Our skin serves as the primary barrier against external environmental insults, the latter of which can cause oxidative stress within cells, while various bioactive peptides sourced from natural resources hold promise in protecting cells against such oxidative stress. In this study, we investigate the efficacy of a low molecular weight extract from the sea cucumber Apostichopus japonicus, denoted as Sample-P, in facilitating cell migration and wound healing under oxidative stress conditions in skin cells. The naturally derived compound is a highly complex mix of peptides exhibiting antioxidative properties, as highlighted through liquid chromatography-mass spectrometry peptide screening and an in vitro antioxidant assay. Our results demonstrate that Sample-P is capable of promoting cell migration while preventing severe stress responses such as visible through mTOR expression. To further identify the molecular pathways underpinning the overall protective mechanism of Sample-P, we have utilised a proteomics approach. Our data reveal that Sample-P regulates protein expression associated with ribosomal pathways, glycolysis/gluconeogenesis and protein processing in the endoplasmic reticulum (ER), which help in preserving DNA integrity and safeguarding cellular organelles, such as mitochondria and the ER, under oxidative stress conditions in skin cells. In summary, in the presence of H2O2, Sample-P exhibits antioxidative properties at both molecular and cellular levels, rendering it a promising candidate for topical skin treatment to wound healing and to address age-related skin conditions.

Background: Head and Neck Squamous Cell Carcinoma (HNSCC) is a malignancy characterized by a high incidence and recurrence rate. 5-methylcytosine (m5C) RNA modification is a common alteration affecting cancer progression; however, how m5C operates within the tumor microenvironment of HNSCC remains to be elucidated. Methods: We conducted Nanopore sequencing on 3 pairs of cancer and paracancerous tissues from mid- and late-stage HNSCC, obtaining 132 upregulated genes (transcriptomically upregulated, m5C elevated) and 129 downregulated genes (transcriptomically downregulated, m5C reduced). Subsequent Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed; a differential gene interaction network (PPI) was constructed, revealing the interactions of each gene with others in the network. Co-expression analysis was performed on the genes within the PPI, unveiling their expression and regulatory relationships. Through GSVA analysis, variations in related pathways under different states were identified. Furthermore, results of m5C in lncRNA were screened, followed by target gene prediction. Results: Sequencing results from the 3 pairs of mid- and late-stage HNSCC cancer and paracancerous tissues demonstrated that RPS27A, RPL8, and the lncRNAs including differentiation antagonizing nonprotein coding RNA (DANCR), DCST1 antisense RNA 1 (CCDC144NL-AS1), Growth Arrest-Specific Transcript 5 (GAS5), Nuclear Paraspeckle Assembly Transcript 1 (NEAT1), and Small Nucleolar RNA Host Gene 3 (SNHG3), etc., under m5Cregulation, have close connections with surrounding genes. The differentially m5Cmodified genes are primarily involved in ribosomal protein synthesis, oxidative stress response, metabolic reprogramming, immunity, and other life processes; pathways like mitochondrial protein import and photodynamic therapy induced unfolded protein response are upregulated in the tumor, while pathways, including the classic P53, are suppressed. Analysis on m5C-regulated long non-coding RNAs (lncRNAs) revealed tight associations with RPS27A and RPL8 as well. Conclusion: Our study identifies the key factors and signaling pathways involving m5C in HNSCC. The findings suggest that ribosome-related genes might regulate ribosomal protein synthesis, oxidative stress response, metabolic reprogramming, and immune response through m5C RNA modification by means like hypoxia and ferroptosis, thereby playing a pivotal role in the onset and progression of HNSCC. Hence, attention should be paid to the role of ribosomes in HNSCC. These findings may facilitate the precision and individualized treatment of patients with mid- and late-stage HNSCC in clinical settings.

As unsafe food production poses global health threats, especially in developing countries, food safety detection always significantly impacts all fields of the food industry, such as production, processing, transportation, storage, and consumption. Many classical food safety detecting methods have been widely used to provide accurate, sensitive, and reliable analytical characteristics, while they rely on clean laboratories, bulky setups, trained personnel, and long-time detection. Because of those conditions, they are not suitable for on-site food safety detecting applications. To solve this problem, various food safety detecting approaches relying on field-portable devices and field-operable techniques have been designed and successfully used, indicating that they are potentially ideal solutions for on-site applications. This review summarizes the progress on on-site food safety detecting approaches and applications for various risk factors, including bacteria, parasites, viruses, toxins, pesticide and veterinary drug residues, illegal food additives, restricted food ingredients, and heavy metals. It outlines recent advances in field-portable devices and field-operable techniques that provide conditions for on-site food safety detection. Recommended on-site food safety detecting approaches for different food safety risk factors are comprehensively suggested considering detecting feasibility, accuracy, and speed. These approaches reveal an attractive and promising route for future practical food safety detecting applications with the merits of portable devices, simple user-friendly operation, and rapid on-site detection. This review further addresses current challenges, and finally discusses future trends and strategies that could be employed in on-site food safety detection.

Background: Postoperative cognitive dysfunction (POCD) is a common postoperative complication in elderly patients. The strong stress response causing by surgical trauma can induce POCD. We hypothesized that stellate ganglion block (SGB) can provide the neuroprotection to POCD by regulating the neuroendocrine response. Methods: Sprague-Dawley male rats, 18–20 months old and weighing 550–650 g were assigned into four groups: sham surgery group (Sham), sham surgery + saline group (Sham + NS), surgery group (Surgery), and surgery + SGB group (Surgery + SGB). The change of body weight, heart rate variability analysis, behavior testing, neuronal damage, inflammatory response, neuroendocrine hormone level were evaluated by their corresponding methods. Results: The results showed that SGB can reduce the number of both types of errors in the postoperative eight-arm maze assay, attenuate neural structural damage, inhibit neuroapoptosis, suppress inflammatory responses, increase the release of neurotrophic factors, accelerate postoperative weight recovery, and promote postoperative recovery in rats. Most importantly, SGB reduced the level of neuroendocrine hormone of TH, Cyp11b1, CRH, and SGB also activated dorsal motor nucleus of vagus (detected by c-fos immunohistochemistry). Conclusions: Our findings indicated that SGB could be a neuroprotective therapy for the cognitive dysfunction induced by exploratory laparotomy model of POCD, which might be attributable for balancing the autonomic nervous system, regulating hypothalamic-pituitary-adrenal (HPA) axis system.

To investigate the anti-obesity effect of a noni (Morinda citrifolia L.) fruit polysaccharide (NFP) and its potential mechanisms, 24 male C57BL/6J mice were randomly divided into three groups—normal control group, high-fat diet (HFD) group, and NFP-administration group and were gavaged once a day for 12 weeks. The body weights, serum lipid parameters, bile acid (BA)-related proteins, fecal BA, and cecal bacteria of the mice were obtained and analyzed. Results indicate that NFP supplementation (200 mg/kg·bw) reduced the body weight, energy intake, and serum and liver lipids of the HFD mice. Further studies found that NFP activated the hepatic and colonic farnesoid X receptor and increased the excretion of fecal-conjugated BAs, particularly tauro-alpha-muricholic acid and tauro-beta-muricholic acid. In addition, NFP modulated the intestinal microbiota and increased the relative abundance of Lactobacillus in the HFD mice. In conclusion, NFP modulated gut microbiota and improved BA metabolism, which may be the main mechanism behind NFP alleviation of fat accumulation and dyslipidemia in mice under HFD.

The clinical efficiency of everolimus, an mammalian target of rapamycin (mTOR) inhibitor, is palliative as sequential or second-line therapy for renal cell carcinoma (RCC). However, the limited response of everolimus in RCC remains uncertain. In the present study, everolimus-resistant RCC models were established to understand the mechanisms and to seek combination approaches. Consequently, the activation of ERK was found to contribute toward everolimus-acquired resistance and poor prognosis in patients with RCC. In addition, the efficacy and mechanism of combination treatment underlying RCC using everolimus and ERK inhibitors was investigated. The ERK inhibitor in combination with everolimus synergistically inhibited the proliferation of RCC cells by arresting the cell cycle in the G1 phase. The combination treatment markedly attenuated the deoxyribonucleoside triphosphate (dNTP) pools by downregulating the mRNA expression of RRM1 and RRM2 through E2F1. The overexpression of E2F1 or supplementation of dNTP rescued the anti-proliferation activity of the everolimus-SCH772984 combination. The antitumor efficacy of combination therapy was reiterated in RCC xenograft models. Thus, the current findings provided evidence that the everolimus-ERK inhibitor combination is a preclinical therapeutic strategy for RCC. Inflammatory bowel disease (IBD) is a chronic relapsing inflammation involving the gut system, and disequilibrium of T helper (Th) cell paradigm has been recognized as critical pathogenesis. Pycnoporus sanguineus (L.) Murrill is a species of the white-rot basidiomycetes listed as food- and cosmetic-grade microorganisms. In this study, anti-inflammatory activity of the ethanol extract from P. sanguineus (PSE) was investigated in dextran sulfate sodium (DSS)-induced experimental colitis model. PSE recovered the DSS-caused weight loss, reversed the colon shortening, and ameliorated the histopathological lesion in colon, resulting in lower disease activity index (DAI). Levels of serumal lipopolysaccharide (LPS), colonic myeloperoxidase (MPO) in the colitis-suffering mice were declined by PSE treatment. PSE also improved the mucosal integrity by enhancing the expression of tight junction and adherens junction proteins in the colon, including ZO-1, occludin, claudin-1, and E-cadherin. Besides, PSE reduced helper T cells (Th) in the colon, together with an evident decrease of several Th cell-related cytokines. Moreover, it was found that in vitro, PSE suppressed T cells and the Th subset upon Concanavalin A (ConA)-stimulation by inducing apoptosis. In summary, PSE displayed a remission on the colitis-related inflammation, which would possibly rely on the epithelial barrier restoration by suppressing Th cells via apoptosis induction, highlighting a promising potential in the treatment for IBD.

This study compared enhanced serotonin (5-HT) synthesis roles of γ-[Glu]n-Trp (EW) and whey protein hydrolysate (WPH, Val-Ala-Gly-Thr-Trp-Tyr, Val-Ala-Gly-Thr-Trp, Ser-Ala-Trp-Tyr, Trp-Tyr-Ser, Gly-Thr-Trp) using anxious depression C57BL/6 mice. As an index of serotonergic activity, we assessed 5-HT levels in response to Trp oligopeptide ingestion. The neuroprotective and hypothalamic-pituitaryadrenal (HPA) axis hormones were examined in order to characterize the suggested improvements of anxious depression observed in mice after ingestion Trp oligopeptide. The 5-HT levels were increasingly enhanced by the EW and WPH diet, with the EW being more effective. But WPH had higher capacity on anti-oxidative than EW. EW and WPH could normalize the levels of HPA axis hormones, and promote brain-derived neurotrophic factor (BDNF)/tyrosine kinase B (TrkB) signaling. In conclusion, Trp oligopeptide ingestion neuroprotective and HPA regulation effects possibly related to serotonergic activation and anti-oxidation. Consumed Trp oligopeptide diets may induce beneficial effects on anxious depression.

The data presented in this article is related to the research article entitled “High-accuracy haplotype imputation using unphased genotype data as the references” which reports the unphased genotype data can be used as reference for haplotyping imputation [1]. This article reports different implementation generation pipeline, the results of performance comparison between different implementations (A, B, and C) and between HiFi and three major imputation software tools. Our data showed that the performances of these three implementations are similar on accuracy, in which the accuracy of implementation-B is slightly but consistently higher than A and C. HiFi performed better on haplotype imputation accuracy and three other software performed slightly better on genotype imputation accuracy. These data may provide a strategy for choosing optimal phasing pipeline and software for different studies.

Summary: In this study, we obtained the whole genome sequence of GCRV-DY197 and investigated the localization, post-translational modifications, and host interactions of the 11 viral proteins encoded by GCRV-DY197 in grass carp ovary (GCO) cells. The whole genome sequence is 24,704 kb and contains 11 segments (S1–S11). Subcellular localization showed that the VP1, VP2, VP3, VP5, VP56, and VP35 proteins were localized in both cytoplasm and nucleus, whereas the NS79, VP4, VP41, VP6, and NS38 proteins were localized in the cytoplasm. The NS79 and NS38 proteins were phosphorylated, and the ubiquitination modification sites were identified in VP41 and NS38. An interaction network containing 9 viral proteins and 140 host proteins was also constructed. These results offer a theoretical basis for an in-depth understanding of the biochemical characteristics and pathogenic mechanism of GCRV-II.

Superchargers have become a focus of much research into new-energy vehicles, for which the cooling of high-current cable cores is a key problem that needs to be solved. To estimate influences of different core structures of liquid-cooled cables on the fluid flow and heat transfer characteristics in circular pipes, nine helical cable core structures with insertion of smooth pipes were designed taking dimethyl silicone oil as the coolant. The fluid flow and heat transfer in the pipes were studied through computational fluid dynamics (CFD) numerical simulation at different inlet velocities (0.2–2.0 m/s), and the models were verified by building an experimental platform. Numerical calculation results show that under conditions of the same Re and pitch of cable cores, the average surface temperature of three-core cables is lower than those of single-core and double-core cables. Moreover, the mean flow velocity v in pipes, heat transfer performance Nu, resistance factor f, evaluation factor for comprehensive heat transfer performance (PEC), and field synergy number Fc all increase with the decrease in the pitch of cable cores. This finding indicates that the heat transfer effect is gradually enhanced. The research implies that when the pitch p is 22.4 mm (cable C6), the velocity field is the most synergetic with the temperature field and the comprehensive heat transfer performance is optimal. This research provides an excellent cooling scheme for cable cores in superchargers under a high current load.

B-cell lymphoma-2 (BCL-2) superfamily molecules play crucial roles in mitochondrial apoptosis induced by Chinese giant salamander iridovirus (GSIV). As an anti-apoptotic molecule in the BCL-2 family, the molecular mechanism of Bcl-w during GSIV infection remains unknown. In this study, we characterized for the first time an amphibian Bcl-w from Chinese giant salamander Andrias davidianus (AdBcl-w), and its function and regulatory mechanism during GSIV infection were investigated. AdBcl-w possesses the conserved structural features of Bcl-w and shares 35–54% sequence identities with other Bcl-w. mRNA expression of AdBcl-w was most abundant in liver and muscle. The AdBcl-w mRNA expression was regulated during GSIV infection. Western blotting assays revealed that the level of Bcl-w protein was downregulated markedly as the infection progresses. Confocal microscopy showed that overexpressed AdBcl-w was translocated to the mitochondria after infection with GSIV. Flow cytometry analysis demonstrated that compared with control, the apoptotic progress in cells transfected with AdBcl-w was reduced while that in cells transfected with AdBcl-w siRNA was enhanced. The number of virus major capsid protein gene copies was lower and protein synthesis was reduced in AdBcl-w overexpressing cells. In addition, AdBcl-w could bind directly to the pro-apoptotic molecule AdBak, while this interaction was weakened with GSIV infection. Moreover, p53 level was reduced and the mRNA expression levels of crucial regulatory molecules in the p53 pathway were regulated in AdBcl-w overexpressing cells during GSIV infection. These results suggested that AdBcl-w inhibit GSIV replication by regulating the virus induced mitochondrial apoptosis.

Fusion-associated small transmembrane (FAST) proteins can promote cell fusion, alter membrane permeability and trigger apoptosis to promote virus proliferation in orthoreoviruses. However, it is unknown whether FAST proteins perform these functions in aquareoviruses (AqRVs). Non-structural protein 17 (NS17) carried by grass carp reovirus Honghu strain (GCRV-HH196) belongs to the FAST protein family, and we preliminarily explored its relevance to virus infection. NS17 has similar domains to FAST protein NS16 of GCRV-873, comprising a transmembrane domain, a polybasic cluster, a hydrophobic patch and a polyproline motif. It was observed in the cytoplasm and the cell membrane. Overexpression of NS17 enhanced the efficiency of cell-cell fusion induced by GCRV-HH196 and promoted virus replication. Overexpression of NS17 also led to DNA fragmentation and reactive oxygen species (ROS) accumulation, and it triggered apoptosis. The findings illuminate the functions of NS17 in GCRV infection, and provide a reference for the development of novel antiviral strategies.

Deciphering biogeographical patterns of alkaline phosphatase (phoD)-harboring bacteria is essential to understand organic phosphorus mineralization. However, it is poorly understood about distribution pattern and diversity maintenance mechanisms of phoD-harboring bacteria (PHB) in watershed ecosystems. Here, we estimated ecological processes shaping landscape of PHB in soils and sediments along the Yangtze River. The PHB community similarity decayed against higher geographical distance at taxonomic and phylogenetic levels, and larger compositional variation in PHB community were found in sediments only. The PHB displayed higher α-diversities, broader environmental breadths, higher community stability, and stronger species replacement in soils. Conversely, PHB showed stronger phylogenetic signals in sediments. Stochastic and differentiating processes dominated community assemblies of PHB in both soils and sediments. Electrical conductivity displayed decisive roles in shaping PHB diversity for soils and sediments at taxonomic and phylogenetic levels. Our results emphasized differences in distribution patterns of PHB between soils and sediments, and highlighted ecological processes shaping landscapes of PHB in soils and sediments along the Yangtze River. The phosphorus cycling-related findings might be helpful to estimate ecological potential of a watershed ecosystem and could provide new insights for ecological protection policy for the Yangtze River.

Summary: The spatial pattern and driving mechanism of biodiversity along elevational gradients are key topics in ecology. However, it is still unclear whether the multidimensional diversity of different types of organisms shows a similar response to elevation changes. Here, we measured the species and phylogenetic diversity of plants, bacteria, fungi, and microbial functional groups (nitrifiers, denitrifiers, methanogens, and methanotrophs) in 36 wetland sites on the Qinghai-Tibetan Plateau. The results showed that both species and phylogenetic diversity of plants, bacteria, and fungi exhibited a significant elevational gradient, in direct contrast to no significant diversity changes observed for denitrifiers, methanogens, and methanotrophs along the same altitude gradient. Our findings suggest that elevation and temperature were more likely to associate with the diversity of plants, bacteria, and fungi than the diversity of microbial functional groups, with important implications for assessing the effect of ongoing climate warming on biodiversity in Qinghai-Tibetan alpine wetlands.

Nanoplastics (NPs) are increasingly perceived as an emerging threat to terrestrial environments, but the adverse impacts of NPs on soil fauna and the mechanisms behind these negative outcomes remain elusive. Here, a risk assessment of NPs was conducted on model organism (earthworm) from tissue to cell. Using palladium-doped polystyrene NPs, we quantitatively measured nanoplastic accumulation in earthworm and investigated its toxic effects by combining physiological assessment with RNA-Seq transcriptomic analyses. After a 42-day exposure, earthworm accumulated up to 15.9 and 143.3 mg kg−1 of NPs for the low (0.3 mg kg−1) and high (3 mg kg−1) dose groups, respectively. NPs retention led to the decrease of antioxidant enzyme activity and the accumulation of reactive oxygen species (O2− and H2O2), which reduced growth rate by 21.3 %–50.8 % and caused pathological abnormalities. These adverse effects were enhanced by the positively charged NPs. Furthermore, we observed that irrespective of surface charge, after 2 h of exposure, NPs were gradually internalized by earthworm coelomocytes (∼0.12 μg per cell) and mainly amassed at lysosomes. Those agglomerations stimulated lysosomal membranes to lose stability and even rupture, resulting in impeded autophagy process and cellular clearance, and eventually coelomocyte death. In comparison with negatively charged nanoplastics, the positively charged NPs exerted 83 % higher cytotoxicity. Our findings provide a better understanding of how NPs posed harmful effects on soil fauna and have important implications for evaluating the ecological risk of NPs.

Intestinal microbiota influence host health. Numerous pathogens have been reported in crayfish (Procambarus clarkii) breeding systems. White spot disease, which is caused by white spot syndrome virus (WSSV), is the most severe disease observed in crayfish aquaculture systems. The present study investigated changes in intestinal morphology and gut microbiota in crayfish under WSSV infection. WSSV infection caused intestinal villi and hepatopancreas damage. In addition, intestinal microbiota diversity and richness decreased significantly under WSSV infection, and some functions of the intestinal microbiota were altered. Proteobacteria and Firmicutes abundance increased and decreased significantly, respectively, under WSSV infection. At the genus level, the relative abundance of an opportunistic pathogen, Aeromonas, increased significantly, and the abundances of Acinetobacter and Kurthia decreased significantly, in WSSV-infected crayfish. Additionally, according to the results of metagenomics analyses using PICRUSt, the altered microbiota was mainly linked to metabolism, diseases, environmental information processing, organismal systems, and cellular processes. Microbiome phenotype analyses based on BugBase revealed that the potential pathogenicity of intestinal flora in WSSV-infected crayfish was higher than in healthy crayfish. Overall, WSSV infection influenced intestinal and hepatopancreatic morphology, in addition to gut microbiota structure in crayfish.

A global survey was performed with 122 aquatic metagenomic DNA datasets (92 lake water and 30 seawater) obtained from the Sequence Read Archive (SRA). Antibiotic resistance genes (ARGs) and metal resistance genes (MRGs) were derived from the dataset sequences via bioinformatic analysis. The relative abundances of ARGs and MRGs in lake samples were in the ranges ND (not detected)–1.34 × 100 and 1.22 × 10−3–1.98 × 10−1 copies per 16S rRNA, which were higher than those in seawater samples. Among ARGs, multidrug resistance genes and bacitracin resistance genes had high relative abundances in both lake and sea water samples. Multi-metal resistance genes, mercury resistance genes and copper resistance genes had the greatest relative abundance for MRGs. No significant difference was found between epilimnion and hypolimnion in abundance or the Shannon diversity index for ARGs and MRGs. Plastics have been accumulated offshore and in the deep oceans at an unprecedented scale. Microbial communities have colonized the plastisphere, which has become a reservoir for both antibiotic and metal resistance genes (ARGs and MRGs). This is the first analysis of the diversity, abundance, and co-occurrence of ARGs and MRGs, and their relationships within the microbial community, using metagenomic data of plastic particles observed in the North Pacific Gyre obtained from the National Centre for Biotechnology Information Sequence Read Archive database. The abundance of ARGs and MRGs in microbial communities on the plastics were in the ranges 7.07 × 10−4–1.21 × 10−2 and 5.51 × 10−3–4.82 × 10−2 copies per 16S rRNA, respectively. Both the Shannon-Wiener indices and richness of ARGs and MRGs in plastics microbiota were significantly greater than those of ARGs and MRGs in seawater microbiota in the North Pacific Gyre via one-way analysis of variance. Multidrug resistance genes and multi-metal resistance genes were the main classes of genes detected in plastic microbiota. There were no significant differences in the abundance or diversity of ARGs and MRGs between macroplastics biota and microplastics biota, indicating that particle size had no effect on resistance genes. Procrustes analysis suggested that microbial community composition was the determining factor of the ARG profile but not for MRG. Some ARGs and MRGs had a higher incidence of non-random co-occurrence, suggesting that the co-effects of selection for antibiotic or metal resistance are important factors influencing the resistome of the microbiota on the plastic particles.
