This study investigates how plants protect themselves, specifically looking at a group of genes called Xyloglucan Endotransglucosylase/Hydrolase (XTH) in cucumber plants. A detailed investigation of CsXTH genes, uncover their evolutionary background, structural motifs, regulatory networks, and functional responsibilities. The investigation utilized various bioinformatics tools and databases to highlight the dynamic interplay between CsXTH genes and abiotic stress responses. The investigation begins with a phylogenetic analysis that reveals the cladding of CsXTH genes into four categories. Further unraveling the conserved motif and domain analyses expose the coordination of xyloglucan modification, highlighting essential motifs and domains shared among these genes. Analyzing cis-regulatory elements helps us understand the complex networks that control the defense mechanisms of cucumber plants. Among these elements, CGTCA-motif and TGACG-motif stand out as key players in interpreting environmental signals and coordinating stress responses. The microRNA Csa-miR169 is becoming a key player in regulating the expression of CsXTH genes, and controlling the biological responses of cucumber plants. Chromosome mapping, Ka/Ks calculations, and protein-protein interaction analysis offer insights into the structural and evolutionary dynamics of CsXTH genes. Additionally, the study explores sub-cellular localization, uncovering the diverse cellular compartments where CsXTH genes are active. Finally, through chrysophanol and physcion, we unveil the adaptable functions of specific CsXTH genes in response to distinct treatments, underscoring their role in orchestrating cucumber's adaptive strategies.

The growth and yield of crops have been significantly affected by abiotic stresses. Although respiratory metabolism is integral to plants, its understanding within the context of stress adaptation remains limited . While the rice genome contains three Aconitase genes, their specific roles in abiotic stress tolerance and plant development remain largely unexplored. This study aimed to characterize the Aconitase gene family in rice, shedding light on the evolution of Aconitase and its involvement in diverse developmental processes, as well as its response under various abiotic stress conditions. Phylogenetic analysis indicated the evolutionary relationship between rice Aconitase genes and those of other plant species. The prediction of promoter cis-elements revealed the presence of diverse cis-acting elements associated with the response to abiotic stress within the Aconitase gene family. In addition, an analysis of public RNA-seq data indicated the potential involvement of Aconitase genes in various developmental processes and mechanisms related to stress tolerance. Further, qRT-PCR confirmed the expression patterns of Aconitase genes. Expression analysis revealed differential expression of Aconitase genes under cold, drought, ABA, and salt stresses. Elevated levels of the OsAco1 transcript resulted in a phenotype that exhibited increased tolerance to salt stress. Biochemical analysis revealed higher Aconitase activity in the overexpressed line compared to the control plant. Furthermore, the overexpressed line exhibited improved growth and lower H2O2 levels than the control plant following a 2-hour exposure to 200 mM salt stress. These findings suggest a potential role of Aconitase in enhancing salt stress tolerance. Moreover, they lay the foundation for further exploration into the precise involvement of Aconitase in plant developmental processes under various stress conditions, with the ultimate aim of developing salt-tolerant rice cultivars.

An array of external factors, an important one being temperature, decide the fate of survival in a microbe. The ability of microbes to sense external cues and to regulate the expression of genes accordingly is critical for its likely survival. Among a myriad of cellular defence mechanisms, a strategy to recuperate stress involves RNA regulatory elements. RNAs own a repertoire of functions in a cell as messengers, for transfer or as a component of ribosomes. A shift from its indigenous role is as regulators of gene expression, where in the cis-encoded RNA termed as "RNA Thermometers" play a pivotal role in translational level of gene expression. In this paper, we review the occurrence, the different types and molecular mechanism of gene regulation by RNATs, with a special focus limited to the domain Bacteria. Apple is the fourth most important fruit crop grown in temperate areas of the world belongs to the family Rosaceae. In the present study, the promoter (∼1000bp) region of atpB gene was used to evaluate the genetic diversity and phylogeny of six local apple varieties. atpB gene is one of the large chloroplastic region which encodes β-subunit of ATP synthase and previously it had been used largely in phylogenetic studies. During the present study, atpB promoter was amplified, sequenced and analyzed using various bioinformatics tools including Place Signal Scan, MEGA6 and BLASTn. During the phylogenetic analysis, obtained phylogram divided the studied varieties into two clusters revealing the monophyletic origin of studied apple varieties. Pairwise distance revealed moderate genetic diversity that ranges from 0.047-0.170 with an average of 0.101. While identifying different cis-acting elements present in the atpB promoter region, results exhibited the occurrence of 56 common and 20 unique cis-regulatory elements among studied varieties. The identified cis-acting regulatory elements were mapped as well. It was observed that Kala Kulu has the highest unique features with reference to the availability of cis-acting elements.
