Background: Breast cancer patients experience acute radiation dermatitis (ARD) during radiation therapy (RT). This study investigated the prophylactic effect of a newly developed xenogeneic platelet-rich plasma (PRP) lotion on ARD for breast cancer patients. Methods: This study enrolled patients with ductal carcinoma in situ and early-stage invasive breast cancers after breast-conserving surgery. Hypofractionated whole-breast RT (42.5 Gy in 16 fractions) followed by tumour bed boost (10 Gy in 5 fractions) was used. The patients were randomly assigned to XONRID® gel (n = 48) or PRP lotion (n = 52) groups. We recorded the skin toxicity weekly during RT and at 2 weeks after RT. ARD was graded on the basis of the RTOG definition by two senior radiation oncologists, and the numerical rating scale (NRS) for pain and Dermatology Life Quality Index (DLQI) were subjectively scored by patients. Results: Grade 3–4 ARD was noted in three (6 %) patients in the XONRID® gel group and no patients in the PRP lotion group (p < 0.001). One patient did not complete RT in the XONRID® gel group due to intolerable pain and refused to complete the weekly questionnaires and follow-ups. Compared with the XONRID® gel group, the PRP lotion group had significantly reduced and delayed progression of mean ARD (p = 0.001), lower mean NRS for pain value (p = 0.021) and lower mean DLQI (p = 0.048). Conclusions: This randomized controlled trial is the first to use xenogeneic PRP lotion for ARD prevention. This lotion has prophylactic effects against ARD and thus improves quality of life of patients undergoing RT.

Background/purpose: Oral cancer is one of the most prevalent malignant tumors in Taiwan. Due to the heterogeneity of oral cancer cells, the five-year survival rate of patients is only 50%. Post-translational modifications contribute to protein diversity and directly influence cell functions. The protein inhibitor of activated signal transducer and activator of transcription 2 (PIAS2) is known to undergo post-translational modifications, yet its impact on oral cancer remains unclear. Materials and methods: PIAS2 expression was modulated by transfecting cells with a PIAS2 expression vector or by knocking down PIAS2 using siRNA with low and high PIAS2 expression, respectively. These cells were subjected to invasion, migration, and proliferation assays to evaluate the effects of PIAS2. Changes in genotype, such as epithelial-mesenchymal transition (EMT) markers, were also examined. Additionally, the effect of cigarette smoke condensate (CSC) on PIAS2 expression in oral squamous cell carcinoma (OSCC) cells was investigated. Results: Overexpression of PIAS2 significantly increased the malignant behaviors of oral cancer cells. In YD38 and SAS cells with low PIAS2 expression, overexpression of PIAS2 enhanced proliferation, invasion, and migration. PIAS2 overexpression also affected EMT gene expression, suppressing E-cadherin and increasing fibronectin expression. Conversely, PIAS2 knockdown in OECM1 and SCC25 cells suppressed malignant behaviors and reversed EMT markers, increasing E-cadherin and decreasing fibronectin expression. Furthermore, a dose-dependent increase in PIAS2 expression was observed when OSCC cells were treated with CSC. Conclusion: PIAS2 functions as an oncogene in oral cancer, and cigarette smoking induces PIAS2 expression. Increased PIAS2 levels lead to enhanced malignancy in oral cancer.

The decline in eggshell quality with increasing hen age may be related to changes in ultrastructure and chemical composition, with matrix proteins playing key roles in these changes. However, research on blue-shelled eggs remains limited. This study investigated the effects of hen age (35, 55, 75, and 85 weeks) on the physical, mechanical, and chemical properties of eggshells in the Xinyang blue-shelled laying hens, as well as their ultrastructural and nanostructural characteristics. Subsequently, a comparative proteomic analysis was performed to elucidate the differential protein profiles in eggshells from hens at 35 and 85 weeks of age. Results showed that egg weight, eggshell weight, and eggshell surface area increased with hen age, whereas eggshell stiffness decreased (p < 0.05). As the age advanced, the eggshell organic matter content declined (p < 0.05). The effective layer ratio, mammillae density, as well as the porosity and total pore area in the mammillary layer also decreased with age, whereas the ratio of the mammillary layer increased (p < 0.05). Compared to eggshells collected from 35-week-old hens, those from 85-week-old hens showed increases in egg weight, eggshell weight, surface area, and both the ratio and thickness of the mammillary layer (p < 0.05). However, significant decreases were observed in eggshell stiffness, organic matter content, phosphorus content, effective layer ratio, mammillae density, as well as the porosity and total pore area in the mammillary layer (p < 0.05). Additionally, eggshell stiffness, phosphorus content, and organic matter content were significantly correlated with each other (p < 0.05). Proteomic analysis identified 37 downregulated and 68 upregulated differentially expressed proteins (DEPs, FC > 1.2 or < 0.83, with a p-value < 0.05) in eggshells from 85-week-old hens compared to those from 35-week-old hens. These DEPs are associated with functions such as biomineralization, calcium transport, immunity, and proteases and protease inhibitors. Mantel and Pearson correlations suggest that these functions may be involved in regulating eggshell stiffness, phosphorus content, and organic matter content. Overall, the eggshell stiffness decreased from 35 to 85 weeks of age, which may be attributed to the reductions in eggshell organic matter and phosphorus contents, as well as the deteriorations in eggshell ultrastructure. The proteins associated with biomineralization, calcium transport, immunity, and proteases and protease inhibitors may contribute to these changes.

Adipose tissue, an important organ involved in energy metabolism and endocrine, is closely related to animal meat quality and human health. Transient receptor potential channel 1 (TRPC1), an ion transporter, is adipocytes’ major Ca2+ entry channel. However, its function in fat deposition is poorly understood, particularly in pigs, which are both an ideal model for human obesity research and a primary meat source for human diets. In the present investigation, our findings demonstrate a prominent expression of TRPC1 within the adipose tissue of pigs with a strong fat deposition ability. Functional analysis showed that TRPC1 promotes primary preadipocyte proliferation and adipogenic differentiation. In vivo, transgenic mice expressing porcine TRPC1 exhibited aggravated high-fat diet–induced obesity, hepatic steatosis, and insulin resistance. Moreover, TRPC1 may facilitate adipogenesis via activating phosphatidylinositol 3 kinase/AKT and β-catenin signaling pathways. Our research underscores the pivotal role of porcine TRPC1 as a positive regulator in adipogenesis and lipid accumulation processes, providing a potential target for improving animal meat quality and treating obesity-related diseases in humans.

Background: This investigation quantifies the mean and median hearing thresholds and assesses the prevalence of age-related hearing loss within the senior population of Taipei. Methods: In a substantive geriatric assessment supported by government initiative, 1696 individuals from a community hospital partook in this cross-sectional study (2016–2018). Detailed audiometric evaluations logged pure-tone thresholds across critical frequencies (0.5k, 1k, 2k, 4k Hz), in conjunction with participant ages, genders, and HHIE-S questionnaire results. Results: The findings indicated mean and median pure tone averages (PTA) of 25.3 ± 15.6 dB HL and 22.5 dB HL, respectively. Gender-based analysis showcased higher PTAs for males than females. The study illuminated a progressive increase in hearing thresholds with age and higher frequencies. A 41 % prevalence of hearing impairment was recorded, with a notable 15.2 % deemed disabling, and a potential candidacy for cochlear implantation in 1.5 % of the study group. Conclusions: Hearing loss was prevalent in 41 % of the non-hospitalized elderly demographic, chiefly characterized as mild. However, older age groups, particularly those over 85, presented an elevated occurrence of moderate hearing loss.

Sheep and goat meat products are becoming increasingly popular among consumers due to their unique flavor derived from intramuscular fat (IMF), which contributes to formation of the distinctive odor. However, there is currently a dearth of reviews on the impact of IMF on the flavor of sheep and goat meat. The present review aims to discuss the relationships between IMF and flavor through lipid composition and fatty acid (FA) distribution, provide an overview of characteristic flavor compounds affecting the flavor of sheep and goat meat, and shed light on the impacts of pre-mortem and post-mortem factors on meat flavor attributed to changes in FAs and flavor compounds. Controlling pre-mortem practices and adjusting post-mortem harvesting methods are key factors in shaping and/or driving the flavor of sheep and goat meat products. This review enhances the comprehensive understanding of the impact of IMF on the flavor of sheep and goat meat.

To investigate the impacts of glycation modification on the structure and flavor of fish viscera-derived protein hydrolysates, channel catfish viscera was utilized and hydrolyzed by Flavourzyme, followed by glycation with glucosamine, xylose, ribose and glucose, respectively. The structural characteristics, taste and odor of glycated products were evaluated. The results revealed that glycation led to an increase in peptides (5 kDa). UV and fluorescence spectra indicated a gradual rise in absorbance and fluorescence intensity. Glucosamine-induced glycation significantly mitigated bitterness, while enhancing saltiness and umami. GC–MS/MS analysis revealed that glycation led to reduced undesirable odor, and formation of aromatic compounds. LC-MS/MS identified Arg and Lys as the main modification sites. Interestingly, aldehydes were found to modify peptides via carbonyl-amine reaction, leading to decreased fishy odor. The present study contributes to flavor improvement of fish viscera-derived peptides through glycation and high-value utilization of fish viscera.

A 73-day feeding trial was conducted to determine the effects of the freshness of poultry by-product meal on the growth performance, immune response and hepatopancreatic health of giant freshwater prawn, Macrobrachium rosenbergii. Three semi-purified diets were formulated containing 2.5 % poultry by-product meal with different TVB-N value, designated as L group (untreated poultry by-product meal, TVB-N 10 mg/100 g, i.e., dietary TVB-N 55 mg/100 g), M group (TVB-N 300 mg/100 g, i.e., dietary TVB-N 60 mg/100 g), and H group (TVB-N 450 mg/100 g, i.e., dietary TVB-N 68 mg/100 g), and fed to three groups of prawns (initial weight: 0.82 ± 0.01 g), respectively. Results showed that the weight gain rate (WGR) and specific growth rate (SGR) of prawns significantly decreased with the TVB-N value increased (P < 0.05). The survival rate (SR) and hepatosomatic index (HSI) of prawns in group M and group H were significantly decreased (P < 0.05), while the feed conversion ratio (FCR) was significantly increased (P < 0.05). In addition, the lipase (LPS) and α-amylase (α-AMS) activity of prawns in group L was significantly higher than that in group H (P < 0.05). With the poultry by-product meal TVB-N value increased, the activity of total superoxide dismutase (T-SOD) and glutathione S-transferase (GST) was significantly decreased. RT-qPCR results showed that the expression of bok in group H was significantly higher than that in group L and group M (P < 0.05), the expressions of caspase3, caspase9, bax, and nf-κb in group H were significantly higher than that in group L (P < 0.05). High temperature stress and ammonia nitrogen stress significantly decreased the cumulative survival rate of prawns in group M and group H. Conclusively, it is necessary to guarantee the freshness of the ingredients in prawn formulated diets, and the TVB-N value of diets is recommended not to exceed 60 mg/100 g.

Purpose: This study aimed to establish a nomogram combining 31-gene signature (31-GS), radiosensitivity index (RSI), and radiation-resistance-related gene index (RRRI) to predict recurrence in prostate cancer (PCa) patients. Methods and Materials: Transcriptome data of PCa were obtained from gene expression omnibus and the cancer genome atlas to validate the predictive potential of 3 sets of published biomarkers, namely, 31-GS, RSI, and RRRI. To adjust these markers for the characteristics of PCa, we analyzed 4 PCa-associated radiosensitivity predictive indices based on 31-GS, RSI, and RRRI by the Cox analysis and least absolute shrinkage and selection operator regression analysis. Time-dependent receiver operating characteristic curves, decision curve analyses, integrated discrimination improvement, net reclassification improvement and decision tree model construction were used to compare the radiosensitivity predictive ability of these 4 gene signatures. Key modules and associated functions were identified through a weighted gene co-expression network analysis and gene function enrichment analysis. A nomogram was built to improve the recurrence-prediction capability. Results: We validated and compared the predictive potential of 2 published predictive indices. Based on the 31-GS, RSI, and RRRI, we analyzed 4 PCa-associated radiosensitivity predictive indices: 14Genes, RSI, RRRI, and 20Genes. Among them, 14Genes showed the most promising predictive performance and discriminative capacity. Genes in the key module defined by the 14Genes model were significantly enriched in radiation therapy-related cell death pathways. The area under receiver operating characteristic curve and decision tree variable importance for 14Genes was the highest in the cancer genome atlas and Gene Expression Omnibus Series (GSE) cohorts. Conclusions: This study successfully established a radiosensitivity-related nomogram, which had excellent performance in predicting recurrence in patients with PCa. For patients who received radiation therapy, the 20Genes and RRRI models can be used to predict recurrence-free survival, whereas 20Genes is more radiation therapy-specific but needs further external validation.

This study aimed to modify plant protein mixture to improve their functionality and digestibility by limited hydrolysis. Soy protein isolate and corn zein were mixed at the ratio of 5:1 (w/w), followed by limited hydrolysis using papain from 15 to 30 min. The structural characteristics, in vitro digestibility, and functional properties were evaluated. Also, DPPH radical scavenging activity was determined. The results indicated that the molecular weight of different modified samples was largely reduced by limited hydrolysis, and the proportion of random coil was significantly increased. Furthermore, the solubility, foaming, emulsifying and water-holding capacity of hydrolyzed protein mixture were significantly improved, which were close to those of whey protein isolate. In vitro digestibility after 30-min limited hydrolysis was remarkably elevated. In addition, the hydrolyzed protein mixture exhibited a higher antioxidant activity than those of untreated proteins. Overall, limited hydrolysis of protein mixture led to improved digestibility, functionality and antioxidant activity.

This study assessed the impacts of monosodium glutamate (MSG) on the growth performance, feed utilization, feed intake, and appetite regulation of Chinese mitten crab, Eriocheir sinensis. A total of 240 juvenile crabs (9.16 ± 0.21 g) were assigned to six experimental diets supplemented with MSG at 0 %, 0.20 %, 0.39 %, 0.59 %, 0.78 % and 0.98 %. The results demonstrated that 0.59 % MSG supplementation significantly enhanced the weight gain rate (WGR), specific growth rate (SGR), and feed utilization of juvenile E. sinensis by increasing feed intake (FI) (p < 0.05). Moreover, dietary moderate MSG improved the mid-intestine morphology, such as height of microvillus (HMV), width of microvillus (WMV), and thickness of muscularis (TM). The qRT-PCR analysis demonstrated that the mRNA expression levels of the appetite-stimulating neural signals agouti related protein (agrp) and ghrelin were increased significantly by dietary moderate MSG (p < 0.05). Conversely, the mRNA levels of protein kinase B (akt), target of rapamycin (tor) and 4E-binding protein (4e-bp), as well as appetite-suppressing neural signals pro-opiomelanocortin (pomc), corticotropin-releasing factor (crf) and peptide yy (pyy) were significantly down-regulated (p < 0.05). The two-slope broken-line regression analysis of SGR in relation to dietary MSG levels suggested that the optimum dietary MSG level for juvenile E. sinensis was 5.6 g/kg. Collectively, MSG may potentially stimulate or suppress the appetite regulation neural signals/hormones, thereby modulating the feeding behavior of E. sinensis.

The antitumor activity of a fermented juice (called Shaduojin, SDJ) made from sea buckthorn, raspberry, etc. was investigated using CT26 bearing mice. The results showed a dose-dependent inhibition of tumor growth in the SDJ groups compared to the model group. The number of CD8+ T and dendritic cells were increased while the Treg cells and monocytes was decreased in both the tumor and spleen in the SDJ groups. The TNF-α and INF-γ increased and the IL-1β, IL-6, and IL-10 decreased in the SDJ groups. Additionally, there was an increase in the abundance of beneficial bacteria (Firmicutes, Coildextribacter, and Lachnospiraceae) which contribute to intestinal health and produce short-chain fatty acids (SCFAs). On the other hand, harmful bacteria (Muribaculaceae and uncultured Bacteroidales) decreased. The SCFA content in the intestine also increased in the SDJ groups. Therefore, it has been showed that SDJ may elicit its effect on preventing colorectal cancer by regulating the immune system and gut microbiota.

During freeze–thaw cycling, aggregates undergo a dynamic change in breakdown–formation (turnover), however, how the turnover occurs between aggregates of various particle sizes is not clear. To clarify the influence of freeze–thaw cycling on the dynamic changes in the particle size of soil aggregates, soil aggregates from the Black Soil Region of Northeast China were selected as the research objects. The study conducted in situ dynamic monitoring experiments, innovatively applying the rare earth oxide (REO) tracer method to natural conditions of freeze–thaw cycles (autumn freeze–thaw period, freezing period, and spring freeze–thaw period), accurately tracking the turnover paths and quantifying the turnover rates between aggregates of various particle sizes. The results revealed that the total value of the formation paths of the 2–5 mm aggregates and 0.25–2 mm aggregates increased during the autumn freeze–thaw period. The number of freeze–thaw cycles and accumulated snowfall were significantly positively correlated with aggregate stability, with an increase in the number of freeze–thaw cycles and accumulated snowfall resulting in an increase in the proportion of aggregates > 0.25 mm, which improved aggregate stability. In addition, the total value of the breakdown path of macro-aggregates increased during the spring freeze–thaw cycling period. Soil moisture was significantly negatively correlated with aggregate stability, with increased soil moisture resulting in a decrease in the percentage of aggregates > 0.25 mm, which resulted in a decrease in aggregate stability. The study can provide a reference understanding for the effects of freeze–thaw cycles on the structure of black soil and provide a theoretical basis for improving the quality of arable land.

The influences of simultaneous precipitation of isothermal ω and O′ on deformation mechanisms and mechanical properties of near β Ti alloy Ti-5Al-3Mo-3V-2Cr-2Zr-1Nb-1Fe with (α + β) hybrid structure are revealed. During continuous heating, the phase transformation sequence in this alloy is β + αp + ω-embryos + O′ → β + αp + ωiso + O′ → β + αp + ωiso + O′ + αs → β + αp + αs. Low-temperature heat treatments from 200 °C to 400 °C are carried out to develop such intermediate phases. Upon aging at 200 °C and 300 °C, the isothermal ω and O′ domains are continuously generated. In comparison with solution state, the deformation behaviors of aged samples are altered, and the yield stress and ductility are simultaneously increased. When aged at 200 °C, the yield strength is increased by 13.8 %, and fracture elongation is increased by 63.2 %. When aged at 300 °C, the yield strength is increased by 18.2 %, and preserves large ductility same to solution state. This is due to the development of isothermal ω and O′ domains suppress long-range stress induced martensitic transformation during loading, and the deformation mechanisms evolve into dislocations slipping gradually. When the aging temperature increases to 400 °C, α precipitation takes place leading to an obvious decrement in ductility. Upon loading the primary α can also accommodate partly the strains and relax local stress concentration. The results in this work clearly proves that better mechanical properties can be achieved through tailoring intermediate phases generation.

Intratumoral microbiota has become research hotspots, and emerges as a non-negligent new component of tumor microenvironments (TME), due to its powerful influence on tumor initiation, metastasis, immunosurveillance and prognosis despite in low-biomass. The accumulations of microbes, and their related components and metabolites within tumor tissues, endow TME with additional pluralistic features which are distinct from the conventional one. Therefore, it's definitely necessary to comprehensively delineate the sophisticated landscapes of tumor microbe microenvironment, as well as their functions and related underlying mechanisms. Herein, in this review, we focused on the fields of tumor microbe microenvironment, including the heterogeneity of intratumor microbiota in different types of tumors, the controversial roles of intratumoral microbiota, the basic features of tumor microbe microenvironment (i.e., pathogen-associated molecular patterns (PAMPs), typical microbial metabolites, autophagy, inflammation, multi-faceted immunomodulation and chemoresistance), as well as the multidisciplinary approach-based intervention of tumor microbiome for cancer therapy by applying wild-type or engineered live microbes, microbiota metabolites, antibiotics, synthetic biology and rationally designed biomaterials. We hope our work will provide valuable insight to deeply understand the interplay of cancer-immune-microbial, and facilitate the development of microbes-based tumor-specific treatments.

Primary graft dysfunction (PGD) is a severe form of acute lung injury resulting from lung ischemia/reperfusion injury (I/R) in lung transplantation (LTx), associated with elevated post-transplant morbidity and mortality rates. Neutrophils infiltrating during reperfusion are identified as pivotal contributors to lung I/R injury by releasing excessive neutrophil extracellular traps (NETs) via NETosis. While alveolar macrophages (AMs) are involved in regulating neutrophil chemotaxis and infiltration, their role in NETosis during lung I/R remains inadequately elucidated. Extracellular histones constitute the main structure of NETs and can activate AMs. In this study, we confirmed the significant involvement of extracellular histone-induced M1 phenotype of AMs (M1-AMs) in driving NETosis during lung I/R. Using secretome analysis, public protein databases, and transwell co-culture models of AMs and neutrophils, we identified Cathepsin C (CTSC) derived from AMs as a major mediator in NETosis. Further elucidating the molecular mechanisms, we found that CTSC induced NETosis through a pathway dependent on NADPH oxidase-mediated production of reactive oxygen species (ROS). CTSC could significantly activate p38 MAPK, resulting in the phosphorylation of the NADPH oxidase subunit p47phox, thereby facilitating the trafficking of cytoplasmic subunits to the cell membrane and activating NADPH oxidase. Moreover, CTSC up-regulated and activated its substrate membrane proteinase 3 (mPR3), resulting in an increased release of NETosis-related inflammatory factors. Inhibiting CTSC revealed great potential in mitigating NETosis-related injury during lung I/R. These findings suggests that CTSC from AMs may be a crucial factor in mediating NETosis during lung I/R, and targeting CTSC inhition may represent a novel intervention for PGD in LTx.

The degradation of cyanobacterial blooms releases hazardous contaminants such as microcystin-LR (MC-LR) and nitrite, which may collectively exert toxicity on various bodily systems. To evaluate their individual and combined toxicity in the kidney, mice were subjected to different concentrations of MC-LR and/or nitrite over a 6-month period in this study. The results revealed that combined exposure to MC-LR and nitrite exacerbated renal pathological alterations and dysfunction compared to exposure to either compound alone. Specifically, the protein and mRNA expression of kidney injury biomarkers, such as kidney injury molecule 1 (KIM-1) and neutrophil gelatinase-associated lipocalin (NGAL), were notably increased in combined exposure group. Concurrently, co-exposure to MC-LR and nitrite remarkedly upregulated levels of proinflammatory cytokines TNF-α, IL-6 and IL-1β, while decreasing the anti-inflammatory cytokine IL-10. Notably, MC-LR and nitrite exhibited synergistic effects on the upregulation of renal IL-1β levels. Moreover, MC-LR combined with nitrite not only elevated mRNA levels of proinflammatory cytokines but also increased protein levels of pyroptosis biomarkers such as IL-1β, Gasdermin D (GSDMD), and Cleaved-GSDMD. Mechanistic investigations revealed that co-exposure to MC-LR and nitrite promoted pyroptosis both in vivo and in vitro, possibly through the activation of the TLR4/NLRP3/GSDMD pathway. Pretreatment with TLR4 inhibitor and NLRP3 inhibitor effectively suppressed pyroptosis induced by the co-exposure of these two toxins in HEK293T cells. These findings provide compelling evidence that MC-LR combined with nitrite synergistically induces pyroptosis in the kidney by activating the TLR4/NLRP3/GSDMD pathway. Overall, this study significantly enhances our comprehension of how environmental toxins interact and induce harm to the kidneys, offering promising avenues for identifying therapeutic targets to alleviate their toxic effects on renal health.

ABSTRACT: Mercuric chloride (HgCl2) is a nephrotoxic contaminant that is widely present in the environment. Selenium (Se) can effectively antagonize the biological toxicity caused by heavy metals. Here, in vivo and in vitro models of Se antagonism to HgCl2-induced nephrotoxicity in chickens were established, with the aim of exploring the specific mechanism. Morphological observation and kidney function analysis showed that Se alleviated HgCl2-induced kidney tissue injury and cytotoxicity. The results showed that ferroptosis was the primary mechanism for the toxicity of HgCl2, as indicated by iron overload and lipid peroxidation. On the one hand, Se significantly prevented HgCl2-induced iron overload. On the other hand, Se alleviated the intracellular reactive oxygen species (ROS) levels caused by HgCl2. Subsequently, we focused on the sources of ROS during HgCl2-induced ferroptosis. Mechanically, Se reduced ROS overproduction induced by HgCl2 through mitochondrial calcium uniporter (MCU)/mitochondrial calcium uptake 1 (MICU1)-mediated mitochondrial calcium ion (Ca2+) overload. Furthermore, a dual luciferase reporter assay demonstrated that MICU1 was the direct target of miR-202-5p. Overall, Se represses miR-202-5p/MICU1 axis to attenuate HgCl2-induced kidney ferroptosis.

A fractal structure featuring symmetric/asymmetric airfoil fins is embedded within the smooth channels of the printed circuit heat exchanger (PCHE), significantly enhancing its heat transfer performance. In this study, a horizontally installed two-layer channel configuration is examined, with hot CO2 flowing through the upper channel and cold CO2 through the lower channel. Two distinct airfoil fin designs, namely NACA 0021 symmetric and NACA 4822 asymmetric, are tested for their channel reinforcement effect. The numerical analysis reveals that channels equipped with these fins exhibit substantially improved heat transfer compared to smooth channels. Notably, the comprehensive evaluation index for the asymmetric airfoil fin channel is enhanced by a factor ranging from 1.05 to 3.76. Moreover, despite a modest increase in pressure drop, indicated by a dimensionless friction factor of 0.67–1.31, the heat transfer performance remains superior. Crucially, optimal heat transfer does not rely solely on high fluid flow rates but instead demands an apt flow matching between the hot and cold fluid channels.

Ultraviolet B (UVB) light exposure accelerates skin photoaging. Human adipose-derived stem cell exosomes (hADSC-Exos) and some antioxidants may have anti-photoaging effects. However, it is unknown whether the combination of hADSC-Exos and antioxidants plays a synergistic role in anti-photoaging. In cellular and 3D skin models, we showed that vitamin E (VE) and hADSC-Exos were optimal anti-photoaging combinations. In vivo, VE and hADSC-Exos increased skin tightening and elasticity in UVB-induced photoaging mice Combined treatment with VE and hADSC-Exos inhibited SIRT1/NF-κB pathway. These findings contribute to the understanding of hADSC-Exos in conjunction with other antioxidants, thereby providing valuable insights for the future pharmaceutical and cosmetic industries.

Natural products (NPs) play an irreplaceable role in the intervention of various diseases and have been considered a critical source of drug development. Many new pharmacodynamic compounds with potential clinical applications have recently been derived from NPs. These compounds range from small molecules to polysaccharides, polypeptides, proteins, self-assembled nanoparticles, and extracellular vesicles. This review summarizes various active substances found in NPs. The investigation of active substances in NPs can potentiate new drug development and promote the in-depth comprehension of the mechanism of action of NPs that can be beneficial in the prevention and treatment of human diseases.

To investigate the fatigue behaviour of stainless steel (SS) reinforcement, monotonic tensile and cyclic fatigue loading tests on 1.4362 duplex SS bars with diameters of 12 mm, 16 mm, and 20 mm are conducted. To keep the characteristics of reinforcement used in engineering construction, the specimens remain unprocessed. Two loading schemes for cyclic tests are considered in this work: (i) constant strain-amplitude, and (ii) variable strain-amplitude. The Ramberg-Osgood model was used to fit the experimental results. The strain-based and energy-based fatigue life equations are obtained. The strain-based fatigue life equation is compared with the present fatigue life estimation equations. Moreover, the ReinforcingSteel material model in OpenSees is calibrated with the experimental data. The results show that SS bars under cyclic experience hardening followed by softening. The bar diameter does not significantly affect the fatigue life prediction. The modified fatigue life equation suggests that SS bars have better fatigue resistance than conventional steel (CS) bars. The numerical analysis indicates that the calibrated steel material model (i.e. ReinforcingSteel) can produce a good simulation of SS bars under cyclic loading. It can improve the accuracy of numerical models of concrete components reinforced by SS bars.

With the rapid development of immunotherapy in recent years, cytokine storm has been recognized as a common adverse effect of immunotherapy. The emergence of COVID-19 has renewed global attention to it. The cytokine storm's inflammatory response results in infiltration of large amounts of monocytes/macrophages in the lungs, heart, spleen, lymph nodes, and kidneys. This infiltration leads to secondary tissue damage, acute respiratory distress syndrome (ARDS), organismal damage, and even death. However, there is currently no designated treatment for cytokine storm and the resulting ARDS. Consequently, there is a pressing need to identify a pharmaceutical agent that can effectively mitigate cytokine storms. Ebosin is a new exopolysaccharide generated by Streptomyces sp.139 and pharmacological activity for cytokine storm is investigated in vivo. The results show that Ebosin significantly augments the survival rates of mice, and its effectiveness increases with higher doses. It significantly inhibited the expression of cytokines IL-5, IL-6, IL-9 and chemokine Eotaxin in serum and lung tissues. Ebosin can alleviate the pathological damage in the lungs, liver, and spleen caused by LPS. Additionally, it can inhibit the phosphorylation of IKKα/β, Stat3 and NF-κB p65 upon LPS stimulation in vitro. We hypothesized that Ebosin may decrease cytokine release by inhibiting the phosphorylation of IKKα/β, Stat3, and NF-κB p65, neutrophil infiltration in animals. The article preliminarily elucidated the activity and mechanism of Ebosin against cytokine storm, which provides a reference for the study of anti-cytokine storm activity of microbial natural products.

A comprehensive study of the overall flavor and taste profile of different radishes is lacking. This study systematically compared the volatile profile of six radish varieties using HS-GC-IMS and their correlation with the E-nose analysis. Organic acids and amino acids were quantified, and their association with the E-tongues analysis was explored. A total of 73 volatile compounds were identified, with diallyl sulfide and dimethyl disulfide being the primary sulfides responsible for the unpleasant flavor in radish. Compared to other varieties, cherry radishes boast a significantly higher concentration of allyl isothiocyanate, which likely contributes to their characteristic radish flavor. Moreover, oxalic acid was identified as the most abundant organic acid in radish, accounting for over 97% of its content, followed by malic acid and succinic acid. In conclusion, the distinct flavor and taste characteristics of different radish varieties partially explain their suitability for diverse culinary preferences.

The purpose of the present work was to encapsulate zingerone (a bioactive compound from ginger) by self-assembling peptides derived from fish viscera. The encapsulation conditions were investigated and the structure of fish peptides-zingerone complex was characterized. The interaction between zingerone and fish peptides was investigated using fluorescence spectroscopy. Further research was performed on the in vitro release of zingerone and fish peptide-zingerone as well as their antiproliferative effects on colon epithelial Caco-2 cells. The results demonstrated that zingerone can be successfully encapsulated by self-assembling peptides derived from fish viscera with high encapsulation efficiency and loading capacity. Furthermore, transmission electron microscope and confocal laser scanning microscope observations revealed the successful encapsulation of zingerone by fish viscera peptides. In addition, in vitro release and antiproliferative activity against Caco-2 cells can be significantly increased by encapsulating zingerone via peptide self-assembly. The current study advances knowledge of encapsulation of bioactive compounds through peptide self-assembly.

With the development of integrated energy systems, the system energy demands become more complicated and the renewable energy proportion becomes higher. Under this background, integrated energy systems gradually present a state of high randomness and strong uncertainty, which affects the system collaborative optimization. In order to handle the multiple uncertainties’ effects effectively, multi-objective optimization considering uncertainties of energy demand and renewable energy using information gap decision theory (IGDT) method was carried out. By analyzing the optimization results and comparing the results of different weight coefficient of multiple uncertainties, the renewable energy uncertainty has a strong effect, while the energy demand uncertainty’s effect is more complex.

Off-grid or grid-connected operation of large-scale photovoltaic system may cause stability problems. When the system performs off-grid operation, energy storage, as balance node, regulates power and frequency, then also influences stability. Under grid-connected operation, the external power system balances power and frequency regulation, and energy storage only equilibrium wave power. Although a detailed model for a single photovoltaic generation system has been established, the connection of numerous photovoltaic units is difficult to model and simulate, and it can easily cause dimension disaster. In order to analyze stability effectively of off-grid/grid-connected operation of large-scale photovoltaic system, we first establish the equivalent model of a photovoltaic farm through the method of dynamic aggregation of wind turbines, and then obtain indicators affecting the stability considering the reactance matrix of the transmission line. When the real part of the oscillation mode sensitivity index is greater than zero, oscillation mode approaches the imaginary axis, indicating that the PV system stability deteriorates. Conversely, the PV system is regionally stable. Finally, the effects of the number of photovoltaic units and transmission line length on stability are theoretically analyzed, and the theoretical derivation is validated through simulations. More PV units and a longer transmission line increase the likelihood of instability through the sensitivity of the oscillation mode.

Alginate is the most abundant polysaccharide compound in brown algae, which is widely used in various fields. At present, the determination of the content of alginate is mostly carried out using sulfuric acid and trifluoroacetic acid hydrolysis followed by the determination of the content, but the results are not satisfactory, and there are problems such as low hydrolysis degree and low recovery rate. Therefore, in this study, based on the optimization of high performance liquid chromatographic conditions for pre-column derivatization of 1-phenyl-3-methyl-5-pyrazolone (PMP), the hydrolysis effects of sulfuric acid, trifluoroacetic acid (TFA), oxalic acid, and formic acid were compared and the hydrolysis conditions were optimized. The results showed that formic acid was the best hydrolyzing acid. The optimal hydrolysis conditions were 95 % formic acid at 110 °C for 10 h. The hydrolysis effect was stable, with high recovery and low destruction of monosaccharides, which made it possible to introduce formic acid into the subsequent polysaccharide hydrolysis. The pre-column derivatization high performance liquid chromatography method established in this study was accurate and reliable, and the hydrolysis acid with better effect was screened, which provided a theoretical basis for the subsequent determination of alginate content.

Summary: Background: Recurrence following radical resection in patients with stage IB gastric cancer (GC) is not uncommon. However, whether postoperative adjuvant chemotherapy could reduce the risk of recurrence in stage IB GC remains contentious. Methods: We collected data on 2110 consecutive patients with pathologic stage IB (T1N1M0 or T2N0M0) GC who were admitted to 8 hospitals in China from 2009 to 2018. The survival of patients who received adjuvant chemotherapy was compared with that of postoperative observation patients using propensity score matching (PSM). Two survival prediction models were constructed to estimate the predicted net survival gain attributable to adjuvant chemotherapy. Findings: Of the 2110 patients, 1344 received adjuvant chemotherapy and 766 received postoperative observation. Following the 1-to-1 matching, PSM yielded 637 matched pairs. Among matched pairs, adjuvant chemotherapy was not associated with improved survival compared with postoperative observation (OS: hazard ratio [HR], 0.72; 95% CI, 0.52–1.00; DFS: HR, 0.91; 95% CI, 0.64–1.29). Interestingly, in the subgroup analysis, reduced mortality after adjuvant chemotherapy was observed in the subgroups with elevated serum CA19-9 (HR, 0.22; 95% CI, 0.08–0.57; P = 0.001 for multiplicative interaction), positive lymphovascular invasion (HR, 0.32; 95% CI, 0.17–0.62; P

There are two cultivated and weedy types of Perilla crop (TCWTPC), and they are widely distributed and cultivated in East Asia, especially in South Korea and Japan. The objective of this study is to create simple sequence repeat (SSR) markers linked to morphological traits that show differences between accessions of the TCWTPC using recently designed SSR primer sets in Perilla crop. Genetic diversity within 52 accessions of the TCWTPC, gathered from South Korea, was assessed using 28 novel Perilla SSR primer sets. Based on the assessment, a collection of 28 Perilla SSR primer sets were shown to exhibit polymorphism and yielded a total of 142 alleles across the 52 accessions of the TCWTPC. Through inspection of a phylogenetic tree and population structure, the 52 accessions of the TCWTPC were classified into three major groups. Although most accessions of the TCWTPC were relatively clearly distinguished, SSR markers failed to distinguish several accessions belonging to the two weedy types of the Perilla crop. By using an association mapping analysis (AMA) of the 28 Perilla SSR markers and seven morphological characteristics in the 52 TCWTPC accessions, we detected that three of the Perilla SSR markers (KNUPF134, KNUPF137, KNUPF149) were associated with plant and seed characteristics. The novel SSR primer sets developed in Perilla crop should be useful in AMA for assessing genetic diversity and relationships between and within TCWTPC accessions, and this information will be helpful for genetic mapping in breeding programs for Perilla crop.

The decomposition of maize residues has a vital influence on soil aggregate buildup–breakdown (turnover) processes, but the following remain unclear: i) which size aggregates of Mollisols are primarily affected by the decomposition of maize residues and ii) the turnover proportions of these aggregates. In the study, rare earth oxides (REOs) were used to trace and quantify the buildup–breakdown paths of soil aggregate fractions during the whole incubation time. The two treatments were designed as follows: i) REO-labelled soil with maize residues addition (maize residue treatment) and ii) REO-labelled soil without maize residues addition (control, CK). The soil respiration, aggregate size distribution, REO concentrations, SOC and pH were measured during 84 days of incubation. The results showed that on days 0–14, the incorporation of maize residues significantly increased soil respiration and promoted the formation of 2–5 mm and 0.25–2 mm aggregate fractions at the expense of

Background: More and more novel anticancer drugs have been approved for patients with hematological malignancies in recent years, but HBV reactivation (HBV-R) data in this population is very scarce. This study aimed to evaluated HBV-R risk in patients with hematological malignancies receiving novel anticancer drugs. Methods: HBV markers and serum HBV DNA levels of patients with hematological malignancies receiving novel anticancer drugs in a tertiary cancer hospital were retrospectively collected. HBV-R risk in the whole cohort and subgroups was described. The relevant literature was reviewed to make a pooled analysis. Results: Of 845 patients receiving novel anticancer drugs, 258 (30.5%) were considered at risk for HBV-R. The median duration of exposure to novel drugs was 5.6 (0.1–67.6) months. The incidence of HBV-R was 2.1% in patients with past HBV infection without prophylactic antiviral treatment (PAT) and 1.2% in all patients at risk of HBV-R. In a pooled analysis of 11 studies with 464 patients, the incidence of HBV-R was 2.4% (95% CI: 1.3–4.2) in all at-risk patients receiving novel anticancer drugs and 0.6% (95% CI: 0.03–3.5) in patients with anticancer drugs plus PAT. The incidence of death due to HBV-R was 0.4% (95% CI: 0.1–1.6) in all at-risk patients and 18.2% (95% CI: 3.2–47.7) in patients with HBV-R. Conclusion: Most episodes of HBV-R are preventable, and most cases with HBV-R are manageable. We recommend that novel anticancer drugs should not be intentionally avoided when treating cancer patients with HBV infection.

Spinal muscular atrophy (SMA) is a neurodegenerative disease characterized by the selective loss of spinal motor neurons (MNs) and concomitant muscle weakness. Mutation of SMN1 is known to cause SMA, and restoring SMN protein levels via antisense oligonucleotide treatment is effective for ameliorating symptoms. However, this approach is hindered by exorbitant costs, invasive procedures, and poor treatment responses of some patients. Here, we seek to circumvent these hurdles by identifying reliable biomarkers that could predict treatment efficacy. We uncovered that MiR34 exhibits consistent downregulation during SMA progression in both human and rodent contexts. Importantly, Mir34 family-knockout mice display axon swelling and reduced neuromuscular junction (NMJ) endplates, recapitulating SMA pathology. Introducing MiR34a via scAAV9 improved the motor ability of SMNΔ7 mice, possibly by restoring NMJ endplate size. Finally, we observed a consistent decreasing trend in MiR34 family expression in the cerebrospinal fluid (CSF) of type I SMA patients during the loading phase of nusinersen treatment. Baseline CSF MiR34 levels before nusinersen injection proved predictive of patient motor skills 1 year later. Thus, we propose that MiR34 may serve as a biomarker of SMA since it is associated with the pathology and can help evaluate the therapeutic effects of nusinersen.

To meet the demand for the track's geometric parameter detection equipment for train speed and high-speed aerodynamics tests, a zero-calibration gauge device is designed with the centering limit, height adjustment and horizontal display function in this paper. The bending situation of the zero-calibration gauge is analyzed and the processing technology is studied, which ensures the rationality and realizability of the design of zero-calibration gauge. Then the gauge zero value and the parallelism of the working surface of zero-calibration gauge have been experimentally tested. The experimental results show that the parameter of gauge zero value is 1434.829 mm with a standard deviation of 1.4 μm. The parallelism of the two upper working surfaces is 1.1 μm, and the parallelism of the two inner working surfaces is 4 μm. Finally, the uncertainty evaluation of zero-calibration gauge is completed. The measurement uncertainty of gauge zero value is 12 μm and the measurement uncertainty of height difference is 6 μm.

Background: We aimed to establish and validate a prognostic nomogram model for improving the prediction of 30-day mortality of gastrointestinal bleeding (GIB) in critically ill patients with severe sepsis. Methods: In this retrospective study, the current retrospective cohort study extracted data from the Medical Information Mart for Intensive Care IV (MIMIC-IV) database, then partitioned the cohort randomly into training and validation subsets. The cohort was partitioned into training and validation subsets randomly. Our primary endpoint was 30-day all-cause mortality. To reduce data dimensionality and identify predictive variables, the least absolute shrinkage and selection operator (LASSO) regression was employed. A prediction model was constructed by multivariate logistic regression. Model performance was evaluated using the concordance index (C-index), receiver operating characteristic (ROC) curve, and decision curve analysis (DCA). Results: The analysis included 1435 total patients, comprising 1005 in the training cohort and 430 in the validation cohort. We found that age, smoking status, glucose, (BUN), lactate, Sequential Organ Failure Assessment (SOFA) score, mechanical ventilation≥48h (MV), parenteral nutrition (PN), and chronic obstructive pulmonary disease (COPD) independently influenced mortality in sepsis patients with concomitant GIB. The C-indices were 0.746 (0.700–0.792) and 0.716 (0.663–0.769) in the training and validation sets, respectively. Based on the area under the curve (AUC) and DCA, the nomogram exhibited good discrimination for 30-day all-cause mortality in sepsis with GIB. Conclusions: For sepsis patients complicated with GIB, we created a unique nomogram model to predict the 30-day all-cause mortality. This model could be a significant therapeutic tool for clinicians in terms of personalized treatment and prognosis prediction.

Rye (Secale cereale L., 2n=2x=14, RR) is a significant genetic resource for improving common wheat because of its resistance to multiple diseases and abiotic-stress tolerant traits. The 1RS chromosome from the German cultivated rye variety Petkus is critical in wheat breeding. However, its weakened disease resistance highlights the need to identify new resources. In the present study, a novel derived line called D27 was developed from common wheat and Mexico Rye. Cytological observations characterized the karyotype of D27 as 2n=42=21 II. Genomic in situ hybridization indicated that a pair of whole-arm translocated Mexico Rye chromosomes were inherited typically in the mitotic and meiosis stages of D27. Experiments using fluorescence in situ hybridization (FISH) and gliadin electrophoresis showed that D27 lacked wheat 1DS chromosomes. They were replaced by 1RS chromosomes of Mexico Rye, supported by wheat simple-sequence repeat markers, rye sequence characterized amplified region markers, and wheat 40K SNP array analysis. The wheat 1DS chromosomes could not be detected by molecular markers and wheat SNP array, but the presence of rye 1RS chromosomes was confirmed. Agronomic trait assessments indicated that D27 had a higher tiller number and enhanced stripe rust and powdery mildew resistance. In addition, dough properties analysis showed that replacing 1DS led to higher viscosity and lower dough elasticity in D27, which was beneficial for cake making. In conclusion, the novel cytogenetically stable common wheat–Mexico Rye T1DL·1RS translocation line D27 offers excellent potential as outstanding germplasm in wheat breeding programs focusing on disease resistance and yield improvement. Additionally, it can be valuable for researching the rye 1RS chromosome’s genetic diversity.

The energy hub (EH) integrates various types of energy supply equipment and loads, improving operational efficiency but bringing uncertainty. For dealing with the uncertainty factors that EH faces, the multi-EHs operation mode has more advantages than the single-EH operation mode. Therefore, a cooperative game-based coordinated model for multiple EHs with different risk preferences is built. The expected cost of all EHs is served as the objective function. Also, all EH’s conditional value at risk (CVaR) does not exceed its acceptable upper limit. The upper limit of CVaR can be obtained by solving a bi-objective optimization model for the independent operation of EH. Especially each EH has a different risk preference in the independent operation model, i.e., the weighting factor between costs and risks. Furthermore, by using scenario-based stochastic programming, the above models are transformed into linear models. Then, a novel two-factor cost allocation method for multi-EH with different risk preferences is proposed. The fixed and proportional allocation coefficient of each EH is obtained by solving a set of allocation equations based on the Shapley value. Finally, numerical studies with three EHs demonstrate the effectiveness of the proposed method. In the case study, the overall expected cost and CVaR of multiple EHs decreased by 4.99 % and 15.88 %, respectively.

ABSTRACT: Skeletal muscle satellite cells (SMSCs), known as muscle stem cells, play an important role in muscle embryonic development, post-birth growth, and regeneration after injury. Selenoprotein K (SELENOK), an endoplasmic reticulum (ER) resident selenoprotein, is known to regulate calcium ion (Ca2+) flux and ER stress (ERS). SELENOK deficiency is involved in dietary selenium deficiency-induced muscle injury, but the regulatory mechanisms of SELENOK in SMSCs development remain poorly explored in chicken. Here, we established a SELENOK deficient model to explore the role of SELENOK in SMSCs. SELENOK knockdown inhibited SMSCs proliferation and differentiation by regulating the protein levels of paired box 7 (Pax7), myogenic factor 5 (Myf5), CyclinD1, myogenic differentiation (MyoD), and Myf6. Further analysis exhibited that SELENOK knockdown markedly activated the ERS signaling pathways, which ultimately induced apoptosis in SMSCs. SELENOK knockdown-induced ERS is related with ER Ca2+ ([Ca2+]ER) overload via decreasing the protein levels of STIM2, Orai1, palmitoylation of inositol 1,4,5-trisphosphate receptor 1 (IP3R1), phospholamban (PLN), and plasma membrane Ca2+-ATPase (PMCA) while increasing the protein levels of sarco/endoplasmic Ca2+-ATPase 1 (SERCA1) and Na+/Ca2+ exchanger 1 (NCX1). Moreover, thimerosal, an activator of IP3R1, reversed the overload of [Ca2+]ER, ERS, and subsequent apoptosis caused by SELENOK knockdown. These findings indicated that SELENOK knockdown triggered ERS driven by intracellular Ca2+ dyshomeostasis and further induced apoptosis, which ultimately inhibited SMSCs proliferation and differentiation.

Soil aggregates swell when infiltrated by water, and their size can increase in two ways. First, aggregates can attach to one another with water acting as a bridge (i.e., adsorption), and water can enter the aggregate pores (i.e., swelling). This is defined as the “adsorption-swelling” effect. Second, clay can fill the pores of macroaggregates and microaggregates (i.e., filling) when adsorption occurs. This is defined as the “adsorption-filling” effect. However, the size range of aggregates affected by these effects and the extent of their influence on aggregate swelling are still unclear. Therefore, different initial size fractions (5 ∼ 2, 2 ∼ 1, 1 ∼ 0.5, 0.5 ∼ 0.25 and 0.25 ∼ 0.053 mm) of soil aggregates from the black soil zone of Northeast China were studied. The design and application of sustainable structures are promising approaches to achieve energy conservation and emission reduction in construction and buildings. Cork stoppers have high recycling value and can be particularly used as the core of eco-friendly sandwich structures. The object of this paper is to assess the eco-mechanical performance of a more sustainable sandwich structure made of eco-friendly cores (natural or agglomerated cork stopper), aluminum alloy skins and resin adhesive. A set of sandwich panels with different packing arrangements are fabricated. Experimental tests are conducted to address the effects of packing topology and cork stoppers’ materials on bending characteristics and failure modes. The life cycle assessment method is adopted to quantify the environmental impacts, and the eco-mechanical performances are further evaluated. The results show that this sandwich structure concept with cork stopper cores has good structural and resource-environment efficiency and could be a promising alternative in construction and buildings.

Background: Metabolic syndrome score in children assesses the risk of developing cardiovascular disease in future. We aim to probe the role of the caudate in relation to the metabolic syndrome score. Furthermore, using both functional and structural neuroimaging, we aim to examine the interplay between functional and structural measures. Methods: A longitudinal birth cohort study with functional and structural neuroimaging data obtained at 4.5, 6.0 and 7.5 years and metabolic syndrome scores at 8.0 years was used. Pearson correlation and linear regression was used to test for correlation fractional anisotropy (FA) and fractional amplitude of low frequency fluctuations (fALFF) of the caudate with metabolic syndrome scores. Mediation analysis was used to test if later brain measures mediated the relation between earlier brain measures and metabolic syndrome scores. Inhibitory control was also tested as a mediator of the relation between caudate brain measures and metabolic syndrome scores. Results: FA at 4.5 years and fALFF at 7.5 years of the left caudate was significantly correlated with metabolic syndrome scores. Post-hoc mediation analysis showed that fALFF at 7.5 years fully mediated the relation between FA at 4.5 years and metabolic syndrome scores. Inhibitory control was significantly correlated with fALFF at 7.5 years, but did not mediate the relation between fALFF at 7.5 years and metabolic syndrome scores. Conclusions: We found that variations in caudate microstructure at 4.5 years predict later variation in functional activity at 7.5 years. This later variation in functional activity fully mediates the relation between microstructural changes in early childhood and metabolic syndrome scores at 8.0 years.

Summary: Polycystic ovary syndrome (PCOS) is an endocrine disorder and the main cause of anovulatory infertility, in which persistent activation of androgen receptor (AR) due to aberrant acetylation modifications of transcription is a potential trigger; however, the precise mechanisms of AR activation are poorly understood. In this study, AR activation in dehydroepiandrosterone- and letrozole-induced rat PCOS ovaries coincided with a marked increase of a chromatin acetylation “reader” BRD4. Further bioinformatic analysis showed that the AR promoter contained highly conserved binding motifs of BRD4 and HIF-1α. BRD4 and HIF-1α inducibly bound to the histone 3/4 acetylation-modified AR promoter, while administration of a BRD4-selective inhibitor JQ1 reduced the binding and AR transcription and improved the adverse expression of the core fibrotic mediators in PCOS ovaries and DHT-treated granulosa cells. Our data indicate that BRD4 upregulation and the resultant AR transcriptional activation constitute an important regulatory pathway that promotes ovarian fibrosis in PCOS.

Backgrounds: Spontaneous preterm birth (SPB) is a global problem. Early screening, identification, and prevention in asymptomatic pregnant women with risk factors for preterm birth can help reduce the incidence and mortality of preterm births. Therefore, this study systematically reviewed prediction models for spontaneous preterm birth, summarised the model characteristics, and appraised their quality to identify the best-performing prediction model for clinical decision-making. Methods: PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure, China Biology Medicine disc, VIP Database, and Wanfang Data were searched up to September 27, 2021. Prediction models for spontaneous preterm births in singleton asymptomatic pregnant women with risk factors were eligible for inclusion. Six independent reviewers selected the eligible studies and extracted data from the prediction models. The findings were summarised using descriptive statistics and visual plots. Results: Twelve studies with twelve developmental models were included. Discriminative performance was reported in 11 studies, with an Area Under the Curve (AUC) ranging from 0.75 to 0.95. The AUCs of the seven models were greater than 0.85. Cervical length (CL) is the most commonly used predictor of spontaneous preterm birth. A total of 91.7% of the studies had a high risk of bias in the analysis domain, mainly because of the small sample size and lack of adjustment for overfitting. Conclusion: The accuracy of the models for spontaneous preterm births in singleton asymptomatic women with risk factors was good. However, these models are not widely used in clinical practice because they lack replicability and transparency. Future studies should transparently report methodological details and consider more meaningful predictors with new progress in research on preterm birth.

Building a robust and capable village power networks plays an important role for responding to national strategic goals of “village revitalization”, “peak carbon dioxide emission” and “carbon neutrality”. At present, user-side distributed photovoltaic (PV) power is encouraged to be implemented in rural areas in China which brings about tremendous pressure to village power grids. Those power grids are already under vulnerable and inferior conditions in reality. Since PV power systems featuring volatility and randomness are commonly implemented in different lines across low-voltage distribution networks (LVDN) in village power networks in Chinese rural area, power imbalance affects the reliability of power supply. There is an urgent need for developing adaptive control strategies for managing this kind of distributed PV systems. This paper firstly studies the topology of a three-phase-four-line LVDN with regulated parameters. Afterwards, by analyzing the operational costs of the power grid and equipment reliability, a two-stage control scheme is proposed. Furthermore, the imbalance of three phase power systems, network losses, transformer losses are considered to form a multi-objective optimization problem which is solved by tuning transformer voltage, reactive power from PV inverters, energy storage system and demand response schemes. An actual LVDN and associated parameters from a real power distribution networks in Bijie region in Guizhou province, China are used in this study. The effectiveness of the proposed method is tested where 32.1% of power loss is reduced with three-phase imbalance being obviously mitigated accordingly.

A 85-day feeding trial was designed to verify the effects of soybean meal on growth, feed utilization, antioxidants, and immune response of tilapia (GIFT Oreochromis spp.), and evaluated the effects of dietary tryptophan on alleviating the negative responses induced by high inclusion of soybean meal. Five experimental diets, two of which substituted 15 % and 45 % of the fish meal with soybean meal, the others supplemented with 0.2 %, 0.4 % and 0.6 % L-tryptophan on the basis of 45 % soybean meal, respectively, were fed 400 juvenile fish (initial weight: 2.98 ± 0.04 g). The results showed that the crude ash content, red blood cell count (RBC), lipase (LPS) and glutathione peroxidase (GSH-Px) activities of tilapia significantly decreased when the soybean meal replaced fish meal by 45 % (p 0.05). Objective: This prospective longitudinal study aimed to investigate changes in sarcopenia, physical activity, and inflammation biomarkers in patients with oral cavity cancer during curative treatment and explore their association with treatment outcomes. Methods: Patients newly diagnosed with oral cavity cancer who underwent primary surgery with (chemo)radiation therapy were included. Along with physical activity and inflammatory markers, sarcopenia was assessed using a 5-time chair stand test, hand grip strength, and skeletal muscle index (SMI). Data were collected before operation and after 3 months (T2) and 6 months after operation. Logistic regression and Cox proportional hazards models were used to identify predictors of treatment outcomes. Results: Out of 56 patients, 21 (37.5%) had sarcopenia. SMI score, physical activity, and neutrophil-to-lymphocyte ratio (NLR) showed significant changes after surgery, with exacerbation at T2. Patients with sarcopenia exhibited a significant decrease in SMI scores at T2. Progression to a severe condition remains a major risk factor for the COVID-19 mortality. Robust models that predict the onset of severe COVID-19 are urgently required to support sensitive decisions regarding patients and their treatments. In this study, we developed a multivariate survival model based on early-stage CT images and other physiological indicators and biomarkers using artificial-intelligence analysis to assess the risk of severe COVID-19 onset. We retrospectively enrolled 338 adult patients admitted to a hospital in China (severity rate, 31.9%; mortality rate, 0.9%). The physiological and pathological characteristics of the patients with severe and non-severe outcomes were compared. Age, body mass index, fever symptoms upon admission, coexisting hypertension, and diabetes were the risk factors for severe progression. Compared with the non-severe group, the severe group demonstrated abnormalities in biomarkers indicating organ function, inflammatory responses, blood oxygen, and coagulation function at an early stage. In addition, by integrating the intuitive CT images, the multivariable survival model showed significantly improved performance in predicting the onset of severe disease (mean time-dependent area under the curve = 0.880). Multivariate survival models based on early-stage CT images and other physiological indicators and biomarkers have shown high potential for predicting the onset of severe COVID-19.

Introduction: Lots of research has revealed that vascular dementia (VD), which is closely related to the metabolic disorders of intestinal microbiota, can be treated through traditional Chinese drug by regulating the brain-gut-microbiome axis. However, the specific interaction between VD and intestinal microflora, as well as the mechanism of Wuzang Wenyang Huayu Tang (WWHT) in treating VD by regulating the brain-gut-microbiome axi remains unknown on the whole and needs to be further studied. Objectives: To explore the effect of WWHT on the learning and memory ability of rats with VD based on brain-gut-microbiome axis. Methods: In this study, taking the specific pathogen-free (SPF) SD rats as the subjects, a vascular dementia rat model was created by using the two-vessel occlusion model (2-VO). The subjects were divided into six groups, namely control group, sham group, VD model group, the piracetam group, the WWHT group, and the WWHT combined with piracetam (WCP) group. After 22 days of the gavage administration method, the Morris water maze experiment was adopted to determine the learning and memory abilities of each group through the water maze, observe the effects of hematoxylin and eosin (HE) staining and transmission electron microscopy in the hippocampal area of each group, and analyze the relevant proteins in the whole brain by Western Blot (WB). In addition, the feces of each group were collected and analyzed by fecal metabolomics combined with 16S rRNA gene sequencing to explore the interaction between VD and intestinal microbiota. Results: Through Morris water maze experiment, it was found that the learning and memory ability of VD rats in WWHT, piracetam and WCP groups was improved. The morphology of hippocampal neuron cells were improved in the WWHT group and the number of hippocampal neuron cells were increased in the VD rats by HE. By observing the ultrastructure of hippocampal cells, it can be found that the VD model group showed cell membrane edema and large organelle lysis. The ultrastructure of the hippocampus of all the administered groups was improved. In terms of the effect on the expression levels of related proteins in the whole brain of rats, compared with the VD model group, the expression levels of Caspase-3 protein decreased in all groups after drug administration. The protein expression levels of APG5L/ATG5, Beclin-1 and LC3A/B were upregulated, indicating that the drug administration group could regulate cellular autophagy in the whole brain to further protect brain tissue. Fecal metabolite profiles revealed 27 different metabolites related to VD. These metabolites are mainly involved in related metabolic pathways, such as vitamin B metabolism, purine metabolism, pyrimidine metabolism, and histidine metabolism. Further analysis of the gut microbiota by 16S rRNA gene sequencing data showed that WWHT-treated VD rats were different in gut microbiota composition. Discussion: Through the effect of brain-gut-microbiome axis, WWHT regulates the metabolism of vitamin B6 and tryptophan in VD rats, which can improve the intestinal microbiota of VD rats and regulate autophagy in the brain to improve cell morphology in the hippocampus, thus increasing the number of hippocampal cells to protect hippocampal neuron cells2510100, and finally improving the learning and memory ability of VD rats.
