in the environment, and is a crucial source of contamination. To examine the occurrence of PPCPs and microbial communities, this study comprehensively investigated the concentrations of PPCPs and the abundance of microorganisms in the leachate from 17 municipal landfills across China. Generally, Lidocaine, Linear alkylbenzene sulfonate-C11, and Triclocarban, which are closely associated with human activities, exhibited a detection frequency of 100 % in the leachate. Driven by consumer demand, analgesic and antipyretic drugs have emerged as the most prominent PPCPs in leachate (accounting for 39.9 %). Notably, the Ibuprofen peaked at 56.3 μg/L. Regarding spatial distribution, the contamination of PPCPs in leachates from the eastern regions of China was significantly higher than that in other regions, owing to the level of economic development and demographic factors. Furthermore, the 16S rRNA results revealed significant differences in microbial communities among the leachates from different areas. Although the impact of PPCPs on microbial communities may not be as significant as that of environmental factors, most positive correlations between PPCPs and microorganisms indicate their potential role in providing nutrients and creating favorable conditions for microbial growth. The particle size distribution in tailings notably influences their physical properties and behavior. Despite this, our understanding of how the distribution of tailings particle sizes impacts in situ pollution and ecological remediation in in-situ environment remains limited. In this study, an iron tailings reservoir was sampled along a particle flow path to compare the pollution characteristic and microbial communities across regions with different particle sizes. The results revealed a gradual reduction in tailings particle size along the flow direction. The predominant mineral composition shifts from minerals such as albite and quartz to layered minerals. Total nitrogen, total organic carbon, and total metal concentrations increased, whereas the acid-generating potential decreased. The region with the finest tailings particle size exhibited the highest microbial diversity, featuring metal-resistant microorganisms such as KD4-96, Micrococcaceae, and Acidimicrobiia. Significant discrepancies were observed in tailings pollution and ecological risks across different particle sizes. Consequently, it is necessary to assess tailings reservoirs pollution in the early stages of remediation before determining appropriate remediation methods. These findings underscore that tailings particle distribution is a critical factor in shaping geochemical characteristics. Acid rain and invasive plants have quintessential adverse impacts on terrestrial ecosystems. As an environmentally safe method for disposal of invasive plants, we tested the effect of biochar produced from these plants in altering soil deterioration under acid rain as compared with lime. Given the impacts of the feedstock type and soil properties on the response of soil to the added biochar, we hypothesized that the microbial community and functions would respond differently to the charred invasive plants under acid rain. A pot experiment was conducted to examine the response of soil microbiomes and functions to the biochar produced from Blackjack (Biden Pilosa), Wedelia (Wedelia trilobata), and Bitter vine (Mikania micrantha Kunth), or quicklime (CaO) at a rate of 1 % (w/w) under acid rain. Like soil pH, the nutrient contents (nitrogen, phosphorus, and potassium), calcium, and cation exchange capacity (CEC) were important as dominant edaphic factors affecting soil microbial community and functionality. In this respect, lime decreased nutrients availability, driven by 11-fold, 44 %, and 2-fold increments in calcium content, pH, and C/N ratio. Meanwhile, biochar improved nutrients availability under acid rain owing to maintaining a neutral pH (∼6.5), increasing calcium (by only 2-fold), and improving CEC, water repellency, and aggregation while decreasing the C/N ratio and aluminum content. Unlike biochar, lime decreased the relative abundance of Nitrosomonadaceae (the dominant ammonia-oxidizing bacteria) while augmenting the relative abundance of some fungal pathogens such as Spizellomycetaceae and Sporormiaceae. Given the highest nitrogen and dissolved organic carbon content than other biochar types, Wedelia-biochar resulted in the greatest relative abundance of Nitrosomonadaceae; thus, the microbial carbon and nitrogen biomasses were maximized. This study outlined the responses of the soil biogeochemical properties and the related microbial community structure and functionality to the biochar produced from invasive plants under acid rain. Dense non-aqueous-phase liquids (DNAPLs) represent one of the most hazardous contaminants of groundwater, posing health risks to humans. Radon is generally used to trace DNAPLs; however, external factors, such as rainfall or stream water, can influence its efficacy. To overcome these limitations, this study pioneered the integration of radon and microbial community structures to explore DNAPL tracing and natural attenuation in the context of seasonal variations for human health risk assessments. The results showed that a radon tracer can estimate DNAPL saturation in the source zone, especially during the dry season when radon deficiency predominates. However, samples exhibited mixing effects during the wet season because of local precipitation. Moreover, bioremediation and low health risks were observed in the plume boundary zone, indicating that microbial dechlorination was a predominant factor determining these risks. The abnormal patterns of radon observed during the wet season can be elucidated by examining microbiological communities. Consequently, a combined approach employing radon and microbial analysis is advocated for the boundary zone, albeit with a less intensive management strategy, compared with that for the source zone. Oil pollution poses a great threat to environments and makes the remediation of oil-contaminated soils an urgent task. Microorganisms are the main biological factor for oil removal in the environment but microbial remediation is greatly affected by environmental factors. For our research, we inoculated three species of bacterivorous nematodes into oil-contaminated soil to explore how bacterivorous nematodes affect soil microbial activities and community structure in contaminated soil, as well as how efficiently different nematodes remove oil pollution from the soil. Six treatments were set in this experiment: sterilized oil-contaminated soil (SOC); nematode-free soil (S); oil-contaminated soil (OC); oil-contaminated soil + Caenorhabditis elegans (OCN1); oil-contaminated soil + Cephalobus persegnis (OCN2); oil-contaminated soil + Rhabditis marina (OCN3) for a 168-day incubation experiment. After the experiment was done, the oil contents in SOC, OC, OCN1, OCN2, and OCN3 were reduced by 6.5%, 32.3%, 38.2%, 42.8%, and 40.2%, respectively, compared with the beginning of the experiment. The amount of phospholipid fatty acids (PLFAs) of Gram-negative bacteria in OC, OCN1, OCN2, and OCN3 was increased by 50.9%, 43.4%, 37.7%, and 47.9%, respectively, compared with that of S. During the 168-day incubation period, the maximum growth of the number of nematodes in OCN1, OCN2, and OCN3 compared with the initial number of the nematodes were 2.25-, 1.52-, and 1.65-fold, respectively. The amount of oil residue in the contaminated soil negatively correlated with the populations of nematodes, total microorganisms, Gram-negative bacteria, actinomycetes, and eukaryotes. Thus, oil pollution increased the number of Gram-negative bacteria, decreased the ratio of Gram-positive bacteria/Gram-negative bacteria and Fungi/Bacteria significantly, and altered the community structure of soil microorganisms. Each species of bacterivorous nematodes has got its unique effect on the microbial activity and community structure in oil contaminated soils, but those tested can promote oil degradation and thus improve the environment of oil contaminated soils.

Acid rain alters soil carbon (C) cycling by influencing the soil microbial community structure and functions. However, the response of soil microbial communities to acid rain with time and underlying mechanisms are still poorly understood. Herein, we conducted a one-year intact soil core experiment to investigate the temporal changes of soil microbial community composition and C sources metabolism under acid rain (pH 5.0, pH 4.0, and pH 3.0) in an agricultural soil of southern China. We found that pH 3.0 acid rain increased the total, bacterial, gram-positive bacterial, and actinomycetal PLFAs at the early stage, but this effect diminished with time. Conversely, the gram-negative bacterial PLFAs contents were reduced under pH 3.0 acid rain at the later stage. Interestingly, pH 5.0 acid rain increased the total, bacterial, gram-positive bacterial, and actinomycetal PLFAs contents at the later stage. In addition, pH 3.0 and pH 5.0 acid rain treatments accordingly altered the soil microbial community structure at the early and later stage. However, acid rain did not change the microbial C sources utilization pattern. The principal response curve analysis revealed that the seasonal variation exerted a greater effect on the overall variance of soil microbial community structure than the acidity of acid rain. Our results demonstrate the asynchronous response of soil microbial community structure and function, which implies that the microbial functional redundancy may exist in the subtropical agricultural soil under acid rain.

Soil microorganisms are key regulators of soil carbon (C) and nutrients cycles in terrestrial ecosystems. However, it remains uncertain how the inter-annual variation in soil microbial community corresponds well with resource availability in the changing environment. Here, we investigated soil microbial community structure and abundance, as well as the associated environmental variables from 2015 to 2017 under different plant detritus input manipulation treatments in a coniferous (Platycladus orientalis (Linn.) Franco) plantation forest ecosystem in subtropical China. Our results showed that the inter-annual variation in soil microbial community was more visible than that caused by detritus input manipulations, owing to the temporal alterations in microclimates and substrate availability. Both aboveground litter removal and root exclusion had more negative effects on the bacterial PLFAs than fungi except half a year after detritus input manipulations. While, soil microbial abundance increased only after three years of litter addition compared to control. Litter removal, especially the no input treatment significantly increased the fungi to bacteria (F:B) and Gram-positive to Gram-negative bacteria (GP:GN) ratios after one and two years of detritus input manipulations. Whereas, the litter addition treatment had minor effects on these parameters. A clear discrimination of microbial community structure among the different detritus input manipulations appeared after two and three years. Both the F:B and GP:GN ratios were positively related to the carbon to nitrogen (C:N) ratio, recalcitrance index of carbon (RIC) and nitrogen (RIN). Overall, our results reveal that the inter-annual variations in soil microbial community are clearly differentiated by the environmental variables and substrate availability that occur in different years and detritus input manipulations. Our results also suggest that due to the vital role of microorganisms in biogeochemical cycling, shifts in the microbial community structure with altered plant detritus input could profoundly affect ecosystem processes in the long run.

Chloropicrin (CP) can cause long-term damage to beneficial microbes which reduces soil health. Biochar (BC) can mitigate against the effects of CP by reducing the time for beneficial microbes to recover after CP fumigation. In this study, we used Real-Time Quantitative PCR to determine the effects of different rates of BC added to CP-fumigated soil on the speed of recovery of bacteria and fungi population and on changes to gene copy number of the target pathogen Fusarium oxysporum. And then we compared the structure and composition of the beneficial microbial community in the different treatments soil by using High throughput Illumina sequencing. As the results shown, adding 1 or 3% BC after CP fumigation accelerated the recovery of bacterial and fungal populations without increasing F. oxysporum abundance. BC also promoted the recovery of beneficial bacteria Rokubacteria and Latescibacteria damaged by CP. And these two bacteria may be related to the immunity of soil to F. oxysporum. In CP-fumigated soil, BC improved the disease resistance of the soil by increasing beneficial microbes, such as Steroidobacter, Sphingomonas, Purpureocillium and Mortierella. This combination of CP and BC is a new concept that could encourages the development of a healthy and sustainable soil ecosystems while controlling plant pathogens.

The increased usage of rare earth elements (REEs) leads to the extensive exploitation of rare earth mines, and the REEs pollution in soil caused by the legacy mine tailings has brought great harm to environment and human health. Although Phytolacca americana can remove REEs from contaminated soil to some extent, there is still an urgent problem to improve its efficiency. Hyperaccumulator extract is a new organic material with potential in metal phytoextraction, but its role in REEs phytoremediation and the related underlying processes remain unclear. In this study, hyperaccumulator extracts from P. americana root (PR), stem (PS), leaf (PL) and EDTA were used to improve the phytoremediation efficiency of REEs with P. americana. Soil zymography was applied to assess the enzyme hotspots' spatial distribution in the rhizosphere, and the hotspots' microbial communities were also identified. The results indicated that the application of hyperaccumulator extracts improved the biomass and REEs uptake of P. americana, and the highest REEs content in plant was observed in the treatment of PS, which increased 299% compared to that of the control. Hotspots area of β-glucosidase, leucine aminopeptidase and acid phosphatase were concentrated in the pant rhizosphere along the roots and increased 2.2, 5.3 and 2.2 times after PS application compared to unamended soils. The PS application increased the relative abundance of Proteobacteria, Cyanobacteria, Bacteroidota and Firmicutes phyla in rhizosphere. Soil fungi have a higher contribution on promoting REEs activation than that of bacteria. Available P and extractable REEs were leading predictors for the plant biomass and REEs concentrations. The co-occurrence network showed that the application of PS creates a more efficient and stable microbial network compared to other treatments. In this study, 16S rDNA high-throughput sequencing, Fourier transform infrared spectroscopy, and two-dimensional correlation spectroscopy techniques were used to analyze the mechanisms driving the sequence of degradation of gummy substances by the microbial community and hydrolytic enzymes during the flax dew degumming process. The results revealed that the inoculation of combined bacteria induced quorum sensing, modulated hydrolytic enzyme production, and reshaped the community structure. Lignin-degraded genera (Pseudomonas and Sphingobacterium) were enriched, and the relative abundances of pectin- and cellulose-degraded genera (Chryseobacterium) decreased in the early degumming stages. Hemicellulose-degraded genera (Brevundimonas) increased over the degumming time. Moreover, the abundance of lignin hydrolytic enzymes improved in the early stages, while the abundance of pectin hydrolytic enzymes increased at the end of degumming. Various types of functional bacteria taxa changed the sequence of substance degradation. Electron scanning microscopy and differential scanning calorimetry results indicated that the degumming, facilitated by the inoculation of combined bacteria, was nearly completed by 21 d. The fibers exhibited smoother and more intact properties, along with higher thermal stability, as indicated by a melting temperature of 71.54 °C. Under the system of full straw returning, the relationship between soil fungal community diversity and soil physiochemical properties, and the combined application of slow-release nitrogen and urea is unclear. To evaluate its effect and provide an effective strategy for sustainable agricultural production, a 2-year field positioning trial was conducted using maize as the research object. The experiment was designed with two factors: straw treatment(S) and nitrogen fertilizer treatment(N),Six experimental treatments were set up,S1N0,S1N1,S1N2,S1N3,S1N4,S0N2,respectively.Analysis of 54 soil samples revealed 15 fungal phyla and 49 fungal classes. The composition of fungal communities in each treatment was basically the same, but there were significant differences in species abundance. Under total straw returning conditions, the combined application of slow-release nitrogen fertilizer and normal nitrogen fertilizer significantly increased the relative abundance of Ascomycota. During the jointing stage, tasseling stage and maturity stage, S1N4, S1N3 and S1N2 increased by 25.76%, 22.97%, 20.74%; 25.11%, 30.02%, 23.64% and 22.47%, 28.14%, 22.71% respectively compared with S0N2.The relative abundance of Basidiomycota was significantly reduced. Alpha diversity analysis showed that the straw returning mode significantly increased the Shannon index and decreased the Simpson index, which was obvious in the jointing stage and tasseling stage. The principal coordinate analysis analysis results showed that the fungal communities formed different clusters in the horizontal and vertical directions at the three growth stages of corn jointing, tasseling and maturity. At the jointing stage and tasseling stage, the communities of the straw return treatment and the straw removal treatment were separated, and the community distribution of each treatment was not significantly different in the mature stage. Total straw returning combined with slow-release fertilizer significantly (P＜0.05) increased the soil organic carbon, nitrate nitrogen and ammonia nitrogen content in each growth period, and increased the soil total nitrogen and hydrolyzable nitrogen content (P＞0.05).After the straw was returned to the field, the combined application of slow-release nitrogen fertilizer and common urea had a significant impact on soil urease, catalase, and sucrase activities. Among them, the three enzyme activities were the highest in the S1N3 treatment at the jointing stage and maturity stage, and the S1N4 treatment at the tasseling stage had the highest enzyme activity. Antibiotic residues in mariculture wastewater seriously affect the aquatic environment. Antibiotic Resistance Genes (ARGs) produced under antibiotic stress flow through the environment and eventually enter the human body, seriously affecting human health. Microalgal-bacterial symbiotic system (MBSS) can remove antibiotics from mariculture and reduce the flow of ARGs into the environment. This review encapsulates the present scenario of mariculture wastewater, the removal mechanism of MBSS for antibiotics, and the biomolecular information under metagenomic assay. When confronted with antibiotics, there was a notable augmentation in the extracellular polymeric substances (EPS) content within MBSS, along with a concurrent elevation in the proportion of protein (PN) constituents within the EPS, which limits the entry of antibiotics into the cellular interior. Quorum sensing stimulates the microorganisms to produce biological responses (DNA synthesis - for adhesion) through signaling. Oxidative stress promotes gene expression (coupling, conjugation) to enhance horizontal gene transfer (HGT) in MBSS. The microbial community under metagenomic detection is dominated by aerobic bacteria in the bacterial-microalgal system. Compared to aerobic bacteria, anaerobic bacteria had the significant advantage of decreasing the distribution of ARGs. Mining tailings containing large amounts of Pb and Cd cause severe regional ecosystem pollution. Soil microorganisms play a regulatory role in the restoration of degraded ecosystems. The remediation of heavy metal-contaminated tailings with amendments and economically valuable Eucalyptus camaldulensis is a research hotspot due to its cost-effectiveness and sustainability. However, the succession and co-occurrence patterns of these microbial communities in this context remain unclear. Tailing samples of five kinds of Cd and Pb were collected in E. camaldulensis restoration models. Physicochemical properties, the proportions of different Cd and Pb forms, microbial community structure, and the co-occurrence network of rhizosphere tailings during different restoration process (organic bacterial manure, organic manure, inorganic fertilizer, bacterial agent) were considered. Organic and organic bacterial manures significantly increased pH, cation exchange capacity, and the proportion of residual Pb. Still, there was a significant decrease in the proportion of reducible Pb. The changes in microbial communities were related to physicochemical properties and the types of amendments. Organic and organic bacterium manures decreased the relative abundance of oligotrophic groups and increased the relative abundance of syntrophic groups. Inorganic fertilizers and bacterial agents decreased the relative abundance of saprophytic fungi. B. subtilis would play a better role in the environment improved by organic manure, increasing the relative abundance of beneficial microorganism and reducing the relative abundance of pathogenic microorganism. pH, cation exchange capacity, and the proportion of different forms of Pb were the main factors affecting the bacterial and fungi variation. All four amendments transformed the main critical groups of the microbial network structure from acidophilus and pathogenic microorganisms to beneficial microorganisms. Heavy metal-resistant microorganisms, stress-resistant microorganisms, beneficial microorganisms that promote nutrient cycling, and copiotrophic groups have become critical to building stable rhizosphere microbial communities. The topological properties and stability of the rhizosphere co-occurrence network were also enhanced. Adding organic and organic bacterium manures combined with E. camaldulensis to repair Cd and Pb tailings improved (1) pH and cation exchange capacity, (2) reduced the biological toxicity of Pb, (3) enhanced the stability of microbial networks, and (4) improved ecological network relationships. Soilless revegetation is a cost-effective and eco-friendly method for the ecological restoration of gold mine tailings. However, due to gold mine tailings are high-salty, alkaline and low-nutrient, little research has been done on soilless revegetation of gold mine tailings. The aim of study was to apply soilless revegetation to gold mine tailings, and investigate the changes of physicochemical properties and microbial communities of tailings after soilless revegetation. Six selected herbaceous plants (Melilotus officinalis, Xanthium sibiricum, Festuca elata, Zoysia japonica, Amaranthus tricolor L., Artemisia desertorum) grew well on the bare tailings, and their heights reached as high as 16.28 cm after 90 days. After soilless revegetation, tailings salinity dramatically dropped from 547.15 to 129.24 μS cm−1, and pH went down from 8.68 to 7.59 at most. The content of available phosphorus (AP), available nitrogen (AN) and organic matter (OM) in tailings gradually improved, especially the content of AP and OM increased 53.36% and 52.58%, respectively. Furthermore, microbial metabolic activity and diversity in tailings obviously increased 70.33–264.70% and 1.64–13.97% respectively. The relative abundance of potential plant growth-promoting bacteria increased 1.40–3.05%, while the relative abundance of opportunistic pathogens and halophilic bacteria decreased 10.58–17.03% and 2.98–6.52% respectively. Such variations of microbial communities were beneficial for tailings restoration. This study provided insight into soilless revegetation and its impact on tailings microorganisms, which could be a new strategy for ecological restoration of gold mine tailings.

In this study, one lab-scale EGSB reactor (1.47 L volume) was designed to treat the antibiotic wastewater under different environmental factors, including the addition of cephalexin (CFX), Temperature (T) and Hydraulic Retention Time (HRT). The microbial community structure in EGSB reactor was analyzed with high-throughput sequencing technology to investigate their response to environmental factors changes, and then the random-matrix-theory (RMT)-based network analysis was used to investigate the microbial community’s molecular ecological network in EGSB systems treating antibiotics wastewater. Moreover, the explanatory value of each environmental factor on the change of microbial community structure was obtained through the result of redundancy analysis (RDA). The results showed that the addition of cephalexin (CFX), decline of T and decline of HRT (8 h) would decrease the removal efficiency of COD decreasing. And the removal efficiency of CFX would not be affected by decline of T and HRT, except the producing and degrading process of CFX by-products was changed obviously. The result of RDA analysis suggested the environmental factors mainly affected bacterial and fungal microbial community structure but not archaeal ones. The result of high-throughput sequencing showed the relative abundance (RA) of Firmicutes had been obviously affected by T and HRT, which might be main reason leading to the decrease of COD removal efficiency. In addition, molecular ecological network analysis showed the growth of Bacteroidetes occupied the niche of functional microorganism and led to the unstable operation of EGSB when T declined. What’s more, the molecular ecological network analysis revealed that Exophiala which belonged to fungi Ascomycota phylum was the hub genus to degrade complex refractory organic pollutants, and Aceticlastic methanogens Methanosaeta was the core functional archaea genus.

In recent years, in order to promote animal growth and reduce the risk of disease, a variety of antibiotics are frequently added to the animal feed of livestock and poultry. However, these antibiotics can not be fully digested by animals and most of them are excreted with feces, consequently causing the enrichment of antibiotic resistance genes (ARGs) and huge environmental risks. Nowadays, composting is a better option to solve these problems. Accordingly, this study explored the effects of co-composting swine manure with different inoculants dominated by Phanerochaete chrysosporium (p), Aspergillus niger (a), and Bacillus licheniformis (b) on the simultaneous removal of multiple antibiotics and resistance genes and evolution of the bacterial community. The results showed that the highest removal extent of tetracycline and oxytetracycline occurred in pile D (p:b:a=1:5:5, biomass) reaching 89.2% and 87.8%, respectively, while the highest removal extent of doxycycline and enrofloxacin occurred in pile A (p:b:a=1:0:0, biomass) reaching 98.6% and 89%, respectively. Compared with the levels in pile B (control check), in pile D, ARGs, except those for sulfonamides, decreased by 1.059 × 10−3–6.68 × 10−2 gene copies/16S rRNA copies. Inoculation with p alone effectively reduced intI1 and intI2. Canonical correspondence analysis (CCA) that microbial community structure evolution had a greater influence on ARGs than environmental factors. In summary, this study provided a feasible way to efficiently remove the antibiotics and antibiotic resistance genes in pig manure.

The return of straw to soil can help closing the nutrient cycle in agricultural ecosystems and is an effective means of improving the physical and chemical properties of soil. The objective of this research was to evaluate the effect of rice straw return on soil organic carbon (SOC) fractions and microbial community with a 10-year plot experiment in Northeast China. The study involved four treatments: no rice straw return (S0), a low amount of rice straw return (S1), a high amount of rice straw return (S2), and abandoned farmland (AL). The results indicated that the labile organic carbon (LOC), particulate organic carbon (POC), dissolved organic carbon (DOC), and microbial biomass carbon (MBC) in the S1, S2, and AL treatments were significantly higher than those in the S0 treatment. Rice straw return increased soil microbial richness but did not affect the soil microbial diversity. Proteobacteria, Bacteroidetes, Firmicutes, Acidobacteria, and Verrucomicrobia were the dominant phyla in all treatments based on their relative abundances. Redundancy analysis indicated that MBC and DOC had relatively large influences on the microbial community.

Choosing a good crop rotation plan helps maintain soil fertility and creates a healthy soil ecosystem. However, excessive fertilization and continuous cultivation of vegetables in a greenhouse results in secondary salinization of the soil. It remains unclear how crop rotation affects Yunnan's main place for vegetable growing in the greenhouse. Six plant cultivation patterns were chosen to determine how different rotation patterns affect the chemical properties and the soil microbial communities with secondary salinization, including lettuce monoculture, lettuce-large leaf mustard, lettuce-red leaf beet, lettuce-cabbage, lettuce-romaine lettuce, and lettuce-cilantro (DZ, A1, A2, A3, A4, and A5). The results showed that all treatments increased the proportion of nutrients available in the soil, and the effect of the A1 treatment was the most significant compared to the monoculture mode. The high-throughput sequencing findings revealed that distinct crop rotation patterns exerted varying effects on the microbial communities. Microbial community diversity was significantly lower in the monoculture than in the other treatments. The number of microbial operational taxonomic units OTUs was significantly higher in the crop rotation modes (P < 0.05), and the A1 treatment had larger numbers and diversity of bacterial and fungal OTUs (Shannon's and Simpson's) than other treatments (P < 0.05). Prominent bacterial and fungal communities were readily observable in the soils planted with rotational crops. Proteobacteria had the highest relative abundance of bacteria, whereas Ascomycota was the most abundant fungus. The principal coordinate analysis at the OTU level separated soil bacterial and fungal growth communities under the different treatments. Among the six treatments, The first two axes (PC1 and PC2) described 46.44 % and 42.42 % of the bacterial and fungal communities, respectively. Network-based analysis showed that Bacteroidota and Gemmatimonadota members of the genus Bacteroidota were positively correlated with Proteobacteria. Members of Ascomycota and Chytridiomycota exhibited positive relationships. These results extend the theoretical understanding of how various crop rotation patterns affect soil chemical properties, microbial community diversity, and metabolic functions. They reveal the beneficial effects of crop rotation patterns on enhanced soil quality. Light radiation can degrade recalcitrant materials like lignocelluloses in litter and serve as a physical condition to accelerate green waste (GW) decomposition, but few studies have considered the microbial effects of light wavelength on GW composting. This study innovatively investigated the effects of different wavelengths of light radiation, including full-spectrum, no blue light, no UV, no UV-A, no UV-B, and dark conditions, on accelerating the GW composting process. Especially, the study explored the dynamic changes in the degradation of lignocelluloses and evaluated the responses of microorganisms throughout the composting process under different light radiation wavelengths. No blue light (where radiation between 400 and 500 nm was blocked by the film) yielded the highest-quality compost within 40 days. In comparison to the dark (control), no blue light exhibited an elevated composting temperature (56.7 °C), an extended thermophilic phase (6 days), and increased degradation rates of lignin, cellulose, and hemicellulose by 13 %, 15 %, and 12 %, respectively. This study revealed that during the composting mesophilic phase, bacterial diversity performed best under no blue light, while fungal diversity excelled under full-spectrum. In the thermophilic phase, microbial diversity exhibited optimal performance under full-spectrum. During the cooling phase, bacterial diversity was highest under no blue light, and fungal diversity excelled under no UV-A. During the mesophilic and cooling phases, the bacterial ACE index for no blue light exceeded that of the other light radiation wavelengths, with values of 418 and 494, respectively. Under no blue light, the Shannon index of microorganisms remained within the range of 2.0-4.8, demonstrating superior performance. Meanwhile, the relative abundances of lignin-degrading microorganisms (Flavobacterium, Acaulium, and Acremoniu) under no blue light has increased, demonstrating improved microbial community structures. Preventing soil nitrogen (N) losses driven by microbial nitrification and denitrification contributes to improving global environmental concerns caused by NO 3 - -N leaching and N 2 O emission. Quorum sensing (QS) signals regulate nitrification and denitrification of N-cycling bacteria in pure culture and water treatment systems, and mediate the composition of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in activated sludge. However, whether disrupting QS could prevent soil N losses remains unclear. This study explored the feasibility of applying quorum sensing inhibitors (QSIs) as an innovative strategy to reduce N losses from agricultural soils. The two QSIs, penicillic acid and 4-iodo-N-[(3S)-tetrahydro-2-oxo-3-furanyl]-benzeneacetamide (4-iodo PHL), were more effective in reducing N losses than traditional inhibitors, including N-(n-butyl) thiophosphoric triamide and 3,4-dimethylpyrazole phosphate. After 36 days of aerobic incubation, penicillic acid and 4-iodo PHL inhibited nitrification by 39% and 68%, respectively. The inhibitory effects are attributed to the fact that 4-iodo PHL decreased the abundance of archaeal and bacterial amoA genes, as well as the relative abundance of Candidatus Nitrocosmicus (AOA), Candidatus Nitrososphaera (AOA), and Nitrospira (nitrite-oxidizing bacteria/comammox), while penicillic acid reduced archaeal amoA abundance and the relative abundance of Nitrosospira (AOB) and the microbes listed above. Penicillic acid also strongly inhibited denitrification (33%) and N 2 O emissions (61%) at the peak of N 2 O reductase gene (nosZ) abundance, respectively. Furthermore, the environmental risks of QSIs to microbial community structure and network stability, CO 2 O reductase gene (nosZ) abundance, respectively. Furthermore, the environmental risks of QSIs to microbial community structure and network stability, CO 2 emissions, and soil animals were acceptable. Overall, QSIs have application potential in agriculture to reduce soil N losses and the associated effect on climate change. Groundwater contamination from abandoned pesticide sites is a prevalent issue in China. To address this problem, natural attenuation (NA) of pollutants has been increasingly employed as a management strategy for abandoned pesticide sites. However, limited studies have focused on the long-term NA process of co-existing organic pollutants in abandoned pesticide sites by an integrated approach. In this study, the NA of benzene, toluene, ethylbenzene, and xylene (BTEX), and chlorobenzenes (CBs) in groundwater of a retired industry in China was systematically investigated during the monitoring period from June 2016 to December 2021. The findings revealed that concentrations of BTEX and CBs were effectively reduced, and their NA followed first-order kinetics with different rate constants. The sulfate-reducing bacteria, nitrate-reducing bacteria, fermenting bacteria, aromatic hydrocarbon metabolizing bacteria, and reductive dechlorinating bacteria were detected in groundwater. It was observed that distinct environmental parameters played a role in shaping both overall and key bacterial communities. ORP (14.72%) and BTEX (12.89%) were the main drivers for variations of the whole and key functional microbial community, respectively. Moreover, BTEX accelerated reductive dechlorination. Furthermore, BTEX and CBs exhibited significant enrichment of 13 C, ranging from +2.9 to +27.3‰, demonstrating their significance in situ biodegradation. Till present, natural wetlands have been continuously subjected to intensive pollution stress in recent years, mainly because of the rapidly growing industrialization and urbanization that are associated with a myriad of anthropogenic activities and land use practices. These man-made sources of pollution change the chemical properties of the natural wetlands, which in turn alter their microbial ecological biodiversity and functions. For the first time, the impact of the current anthropogenic activities and land use practices on the Blesbokspruit wetland chemical status and their consequential effect on the microbial structure and functions were investigated. Sites of high pollution intensity were identified using geographic information systems mapping (GISMapping) and the wetland microbiome and functional profile were studied through the use of high throughput shotgun metagenomics sequencing analysis. The predominant phyla that stemmed along the Blesbokspruit wetland were found to be Proteobacteria which was more dominant in water (93 %) than in the sediments (89 %), followed by firmicutes which was more abundant in sediments (9 %) than in water (6 %), and Bacteroidetes were relatively low in abundance within both the sediments (2 %) and the overlying water (1 %). The genera Klebsiella (70.4 %-28.2 %), Citrobacter (52.0 %-30.6 %), Escherichia (51.0 %-8.4 %), and Lynsinibacillus (9.3 %-1.5 %) were observed in most water and sediment samples. Within the six polluted sites, Site 2 was found to be the most highly polluted site in the Blesbokspruit wetland with very high COD (900 mg/L), TOC (11.60 mg/L), NO 3 - (39.74 mg/L), NO 2 - (12.64 mg/L), PO 4 (145.95 mg/L) concentrations recorded in the water and high levels of TOC (0.37 mg/L), TC (6.92 %), TN (1.82 %), TS (0.53 %) in sediments. The microbial community structure and functions were found to be strongly influenced by the high organic content from the intense agricultural activities and sewage spillages and heavy metals from the mining activities nearby.3 (4.14 mg/L), Fl - (143.88 mg/L), Cl - (145.95 mg/L) concentrations recorded in the water and high levels of TOC (0.37 mg/L), TC (6.92 %), TN (1.82 %), TS (0.53 %) in sediments. Aerobic composting increases the content of soluble nutrients and facilitates the safe treatment of livestock manure. Although different taxa play crucial roles in maintaining ecological functionality, the succession patterns of community composition and assembly of rare and abundant subcommunities during aerobic composting under antibiotic stress and their contributions to ecosystem functionality remain unclear. Therefore, this study used 16 S rRNA gene sequencing technology to reveal the response mechanisms of diverse microbial communities and the assembly processes of abundant and rare taxa to amoxicillin during aerobic composting. The results indicated that rare taxa exhibited distinct advantages in terms of diversity, community composition, and ecological niche width compared with abundant taxa, highlighting their significance in maintaining ecological community dynamics. In addition, deterministic (heterogeneous selection) and stochastic processes (dispersal limitation) play roles in the community succession and functional dynamics of abundant and rare subcommunities. Understory vegetation has an important impact on soil organic carbon (SOC) accumulation. However, little is known about how understory vegetation alters soil microbial community composition and how microbial diversity contributes to SOC chemical composition and persistence during subtropical forest restoration. In this study, removal treatments of an understory fern (Dicranopteris dichotoma) were carried out within pine (Pinus massoniana) plantations restored in different years in subtropical China. Soil microbial community composition and microbial diversity were measured using phospholipid fatty acids (PLFAs) biomarkers and high-throughput sequencing, respectively. The chemical composition of SOC was also measured via solid-state 13 C nuclear magnetic resonance ( 13 C NMR). Our results showed that fern removal decreased alkyl C by 4.2 % but increased O-alkyl C by 15.6 % on average, leading to a decline of alkyl C/O-alkyl C ratio, suggesting altered chemical composition of SOC and lowered SOC recalcitrance without fern. Fern removal significantly lowered the fungi-to-bacteria ratio, and it also reduced fungal and bacterial diversity. Partial correlation analysis revealed that soil nitrogen availability was a key factor influencing microbial diversity. Bacterial diversity showed a close relationship with the Alkyl C/O-alkyl C ratio following fern removal. Furthermore, the microbial community structure and bacterial diversity were responsible for 18 % and 55 % of the explained variance in the chemical composition of SOC, respectively. Taken together, these analyses jointly suggest that bacterial diversity exerts a greater role than microbial community structure in supporting SOC persistence during understory fern removal. Livestock manure is a major source of veterinary antibiotics and antibiotic resistance genes (ARGs). Elucidation of the residual characteristics of ARGs in livestock manure following the administration of veterinary antibiotics is critical to assess their ecotoxicological effects and environmental contamination risks. Here, we investigated the effects of enrofloxacin (ENR), a fluoroquinolone antibiotic commonly used as a therapeutic drug in animal husbandry, on the characteristics of ARGs, mobile genetic elements, and microbial community structure in swine manure following its intramuscular administration for 3 days and a withdrawal period of 10 days. The results revealed the highest concentrations of ENR and ciprofloxacin (CIP) in swine manure at the end of the administration period, ENR concentrations in swine manure in groups L and H were 88.67 ± 45.46 and 219.75 ± 88.05 mg/kg DM, respectively. Approximately 15 fluoroquinolone resistance genes (FRGs) and 48 fluoroquinolone-related multidrug resistance genes (F-MRGs) were detected in swine manure; the relative abundance of the F-MRGs was considerably higher than that of the FRGs. On day 3, the relative abundance of qacA was significantly higher in group H than in group CK, and no significant differences in the relative abundance of other FRGs, F-MRGs, or MGEs were observed between the three groups on day 3 and day 13. The microbial community structure in swine manure was significantly altered on day 3, and the altered community structure was restored on day 13. The FRGs and F-MRGs with the highest relative abundance were qacA and adeF, respectively, and Clostridium and Lactobacillus were the dominant bacterial genera carrying these genes in swine manure.
