This study demonstrates the feasibility and effectiveness of utilizing native soils as a resource for inocula to produce n-caproate through the chain elongation (CE) platform, offering new insights into anaerobic soil processes. The results reveal that all five of the tested soil types exhibit CE activity when supplied with high concentrations of ethanol and acetate, highlighting the suitability of soil as an ideal source for n-caproate production. Compared with anaerobic sludge and pit mud, the native soil CE system exhibited higher selectivity (60.53%), specificity (82.32%), carbon distribution (60.00%), electron transfer efficiency (165.00%), and conductivity (0.59 ms∙cm−1). Kinetic analysis further confirmed the superiority of soil in terms of a shorter lag time and higher yield. A microbial community analysis indicated a positive correlation between the relative abundances of Pseudomonas, Azotobacter, and Clostridium and n-caproate production. Moreover, metagenomics analysis revealed a higher abundance of functional genes in key microbial species, providing direct insights into the pathways involved in n-caproate formation, including in situ CO2 utilization, ethanol oxidation, fatty acid biosynthesis (FAB), and reverse beta-oxidation (RBO). The numerous functions in FAB and RBO are primarily associated with Pseudomonas, Clostridium, Rhodococcus, Stenotrophomonas, and Geobacter, suggesting that these genera may play roles that are involved or associated with the CE process. Overall, this innovative inoculation strategy offers an efficient microbial source for n-caproate production, underscoring the importance of considering CE activity in anaerobic soil microbial ecology and holding potential for significant economic and environmental benefits through soil consortia exploration.

The current interaction of pedestrian flow and environmental pollutants in high-occupancy public areas of apartment and the risks of residents being exposed to environmental pollutants are issues that are often overlooked but urgently need to be addressed. In this study, we provide a comprehensive of pedestrian flow-environmental pollutants interactions and health risks to residents in first-floor public areas of apartment with high-occupancy. The main findings indicate that under closed management conditions, there is a significant increase in TVOC and noise levels during the peak periods of nighttime pedestrian flow. In the correlation analysis, the significant impact of time granularity selection in clarifying the correlation between pedestrian flow and environmental pollutants has been highlighted, with larger time granularities generally showing stronger correlations, while finer time granularities may help identify specific risks in areas directly connected to the external environment. There is a significant correlation exists between pedestrian flow and environmental pollutants (TVOC, ozone, and noise), with higher concentrations of these pollutants observed during peak pedestrian flow periods, thereby increasing the risk of residents being exposed to adverse environmental conditions. To mitigate the risks associated with TVOC pollution and noise exposure, it is crucial to maintain proper ventilation, avoid conducting cleaning or maintenance activities during peak hours, and implement noise-reducing measures, such as distancing noise sources from residential areas or installing soundproofing barriers. Additionally, the study identifies total volatile organic compounds originating from property maintenance activities and clarifies their dispersion patterns, emphasizing the importance of developing robust, standardized maintenance protocols for indoor environmental quality assurance. This research can improve the environmental sustainability of apartment buildings and provide a theoretical basis for the development of environmental health strategies for high-occupancy public areas of apartment buildings.

Urban areas are currently facing the increasingly pressing issue of urban heat worldwide, which is being worsened by climate change and rising urbanization. As a result, there is a growing need for new approaches to enhance urban resilience and adapt to these challenges. The escalating occurrence and severity of urban heat events provide notable hazards, particularly to susceptible groups, necessitating proactive efforts to alleviate detrimental consequences. Therefore, this research addresses the inquiry, “What strategic approaches can be effectively employed to mitigate vulnerability and strengthen urban resilience in response to urban heat?” Thus, this study ascertains and examines approaches to enhance urban resilience, mitigate susceptibility, and implement adaptation strategies to combat urban heat. Utilizing the content analysis method, a comprehensive assortment of documents encompassing academic publications, policy documents, and reports was subjected to a systematic analysis employing the MAXQDA software. Databases searched included Web of Science, Scopus, and Google Scholar, and a total of 72 studies were included in the final analysis. The research reveals a wide range of novel ideas and practical measures that can be implemented to improve urban resilience and mitigate vulnerability to urban heat. Urban greening strategies, heatwave early warning sys-tems, and community involvement projects have exhibited differing effectiveness, application, and adaptation levels in many urban landscapes and socio-economic circumstances. Additionally, this research emphasizes the value of using multidimensional, context-specific strategies to address the unique challenges and needs of diverse urban regions and marginalized communities. Furthermore, structural changes, legislative reforms, and community-based solutions may be necessary to manage complex issues posed by urban heat. Therefore, effectively implementing adaptation strategies is vital to effectively combating challenges caused by urban heat in urban areas.

The Connected Sensor Problem (CSP) presents a prevalent challenge in the realms of communication and Internet of Things (IoT) applications. Its primary aim is to maximize the coverage of users while maintaining connectivity among K sensors. Addressing the challenge of managing a large user base alongside a finite number of candidate locations, this paper proposes an extension to the CSP: the h-hop independently submodular maximization problem characterized by curvature α. We have developed an approximation algorithm that achieves a ratio of 1−e−α(2h+3)α. The efficacy of this algorithm is demonstrated on the CSP, where it shows superior performance over existing algorithms, marked by an average enhancement of 8.4%.

The collaborative governance of inclusive preschool education (IPE) is a process in which multiple subjects work together to promote the advancement and development of IPE by taking a series of governance measures. Based on precise identification of multiple stakeholders in collaborative governance of integrated preschool education, it applied the quadrilateral evolutionary game model and Lyapunov's first law to analyze the stability of behavioral strategy choice by multiple subjects in promoting the practice of collaborative governance of IPE and the possible equilibrium points in the system. The influence of key elements on the system evolution is analyzed using MATLAB 2019 simulation. The study reveals that the evolution and trend of one game agent's behavior are closely related to the strategy choice of others. The willingness of the main body of kindergarten, social organization, and parents to participate in the collaborative governance of IPE are related to the government's active promotion to perform its leading functions at the government level. The willingness for collaborative governance of integrated education can help improve mutual trust and the main body's benefits, thus promoting the collaborative behavior of multiple subjects. Complementary resources, strict supervision, reasonable rewards and punishments can promote the collaborative behavior of multiple subjects.

Due to increasingly strict global emission regulations and energy conservation policies, gasoline engines are facing significant challenges. Achieving high efficiency and ultra-low emissions in gasoline engines has become a critical technical challenge that requires attention. Pre-chamber (PC) Turbulent Jet Ignition (TJI) is a highly promising technology for improving thermal efficiency and reducing emissions in gasoline engines. This study employs a three-dimensional flow simulation analysis combined with a detailed chemical reaction mechanism. The investigation is based on eight distinct passive PC structural configurations. The effects of various structural parameters, including the orifice number (ON), orifice axis angle (OAA), orifice diameter (OD), and volume ratio (VR), on the in-cylinder combustion process and emission characteristics are investigated, in which the formation of the fire nucleus and the propagation of the flame are intensively involved. The investigation is conducted under equivalence ratio working condition. Among these, the ITE of the best configuration is relatively 5.46% higher than the baseline engine. The combustion duration decreased by 55.95% to 7.4 °CA compared to the baseline engine, while particulate soot emissions also decreased by 51.22%.

The LiFePO4 material continue to suffer from poor rate performance. Nanosized LFP can improve rate performance; however, nanofabrication typically requires expensive organic matter and high-pressure-resistant equipment. The current widely used solid-phase sintering methods employ FePO4 as the precursor, but it is difficult to suppress grain growth. Here, we propose a W–Ti codoping strategy to suppress grain growth during sintering, thereby improving rate performance of Li-ion batteries. W and Ti doping shrinks grains during the sintering process and make the Li-O bond more ionized. Li0.36WO3 accumulates at grain boundaries, hindering grain boundary diffusion and inhibiting grain growth during sintering. The resulting material exhibits refined grains and suppressed polarization growth, resulting in improved rate performance while maintaining a specific capacity of 117 mAh g−1 at a rate of 10 C. After 1000 cycles at a rate of 1 C, 91.0 % of the initial capacity was retained. In summary, this research provides a method for large-scale production of LFP with excellent rate performance.

In response to the epochal demand for high-quality development in early childhood education in China, it is imperative and necessary to improve the competency level of early childhood educators. The study aims to investigate the relationship between psychological contracts and teacher competency, and to verify the mediating roles of job crafting and professional identity in the relationship between psychological contracts and teacher competency. This study employed validated measurement scales regarding psychological contracts, teachers' professional identity, job crafting, and teacher competency. Each of these scales has established internal consistency coefficients. Data were collected from 318 early childhood teachers in Sichuan, China. The results highlight the significant impact of psychological contracts on the prediction of teacher competency. It is worth mentioning that the psychological contracts, together with their distinct components such as normative responsibility and development responsibility, have a direct and favorable impact on teacher competency. This implies that developing the psychological contracts might be an effective technique for improving teacher competence. The individual mediation of job crafting and professional identity in the link between psychological contracts and teacher competency has been well-established. However, the combined or chain mediating influence of these factors provides a unique and valuable perspective on the phenomenon of job crafting leading to professional identity, which in turn impacts teacher competency. The study found that psychological contracts have a positive predictive effect on teacher competency, while job crafting and professional identity both have independent and chain mediating roles in the relationship between psychological contracts and teacher competency. Therefore, this study suggests a comprehensive enhancement of the psychological contracts level from aspects such as normative responsibility, interpersonal responsibility, and development responsibility. By stimulating job crafting and professional identity levels in both internal and external environments, we can improve the competency level of early childhood educators.

The successful implementation of urban resilience strategies is of utmost importance in order to reduce susceptibility and bolster resilience in the face of climate change consequences. The current understanding of the efficacy of different resilience strategies in mitigating vulnerability and bolstering urban resilience is lacking, despite its significance. This study assesses the efficacy of resilience strategies in mitigating vulnerability and enhancing urban resilience. We conducted a comprehensive analysis of scholarly literature published in English following PRISMA criteria from January 2001 to July 2023. Finally, 116 articles met the inclusion criteria and were selected for in-depth analysis. Results indicate that while resilience strategies have the potential to reduce susceptibility and enhance urban resilience, the effectiveness of resilience techniques is contingent upon various factors, such as the type of hazard, urban setting, and implementation process. The study also highlights the significance of stakeholder involvement, community participation, and adaptive management as essential components for effectively implementing resilience measures. Integrating physical, social, and institutional components in resilience practices demonstrated notable effectiveness. This study also reveals that improving the physical resilience of urban areas and strengthening their social and institutional capabilities to address and learn from disruptive events and pressures can decrease their vulnerability. The research also exposes those strategies focusing solely on mitigating a single issue, such as physical infrastructure, while neglecting social or institutional elements, which prove less effective. A comprehensive approach, incorporating institutional, social, and physical measures, should be designed to achieve maximal efficacy in mitigating vulnerability and strengthening urban resilience.

Background: Identifying biomarkers may lead to easier detection and a better understanding of pathogenesis of pancreatic ductal adenocarcinoma (PDAC). Methods: Plasma small extracellular vesicles (sEV) from 106 participants, including 20 healthy controls (HC), 12 chronic pancreatitis (CP) patients, 12 benign pancreatic tumour (BPT) patients, and 58 PDAC patients, were profiled for microRNA (miRNA) sequencing. Three machine learning methods were applied to establish and evaluate the diagnostic model. Results: The plasma sEV miRNA diagnostic signature (d-signature) selected using the three machine learning methods could distinguish PDAC patients from non-PDAC individuals, HC, and benign pancreatic disease (BPD, CP plus BPT) both in training and validation cohort. Combining the d-signature with carbohydrate antigen 19-9 (CA19-9) performed better than with each model alone. Plasma sEV miR-664a-3p was selected by all methods and used to predict PDAC diagnosis with high accuracy combined with CA19-9. Plasma sEV miR-664a-3p was significantly positively associated with the presence of vascular invasion, lower surgery ratio, and poor differentiation. MiR-664a-3p was mainly distributed in the PDAC cancer stroma, including fibers and vessels, and was accompanied by VEGFA expression. Overexpression of miR-664a-3p could promote the epithelial-mesenchymal transition (EMT) and angiogenesis. Conclusion: In conclusion, our study demonstrated the potential utility of the sEV-miRNA d-signature in the diagnosis of PDAC via machine learning methods. A novel sEV biomarker, miR-664a-3p, was identified for the diagnosis of PDAC. It can also potentially promote angiogenesis and metastasis, provide insight into PDAC pathogenesis, and reveal novel regulators of this disease.

The Technical Note aims to present an arthroscopic capsular closure technique at the end of the hip arthroscopy. The technology employs a dual-channel approach and modified shoelace suture technique to continuously suture the hip capsule. Recent studies have indicated that routine intraoperative repair of the articular capsule at the end of the hip arthroscopy is advocated. However, the majority of the hip capsular closure techniques are relatively complex, time-consuming, and bring many complications, which has hindered their widespread use in clinical practice. Herein, we provide an arthroscopic capsular closure technique using the modified shoelace continuous suture in combination with a dual cannula for correcting hip instability during hip arthroscopic surgery of femoroacetabular impingement.

Resilience is a prior term of debate in vulnerability research irrespective of discipline. Nowadays the resilience concept has been used abnormally in many cases without realizing its intended meaning. Therefore, this study clarifies the resilience concept in the context of climate vulnerability. An integrated review of the literature has been done by the PRISMA approach to identify the most relevant documents. Besides Walker and Avant Method has been used to analyze the concept of resilience. The use of concept in different disciplines, historical perspectives, dimensions, attributes, consequences, and relationship with vulnerability and adaptability have been reviewed to clarify it. This study also provides a theoretical and workable definition of resilience from the perspective of climate change and natural hazards. A conceptual framework has been developed with concept mapping, contributing to the existing debate on resilience study. A case analysis has been done by focusing on a climate-vulnerable community living in the riverine islands in Bangladesh. This article shows that socio-ecological systems approach to defining small island disaster resilience has shortcomings. Additionally, the research-policy divide in resilience studies is exacerbated by the paucity of studies that use the participatory approach to identify disaster resilience indicators. The study makes a compelling case for further research on how to turn academic discoveries into a resource that might benefit local people, particularly those on small islands. The study suggests that social community-based resilience should be developed to protect geographically isolated communities.

The continuous development of large-scale electric vehicles (EVs) has provided abundant dispatchable resources for the power grid. However, the existing control methods are not enough to support the effective interaction of the EVs and the grid. This paper proposes a method for generating a charging plan of EV clusters considering the stable operation of the regional distribution network, which fully considers the load regulation requirements of the grid and the charging economy of the EV clusters. Combined with the improved Particle Swarm Optimization based on Genetic Algorithm, a day-ahead charging plan for multiple EV clusters can be figured out. It can be proved by the case analysis that, compared with the disordered charging behavior and the participation of demand response with peak and valley electricity price, the charging plan generated by the method proposed can reduce the comprehensive cost of EV charging while optimizing the operation parameters of the distribution network, and it can reduce the calculation dimension in the process of plan generation.

The prediction of the orderly charging load of electric vehicles is of great significance for planning of charging facilities, analysis of bearing capacity of the distribution network and later rectification and reconstruction. This article proposes a method of charging load prediction based on characteristics of EV charging behavior and nonlinear programming. This article uses Monte Carlo algorithm to predict charging probability and disorderly charging load of EVs, and uses non -linear programming algorithm to solve the optimized target function of orderly charging to achieve the prediction of the orderly charging load of EVs in the region.

Summary: The type V-K CRISPR-associated transposons (CASTs) allow RNA-guided DNA integration and have great potential as a programmable site-specific gene insertion tool. Although all core components have been independently characterized structurally, the mechanism of how the transposase TnsB associates with AAA+ ATPase TnsC and catalyzes donor DNA cleavage and integration remains ambiguous. In this study, we demonstrate that TniQ-dCas9 fusion can direct site-specific transposition by TnsB/TnsC in ShCAST. TnsB is a 3′-5′ exonuclease that specifically cleaves donor DNA at the end of the terminal repeats and integrates the left end prior to the right end. The nucleotide preference and the cleavage site of TnsB are markedly different from those of the well-documented MuA. We also find that TnsB/TnsC association is enhanced in a half-integration state. Overall, our results provide valuable insights into the mechanism and application expansion of CRISPR-mediated site-specific transposition by TnsB/TnsC.

In this paper, the rice hush ash (RHA) with different particle size were applied to explore its effect on the autogenous shrinkage and hydration property of cement pastes. The water absorption/release behavior of RHA in cement pastes were characterized by using 1H nuclear magnetic resonance. Additionally, the microstructure and compressive strength of cement pastes containing RHA were also measured. Experimental results showed that water absorption capacity of raw RHA in cement pastes reached 0.89 g/g due to its porous structure, but reduced with the decrease of RHA particle size. The addition of RHA could mitigate the development of self-desiccation, being ascribed to the release of water from RHA after about 12 h which decreased the consumption of capillary water. The autogenous shrinkage of cement pastes containing RHA was reduced by about 41.6 %, 35.6 % and 23.8 %, respectively when the mean particle size of RHA decreased from 43.6 µm to 8.5 µm. Furthermore, the addition of RHA could shorten the dormant period and greatly enhance early-age compressive strength of cement pastes due to its water absorption effect. At later ages, the hydration degree of cement was continuously increased under the double effect of internal curing and pozzolanic reaction of RHA, resulting in the formation of the dense interfacial transition zone (ITZ) between matrix and RHA particles, ultimately improved long-term strength.

Under the action of repeated load of vehicles, asphalt pavement tends to suffer rutting and other failures at high temperatures, which shortens the service life of the pavement. Meanwhile, asphalt pavement aggravates the urban heat island (UHI) effect, reducing the livability of the city. Research shows that lower road temperatures can mitigate the UHI effect. For this reason, researchers have launched lots of studies to cool asphalt pavement under high temperatures, such as reflective pavement and phase change pavement, etc. Although these technologies are effective at reducing the temperature of asphalt pavement, there are still some limitations. This paper mainly discusses the latest development of asphalt pavement temperature reduction strategies, classifies them according to different action mechanisms, and discusses the advantages and limitations of each strategy in detail. Suggestions for future research are given to address the limitations of each strategy, such as passive radiation technology can provide new ideas for reflective pavement, planting trees should consider plant physiological parameters and spatial distribution of planting, and phase change materials can be encapsulated by high thermal conductivity materials, etc. This study can provide a guidance for further development of asphalt pavement cooling technologies.

The quality of a country's education system is both an indicator of its present level of development and a predictor of future economic advancement. This study intended to explore the impact of the “expanding power and strengthening counties” reform on the supply of basic public education resources. A combination of qualitative and quantitative methods was used to analyze the panel data of 114 counties in Sichuan Province from 2005 to 2017, including 78 counties with expanded powers. The study found that the “expanded power strong counties (EPSC)'' had a significant positive impact on the supply level of funding resources measured by per capita public education expenditure. The time effect of the EPSC on the supply of teacher resources for county public education was manifested as a sharp increase in the supply level in the three years before the reform. The supply level tended to be stable after four years of reform. The economic resources supply level was relatively stable in the first three years and rose sharply after four years. There was a downward trend after the reform for 8 years. The structural effect has shown a more significant incentive effect on the supply of public education resources to strong and weak counties. But compared with the weak counties with lower economic development levels, the strong counties with higher economic development levels have a more noticeable effect of their decentralization reform on improving the supply level of public education resources.

Previous research on antibiotic resistance genes and microorganisms centered on those in urban sewage treatment plants, breeding farms, hospitals and others with serious antibiotic pollution. However, at present, there are evident proofs that antibiotic resistance genes (ARGs) indeed exist in a primitive environment hardly without any human’s footprints. Accordingly, an original karst mountain swamp ecosystem in Niangniang Mountain, Guizhou, China, including herbaceous swamp, shrub swamp, sphagnum bog and forest swamp, was selected to analyze the physical and chemical parameters of sediments. Moreover, microbial compositions, functions, as well as their connections with ARGs were assayed and analyzed using metagenomic technology. The results showed that there was no significant difference in the dominant microorganisms and ARGs in the four marshes, in which the dominant bacteria phyla were Proteobacteria (37.82 %), Acidobacteriota (22.17 %) and Actinobacteriota (20.64 %); the dominant archaea Euryarchaeota. (1.00 %); and the dominant eukaryotes Ascomycota (0.07 %), with metabolism as their major functions. Based on the ARDB database, the number of ARGs annotated reached 209 including 30 subtypes, and the dominant ARGs were all Bacitracin resistance genes (bacA, 84.77 %). In terms of the diversity of microorganisms and ARGs, the herbaceous swamp ranked the top, and the shrub swamp were at the bottom. Correlation analysis between microorganisms and resistance genes showed that, apart from aac2ic, macB, smeE, tetQ, and tetL, other ARGs were positively correlated with microorganisms. Among them, baca coexisted with microorganisms. Pearson correlation analysis results showed that contrary to ARGs, microorganisms were more affected by environmental factors.

Compression ignition-internal combustion Rankine cycle (CI-ICRC) concept implement oxy-fuel combustion, direct water injection (DWI) and waster heat recovery into traditional diesel cycle to realize ultra-high thermal efficiency and low emission powertrain. As indicated by ICRC concept, the DWI process show critical impact on its combustion process and thermal efficiency. This paper dedicated in investigating the effect of different DWI temperature on cycle performance and thermal efficiency within prototype CI-ICRC engine, in order to do so, independent DWI system, O2/CO2 intake system and reconfigurable electronic controller is established. Combustion characteristics including in-cylinder pressure, heat release rate, combustion stability, brake thermal efficiency are experimentally investigated. The cycle performance and thermal efficiency are proved to be optimized under elevated DWI temperature, an optimum 46.6% brake thermal efficiency is achieved, while the coefficient of variation remains around 1%. The NOX and soot emissions are both reduced under DWI utilization, the mechanism of soot reduction is highly related to thermolysis of water or carbon-water reactions which further chemical kinetic analysis is needed. The result of this study provides fundamental information for future CI-ICRC engine optimization, and can also be utilized in providing reference guidance for DWI implementation within modern internal combustion engines.

We report the synthesis of β-palladium hydride nanodendrites (PdH0.33 NDs). The uniform PdH0.33 NDs are 36.3 ± 5.0 nm in diameter and selectively expose (111) planes decorated with (221) and (331) high-indexed steps. The PdH0.33 NDs show a high mass activity of 0.719 A/mgPd at 0.9 V (vs. reversible hydrogen electrode-RHE) toward alkaline oxygen reduction reaction (ORR), which is 3.7 and 6.3 times that of commercial Pt/C and house-made Pd/C, respectively. This study exemplifies the possibility of using special morphology and lattice H to modify the strain and electronic effect of metal for the optimization of functionalities. Metabolomics is an implement for testing the toxicity of antibiotics, and provides a comprehensive view of the overall response to stress; however, the connections between metabolites and biologic endpoints keep unclear in response to antibiotics. In this study, wheat seeds were exposed to tetracycline for 5 days. The results proved that tetracycline restrained growth, reduced chlorophyl and carotinoid contents and cell permeability, and increased reactive oxygen species (ROS) levels and malondialdehyde (MDA) content. Orthogonal partial least squares (OPLS) was used to analyze the connections between metabolites and biologic endpoints, which discovered that 11 metabolic pathways were significantly affected by tetracycline, and amino acid metabolism could largely apply to root growth and ROS accumulation, while carbohydrate metabolism could have a ruling effect on tetracycline-induced cell permeability. 13 metabolites all played active roles in mediating tetracycline’s effects on root length, root fresh weight and cell permeability but had no significant effects on ROS levels. The majority of metabolites with passive effects on root length, root fresh weight and cell permeability had active effects on ROS levels. These results offer a view about stress reaction of wheat to tetracycline.

The tensile properties of the lightweight titanium-based carbon-fibre/epoxy laminates (TI-CF FMLs) were investigated under quasi-static loading. A comprehensive experimental study was conducted for eleven different types of the TI-CF FMLs (considering various metal sheet thicknesses, metal volume fractions and fibre orientations) as well as their associated constituent materials (i.e., Ti-6Al-4V and carbon-fibre/epoxy laminates). Specimens were fabricated using vacuum bagging technique in an autoclave. Summary: The accurate control of early cell fate specification during differentiation of human embryonic stem cells (hESCs) is critical for acquiring pure therapeutic cell populations of interest. Bone morphogenetic protein 4 (BMP4) is a key mesoderm inducer from ESCs. However, the molecular mechanism of the mesodermal cell fate decision induced by BMP4 remains unclear. Here, we demonstrate the requirement of a bioactive lipid, prostaglandin E2 (PGE2), for the mesoderm specification from hESCs by BMP4 induction. We show that BMP4 directly regulates the expression of the key enzyme for PGE2 synthesis, COX-1, and promotes PGE2 production. More importantly, in the absence of BMP4, forced COX-1 expression or PGE2 treatment is sufficient to initiate mesoderm specification of hESCs by activation of EP2-PKA signaling and modulation of nuclear translocation of β-catenin. Together, our findings provide insights into the critical role of BMP regulation of PGE2 synthesis and its downstream signaling in initiating mesoderm commitment of hESCs. : In this article, Pei and colleagues demonstrate the requirement of prostaglandin E2 for the mesoderm specification from hESCs by BMP4 induction. The purpose of this study was to investigate the effects of salvianolic acid A (SAA) in systemic lupus erythematosus (SLE) induced by pristane in BALB/c mice. Lupus mice were established by confirming elevated levels of autoantibodies and IL-6 after intraperitoneal injection of pristane. Mice were then treated with daily oral doses of SAA for 5 months in parallel with mice treated with prednisone and aspirin as positive controls. The levels of autoantibodies were monitored at monthly intervals and nephritic symptoms observed by hematoxylin and eosin (H&E) and periodic acid–Schiff (PAS) staining. Western blot analysis of renal tissue was also employed. SAA treatment caused a significant reduction in the levels of anti-Sm autoantibodies and reduced renal histopathological changes and pathological effects. SAA treatment also significantly inhibited the phosphorylation of IKK, IκB and NFκB in renal tissues of lupus mice. In conclusion, the results suggest that SAA alleviates renal injury in pristane-induced SLE in BALB/c mice through inhibition of phosphorylation of IKK, IκB and NFκB.

This study compares the results of three certified methods, namely differential scanning calorimetry (DSC), the mass balance (MB) method and coulometric titrimetry (CT), in the purity assessment of ferulic acid certified reference material (CRM). Purity and expanded uncertainty as determined by the three methods were respectively 99.81%, 0.16%; 99.79%, 0.16%; and 99.81%, 0.26% with, in all cases, a coverage factor (k) of 2 (P=95%). The purity results are consistent indicating that the combination of DSC, the MB method and CT provides a confident assessment of the purity of suitable CRMs like ferulic acid.

By means of thermogravimetric analysis (TGA), influences of organic acids such as citric acid, oxalic acid and tartaric acid on decomposition process and thermal behaviors of calcium carbonate were investigated in non-isothermal condition. Experiments were conducted from the ambient temperature to 1273 K at a heating rate of 10 K min−1. Moreover, the kinetic parameters (activation energy and pre-exponential factor) of the mixtures of calcium carbonate and organic acids were determined using the Coats–Redfern method. It is indicated that the decomposition process of calcium carbonate containing different organic acids includes three or four weight loss processes. In the zone of 370–450 °C, energy is released due to the combustion of organic products decomposed from organic salts, and energy increases with the increase of molecular weight of organic acid. Activation energy of the samples containing citric acid, oxalic acid and tartaric acid is decreased by 34.9%, 28.8% and 31.9%, respectively. The results may be used to provide a useful basis for further applying industrial wastes containing different organic acids in cement industries with high-efficiency.

Background: Current management guidelines for gallbladder polyps (GBPs) focus on a diameter more than 1 cm as an indication for cholecystectomy. Since most GBPs are not malignant, unnecessary cholecystectomies can lead to unnecessary complications and costs. We developed a score to identify true polyps focusing on their cross-sectional area (CSA). Methods: We retrospectively analyzed the demographic, clinical, laboratory, and sonographic characteristics of 522 patients with GBPs who had undergone cholecystectomy at our hospital between January 2010 and July 2020 (reference group). We used univariate analysis to compare these parameters between 88 true polyps and 434 pseudopolyps and multivariate logistic regression analysis to identify parameters to include in our scoring model. Receiver operating characteristics and area under the curve were used to identify cut-off values. The model was tested on a validation group of 98 patients. Results: In the multivariate analysis, a CSA >123 mm2, positive blood flow signal, age >55.5 years, alanine aminotransferase (ALT) levels > 50 U/L, and an ALT/aspartate aminotransferase ratio > 0.77 were significantly associated with true polyps (odds ratio 6.528, 2.377, 2.617, 2.445, and −0.372, respectively). Metallic pollutants can have harmful impacts on ant morphology and physiology. We studied the occurrence of labial gland disease in Camponotus japonicus from two polluted areas (traffic pollution and industrial pollution) and one non-polluted area. We further analyzed the metal levels (Al, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb and Zn) and morphological characteristics (head width, body mass, and other morphological traits) of both diseased and healthy workers. Our results showed that labial gland disease was only present in polluted areas, indicating that pollution stress makes ants more vulnerable to infections. Our research revealed that diseased ants in polluted areas accumulate higher levels of metals in their bodies and have lower dry weight and residual body mass compared to healthy ants in non-polluted environments, negatively impacting their development. We evaluated the importance of these elements and found that Cu has the greatest impact on the health risk of C. japonicus. Natural water pollution and eutrophication are environmental problems that urgently need to be solved. Porous ceramsite could be applied for both water storage and water purification. This research used biomass and dredged silt to prepare water storage ceramsite (WSC), and investigated the adsorption and removal effects of WSC on phosphorus (P), nitrogen ((NH 4+ )N) and chemical oxygen demand (COD). The results showed that the biomass was mostly burned and partially carbonized during the high-temperature sintering process to form a rich pore structure inside the material. The rich pore structure effectively improved the water absorption to 105.58 %. The abundant specific surface area could provide many attachment sites, which is conducive to the adsorption of target ions by WSC. Further testing showed that WSC could adsorb ions with different charges in different pH solutions. Background: Osteonecrosis of femoral head (ONFH) is a pathological state caused by lack of blood supply in femoral head. This study aimed to explore the function of Lycium barbarum polysaccharide (LBP), an antioxidant agent extracted from L. barbarum, on ONFH., Methods: Osteonecrosis rat model was generated using lipopolysaccharide (LPS) and methylprednisolone followed by examination of body weight, blood glucose, morphology, and BMSC osteoblast differentiation. The effect and underlying mechanism of LBP on the proliferation, apoptosis, and osteoblast differentiation of BMSC were determined with or without LPS or hypoxia treatment using CCK-8. Alizarin Red S staining, flow cytometry, and western blot, respectively., Result: LBP could protect against glucocorticoid-induced ONFH in rats, resulting in improved sparse trabecular bone, empty lacunae and bone cell coagulation. Moreover, LBP promoted the proliferation and osteoblast differentiation of bone mesenchymal-derived stem cells (BMSCs) in a dose-dependent manner. Furthermore, LBP enhanced osteoblast differentiation of BMSCs under hypoxia condition. Mechanistically, we found that LBP treatment enhanced Runx2 and ALP expression in BMSCs. LBP restored the expression of Runx2 and ALP under hypoxia, suggesting that LBP might be involved in regulating Runx2/ALP expression and contributed to osteoblast differentiation. GPR65 (TDAG8) is a proton-sensing G protein-coupled receptor predominantly expressed in immune cells. Genome-wide association studies (GWAS) have identified GPR65 gene polymorphisms as an emerging risk factor for the development of inflammatory bowel disease (IBD). Patients with IBD have an elevated risk of developing colorectal cancer when compared to the general population. To study the role of GPR65 in intestinal inflammation and colitis-associated colorectal cancer (CAC), colitis and CAC were induced in GPR65 knockout (KO) and wild-type (WT) mice using dextran sulfate sodium (DSS) and azoxymethane (AOM)/DSS, respectively. Disease severity parameters such as fecal score, colon shortening, histopathology, and mesenteric lymph node enlargement were aggravated in GPR65 KO mice compared to WT mice treated with DSS. Elevated leukocyte infiltration and fibrosis were observed in the inflamed colon of GPR65 KO when compared to WT mice which may represent a cellular mechanism for the observed exacerbation of intestinal inflammation. In line with high expression of GPR65 in infiltrated leukocytes, GPR65 gene expression was increased in inflamed intestinal tissue samples of IBD patients compared to normal intestinal tissues. Moreover, colitis-associated colorectal cancer development was higher in GPR65 KO mice than WT mice when treated with AOM/DSS. Altogether, our data demonstrate that GPR65 suppresses intestinal inflammation and colitis-associated tumor development in murine colitis and CAC models, suggesting potentiation of GPR65 with agonists may have an anti-inflammatory therapeutic effect in IBD and reduce the risk of developing colitis-associated colorectal cancer., (Copyright © 2021 Elsevier B.V. Enucleated mammalian cells (cytoplasts) have been widely used for studying differential roles of the cytoplasm and nucleus in various cellular processes. Here, we reported an improved enucleation protocol, in which cells were seeded in extracellular matrix (ECM)-coated 24-wells and spun at 4600 g and 35 °C for 60 min in the presence of cytochalasin B and colchicine. When glass-bottom wells were used, cellular structures and organelles in cytoplasts could be examined directly by confocal microscopy. Nuclear envelope rupture did not occur probably due to mild centrifugation conditions used in this study. Addition of paclitaxel or doxorubicin completely blocked proliferation of residual nucleated cells; however, to our surprise, paclitaxel dramatically prolonged the survival of cytoplasts. Results from Annexin V and Propidium Iodide staining showed that cytoplasts died predominantly by apoptosis, which was partially inhibited by ECM and further by paclitaxel. Mitochondria were mostly rod-shaped and formed a connected network in paclitaxel-treated cytoplasts, indicating lack of fusion and fission dynamics. Moreover, paclitaxel increased mitochondrial membrane potential, suggesting that perturbation of mitochondria might be critical to the survival of cytoplasts. Pancreatic ductal adenocarcinoma (PDAC) is an aggressive, highly metastatic malignancy and accounts for 85% of pancreatic cancers. PDAC patients have poor prognosis with a five-year survival of only 5-10%. Mutations at the TP53 gene are readily detected in pancreatic tumors isolated from PDAC patients. We have investigated the effects of restoration of wild-type (WT) TP53 activity on the sensitivity of pancreatic cancer cells to: chemotherapy, targeted therapy, as well as, nutraceuticals. Upon introduction of the WT-TP53 gene into the MIA-PaCa-2 pancreatic cancer cell line, the sensitivity to drugs used to treat pancreatic cancer cells such as: gemcitabine, fluorouracil (5FU), cisplatin, irinotecan, oxaliplatin, and paclitaxel increased significantly. Likewise, the sensitivity to drugs used to treat other cancers such as: doxorubicin, mitoxantrone, and 4 hydroxy tamoxifen (4HT) also increased upon introduction of WT-TP53 into MIA-PaCa-2 cells. Furthermore, the sensitivity to certain inhibitors which target: PI3K/mTORC1, PDK1, SRC, GSK-3, and biochemical processes such as proteasomal degradation and the nutraceutical berberine as increased upon introduction of WT-TP53. Furthermore, in some cases, cells with WT-TP53 were more sensitive to the combination of drugs and suboptimal doses of the MDM2 inhibitor nutlin-3a. However, TP53-independent effects of nutlin-3a were observed upon treatment with either a proteasomal or a PI3K/mTOR inhibitor. GPR4 is a proton-sensing G protein-coupled receptor that can be activated by extracellular acidosis. It has recently been demonstrated that activation of GPR4 by acidosis increases the expression of numerous inflammatory and stress response genes in vascular endothelial cells (ECs) and also augments EC-leukocyte adhesion. Inhibition of GPR4 by siRNA or small molecule inhibitors reduces endothelial cell inflammation. As acidotic tissue microenvironments exist in many types of inflammatory disorders, including inflammatory bowel disease (IBD), we examined the role of GPR4 in intestinal inflammation using a dextran sulfate sodium (DSS)-induced acute colitis mouse model. We observed that GPR4 mRNA expression was increased in mouse and human IBD tissues when compared to control intestinal tissues. To determine the function of GPR4 in intestinal inflammation, wild-type and GPR4-deficient mice were treated with 3% DSS for 7days to induce acute colitis. Our results showed that the severity of colitis was decreased in GPR4-deficient DSS-treated mice in comparison to wild-type DSS-treated mice. Clinical parameters, macroscopic disease indicators, and histopathological features were less severe in the DSS-treated GPR4-deficient mice than the DSS-treated wild-type mice. Endothelial adhesion molecule expression, leukocyte infiltration, and isolated lymphoid follicle (ILF) formation were reduced in intestinal tissues of DSS-treated GPR4-null mice. TP53 is a critical tumor suppressor gene that regulates cell cycle progression, apoptosis, cellular senescence and many other properties critical for control of normal cellular growth and death. Due to the pleiotropic effects that TP53 has on gene expression and cellular physiology, mutations at this tumor suppressor gene result in diverse physiological effects. T53 mutations are frequently detected in numerous cancers. The expression of TP53 can be induced by various agents used to treat cancer patients such as chemotherapeutic drugs and ionizing radiation. Radiation will induce Ataxia telangiectasia mutated (ATM) and other kinases that results in the phosphorylation and activation of TP53. TP53 is also negatively regulated by other mechanisms, such as ubiquitination by ligases such as MDM2. While TP53 has been documented to control the expression of many "classical" genes (e.g., p21 Cip-1 , PUMA, Bax) by transcriptional mechanisms for quite some time, more recently TP53 has been shown to regulate microRNA (miR) gene expression. Different miRs can promote oncogenesis (oncomiR) whereas others act to inhibit tumor progression (tumor suppressor miRs). Targeted therapies to stabilize TP53 have been developed by various approaches, MDM2/MDM4 inhibitors have been developed to stabilize TP53 in TP53-wild type (WT) tumors. In addition, small molecules have been isolated that will reactivate certain mutant TP53s. Both of these types of inhibitors are in clinical trials. Objective: To reveal the characteristic and distribution of length of hospital stay (LOS) and direct hospitalisation costs of paediatric scald., Methods: A prospective case series observation was performed from January 2005 to December 2006 at the Burn Center, Changhai Hospital, Shanghai, China. The information, such as demographics, clinical diagnosis and treatments since admission, of the paediatric scald patients included in the series was recorded. The direct cost of a treatment event was recorded into the price system when it was incurred. All cost data were summarised on completion of the study. The distribution of LOS and the hospitalisation costs were recorded by gender, age, total burn area, depth of burn, blood transfusion and patterns of treatment. Mann-Whitney signed-rank test was used to assess the differences between continuous, non-normally distributed variables, and multiple linear regression was used to model LOS and direct hospitalisation costs. Statistical analyses were undertaken with SPSS 15.0 statistical software., Results: Patients aged 3 years or less accounted for more than half of the total LOS and hospitalisation costs, patients with burn area less than 10%TBSA (total burn surface area) accounted for more than 70% of the total LOS and more than half of the hospitalisation costs and patients with second-degree burn accounted for more than 78% of the total LOS and hospitalisation costs. Depth of burn, area of burn, patterns of treatment and blood transfusion were independent predictors of LOS; whereas LOS, area of burn and blood transfusion were independent predictors of hospitalisation costs., Conclusion: Paediatric scalds have particular characteristics in terms of distribution of LOS and direct hospitalisation costs and the factors influencing them. The data presented in this study should assist burn care practitioners and hospital epidemiologists estimate and compare the economic burden of paediatric burns at other institutions; it may also be useful in resource allocation and cost-effectiveness analysis of treatment versus prevention strategies.

G2A is a G-protein-coupled receptor (GPCR) involved in immune regulation. Previous studies have shown that lysophosphatidylcholine (LPC), a bioactive lipid associated with atherosclerosis and autoimmunity, acts through G2A to induce diverse biologic effects. Production of LPC during cell apoptosis serves as a chemotactic signal for macrophage recruitment. Here we demonstrate that macrophage chemotaxis to LPC is dependent on G2A function. Wild-type but not G2A-deficient mouse peritoneal macrophages migrated toward LPC. RNAi-mediated knockdown of G2A in J774A.1 macrophages abolished LPC-induced chemotaxis, whereas overexpression of G2A significantly enhanced this process. Mutation of the conserved DRY motif of G2A resulted in loss of chemotaxis to LPC, suggesting a requirement for G-protein signaling. Unlike most GPCRs, including the chemokine receptors, coupling to G(i) is not required for LPC/G2A-mediated chemotaxis, but coupling to G(q/11) and G(12/13) is necessary as judged by inhibition with dominant negative forms of these alpha subunits or with regulators of G-protein signaling (RGS) constructs. Collectively, these data establish that pertussis toxin-insensitive G2A signaling regulates macrophage chemotaxis to LPC. Defects in this signaling pathway may be related to the pathogenesis of systemic autoimmune disease.

We cloned a novel murine gene, designated Hemogen (hemopoietic gene), which was sequentially expressed in active hematopoietic sites and downregulated in the process of blood cell differentiation. Hemogen transcripts were specifically detected in blood islands, primitive blood cells and fetal liver during embryogenesis, and then remained in bone marrow and spleen in adult mice. Immunostaining demonstrated that Hemogen was a nuclear protein. We also identified a human homologue of Hemogen, named EDAG, which was mapped to chromosome 9q22, a leukemia breakpoint. Like Hemogen, EDAG exhibited specific expression in hematopoietic tissues and cells. Taken together, these data are consistent with Hemogen and EDAG playing an important role in hematopoietic development and neoplasms.
