The spreading of coronavirus from contacting surfaces and aerosols created a pandemic around the world. To prevent the transmission of SARS-CoV-2 virus and other contagious microbes, disinfection of contacting surfaces is necessary. In this study, a disinfection box equipped with infrared (IR) radiation heating and ultraviolet-C (UV-C) radiation is designed and tested for its disinfection ability against pathogenic bacteria and SARS-CoV-2 spike protein. The killing of a Gram-positive, namely, S. aureus and a Gram-negative namely, S. typhi bacteria was studied followed by the inactivation of the spike protein. The experimental parameters were optimized using a statistical tool. For the broad-spectrum antibacterial activity, the optimum condition was holding at 65.61 °C for 13.54 min. The killing of the bacterial pathogen occurred via rupturing the cell walls as depicted by electron microscopy. Further, the unfolding of SARS-CoV-2 spike protein and RNase A was studied under IR and UV-C irradiations at the aforesaid optimized condition. The unfolding of both the proteins was confirmed by changes in the secondary structure, particularly an increase in β-sheets and a decrease in α-helixes. Remarkably, the higher penetration depth of IR waves up to subcutaneous tissue resulted in lower optimum disinfection temperature, <70 °C in vogue. SARS-CoV-2 virus and other pathogenic microbes are transmitted to the environment through contacting surfaces, which need to be sterilized for the prevention of COVID-19 and related diseases. In this study, a prototype of a cost-effective sterilization box is developed to disinfect small items. The box utilizes ultra violet (UV) radiation with heat. For performance assessment, two studies were performed. First, IgG (glycoprotein, a model protein similar to that of spike glycoprotein of SARS-COV-2) was incubated under UV and heat sterilization. An incubation with UV at 70 °C for 15 min was found to be effective in unfolding and aggregation of the protein. At optimized condition, the hydrodynamic size of the protein increased to ~171 nm from ~5 nm of the native protein. Similarly, the OD 280 values also increased from 0.17 to 0.78 indicating the exposure of more aromatic moieties and unfolding of the protein. The unfolding and aggregation of the protein were further confirmed by the intrinsic fluorescence measurement and FTIR studies, showing a 70% increase in the β-sheets and a 22% decrease in the α-helixes of the protein. The designed box was effective in damaging the protein's native structure indicating the effective inactivation of the SARS-COV-2. Furthermore, the incubation at 70 °C for 15 min inside the chamber resulted in 100% antibacterial efficacy for the clinically relevant E.coli bacteria as well as for bacteria collected from daily use items. Exclusive magnetocaloric properties of orthoferrites offer advantages for their application in the magnetic hyperthermia as well as imaging applications. In the present study, the effect of yttrium concentration on the magnetic characteristics of the iron oxide based nanomaterials was analyzed to assess their potential for the hyperthermia applications. The Sol-gel method was used to synthesize the Yttrium Iron Garnet (YIG) based nanoparticles, using different molar ratios of Fe and Y precursors, followed by the calcination at 900, 1000 and 1100 °C. XRD analysis determined the formation of the pure phase of yttrium iron garnet Y 3 Fe 5 O 12 (YIG) at 0.5 molar ratio of yttrium at all the calcination temperatures and pure phase of yttrium iron perovskite YFeO 3 (YIP) for 1 molar ratio of yttrium at 1000 and 1100 °C. The mean particle size was observed in the range of 100 to 400 nm. The magnetic characterization studies showed the highest saturation magnetization for the sample containing 0.5 molar ratio of the yttrium calcinated at 1000 °C. The magnetization values were linearly related to the contents of YIG phases in the synthesized samples. Induction heating of YIG resulted in the hyperthermia temperature (42 to 44 °C) in 13 min with the SAR values 114.65 W/g at 1 mg/ml. The prepared samples showed no in-vitro toxic effects on the MG63 cells (>90% cell viability). In addition, in-vitro treatment at hyperthermia temperature for 15 min reduced cell viability of cancer cells (A549) to 55%, while no toxic effect was observed on MG 63 cells. Amyloid fibrils are the hallmarks of neurodegenerative diseases like Alzheimer's, Parkinson's and other proteopathies. Inhibition of fibrillation is a potential strategy to check the progress of amyloid associated diseases and further allied deterioration. In this study, we have synthesized proline functionalized gold nanoparticles (Pro-AuNPs) and scrutinized its antifibrillation property towards Hen Egg White Lysozyme (HEWL) aggregation. Pro-AuNPs were characterized using various biophysical methods like ultraviolet-visible spectroscopy, fourier transform infra-red spectroscopy, zeta potential measurement, dynamic light scattering and transmission electron microscopy. The effect of Pro-AuNPs on HEWL fibrillation was analyzed employing thioflavin T (ThT) and 8-Anilino-1-naphthalenesulfonic acid (ANS) assays. The kinetics of HEWL exhibited a typical sigmoidal nature of protein aggregation and was fitted to Boltzmann model. HEWL in the presence of bare gold nanoparticles (bAuNPs) exhibited similar aggregation kinetics as HEWL alone. However, HEWL fibrillation substantially reduced upon co-incubation with proline and Pro-AuNPs, and two slightly different intermediate species were formed with these two systems as predicted by CD spectroscopy. TEM images also supported the above observation displaying different morphological states of HEWL aggregates in the presence of proline and Pro-AuNPs. Using computational methods, the nature of interaction of HEWL and proline was found to be hydrogen bonding and hydrophobic interaction in multiple amyloidogenic regions. These interactions inhibited the formation of prefibrils (β-sheet rich intermediates) and also found to disintegrate fibrils. Furthermore, HEWL-Pro-AuNPs system resulted HEWL adsorption through hydrophobic patches, which blocked the intermolecular β-sheet formation. The use of gadolinium orthoferrite for biomedical application like as contrast agents for magnetic resonance imaging (MRI) has been found to be very promising due to its fascinating properties. The present study focuses on the determination of the effect of the gadolinium concentration in the formation biphasic α-Fe 2 O 3 -GdFeO 3 for hyperthermia applications. An in-situ sol-gel technique was adopted for the synthesis of biphasic orthoferrites with four different gadolinium concentrations. The XRD analysis confirmed the formation of gadolinium orthoferrites after heat treatment at 1000 °C, 1100 °C, and 1200 °C. The presence of α-Fe 2 O 3 in trace amounts was observed in the materials with low gadolinium concentrations. VSM (Vibrating-sample magnetometer) analysis was performed to ensure the magnetic properties of the materials, which were found to be weakly ferromagnetic. The biocompatibility of the materials was investigated through MTT assay and no cytotoxic effect was observed. The assessment of heating ability of the materials was performed under an alternating magnetic field using an induction heating instrument and all the samples showed temperature rise in the range of hyperthermia temperature with a maximum temperature of 55.71 °C in 6 min. In the present work, we aimed to synthesize highly efficient nano-composite polymeric scaffolds with controllable pore size and mechanical strength. We prepared nanocomposite (CCNWs-AgNPs) of silver nanoparticles (AgNPs) decorated on carboxylated CNWs (CCNWs) which serves dual functions of providing mechanical strength and antimicrobial activity. Scaffolds containing chitosan (CS) and carboxymethyl cellulose (CMC) with varying percent of nanocomposite were fabricated using freeze drying method. XRD and FESEM analysis of nanocomposite revealed highly crystalline structure with AgNPs (5.2 nm dia) decorated on ~200 nm long CCNWs surface. FTIR analysis confirmed the interaction between CCNWs and AgNPs. Incorporation of nanocomposite during scaffolds preparation helped in achieving the desirable 80-90% porosity with pore diameter ranging between 150 and 500 μm and mechanical strength was also significantly improved matching with the mechanical strength of cancellous bone. The swelling capacity of scaffolds decreased after the incorporation of nanocomposite. In turn, scaffold degradation rate was tuned to support angiogenesis and vascularization. Scaffolds apart from exhibiting excellent antimicrobial activity, also supported MG63 cells adhesion and proliferation. Incorporation of CCNWs also resulted in improved biomineralization for bone growth. Overall, these studies confirmed excellent properties of fabricated scaffolds, making them self-sustained and potential antimicrobial scaffolds (without any loaded drug) to overcome bone related infections like osteomyelitis., (Copyright © 2018 Elsevier B.V. Various metal ions are recently implicated in protein aggregates and are associated with numerous neurodegenerative diseases. In the present work, we have scrutinized the effect of stoichiometric variation of Cu(II) on BSA fibrillation at physiological pH 7.4 through Thioflavin-T dye binding study, residual protein concentration, Fourier Transform Infrared spectroscopy (FTIR), Dynamic Light Scattering (DLS), Atomic Force Microscopy (AFM) and Isothermal Titration Calorimetry (ITC). Fibrillation kinetics was studied through ThT fluorescence, which illustrated dependency on stoichiometric ratio of Cu(II). The ThT intensity data were fitted to a single exponential expression to determine the aggregation rate, k, which revealed a slight lower k value for 1:1 ratio of BSA-Cu(II) reaction as compared to BSA alone whereas k value for 1:2 ratio of BSA-Cu(II) reaction was higher. Higher equilibrium residual concentration in case of 1:1 ratio of BSA-Cu(II) agreed to the lower aggregation rate. IR spectroscopy revealed the presence of increased β-sheet proportion to the detriment of α-helix conformation with increasing concentration of Cu(II) and illustrated maximum β-sheet proportion in 1:2 ratio of BSA-Cu(II). These results were combined with scattering results that showed the higher average hydrodynamic radius (Z-average value) of aggregates in 1:2 ratio of BSA-Cu(II) with respect to BSA and 1:1 ratio of BSA-Cu(II). AFM analysis confirmed the fibril formation. ITC analysis has shown the presence of two binding sites with many fold difference in binding affinity at prescribed in vitro conditions. An N-terminal binding site with many fold higher binding affinity was found as the first Cu(II) binding site. The free thiol group of the cysteine residue positioned at 34 (cys-34) in BSA was covalently capped and this modified BSA also showed the presence of two binding sites, which declined the cys-34 site as the second Cu(II) bind site. Therefore Zn(II) binding site was predicted as the second Cu(II) binding site. The binding of Cu(II) at N-terminal binding site marks a square planner geometry; while that at zinc binding site marks a rigid coordination geometry. Conclusively, analyzed data has concluded that the variation in Cu(II) efficacy in the different stoichiometric ratio results because of two different coordination types in different stoichiometric ratios., (Copyright © 2017 Elsevier B.V. The present study deals with kinetic studies of the chemical modification for synthesizing a hydrophobic silica surface. Surface silanization (modification) via formation of Self-Assembled Monolayer (SAM) using a short chain triethoxyoctylsilane (TEOS) was carried out under inert atmosphere at room temperature. Fourier transmission infrared (FTIR) spectroscopy, water contact angle (WCA) and atomic force microscopy (AFM) were employed to investigate surface modification. FTIR analysis in the range from 900-1200cm(-1) and 2850-3000cm(-1) confirmed surface modification and re-orientation of the attached molecules. Kinetic studies of TEOS SAM formation were fitted by Exponential Association function. Kinetic fitting of FTIR data in the range from 900-1200cm(-1) revealed a very fast attachment of TEOS molecules resulting in total surface coverage within 16min whereas re-orientation rate was slow and continued till 512min. Further, change in orientation from lying-down to standing-up state was supported by contact angle analysis. AFM images initially showed small islands of ~20nm, which in-fill with time indicating formation of a smooth monolayer. Our findings indicate that formation of octyl SAM is fast process and completes within 8.5h in contrary to reported 24h in conventional SAM formation protocols. We synthesized surfaces with different hydrophobicities and roughness by forming self-assembled monolayers (SAMs) of mixed amine and octyl silanes. Insulin aggregation kinetics in the presence of the above surfaces is characterized by a typical lag phase and growth rate. We show that the lag time but not the growth rate varies as a function of the amine fraction on the surface. The amount of adsorbed protein and the adsorption rate during the aggregation process also vary with the amine fraction on the surface and are maximal for equal parts of amine and octyl groups. For all surfaces, the growth phase starts for identical amounts of adsorbed insulin. The initial surface roughness determines the rate at which protein adsorption occurs and hence the time to accumulate enough protein to form aggregation nuclei. In addition, the surface chemistry and topography influence the morphology of aggregates adsorbed on the material surface and the secondary structures of final aggregates released in solution., (Copyright © 2012 Elsevier B.V.
