Abstract

The study of tropical diseases like leishmaniasis, a parasitic disease, has not received much attention even though it is the second-largest infectious disease after malaria. As per the WHO report, a total of 0.7–1.0 million new leishmaniasis cases, which are spread by 23 Leishmania species in more than 98 countries, are estimated with an alarming 26,000–65,000 death toll every year. Lack of potential vaccines along with the cost and toxicity of amphotericin B (AmB), the most common drug for the treatment of leishmaniasis, has raised the interest significantly for new formulations and drug delivery systems including nanoparticle-based delivery as anti-leishmanial agents. The size, shape, and high surface area to volume ratio of different NPs make them ideal for many biological applications. The delivery of drugs through liposome, polymeric, and solid-lipid NPs provides the advantage of high biocomatibilty of the carrier with reduced toxicity. Importantly, NP-based delivery has shown improved efficacy due to targeted delivery of the payload and synergistic action of NP and payload on the target. This review analyses the advantage of NP-based delivery over standard chemotherapy and natural product-based delivery system. The role of different physicochemical properties of a nanoscale delivery system is discussed. Further, different ways of nanoformulation delivery ranging from liposome, niosomes, polymeric, metallic, solid-lipid NPs were updated along with the possible mechanisms of action against the parasite. The status of current nano-vaccines and the future potential of NP-based vaccine are elaborated here.

Abstract

Recent research in graphene nanomaterials has witnessed significant contribution in biomedical science. With functional dopants, the inherent properties of graphene are extensively tuned for various applications. With a two-dimensional planar structure, large surface area with functional dopant creates a hybrid graphene as a new amalgam; with a high-performance material for bio-electronics and biomedical. Here, we present an overview of recent advances in the field of hybrid graphene derivatives, focused on drug delivery, cyto-compatabilitys and electrochemical biosensors. The physicochemical properties of graphene and surface functionalization strategies employed for loading of active pharmaceuticals toward drug delivery are discussed. Studies related to the biocompatibility of functionalized graphene for cell proliferation-associated applications are also reviewed. Furthermore, electrochemical biosensors based on hybrid graphene for the detection of metabolic biomarkers, infectious agents and active pharmaceuticals/nutraceuticals are highlighted. In summary, the integration feasibility of hybridized graphene nanomaterials in the subdisciplines of nanotechnology, viz, microelectromechanical system, microfluidics, and biomedical engineering toward its translational scope are provided.

Abstract

Inflammatory conditions associated with kidney diseases are a global public health issue that requires efficient and safe therapies. Current treatment of renal inflammatory conditions using modern medicines has limitations such as undesired adverse effects, especially in long-term use. Combinations of herbs derived from historical medicinal practice such as Kampo (Japanese herbal formulation) and traditional Chinese medicines were found to synergistically elevate the therapeutic efficacy, and lowering the dose of specific herbal extracts could reduce the side effects if there were any. The clinical practice of traditional Kampo medicine is often followed in Japan for a range of health issues, including acute and chronic renal diseases. Owing to its well-known clinical tradition there has been a significant effort since 1995 devoted to evaluating the individual mechanistic properties of common Kampo formulas. In this chapter, the characteristics of Kampo medicine, especially in renal inflammatory diseases, are briefly reviewed. The pharmacological activity of important Kampo formulations, supported by either in vitro or animal studies, is presented. Reports on randomized clinical trials of traditional herbal medicines for kidney disease and required future directions are also discussed.

Abstract

Visceral leishmaniasis (VL) is the most lethal among all leishmaniasis diseases and remains categorized as a neglected tropical disease (NTD). This study aimed to develop a peptide-based multi-epitope vaccine construct against VL using immunoinformatics methodologies. To achieve this, four distinct proteins were screened to identify peptides consisting of 9-15 amino acids with high binding affinity to toll-like receptors (TLRs), strong antigenicity, low allergenicity, and minimal toxicity. The resulting multi-epitope vaccine construct was fused in a tandem arrangement with appropriate linker peptides and exhibited superior properties related to cytotoxic T lymphocytes (CTLs), helper T lymphocytes (HTLs), and B-cell epitopes. Subsequently, a three-dimensional (3D) model of the vaccine construct was generated, refined, and validated for structural stability and immune response capabilities. Molecular docking and simulations confirmed the vaccine construct's stability and binding affinities with TLRs, with TLR4 displaying the highest binding affinity, followed by TLR2 and TLR3. Additionally, simulations predicted robust cellular and humoral antibody-mediated immune responses elicited by the designed vaccine construct. Notably, this vaccine construct includes proteins from various pathways of Leishmania donovani (LD), which have not been previously utilized in VL vaccine design. Abstract

Diabetic nephropathy (DN), a persistent microvascular problem of diabetes mellitus is described as an elevated level of albumin excretion in urine and impaired renal activity. The morbidity and mortality of type-1 diabetics and type-2 diabetics due to end stage renal disease is also a result of the increased prevalence of DN. DN typically occurs as a consequence of an association among metabolic and hemodynamic variables, activating specific pathways leading to renal injury. According to current interventions, intensive glucose regulation decreases the threat of DN incidence and growth, and also suppressing the renin-angiotensin system (RAS) is a significant goal for hemodynamic and metabolism-related deformities in DN. However, the pathogenesis of DN is multifactorial so novel approaches other than glucose and blood pressure control are required for treatment. Abstract

Obesity induced insulin resistance is primarily regulated by the inhibitory phosphorylation of peroxisome proliferator-activated receptor γ at serine 273 (PPARγS273) which has been shown to be regulated by MEK and ERK. An upstream regulatory molecule of this pathway could be a therapeutic option. Here we analyzed the involvement of Fetuin-A (FetA), a key hepato-adipokine implicated in insulin resistance, as an upstream regulator molecule for the regulation of PPARγ inhibitory phosphorylation. Mice fed with standard diet (SD), high fat diet (HFD) and HFD with FetA knockdown (HFD-FetA KD ) were used to examine the role of FetA on PPARγS273 phosphorylation in adipocytes. The mechanism of regulation and its effect on skeletal muscle were studied using primary adipocytes, 3T3-L1 (preadipocyte) and C2C12 (myotube) cell lines. Increased FetA in HFD mice strongly correlated with augmentation of PPARγS273 phosphorylation in inflamed adipocytes while knockdown of FetA suppressed it. This effect of FetA was mediated through the activation of Ras which in turn activated MEK and ERK. On addressing how FetA could stimulate activation of Ras, we found that FetA triggered TNFα in inflamed adipocytes which induced Ras activation. The ensuing sharp fall in adiponectin level attenuated AMPK activation in skeletal muscle cells affecting mitochondrial ATP production. Our data reveal the essential role of FetA induced activation of Ras in regulating PPARγ inhibitory phosphorylation through Ras-MEK-ERK pathway which downregulates adiponectin disrupting skeletal muscle mitochondrial bioenergetics. Thus, FetA mediated PPARγ inactivation has adverse consequences upon adipocyte-myocyte crosstalk leading to disruption of energy homeostasis and loss of insulin sensitivity., (Copyright © 2020 Elsevier B.V. Abstract

Basidiomycetes-X (BDM-X) is a novel edible mushroom recently identified as a new fungi species and is effective against oxidative stress and anti-inflammation associated with immune response. However the effect of BDM-X on atopic dermatitis (AD) has not been elucidated. In this study, we have investigated the effect of BDM-X on AD skin lesions in NC/Nga mouse model. AD-like lesion was induced by the application of house dust mite extract (DfE) to the dorsal skin of NC/Nga mouse. After AD induction, BDM-X was administered once daily for 2 weeks. We have analyzed the effects of BDM-X on dermatitis severity, histopathological changes and changes in inflammatory and proinflammatory proteins expressions in DfE induced AD mice skin. Treatment with BDM-X attenuated the development of AD-like clinical symptoms and effectively inhibited hyperkeratosis, parakeratosis, acanthosis and mast cells in AD mice skin. Furthermore, BDM-X treatment inhibited DfE induced tumor necrosis factor (TNF)α, high mobility group protein (HMG)B1, nuclear factor kappa (NFκ)B and inflammatory cytokines. These results indicate that BDM-X inhibits AD through modulating Th1 and Th2 responses and diminishing the mast cells infiltration in the skin lesions in NC/Nga mice., (Copyright © 2018 Elsevier Inc.
