Emerging as a promising drug target for Alzheimer's disease (AD) therapy, glycogen synthase kinase 3β (GSK-3β) has garnered attention. This study sought to rigorously scrutinize a compendium of natural compounds retrieved from the ZINC database through pharmacodynamic experiments, employing a 1 H-indazole-3-carboxamide (INDZ) scaffold, to identify compounds capable of inhibiting the GSK-3β protein. Utilizing a multi-step approach, the study involved pharmacophore analysis, followed by molecular docking to select five promising ligands for further investigation. Subsequently, ESMACS simulations were employed to assess the stability of the ligand-protein interactions. Evaluation of the binding modes and free energy of the ligands revealed that five compounds (2a-6a) exhibited crucial interactions with the active site residues. Furthermore, various methodologies, including hydrogen bond and clustering analyses, were utilized to ascertain their inhibitory potential and elucidate the factors contributing to ligand binding in the protein's active site. The findings from MMPBSA/GBSA analysis indicated that these five selected small molecules closely approached the IC50 value of the reference ligand (OH8), yielding energy values of -34.85, -32.58, -31.71, and -30.39 kcal/mol, respectively. Additionally, an assessment of the interactions using hydrogen bond and dynamic analyses delineated the effective binding of the ligands with the binding pockets in the protein. Background: With the progress and prevalence of COVID-19, concerns have arisen regarding its impact on men's sexual health. Therefore, this study was conducted with the aim of examining the effects of COVID-19 on serum levels of sex hormones and semen., Methods: Sixty participants who met the study inclusion criteria enrolled in this study between January and April 2022. The individuals were divided into three groups (n = 20): healthy, COVID-19 positive, and recovered from COVID-19. Blood and semen samples were collected from the participants. Serum levels of sex hormones and semen were evaluated both macroscopically and microscopically., Results: Our study results showed that the most common symptoms observed in the COVID-19 group were cough (100 %), fever (100 %), fatigue (95 %), and runny nose (90 %). Serum levels of sex hormones (testosterone, FSH, and prolactin) in the COVID-19 group were significantly decreased compared to the healthy group. Microscopic examination of semen revealed significant differences in vitality, progressive, and motile parameters among the three groups, with a decrease observed in the COVID-19 group., Conclusion: These results indicate that COVID-19 may have a negative impact on men's sexual health, potentially affecting hormone production and sperm quality. Introduction: Pain and anxiety are a major problem of burn patients. Despite the use of different medications, patients still suffer from these two problems. Aromatherapy along with medication may help in alleviating these symptoms. This study aimed to investigate the effect of inhalation aromatherapy with damask rose essence on pain and anxiety in burn patients., Methods: This three group clinical trial was conducted on 120 patients with burns less than 30% TBSA. The patients were randomly allocated into three groups, aromatherapy damask rose essence, placebo, and control. The pain intensity was assessed using visual analogue scale prior to intervention, immediately before, and 15 min after dressing. Anxiety was measured using Spielberger Inventory at before intervention and 15 min after dressing, also the prolonged effect of intervention on pain was assessed by number of the analgesics drugs received for four hours after dressing change. The intervention included inhalation of 6 drops of 40% damask rose essential oil in the damask group, and six drops of distilled water in placebo group one hour before dressing change. The control group received no intervention., Results: Baseline state-trait anxiety and pain intensity were similar in these three groups (p > 0.05). We found a significant reduction in pain intensity immediately before and after dressing and state anxiety after dressing in the damask group compared to the placebo and control groups (p < 0.001). However, we found no significant difference between the placebo and control groups in terms of these variables at these times (p > 0.05). Also, we found no significant difference among three groups in frequency of analgesics drugs and trait anxiety after intervention (p > 0.05)., Conclusion: Inhaled aromatherapy with Damask rose essence reduces subjective pain intensity and state anxiety in burned patients. Nanoparticles (NPs) are of the most interesting novel vehicles for effective drug delivery to humans. Freeze drying is known as an engaging process to improve the long lasting stability of NPs formulations. This study aims to elucidate the importance of various parameters involving in freeze-drying of the most common pharmaceutical/nutraceutical NPs including nanosuspensions, nanocrystals (NCs), cocrystals/nanococrystals, nanoemulsions (NEs), nanocapsules (NCPs) and nanospheres (NSPs). Regarding this, the therapeutic goals of NPs and specifications of drug must be considered. According to our survey, the most influential factors for achieving optimum results include type and concentration of cryoprotectant/lyoprotectant, stabilizer structure and concentration, the NPs concentration in solution, freezing, annealing, and drying rate, the interaction between protectants and stabilizer, solvent type and antisolvent to solvent ratio. The study shows that for each class of NPs, specific variables are of highest significance and should be optimized. For instance, about NCs, freezing rate and antisolvent/solvent ratio should be particularly considered and for emulsified NPs, the best results have been obtained by 5-20% of saccharides as cryoprotectants. These findings suggest that to obtain a product with the lowest aggregation and particle size (PS), optimization of the effective factors in formulation and lyophilization process are essential., (Copyright © 2020 American Pharmacists Association®. Introduction: Herpes simplex virus (HSV) is among the most common viruses in humans. HSV1 is often responsible for oral and perioral herpetic lesions. Photodynamic therapy (PDT) is a novel antimicrobial modality that involves the use of laser and a photosensitizer with a specific wavelength. This study aimed to assess and compare the effect of PDT with 810 and 940 nm diode laser and indocyanine green (ICG) photosensitizer on HSV1., Methods: In this in vitro study, HSV1 isolated from herpes labialis and there were 6 experimental groups.The irradiation parameters were the same for all groups. Number of remaining viruses per milliliter in each group was determined using real-time polymerase chain reaction (PCR) and statistically analyzed by ANOVA., Results: The virus count in all groups significantly decreased compared to the control group (P < 0.05) except in group ICG- without irradiation (P > 0.05). Comparison of groups 810- and 940- (use of each laser alone) with groups 810+ and 940+ (use of each laser plus ICG) revealed that reduction in virus count in groups 810+ and 940+ was significantly greater than that in groups 810- and 940-., Conclusion: 810 nm diode laser irradiation and ICG causes the greatest reduction in number of HSV1 compared to all the other groups. Introduction: Schizophrenia is a disabling psychiatric disorder. The role of Borna Disease Virus (BDV) in the etiology of schizophrenia has been suggested by several studies. However, the existence of such association remained controversial. The present meta-analysis was conducted to evaluate this association., Method: This systematic review and meta-analysis was conducted using preferred reporting items for systematic reviews and meta-analysis (PRISMA). Online databases including Scopus, PubMed, Science direct, Embase, PsycINFO, Web of Science and Google scholar search engine were searched until January 15, 2017. The heterogeneity of the studies was evaluated using Cochran's Q test and I 2 statistic. Finally, random effects model was used for combining the results using Stata software version 11.1., Result: Overall, 30 studies containing 2533 cases and 4004 controls were included in the meta-analysis. The combined odds ratio (OR) for the relationship between BDV and schizophrenia was estimated to be 2.72 (95%CI: 1.75-4.20). This association based on RT-PCR, WB, IFA, EIA, RLA, ECLIA methods was estimated to be 3.83 (95%CI: 1.59-9.20), 4.99 (95%CI: 1.80-13.85), 1.27 (95%CI: 0.23-7.12), 2.26 (95%CI: 0.48-10.64), 1.67 (95%CI: 0.50-5.56) and 2.88 (95%CI: 1.38-6.01), respectively. Subgroup analysis according to WBC, serum and plasma samples was estimated to be 3.31 (95%CI: 1.19-9.25), 2.21 (95% CI: 1.17-4.17), 2.21 (95%CI: 1.03-4.73) and 7.89 (95%CI: 1.75-35.53), respectively., Conclusion: The results indicated the role of BDV in the etiology of schizophrenia., (Copyright © 2017. Substance use disorder is one of the main mental health problems in Iran. In this paper, the six-monthly counts of deaths due to substance abuse in Iran at provincial level between March 21, 2005, and March 20, 2014, were modeled using a log-Gaussian Cox point process model. By assuming population density as the exposure variable, the considered model incorporated known and unknown influential factors in order to describe spatio-temporal variations in the relative risk of substance abuse mortality. We found evidence of spatial heterogeneity and inequality by gender in deaths related to substance abuse across Iran. The present study aimed to measure the concentrations of Sn, Pb, Zn, Hg, Cu, Ni and Cd in the muscle and liver of 40 Common Moorhens (Gallinula chloropus) hunted from four stations in Anzali Wetland (Pirbazar, Ghalam-Koudeh, Selkeh and Abkenar). The histopathologic alteration index (HAI) of liver and kidney was also assessed in these birds. The highest concentrations of selected metals were measured in the liver of birds collected from Ghalam-Koudeh (Pb: 4.59±0.21, Sn: 6.663±0.282, Zn: 29.867±2.011, Cu: 24.07±1.84, Hg: 7.5±0.257, Ni: 6.85±0.52, Cd: 1.879±0.4mg kg(-1) dw). The lowest concentrations of metals were measured in the muscle of birds caught from Abkenar (Pb: 0.799±0.207, Sn: 1.873±0.066, Zn: 18.533±1.582, Hg: 0.86±0.08, Ni: 0.53±0.117, Cu: 6.63±1.114, Cd: 0.08±0.002mg kg(-1) dw). Also the highest and lowest concentrations of metals were recorded in sediment of Ghalam-Koudeh and Abkenar stations, respectively. These stations were located next to multi-industry Anzali Port. However, the concentration of Sn and Zn in sediment and tissues of Common Moorhens collected from different stations was lower than the permissible limit suggested by WHO and Canadian Council of Ministers of the Environment (CCME). But, Pb, Hg and Ni concentration in sediment and birds caught from all stations was higher than the permissible limit defined by WHO and CCME. Cu and Cd concentration in tissue samples and sediment of Ghalam-Koudeh and Pirbazar was also higher than the permissible limit defined by WHO and CCME. Hemorrhage, melanomacrophage aggregations, sinusoidal congestion and hepatocyte vacuolation were the most pathological changes found in the liver. Reduction of the Bowman space, melanomacrophage aggregations and hemorrhage also were observed in the kidney. The HAI means of G. chloropus collected from Ghalam-Koudeh and Pirbazar were significantly higher than other sites. Based on the HAI values and metal bioaccumulation in the tissues of G. chloropus, Ghalam-Koudeh and Pirbazar could be considered as having the worst environmental quality., (Copyright © 2014 Elsevier Inc.
