The development of a simple and effective method for the highly sensitive and selective discrimination of proteins is a subject of enormous interest. Herein, we report the construction of a novel fluorescence detection method based on a perylene probe for the highly efficient discrimination of multiple proteins. Single-stranded DNA (ssDNA) could induce aggregation of the perylene probe which caused quenching of probe fluorescence. After the addition of a protein, the protein could interact with the ssDNA-probe assembly complex with "turn-on" or further "turn-off" fluorescence response. A sensor array was designed based on the above phenomena which could realize the successful discrimination of proteins with 100% accuracy of cross validation. Nine representative proteins were successfully recognized. Moreover, it was observed that a protein could induce characteristic effect on the DNA-probe assembly with varying pH of assay buffer. Thus, different proteins showed unique fluorescence response towards assay buffers having different pH values. The assay buffer pH was then utilized as a sensing channel. Based on Linear Discriminant Analysis (LDA) nine proteins were successfully discriminated at the nanomolar concentration with 100% accuracy of cross validation. Furthermore, the sensor array also demonstrated differentiation of the nine proteins regardless of their concentration. The developed sensor array could also detect the proteins with great precision in human urine sample at a quite low concentration, which suggests its practical applicability for analysis of biological fluids., (Copyright © 2020 Elsevier B.V. Silver nanoparticles (Ag NPs) enhanced perylene probe excimer emission is reported for the first time. It was observed that strong interactions between the perylene probe and the Ag NPs induced co-aggregation. As a result, a new in situ generated plasmonic absportion band of the Ag NPs at longer wavelength emerged. The monomer emission of the perylene probe was efficiently quenched, and dramatically enhanced probe excimer emission was observed. A remarkable emission enhancement of over 1000 fold was obtained compared to anionic polymers and other nanoparticles. The excimer emission intensity could be finely modulated by the size of the Ag NPs and the functionalities of the perylene probe. The observed Ag NPs enhanced perylene probe excimer emission shows good potential for the development of novel sensing techniques for various bioanalytical applications., (Copyright © 2018 Elsevier B.V. A novel and efficient approach has been established for the synthesis of silver nanoclusters capped silica nanoparticles (SiO 2 @AgNCs). These nanoclusters (AgNCs) capped silica nanoparticles were utilized as a novel ratiometric photoluminescence (PL) nanosensor for extremely sensitive and selective detection of I - and S 2- ions. The AgNCs were prepared in situ on the silica nanoparticles through polyethyleneimine (PEI) template approach. While dual PL emissions of AgNCs (at 500 nm) and luminescent silica nanoparticles (at 602 nm) formed the basis for the ratiometric sensing. The PL emission of AgNCs was strongly quenched by I - (or S 2- ), while that of luminescent silica nanoparticles was hardly affected. The PL emission intensity ratio of AgNCs and the luminescent silica nanoparticles was defined as I 500 /I 602 . A good linear relationship between the I 500 /I 602 value and the concentration of I - (or S 2- ) was observed, and the limit of detection (LOD) was estimated to be 57 nM for I - and 62 nM for S 2- . In addition, the fluorescence images of the SiO 2 @AgNCs nanosensor changed from white to orange upon exposure to different concentrations of I - (or S 2- ) (0-250 μM), which could be clearly distinguished by the naked eye. The SiO 2 @AgNCs nanosensor exhibited good selectivity against other analytes, and I - or S 2- ions could be separately detected via the introduction of proper masking agents. A novel label-free fluorescence recovery assay for the sensing of a DNA binding protein has been developed. A transcription factor c-Jun protein, and a 21 base pair duplex DNA containing the c-Jun protein binding site (J-DNA) were selected. J-DNA was mixed with a cationic fluorescent perylene probe (compound 1), and induced aggregation of the probe. Quenching of the probe's fluorescence was observed. However, when c-Jun protein was mixed with the J-DNA, c-Jun bound to the duplex DNA, which reduced the degree of the induced perylene probe aggregation, and a turn on fluorescence signal was observed. The recovered fluorescence intensity was directly related to the amount of c-Jun added. The method is highly selective, six non-DNA binding proteins and one randomly selected 21 base pair duplex DNA (con-1) were tested. No noticeable compound 1 fluorescence recovery was observed. Mutations were also introduced to the c-Jun recognition sequence and much reduced fluorescence recovery was observed. Our assay is label-free, convenient, inexpensive, and fast. To meet the need for rapid and low-cost chemical sensing of explosive, new fluorescence chemosensors based on oligophenothiazines for probing vapor-phase nitro compounds have been developed. The phenothiazine-based trimer P3 and pentamer P5 have been synthesized via Heck and Wittig reactions by convergent approach. It was found that they can detect the vapors of nitro compounds, including p-nitrotoluene (p-NT), 2,4-dinitrotoluene (DNT), 2,4,6-trinitrotoluene (TNT) with good sensitivity and reversibility. And the sensor of P3 film gave a linear fluorescence quenching response to 7-800 ppb TNT with the detection limit of 4 ppb. For DNT vapor, a linear working range of the sensor was 2-24 ppm with the detection limit of 40 ppb. Meanwhile, the interferents, including common organic solvents, p-nitrophenol and 2,4-dinitrophenylhydrazine cannot lead to obvious fluorescence quenching, meaning that the film based on oligophenothiazines exhibited good specificity of fluorescence response to explosive. Based on the fluorescence lifetime and UV-vis absorption measurements, we suggested that the fluorescence quenching of oligophenothiazine-based films exposed to the vapors of nitro compounds was due to the formation of non-fluorescent charge-transfer complex between oligophenothiazine and nitro compounds., (Copyright © 2010 Elsevier B.V.
