Even though researches have shown that biochar can improve soil-health and plant-growth even in harsh environments and get rid of harmful heavy metals and new contaminants, it is still not sustainable, affordable, or effective enough. Therefore, scientists are required to develop nanomaterials in order to preserve numerous aquatic and terrestrial species. The carbonaceous chemical known as nano-biochar (N-BC) can be used to get rid of metal contamination and emerging contaminants. However, techniques to reduce hetero-aggregation and agglomeration of nano-biochar are needed that lead to the emergence of emerging nano-biochar (EN-BC) in order to maximise its capacity for adsorption of nano-biochar. To address concerns in regards to the expanding human population and sustain a healthy community, it is imperative to address the problems associated with toxic heavy metals, emerging contaminants, and other abiotic stressors that are threatening agricultural development. Nano-biochar can provide an effective solution for removal of emerging contaminants, toxic heavy metals, and non-degradable substance. This review provides the detailed functional mechanistic and kinetics of nano-biochar, its effectiveness in promoting plant growth, and soil health under abiotic stress. The whey protein β-lactoglobulin (βLG) forms fibrils similar to the amyloid fibrils in the neurodegenerative diseases due to its higher predisposition of β-sheets. This study shed light on the understanding different inorganic Keggin polyoxometalates (POMs) interaction with the protein βLG fibrils. POMs such as Phosphomolybdic acid (PMA), silicomolybdic acid (SMA), tungstosilicic acid (TSA), and phosphotungstic acid (PTA) were used due to their inherent higher anionic charges. The interaction studies were monitored with fluorescence spectra and Thioflavin T assay for both the βLG monomers and the fibrils initially to elucidate the binding ability of the POMs. The binding of POMs and βLG is also demonstrated by molecular docking studies. Zeta potential studies showed the electrostatic mediated higher interactions of the POMs with the protein fibrils. Isothermal titration calorimetry (ITC) studies showed that the molybdenum containing POMs have higher affinity to the protein fibrils than the tungsten. Climate change, food insecurity, and agricultural pollution are all serious challenges in the twenty-first century, impacting plant growth, soil quality, and food security. Innovative techniques are required to mitigate these negative outcomes. Toxic heavy metals (THMs), organic pollutants (OPs), and emerging contaminants (ECs), as well as other biotic and abiotic stressors, can all affect nutrient availability, plant metabolic pathways, agricultural productivity, and soil-fertility. Comprehending the interactions between root exudates, microorganisms, and modified biochar can aid in the fight against environmental problems such as the accumulation of pollutants and the stressful effects of climate change. Microbes can inhibit THMs uptake, degrade organic pollutants, releases biomolecules that regulate crop development under drought, salinity, pathogenic attack and other stresses. However, these microbial abilities are primarily demonstrated in research facilities rather than in contaminated or stressed habitats. Despite not being a perfect solution, biochar can remove THMs, OPs, and ECs from contaminated areas and reduce the impact of climate change on plants. We hypothesized that combining microorganisms with biochar to address the problems of contaminated soil and climate change stress would be effective in the field. Despite the fact that root exudates have the potential to attract selected microorganisms and biochar, there has been little attention paid to these areas, considering that this work addresses a critical knowledge gap of rhizospheric engineering mediated root exudates to foster microbial and biochar adaptation. Climate change imposes various environmental stresses which substantially impact plant growth and productivity. Salinity, drought, temperature extremes, heavy metals, and nutritional imbalances are among several abiotic stresses contributing to high yield losses of crops in various parts of the world, resulting in food insecurity. Many interesting strategies are being researched in the attempt to improve plants' environmental stress tolerance. These include the application of nanoparticles, which have been found to improve plant function under stress situations. Nanotechnology will be a key driver in the upcoming agri-tech and pharmaceutical revolution, which promises a more sustainable, efficient, and resilient agricultural and medical system Nano-fertilizers can help plants utilise nutrients more efficiently by releasing nutrients slowly and sustainably. Plant physiology and nanomaterial features (such as size, shape, and charge) are important aspects influencing the impact on plant growth. Here, we discussed the most promising new opportunities and methodologies for using nanotechnology to increase the efficiency of critical inputs for crop agriculture, as well as to better manage biotic and abiotic stress. Potential development and implementation challenges are highlighted, emphasising the importance of designing suggested nanotechnologies using a systems approach. Finally, the strengths, flaws, possibilities, and risks of nanotechnology are assessed and analysed in order to present a comprehensive and clear picture of the nanotechnology potentials, as well as future paths for nano-based agri-food applications towards sustainability. Recent efforts have focused on developing improved drug delivery systems with enhanced therapeutic efficacy and minimal side effects. Micelles, self-assembled from amphiphilic block copolymers in aqueous solutions, have gained considerable attention for drug delivery. However, there is a need to further enhance their efficiency. These micelles offer benefits like biodegradability, biocompatibility, sustained drug release, and improved patient compliance. Yet, researchers must address stability issues and reduce toxicity. Nanoscale self-assembled structures have shown promise as efficient drug carriers, offering an alternative to conventional methods. Fine-tuning at the monomeric and molecular levels, along with structural modifications, is crucial for optimal drug release profiles. Various strategies, such as entrapping hydrophobic drugs and using polyethylene oxide diblock copolymer micelles to resist protein adsorption and cellular adhesion, protect the hydrophobic core from degradation. The polyethylene oxide corona also provides stealth properties, prolonging blood circulation for extended drug administration. Amphiphilic copolymers are attractive for drug delivery due to their adjustable properties, allowing control over micelle size and morphology. Emerging tools promise complex and multifunctional platforms. In the present farming era, rhizobacteria as beneficial biofertilizers can decrease the negative effects of Zinc (Zn) agrochemicals. However, their commercial viability and utility are constrained by their instability under field conditions. Thus, to enhance their stability, microbial formulations are considered, which will not only offer an appropriate microenvironment, and protection but also ensure a high rate of rhizospheric-colonization. The goal of this study was to create a new formulation for the Zn-solubilizing bacteria E. ludwigii-PS10. The studied formulation was prepared using the extrusion technique, wherein a composite solution containing alginate, starch, zinc oxide, and poultry waste was uniformly mixed with the bacterial strain PS10 to develop low-cost, eco-friendly, and slow-release microbeads. The produced microbead was spherical, and characterized by SEM, FTIR, and XRD. Further, the microbeads were analyzed for their survival stability over 3 months of storage at room temperature and 4 °C. The effect of the microbead on the vegetative growth of tomato plants was investigated. Results showed that 94 % of the encapsulated microbial beads (EMB) matrix was able to encapsulate the bacterial strain PS10. The dried EMB demonstrated a moisture content of 2.87 % and was able to preserve E. ludwigii-PS10 survival at room temperature at the rate of 85.6 %. The application of the microbead to the tomato plants significantly increased plant biomass and Zn content. Soil salinity is progressively affecting global agriculture area, and act as a brutal environmental factor for the productivity of plants, therefore, sustainable remediation of the saline soil is urgently required. In this study, we tested the effectiveness of PM (poultry manure), SMS (spent mushroom substrate), and CD (cow dung) for the recovery of salt soil and the optimization of the productivity of the maize plant. PM and SMS showed the valuable source of OC, N, P, K as the CD. The HCA analysis showed that 47% of the bacterial population from PM, SMS, and CD survived at 6% NaCl (w/v), which had PGP attributes such as IAA, P-solubilizers, and siderophore activity. The results from pot experiments of plant growth and PCA analysis of bacterial PGP attributes reveled re formulation of PM, SMS, and CD, which were further optimized at the saline field level. T-2 treated plant increased their shoot length, chlorophyll content, reducing sugar, nitrogen, phosphorus, and potassium levels significantly after 30 and 60 days, followed by T-4 and T-3 as the control. A significant (P < 0.01) increase in particle density and decrease in bulk density was observed for all combinations treated (T-2 to T-7). A two-year field study revealed that the T-2 combination increased 43% OC, 57% N, 66% P, 48% K, 32% DHA, 76% PPO in the soil than the control after 60 days. T2-combination decreased ≈50% of Na content in root and shoot, and increased 27% of maize crop yield. Fly ash (FA) management is a key concern of ecologists around the world, so its potential as a nutritional supplement for agro-ecosystems needs to be explored. Therefore, alternate techniques that are eco-friendly to manage this emerging dual-edged waste are preferable in this field. The current study sought to determine the soil-modifying, crop yield improvement, and nematicidal properties of FA. In this study, beetroot seeds were sown in pots comprising field soil amended with differing proportions of FA (w/w) revealed the bio-fold properties of FA. Biomineralization and mapping of elements revealed that increased nutritional elements in soil supplemented with 15% FA induced growth-performance and yield of beetroot. Molecularly and morphologically characterized Meloidogyne incognita was used as nematode in this study for optimization of nematicidal properties FA. Plant growth performance, photosynthetic pigments, and yield of beetroot were significantly reduced owing to M. incognita as compared to control (un-treated and un-inoculated), and 15% FA reversed the negative effect of M. incognita significantly (P < 0.05 ) as compared to control plants. Confocal laser microscopy confirmed that 15% FA augmented in soil reduced nematode-juvenile invasion in beetroot as compared with control. The PCA (principal component analysis) accounted for 98.63% and 98.8% for the total-data variability in plants without nematodes and total data variability in treated plants (M. incognita + FA) respectively, which showed fit for a significant correlation between the various studied parameters in present study., (Copyright © 2022 Elsevier Inc. Catalases are crucial antioxidant enzymes that reduce the excessive level of H 2 O 2 caused by various environmental stresses and metal toxicity and hence protect the plant cells. In this study, a total of ten TaCAT genes, forming three homeologous groups, were identified in the genome of bread wheat (Triticum aestivum L.) and named as per the wheat gene symbolization guidelines. The identified catalases were characterized for various structural and physicochemical features. The proximal active-site (F(D/A)RERIPERVVHAKGASA) and heme-ligand (R(I/V)F(S/A)Y(A/S)DTQ) signature motifs, catalytic residues and peroxisomal targeting peptides were found conserved. Phylogenetic analysis clustered TaCATs into three classes, which showed conserved functional specialization based on their tissue specific expression. Modulated spatio-temporal expression of various TaCAT genes and alteration in total catalase enzyme activity during heat, drought, salt and arsenic (As III and As V ) treatment suggested their roles in abiotic stress response and arsenic tolerance. Molecular cloning and overexpression of TaCAT3-B gene in Escherichia coli showed tolerance against heat, drought, salt and varied concentrations of As III and As V treatments. All mRNAs cannot be translated into full-length proteins due to ribosome-stalling that leads to release of peptidyl-tRNA which can be lethal for bacterial survival. The enzyme peptidyl-tRNA hydrolase (PtH) hydrolyses the ester bond between nascent peptide and tRNA of peptidyl-tRNA and rescues the cells from toxicity. PtH is an essential enzyme in bacteria and inhibiting this crucial enzyme can serve to combat bacterial diseases. But due to lack of understanding about the catalytic mechanism of PtH, its inhibitors have not been developed. In this work, we have carried out the binding studies of M. tuberculosis and E. coli PtH with the peptidyl-tRNA analogue (puromycin) using ITC, FTIR, CD experiments followed by docking and MD simulations to identify the potential active site residues that would help to design PtH inhibitors. Binding studies of puromycin with both PtH by ITC experiments demonstrate similar thermodynamic parameters and three fold difference in their K D . CD and FTIR studies detected changes in secondary structure composition of PtH in the presence of puromycin with different degree of perturbation. Somatic embryogenesis receptor kinases (SERKs) play a significant role in morphogenesis, stress/defense and signal transduction. In the present study, we have identified two SERK and 11 SERK-like (SERL) genes in Phalaenopsis equestris, two SERK and 11 SERL genes in Dendrobium catenatum, and one SERK and eight SERL genes in Apostasia shenzhenica genome. Characterization of the SERK proteins revealed the presence of a signal peptide, a leucine zipper, five leucine-rich repeats (LRRs), a serine proline proline (SPP) motif, a transmembrane region, a kinase domain, and a C-terminus. Most of the SERK/SERL proteins were characterized with similar physicochemical properties. The presence of transmembrane region predicted their membranous localization. Tertiary structure prediction of all the five identified SERK proteins had sequence identity with BAK1 protein of Arabidopsis thaliana. Generally, all the SERK/SERL genes shared similar gene architecture and intron phasing. Gene ontology analysis indicated the role of SERKs in receptor and ATP binding, signal transduction, and protein phosphorylation. Phylogenetic analysis revealed the clustering of SERKs and SERLs in distinct clades. Expression of SERKs in reproductive tissues like floral bud, floral stalk, whole flower and pollen was reported to be higher than their expression in vegetative tissues with an exception of PeSERK1 and DcSERK1 which showed higher expression in leaves and roots, respectively. Likewise, a higher expression of AsSERK1 was observed in tubers. However, lower expression of SERLs was observed in majority of tissues studied irrespective of their vegetative or reproductive origin. Cysteine-rich receptor-like kinases (CRK) constitute one of the largest subfamily of receptor-like kinases, which play crucial roles in plant development and stress response. In total, 43, 37, 36, 38 and 170 CRK genes including duplicated genes were identified in the genome of Brachypodium distachyon, Hordeum vulgare, Oryza sativa, Sorghum bicolor and Triticum aestivum, respectively. These CRK proteins were tightly clustered into four phylogenetic groups and exhibited close syntenic relationship among orthologous genes. Majority of CRK proteins contain a transmembrane domain for plasma membrane localization. The organization of exon/intron, domains and motifs were variably conserved. Tissue-specific expression suggested the involvement of certain CRK genes in plant development. Modulated expression revealed their specific stress-responsive functions. Co-expression and interaction analysis indicated their role in signaling. Ks value and divergence time analysis suggested duplication of TaCRK genes before the hybridization of T. aestivum sub-genomes. Expression comparison of duplicated TaCRK genes revealed functional retention, neofunctionalization or pseudo-functionalization. Recombinant expression of a stress-responsive gene TaCRK68-A in Escherichia coli and Saccharomyces cerevisiae displayed enhanced tolerance against heat, drought, cold and salinity stresses. Objective: Pharmacologic approaches to study brown adipocyte activation in vivo with a potential of being translational to humans are desired. The aim of this study was to examine pre- and postsynaptic targeting of adrenergic system for enhancing brown adipose tissue (BAT) metabolism quantifiable by [(18)F]fluoro-2-deoxyglucose ([(18)F]FDG) positron emission tomography (PET)/computed tomography (CT) in mice., Methods: A β₃-adrenoreceptor selective agonist (CL 316243), an adenylyl cyclase enzyme activator (forskolin) and a potent blocker of presynaptic norepinephrine transporter (atomoxetine), were injected through the tail vein of Swiss Webster mice 30 minutes before intravenous (iv) administration of [(18)F]FDG. The mice were placed on the PET/CT bed for 30 min PET acquisition followed by 10 min CT acquisition for attenuation correction and anatomical delineation of PET images., Results: Activated interscapular (IBAT), cervical, periaortic and intercostal BAT were observed in 3-dimentional analysis of [(18)F]FDG PET images. CL 316243 increased the total [(18)F]FDG standard uptake value (SUV) of IBAT 5-fold greater compared to that in placebo-treated mice. It also increased the [(18)F]FDG SUV of white adipose tissue (2.4-fold), and muscle (2.7-fold), as compared to the control. There was no significant difference in heart, brain, spleen and liver uptakes between groups. Forskolin increased [(18)F]FDG SUV of IBAT 1.9-fold greater than that in placebo-treated mice. It also increased the [(18)F]FDG SUV of white adipose tissue (2.2-fold) and heart (5.4-fold) compared to control. There was no significant difference in muscle, brain, spleen, and liver uptakes between groups. Atomoxetine increased [(18)F]FDG SUV of IBAT 1.7-fold greater than that in placebo-treated mice. There were no significant differences in all other organs compared to placebo-treated mice except liver (1.6 fold increase). A positive correlation between SUV levels of IBAT and CT Hounsfield unit (HU) (R(2)=0.55, p<0.001) and between CT HU levels of IBAT and liver (R(2)=0.69, p<0.006) was observed., Conclusions: The three pharmacologic approaches reported here enhanced BAT metabolism by targeting different sites in adrenergic system as measured by [(18)F]FDG PET/CT., (© 2013 Elsevier Inc. A new benzimidazole-based diamide ligand-N,N'-bis(alanine-2-benzimidazolyl) hexanediamide (ABHA) has been synthesized and utilized to prepare Cu(II) complexes of general composition [Cu(ABHA)X2].nH2O,where X is an exogenous anionic ligand (X=Cl-,NO3-). Low temperature EPR spectra has been obtained that shows gparallel>gperpendicular>2.0024, indicating a tetragonal geometry in the solution state.
