Purpose: The purpose of this study was to review the seasonal variation of Kawasaki disease (KD) by an age-specific analysis to clarify the distribution of infectious agents., Methods: Data obtained from nationwide surveys of KD in Japan, which targeted patients for 12 years (2003-2014), were analyzed. The monthly numbers of patients were classified into the following age groups: 0-11 months, 1 year, 2-3 years 5 months, and 3 years 6 months-4 years. Factors associated with disease onset were analyzed using a 12-month moving average method., Results: In winter, a sharp peak was observed in all age groups, but this was notably sharper in the 1-year age group. Plateaus in disease occurrence were observed in two periods: from March to May in the 2- to 4-year age group and from June to August in the 0- to 11-month age group. Seasonal index was analyzed into two factors that differed depending on the age group., Conclusions: The age-specific analysis of KD clearly identified age-related differences in the seasonal occurrence of this disease., (Copyright © 2018 Elsevier Inc. Some Listeria monocytogenes strains, termed persistent strains, originate from the same processing plant and have the ability to survive and grow over extended periods of time at contamination sources. In order to evaluate biofilm formation by such persistent strains, we isolated the pathogen from chicken samples collected from the same retail shop in repeated visits over 6 months. Strains that were of serotype 1/2b and were assigned to the same genotype by multi-virulence-locus sequence typing analysis were isolated on repeated occasions from December 1997 to June 1998 and thus were defined as persistent strains. In the present study, biofilm formation by the persistent strains was evaluated using microplates at 30 and 37°C. The biofilm-forming capability was measured after cells attaching to the microplate well were stained with crystal violet. Comparison of biofilm formation at 30°C among the persistent strains showed that a significantly higher amount of the stain was obtained from the persistent strains isolated from December to March than from those isolated from April to June. However, no significant difference in biofilm formation at 30°C was observed between persistent and nonpersistent groups of L. monocytogenes strains. In contrast, biofilm formation at 37°C was consistent among the persistent strains, and they produced significantly more biofilm at 37°C than did the nonpersistent strains. The persistent strains were also found to change their biofilm-forming ability in a temperature-dependent manner, which may suggest that the persistent strains alter their biofilm formation in response to changing environmental factors.

This study was conducted to determine the prevalence of Listeria monocytogenes in retailed meats, comprising beef, chicken, and pork, in the Tokyo metropolitan area. A total of 379 samples of retailed meat were collected from 1998 to 2003, most of which were obtained by simultaneously purchasing the three classes of meat from a shop and then making another simultaneous purchase of meat from the same shop a few weeks later. The prevalence of L. monocytogenes was 28.0%, and the serotypes isolated were mainly 1/2a, 1/2b, 1/2c, and 4b. Comparison of the prevalence of each serotype among the classes of meat showed a predominant distribution of serotypes 1/2a, 1/2b, and 4b in chicken, while serotype 1/2c was dominant in pork. A total of nine cases considered to be due to persistence and/or cross-contamination were found. Most of the strains involved in persistence and/or cross-contamination were of serotypes 1/2c or 4b. These results suggest that contamination in retailed meat in Japan is at almost the same level as in other countries and that chicken has the highest potential as a source of contamination and infection. In addition, we suggest that the ecological niche of serotype 1/2c is distinct from those of 1/2a, 1/2b, and 4b, which may explain why human hosts have less opportunity to be exposed to serotype 1/2c and why there is a lower rate of isolation of this serotype from cases of human listeriosis.

PDGF promotes growth of smooth muscle cells (SMCs) and induces morphological changes. We investigated the effects of PDGF on cell-matrix adhesion of SMCs. PDGF reorganized integrin distribution as well as actin filament structure. PDGF-treated cells attached more loosely to fibronectin than non-treated cells. Decreased expression of alpha-actin by PDGF was antagonized by coating culture dishes with fibronectin but not with vitronectin and laminin. The inhibition of cell adhesion by GRGDSP oligopeptide and anti-fibronectin receptor antibody led to the reduction in alpha-actin expression. These results suggest that PDGF may modulate phenotype of SMCs by altering fibronectin-integrin interaction.

Platelet-derived growth factor (PDGF) plays important roles in proliferation of smooth muscle cells (SMCs). PDGF is also known to induce the cytoskeletal reorganization as a chemoattractant. In this communication, we demonstrate that the tyrosine kinase inhibitor herbimycin-A (HM-A) inhibited the effects of PDGF on SMCs in proliferation, phenotypic modulation, and reorganization of F-actin. Another tyrosine kinase inhibitor genistein also inhibited reorganization of F-actin. Since both tyrosine kinase inhibitors reduced PDGF-dependent tyrosine phosphorylation, tyrosine kinase activities may be involved not only in proliferation but also in reorganization of F-actin.

The nucleotide sequence of the import precursor of subunit b of rat liver H(+)-ATP synthase has been determined from a recombinant cDNA clone isolated by screening a rat liver cDNA library with a probe DNA. The sequence was composed of 1,124 nucleotides including a coding region for the import precursor of subunit b and noncoding regions of both the 5'- and 3'-sides. The import precursor of subunit b and its mature polypeptide deduced from the open reading frame consisted of 256 and 214 amino acid residues with a molecular weight of 28,867 and 24,628, respectively. The presequence of 42 amino acids could be the import signal peptide which serves to direct the protein into the mitochondrial matrix.

The mutagenicity of fenitrothion was determined in strains of Salmonella typhimurium and Escherichia coli. Fenitrothion was found to be non-mutagenic in Salmonella typhimurium strains of TA98, TA1535 and TA1537 and in Escherichia coli WP2uvrA both with and without S9 mix, while weak mutagenicity was observed only in Salmonella typhimurium TA100 and enhanced by the addition of S9 mix. The mutagenicity observed in the TA100 strain was not expressed in a nitroreductase-deficient strain, TA100 NR, and decreased in a transacetylase-deficient strain, TA100 1,8-DNP6. The mutagenicity of fenitrothion was also examined by a gene mutation assay using the gene for hypoxanthine-guanine phosphoribosyltransferase (hgprt) in V79 Chinese hamster lung cells. Fenitrothion did not induce any increment of 6-thioguanine-resistant mutant cells at doses ranging from 0.01 to 0.3 mM regardless of the presence or absence of S9 mix. These results suggest that reduction of fenitrothion by a bacterial nitroreductase of TA100 to an active form is essential for the expression of the mutagenicity of fenitrothion in TA100 and that a bacterial transacetylase of TA100 also has an important role in the process of mutagenic activation.

The antimutagenicity of extracts from crude drugs was studied by the Ames bioassay system. The crude drugs chosen were medical plants used very frequently as Chinese medicines. Each crude drug was extracted with hot water similar to the method of Chinese medical treatment. Antimutagenicity of the extract was found with 4 kinds of crude drugs, Paeoniae radix, Bupleuri radix, Hoelen and Glycyrrhizae radix. Each extract of the crude drug showed a different type of antimutagenic action from the others.

A simple and practical method for the determination of very small amounts of nitrite in human, cow and market milks is described. It is based on the reaction of nitrate with 1-hydrazinophthalazine in acidic solution to form tetrazolophthalazine, a stable compound which can be extracted with an organic solvent and then determined by gas-liquid chromatography with electron-capture detection using a column of 3% OV-225 on Chromosorb W HP; the detection limit for nitrite was 2 ng/ml. The procedure for determining nitrite in milks involves extraction with solvent, followed by further clean-up by alumina column chromatography; the detection limit is about 4 ppb and recovery in human, cow and market milks was satisfactory. The method makes possible a micro-assay for nitrite. The tetrazolophthalazine was identified by its elemental composition, melting point and by combined gas chromatography-mass spectrometry.
