Background: The Spinal Instability Neoplastic Score (SINS) classification system is a validated and the most widely accepted instrument for defining instability in vertebral metastasis (VM), in which lesions scoring between 7 and 12 are defined as indeterminate and the treatment is controversial. This study aimed to determine which variables more frequently are considered by spine surgeons for choosing between the conservative and the surgical treatment of VMs among patients with an indeterminate SINS., Methods: A single-round online survey was conducted with 10 spine surgeons with expertise in the management of VMs from our AO Spine Region. In this survey, each surgeon independently reviewed demographic and cancer-related variables of 36 real-life cases of patients with vertebral metastases scored between 7 and 12 in the SINS. Bivariate and multivariate analyses were performed to identify significant SINS and non-SINS variables influencing the decision-making on surgical treatment., Results: The most commonly variables considered important were the SINS element "mechanical pain", rated important for 44.4% of the cases, "lesion type" for 36.1%, and "degree of vertebral collapse" and the non-SINS factor "tumor histology" rated for 13.9% of cases. By far the factor most commonly rated unimportant was "posterior element compromise" (in 72.2% of cases)., Conclusions: Surgeons relied on mechanical pain and type of metastatic lesion for treatment choices. Vertebral collapse, spinal malalignment, and mobility were less influential. Spinal mobility was a predictor of surgical versus non-surgical treatment. The only variables not identified either by surgeons themselves or as a predictor of surgery selection was the presence/degree of posterolateral/posterior element involvement., (Copyright © 2024 Elsevier Inc. Wooden breast (WB) myopathy was investigated in broilers fed varying energy and protein at early ages. Correlation analyses were conducted between echogenicity of ultrasound images (US) of breast muscle from live birds and WB after slaughter. A total of 1,000 Cobb 500 one-day-old male chicks were fed on five dietary programs with eight replicates of 25 birds each, in a completely randomized design. Control feeds (commercially used ME and ideally balanced amino acids) or low-density feeds (low EP, with reductions of 50 kcal/kg ME and 0.20% dig. Lys compared to the control) were formulated. Feeds were provided in different periods: 1 to 7 d, 8 to 14 d, 15 to 21 d or 22 to 28 d. All broilers were fed a common basal diet thereafter until 49 d. Images using US were obtained once a week from all individuals and WB scored from one slaughtered bird per replication (0, normal; 1, mild hardening in the upper breast muscle; 2; moderate hardening in the upper and/or lower breast muscle; 3, severe hardening; 4, severe hardening with hemorrhagic lesions and yellow fluid). Blood was collected for enzyme investigation from the weekly slaughtered bird. Broilers had lower BWG and higher FCR when fed low EP feeds, regardless of the period fed when compared to the control (P < 0.001). Growth compensation, however, occurred afterwards such that all birds presented similar performance at the end. At 14, 21, and 28 d, broilers previously fed low EP feeds had lower WB scores (P < 0.001) compared to birds fed the control; however, both groups presented increased WB scores after 28 d. Wooden breast was positively correlated with breast echogenicity at 21 d (r = 0.31), 28 d (r = 0.43), 35 d (r = 0.21) and 42 d (r = 0.39). Introduction: In Portugal, data on the role of Der p 1 and Der p 2 in patients with house dust mite (HDM) allergy are scarce. Allergen-specific immunotherapy (sIT) is the only treatment that improves symptoms, reduces the need for pharmacological therapy and modifies the natural history of the disease. With this study, the authors aim to understand the local epidemiology and to clarify if the molecular assay of major allergens is advantageous in deciding and/or modifying the decision to initiate sIT in children with clinical indication which are sensitized to Dermatophagoides pteronyssinus., Methods: Retrospective study with analysis of patients with asthma and/or rhinitis., Study Period: January/2013-December/2016., Inclusion Criteria: 1) positive prick-test to Dermatophagoides pteronyssinus; and 2) clinically relevant disease under treatment. Assay Der p 1 and Der p 2 values ≥0.35 kUA/L were considered positive. Statistical significance was set at P<.05., Results: The clinical files of 279 patients. Mean ages 9.55 years (min.4-max.17). Asthma was present in 199 children (71.3%) and rhinitis in 245 (87.8%). Der p 1 and Der p 2 was <0.35kUA/L in 29 (10,4%) patients. The value of Der p 1/Der p 2 correlated with the size of the prick-test papule, the value of the eosinophils and the total IgE., Conclusions: Der p 1 and Der p 2 are dominant allergens in our population and there may be benefits in determining these molecular allergen levels in patients with a positive prick-test and a clinical indication for sIT prior to a decision of initiating sIT or not., (Copyright © 2017 Asociación Española de Pediatría. As the first limiting amino acid in corn-soy broiler diets, methionine (Met) is supplemented using commercial synthetic sources as demanded to obtain economic feed formulations. The Met analogue DL-2-hydroxy-4-(methylthio)-butanoic acid (HMTBA) is largely utilized with that objective. This study intended to obtain responses of broilers fed with increasing levels of HMTBA, from 28 to 42 d, such that economic returns can be calculated. A total of 2,106 Cobb × Cobb 500 one-day-old male broilers was randomly placed in 81 floor pens (2.7 m2 each). Birds were fed conventional starter (zero to 14 d) and grower (14 to 28 d) diets. Starting at 28 d of age, pens of 26 birds were randomly allocated into 9 feed treatments with 9 replications having increasing supplementations with HMTBA (0.00, 0.07, 0.14, 0.21, 0.28, 0.35, 0.42, 0.49 and 0.56%). These were prepared by mixing different proportions of corn-soy dilution and summit diets, which had the same formulated concentration of nutrients and energy [19.7% CP, 0.90% Ca, 0.45% Av. P, 0.95% digestible Lys, and 3,150 kcal/kg AMEn], with the exception of HMTBA [0.56% in the summit but not supplemented in the corn-soy dilution diet (0.52% digestible TSAA)]]. Growth performance was evaluated until 42 d when carcass yield and commercial cuts were evaluated using 6 birds randomly taken from each pen. Body weight gain (BWG), feed conversion ratio (FCR), proportion of breast fillets, and abdominal fat were adjusted using linear broken-line, exponential asymptotic and quadratic polynomial regression models (P < 0.05). Estimations of maximum responses for supplemented HMTBA by the linear broken-line model were 0.17% for BWG, 0.14% for FCR, and 0.29% for breast fillets. Using exponential and quadratic regressions, optimized HMTBA supplementations were obtained at 0.34 and 0.35% for BWG, 0.20 and 0.33% for FCR, and 0.31 and 0.36% for breast fillets, respectively. Supplemental levels of HMTBA that optimize growth performance and breast meat in male broilers from 28 to 42 d, using different regression models, varied from 0.14 to 0.36%.

Lipid homeostasis in liver is known to be altered with diabetes mellitus, ultimately leading to liver damage and related complications. The present work aimed to evaluate changes in the liver phospholipid profile after 4 months of uncontrolled hyperglycemia. Twenty Wistar rats were divided into two groups: control and streptozotocin-treated (T1DM). After 4 months, animals were sacrificed and morphological characterization of liver was performed and related with serum markers of hepatic damage. Lipid extracts were obtained from liver and phospholipid (PL) classes were quantified. Lipid molecular species were determined by LC-MS and LC-MS/MS, and fatty acids by GC-MS. Concomitantly with signs of hepatic damage we found variations in the relative amount of phospholipid classes in T1DM, characterized by a decrease in PLs with choline head group, and by an increase in the relative content of other PL classes. A remodeling in PL fatty acyl chains was observed in T1DM liver, with a similar pattern to all the PL classes, and consisting in the reduction of 16:0 and an increase of 18:0 and 18:2 acyl chains. The observed changes in T1DM lipid profile may contribute to the altered membrane properties underlying hepatic damage, worsening the metabolic alterations that characterize T1DM., (Copyright © 2013 Elsevier Inc. Agaricus brasiliensis cell-wall polysaccharides isolated from fruiting body (FR) and mycelium (MI) and their respective sulfated derivatives (FR-S and MI-S) were chemically characterized using elemental analysis, TLC, FT-IR, NMR, HPLC, and thermal analysis. Cytotoxic activity was evaluated against A549 tumor cells by MTT and sulforhodamine assays. The average molecular weight (Mw) of FR and MI was estimated to be 609 and 310 kDa, respectively. FR-S (127 kDa) and MI-S (86 kDa) had lower Mw, probably due to hydrolysis occurring during the sulfation reaction. FR-S and MI-S presented ~14% sulfur content in elemental analysis. Sulfation of samples was characterized by the appearance of two new absorption bands at 1253 and 810 cm(-1) in the infrared spectra, related to S=O and C-S-O sulfate groups, respectively. Through (1)H and (13)C NMR analysis FR-S was characterized as a (1→6)-(1→3)-β-D-glucan fully sulfated at C-4 and C-6 terminal and partially sulfated at C-6 of (1→3)-β-D-glucan moiety. MI-S was shown to be a (1→3)-β-D-gluco-(1→2)-β-D-mannan, partially sulfated at C-2, C-3, C-4, and C-6, and fully sulfated at C-6 of the terminal residues. The combination of high degree of sulfation and low molecular weight was correlated with the increased cytotoxic activity (48 h of treatment) of both FR-S (EC₅₀=605.6 μg/mL) and MI-S (EC₅₀=342.1 μg/mL) compared to the non-sulfated polysaccharides FR and MI (EC₅₀>1500 μg/mL)., (Copyright © 2013 Elsevier B.V. Oxidized phospholipids are known to be key signaling molecules in the onset of several diseases involving inflammation. The aim of this study was to evaluate the effect of oxidized phosphatidylserines (oxPS) in modulating the immune system, through cytokine production. Flow cytometry analysis was used to evaluate the oxPS capacity to induce the expression of different cytokines by monocytes, myeloid dendritic cells (mDCs) and DCs CD14(-/low)CD16(+). oxPS were formed during oxidation induced by the hydroxyl radical. Among the four families of oxPS studied, only oxPS modified in the polar head with formation of a terminal hydroperoxyacetaldehyde upregulated the production of cytokines IL-8 and TNF-α by monocytes and DCs subsets (mDCs and CD14(-/low)CD16(+) DCs). This family of oxPS showed the capacity to upregulate the production of IL-1β, IL-6, and MIP-1β from the same type of cells. A significant raise in the percentage of monocytes and dendritic cells producing the studied cytokines was observed, when compared with basal control. Oxidation products modified in the fatty acyl chain did not upregulate TNF and IL-8. oxPS with terminal hydroperoxyacetaldehyde has pro-inflammatory properties. This outcome may help to understand the biological role of phosphatidylserine oxidation products in inflammatory processes and in dysfunctions of immune system., (Copyright © 2012 Elsevier Inc. The aim of this study was to assess the impact of sewage discharge on coastal waters by evaluating the influence of physicochemical parameters on the presence of enteric microorganisms in seawater samples collected from 11 beaches in Florianopolis, Santa Catarina, Brazil, over a one-year period (August 2009 to July 2010). Samples were assessed for the presence of human adenoviruses (HAdV), polyomavirus (JCPyV), hepatitis A virus (HAV), and noroviruses (HuNoV GI and GII). Escherichia coli and physicochemical parameters (salinity, temperature, pH and dissolved oxygen) were also evaluated. From the 132 samples analyzed, 55% were positive for HAdV, 51.5% for HAV, 7.5% for HuNoV GI, 4.5% for HuNoV GII, and 3% for JCPyV. E. coli levels ranged from 8 to 1325 CFU/100mL at all sites. The overall results highlight the problem of sewage discharge into coastal waters and confirm that there is no correlation between viral presence and bacterial contamination., (Copyright © 2011 Elsevier Ltd. Methanolic extracts obtained from different organs of Cleome rosea, collected from its natural habitat and from in vitro-propagated plants, were submitted to in vitro biological assays. Inhibition of nitric oxide (NO) production by J774 macrophages and antioxidant effects by protecting the plasmid DNA from the SnCl(2)-induced damage were evaluated. Extracts from the stem of both origins and leaf of natural plants inhibited NO production. The plasmid DNA strand breaks induced by SnCl(2) were reduced by extracts from either leaf or stem of both sources. On the other hand, root extracts did not show any kind of effects on plasmid DNA, and presented significant toxic effects to J774 cells. The results showed that C. rosea presents medicinal potential and that the acclimatization process reduces the plant toxicity both to plasmid DNA and to J774 cells, suggesting the use of biotechnology tools to obtain elite plants as source of botanical material for pharmacological and phytochemical studies.

The antiviral activity of six medicinal plants from Brazilian Atlantic Tropical Forest was investigated against two viruses: herpes simplex virus type 1 (HSV-1) and poliovirus type 2 (PV-2). Cuphea carthagenensis and Tillandsia usneoides extracts showed the best antiherpes activity. T. usneoides dichloromethane, ethyl acetate and n-butanol extracts, and Lippia alba n-butanol extract showed inhibition of HSV-1, strain 29R/acyclovir resistant. In addition, only L. alba ethyl acetate extract showed antipoliovirus activity. These results corroborate that medicinal plants can be a rich source of potential antiviral compounds.

Shellfish are readily contaminated with viruses present in water containing sewage because of the concentration effect of filter feeding. Hepatitis A virus (HAV) is the main cause of acute hepatitis worldwide and may lead to severe illness or even death. It is transmitted through fecal and oral routes and causes widespread endemic and asymptomatic infections in young children. Here we describe a method for the detection of HAV RNA in shellfish involving the extraction of total RNA from oyster meat followed by reverse transcription-polymerase chain reaction (RT-PCR). Virus recovery from oyster extracts artificially seeded with HAV strain HM 175 was examined by RT-PCR. The minimum detection limit was 3.3 focus-forming units of HAV, and the recovery rate was 75.7%. This method was used to assess the viral contamination of four shellfish beds in Santa Catarina State, Brazil, over a 1-year period. Six (22%) of 27 samples collected in autumn and winter from one shellfish bed tested positive for HAV.
