Toxoplasmosis has been recognized as a major public health problem worldwide. The consumption of uncooked or undercooked meat infected with Toxoplasma gondii tissue cysts is one of the main means of transmission of this parasite. Although sheep, goats, and pigs are commonly infected with T. gondii, little information is available on the distribution of T. gondii tissue cysts in naturally infected meat. In this study, we investigated the distribution of viable T. gondii tissue cysts in shoulder muscles of naturally infected lambs and goats. Hearts and shoulders of 46 lambs and 39 goats from a local grocery store were tested for T. gondii infection. Animals were evaluated for the presence of anti–T. gondii antibodies in heart blood and clots by the modified agglutination test. Fourteen of the 85 animals (seven lambs and seven goats) were seropositive. Six to 12 samples weighing 5, 10, and 50 g were obtained from shoulder muscles of each seropositive animal and used for bioassay in mice. The distribution of viable T. gondii differed according to the size of the sample analyzed, but in general larger sample sizes resulted in higher isolation rates (P , 0.05). Results of the study revealed an uneven distribution of T. gondii in muscle samples of lambs and goats and that T. gondii can be transmitted by consumption of very small servings (5 and 10 g) of meat when it is consumed raw or is undercooked. Kinetic growth data for Bacillus cereus grown from spores were collected in cooked beans under several isothermal conditions (10 to 49°C). Samples were inoculated with approximately 2 log CFU/g heat-shocked (808C for 10 min) spores and stored at isothermal temperatures. B. cereus populations were determined at appropriate intervals by plating on mannitol-egg yolk-polymyxin agar and incubating at 308C for 24 h. Data were fitted into Baranyi, Huang, modified Gompertz, and three-phase linear primary growth models. All four models were fitted to the experimental growth data collected at 13 to 46°C. Performances of these models were evaluated based on accuracy and bias factors, the coefficient of determination (R2), and the root mean square error. Based on these criteria, the Baranyi model best described the growth data, followed by the Huang, modified Gompertz, and three-phase linear models. The maximum growth rates of each primary model were fitted as a function of temperature using the modified Ratkowsky model. The high R2 values (0.95 to 0.98) indicate that the modified Ratkowsky model can be used to describe the effect of temperature on the growth rates for all four primary models. The acceptable prediction zone (APZ) approach also was used for validation of the model with observed data collected during single and two-step dynamic cooling temperature protocols. When the predictions using the Baranyi model were compared with the observed data using the APZ analysis, all 24 observations for the exponential single rate cooling were within the APZ, which was set between-0.5 and 1 log CFU/g; 26 of 28 predictions for the two-step cooling profiles also were within the APZ limits. The developed dynamic model can be used to predict potential B. cereus growth from spores in beans under various temperature conditions or during extended chilling of cooked beans. Leafy green vegetables, including lettuce, are recognized as potential vehicles for foodborne pathogens such as Escherichia coli O157:H7. Fresh-cut lettuce is potentially at high risk of causing foodborne illnesses, as it is generally consumed without cooking. Quantitative microbial risk assessments (QMRAs) are gaining more attention as an effective tool to assess and control potential risks associated with foodborne pathogens. This study developed a QMRA model for E. coli O157:H7 in fresh-cut lettuce and evaluated the effects of different potential intervention strategies on the reduction of public health risks. The fresh-cut lettuce production and supply chain was modeled from field production, with both irrigation water and soil as initial contamination sources, to consumption at home. The baseline model (with no interventions) predicted a mean probability of 1 illness per 10 million servings and a mean of 2,160 illness cases per year in the United States. All intervention strategies evaluated (chlorine, ultrasound and organic acid, irradiation, bacteriophage, and consumer washing) significantly reduced the estimated mean number of illness cases when compared with the baseline model prediction (from 11.4- to 17.9-fold reduction). Sensitivity analyses indicated that retail and home storage temperature were the most important factors affecting the predicted number of illness cases. The developed QMRA model provided a framework for estimating risk associated with consumption of E. coli O157:H7–contaminated fresh-cut lettuce and can guide the evaluation and development of intervention strategies aimed at reducing such risk. A recent study by the Centers for Disease Control and Prevention reported that between 1998 and 2008, leafy greens outbreaks accounted for 22.3% of foodborne outbreaks in the United States. Several studies on the growth of bacteria at different temperatures have been conducted; however, there is a need for the prediction of bacterial growth when leafy greens are transported without temperature control. Food products, when taken out of refrigeration, undergo a temperature change, with the rate of temperature change being proportional to the difference in the temperature of food and its surroundings. The objective of this study was to estimate the growth of Escherichia coli O157:H7, Salmonella enterica, and L. monocytogenes in leafy greens during transportation from retail to home at ambient temperatures ranging from 10 to 40°C for up to 10 h. Experiments were conducted to monitor the temperature increase in fresh spinach taken from refrigeration temperature to ambient temperature. The growth of pathogens was predicted using these changing temperature profiles with the three-phase linear model as a primary model and the square root model as the secondary model. The levels of E. coli O157:H7, S. enterica, and L. monocytogenes increased by 3.12, 2.43, and 3.42 log CFU at 40°C for the 10-h period, respectively, when no lag phase was assumed. If leafy greens are not kept out of refrigeration for more than 3 h, when the air temperature is 40°C or more, pathogen growth should be less than 1 log CFU. Toxoplasma gondii is a prevalent protozoan parasite worldwide. Human toxoplasmosis is responsible for considerable morbidity and mortality in the United States, and meat products have been identified as an important source of T. gondii infections in humans. The goal of this study was to develop a farm-to-table quantitative microbial risk assessment model to predict the public health burden in the United States associated with consumption of U.S. domestically produced lamb. T. gondii prevalence in market lambs was pooled from the 2011 National Animal Health Monitoring System survey, and the concentration of the infectious life stage (bradyzoites) was calculated in the developed model. A log-linear regression and an exponential doseresponse model were used to model the reduction of 7. gondii during home cooking and to predict the probability of infection, respectively. The mean probability of infection per serving of lamb was estimated to be 1.5 cases per 100,000 servings, corresponding to ~6,300 new infections per year in the U.S. population. Based on the sensitivity analysis, we identified cooking as the most effective method to influence human health risk. This study provided a quantitative microbial risk assessment framework for T. gondii infection through consumption of lamb and quantified the infection risk and public health burden associated with lamb consumption. Checkpointing mechanism is used to tolerate the impact of transient faults by rollback operation. Recently, it has also been used as a mechanism to enhance system's lifetime by identifying and tolerating permanent fault 5,19,10,12. However, equidistant checkpoint interval may cause task deadline violation in the system. Here, we propose an adaptive checkpoint interval placement algorithm (ADeLiRACI) that meets all tasks deadline. The checkpoint intervals are adjusted to minimize the impact of stresses and permanent faults on the running hosts. This novel mechanism allows greater applicability in real time systems with hard deadline such as weather prediction, financial transactions etc. We compare the estimated completion time for increasing fault-rate in the system against five existing algorithms. Toxoplasma gondii is a global protozoan parasite capable of infecting most warm-blooded animals. Although healthy adult humans generally have no symptoms, severe illness does occur in certain groups, including congenitally infected fetuses and newborns, immunocompromised individuals including transplant patients. Epidemiological studies have demonstrated that consumption of raw or undercooked meat products is one of the major sources of infection with T. gondii. The goal of this study was to develop a framework to qualitatively estimate the exposure risk to T. gondii from various meat products consumed in the United States. Risk estimates of various meats were analyzed by a farm-to-retail qualitative assessment that included evaluation of farm, abattoir, storage and transportation, meat processing, packaging, and retail modules. It was found that exposure risks associated with meats from free-range chickens, nonconfinement-raised pigs, goats, and lamb are higher than those from confinement-raised pigs, cattle, and caged chickens. For fresh meat products, risk at the retail level was similar to that at the farm level unless meats had been frozen or moisture enhanced. Our results showed that meat processing, such as salting, freezing, commercial hot air drying, long fermentation times, hot smoking, and cooking, are able to reduce T. gondii levels in meat products, whereas nitrite and/or nitrate, spice, low pH, and cold storage have no effect on the viability of T. gondii tissue cysts. Raw-fermented sausage, cured raw meat, meat that is not hot-air dried, and fresh processed meat were associated with higher exposure risks compared with cooked meat and frozen meat. This study provides a reference for meat management control programs to determine critical control points and serves as the foundation for future quantitative risk assessments. Toxoplasma gondii is a protozoan parasite that is responsible for approximately 24% of all estimated deaths attributed to foodborne pathogens in the United States. Human infection results from accidental ingestion of oocysts from the environment, in water, or on insufficiently washed produce or from consumption of raw or undercooked meat products that contain T. gondii tissue cysts. This review focused on studies of T. gondii in meat because many human T. gondii infections are acquired through consumption of raw or undercooked meat. Prevalence of T. gondii is higher in conventionally reared pigs, sheep, and poultry than in cattle and is greater in meat products from organic than from conventionally reared meat animals because of outdoor access, which poses substantially greater opportunities for exposure to infected rodents, wildlife, and oocyst-contaminated feed, water, or environmental surfaces. Risk factors related to T. gondii exposure for livestock include farm type, feed source, presence of cats, methods of rodent and bird control, methods of carcass handling, and water quality. This review serves as a useful resource and information repository for informing quantitative risk assessment studies for T. gondii infection in humans through meat consumption. Robot simulators have supported the science of robotics for a long period and have been an indispensable tool for understanding, planning, designing and programming of educational and industrial robots. These soft tools allows the users to program and test their applications without using the real hardware or even building it since such tools allow the analysis of behaviours and performance beforehand. Some of these soft tools have already been designed and developed for professional as well as educational and research purposes by researchers. However, the majority of these tools are specifically designed for serial manipulators and simulation tools for robotic grasping is rather limited e.g Graspit!, OpenGRASP, SynGrasp, and hence there is a need to develop a general purpose software package for performing different analyses on dexterous robotic hands. In this research work, a new MATLAB based dexterous robotic gripper toolbox is developed with GUI based simulating environment for Kinematic analysis and additional features with inbuilt library of various dexterous grippers of Robot. This toolbox has capabilities like Hand modeling, Grasp definition, Grasp modeling, Grasp analysis and Graphics to support representation of various commercially available existing dexterous grippers and has provision and flexibility to accommodate any newly developed hand. The library of toolbox has different parameters like Denavit - Hartenberg parameters, finger size, and joint angles etc. which are used for kinematics of various dexterous grippers. Form closure analysis is a special feature that has been included for understanding the actual positioning and orientation of fingers of dexterous grippers according to the object position. This paper employs a 2-DOF (degree of freedom) PID controller for compensating a physical magnetic levitation system. It is shown that because of having a feedforward gain in the proposed 2-DOF PID control, the transient performance of the compensated system can be changed in a desired manner unlike the conventional 1-DOF PID control. It is also shown that for a choice of PID parameters, although the theoretical loop robustness is the same for both the compensated systems, in real-time, 2-DOF PID control may provide superior robustness if a suitable choice of the feedforward parameter is made. The potential of food-borne pathogens to survive and grow during refrigerated and frozen storage has raised serious concerns over the safety of stored poultry products. In this study, the effect of refrigeration and freezing temperatures (-20, -12, 0, 4, and 8°C) on growth and survival of Liste'ria innocua and Salmonella enterica serovar Typhimurium in raw chicken breasts for storage times of 3, 7, 10, 14, and 21 d were investigated. A modified Weibull model was also developed to analyze the microbial behavior of both microorganisms in raw chicken breasts under different refrigerated storage conditions over time. The results showed that the bacterial loads of L. innocua at 4 and 8°C and Salmonella Typhimurium at 8°C were significantly different (P < 0.05) from those at other refrigerated and frozen storage temperatures over storage times. The loads of both bacteria at frozen storage temperatures did not change significantly over time. At a storage time of 7 d, the increase in bacterial loads of L. innocua at 4 and 8°C was 2.1 log cfu/g and 3.7 log cfu/g, respectively, and that of Salmonella Typhimurium at 8°C was 1.2 log cfu/g. The root mean square errors, median relative error, mean absolute relative error, and the plot of predicted versus observed bacterial loads showed a good performance of the model. The results from this study provided useful information regarding the behavior of Listeria and Salmonella in raw chicken breast meat during refrigerated and frozen storage, which would be helpful in giving insight over the safety of poultry products storage. This study compared the relative public health impact in deli meats at retail contaminated with Listeria monocytogenes by either (i) other products or (ii) the retail environment. Modeling was performed using the risk of listeriosis-associated deaths as a public health outcome of interest and using two deli meat products (i.e., ham and turkey, both formulated without growth inhibitors) as model systems. Based on reported data, dell meats coming to retail were assumed to be contaminated at a frequency of 0.4%. Three contamination scenarios were investigated: (i) a baseline scenario, in which no additional cross-contamination occurred at retail, (ii) a scenario in which an additional 2.3% of products were cross-contaminated at retail due to transfer of L. monocytogenes cells from already contaminated ready-to-eat deli meats, and (iii) a scenario in which an additional 2.3% of products were contaminated as a result of cross-contamination from a contaminated retail environment. By using a previously reported L. monocytogenes risk assessment model that uses product-specific growth kinetic parameters, cross-contamination of deli ham and turkey was estimated to increase the relative risk of listeriosis-associated deaths by 5.9- and 6.1-fold, respectively, for contamination from other products and by 4.9- and 5.8-fold, respectively, for contamination from the retail environment. Sensitivity and scenario analyses indicated that the frequency of cross-contamination at retail from any source (other food products or environment) was the most important factor affecting the relative risk of listeriosis-associated deaths. Overall, our data indicate that retail-level cross-contamination of ready-to-eat deli meats with L. monocytogenes has the potential to considerably increase the risk of human listeriosis cases and deaths, and thus precise estimates of cross-contamination frequency are critical for accurate risk assessments. The objective of this study was to estimate the relative risk of listeriosis-associated deaths attributable to Listeria monocytogenes contamination in ham and turkey formulated without and with growth inhibitors (GIs). Two contamination scenarios were investigated: (i) prepackaged deli meats with contamination originating solely from manufacture at a frequency of 0.4% (based on reported data) and (ii) retail-sliced deli meats with contamination originating solely from retail at a frequency of 2.3% (based on reported data). Using a manufacture-to-consumption risk assessment with product-specific growth kinetic parameters (i.e., lag phase and exponential growth rate), reformulation with GIs was estimated to reduce human listeriosis deaths linked to ham and turkey by 2.8- and 9-fold, respectively, when contamination originated at manufacture and by 1.9- and 2.8-fold, respectively, for products contaminated at retail. Contamination originating at retail was estimated to account for 76 and 63% of listeriosis deaths caused by ham and turkey, respectively, when all products were formulated without GIs and for 83 and 84% of listeriosis deaths caused by ham and turkey, respectively, when all products were formulated with GIs. Sensitivity analyses indicated that storage temperature was the most important factor affecting the estimation of per annum relative risk. Scenario analyses suggested that reducing storage temperature in home refrigerators to consistently below 7°C would greatly reduce the risk of human listeriosis deaths, whereas reducing storage time appeared to be less effective. Overall, our data indicate a critical need for further development and implementation of effective control strategies to reduce L. monocytogenes contamination at the retail level. Foodborne disease associated with consumption of ready-to-eat foods contaminated with Listeria monocytogenes represents a considerable pubic health concern. In a risk assessment published in 2003, the U.S. Food and Drug Administration and the U.S. Food Safety and Inspection Service estimated that about 90% of human listeriosis cases in the United States are caused by consumption of contaminated deli meats. In this risk assessment, all deli meats were grouped into one of 23 categories of ready-to-eat foods, and only the postretail growth of L. monocytogenes was considered. To provide an improved risk assessment for L. monocytogenes in deli meats, we developed a revised risk assessment that (i) models risk for three subcategories of deli meats (i.e., ham, turkey, and roast beef) and (ii) models L. monocytogenes contamination and growth from production to consumption while considering subcategory-specific growth kinetics parameters (i.e., lag phase and exponential growth rate). This model also was used to assess how reformulation of the chosen deli meat subcategories with L. monocytogenes growth inhibitors (i.e., lactate and diacetate) would impact the number of human listeriosis cases. Use of product-specific growth parameters demonstrated how certain deli meat categories differ in the relative risk of causing listeriosis; products that support more rapid growth and have reduced lag phases (e.g., turkey) represent a higher risk. Although reformulation of deli meats with growth inhibitors was estimated to reduce by about 2.5- to 7.8-fold the number of human listeriosis cases linked to a given deli meat subcategory and thus would reduce the overall risk of human listeriosis, even with reformulation deli meats would still cause a considerable number of human listeriosis cases. A combination of strategies is thus needed to provide continued reduction of these cases. Endemic infectious diseases in dairy cattle are of significant concern to the industry as well as for public health because of their potential impact on animal and human health, milk and meat production, food safety, and economics. We sought to provide insight into the dynamics of important endemic infectious diseases in 3 northeastern US dairy herds. `Fecal samples from individual cows and various environmental samples from these farms were tested for the presence of major zoonotic pathogens (i.e., Salmonella, Campylobacter, and Listeria) as well as commensal bacteria Escherichia coli and enterococci. Additionally, the presence of Mycobacterium avinin ssp. paratuberculosis was tested in fecal and serum samples from individual cows. Test results and health and reproductive records were maintained in a database, and fecal, plasma, DNA, and tissue samples were kept in a biobank. All bacteria of interest were detected on these farms and their presence was variable both within and between farms. The prevalence of Listeria spp. and L. monocytogenes in individual fecal samples within farm A ranged from 0 to 68.2% and 0 to 25.5%, respectively, over a period of 3 yr. Within farm B, continuous fecal shedding of Salmonella spp. was observed with a prevalence ranging from 8 to 88%; Salmonella Cerro was the predominant serotype. Farm C appeared less contaminated with Salmonella and Listeria, although in the summer of 2005, 50 and 19.2% of fecal samples were positive for Listeria and L. monocytogenes, respectively. The high prevalence of E. coli (89 to 100%), Enterococcus (75 to 100%), and Campylobacter (0 to 81%) in feces suggested they were ubiquitous throughout the farm environment. Fecal culture and ELISA results indicated a low prevalence of Mycobacterium avium ssp. paratuberculosis infection in these farms (0 to 13.6% and 0 to 4.9% for culture-positive and ELISA-positive, respectively), although the occasional presence of high shedders was observed. Results have major implications for food safety and epidemiology by providing a better understanding of infectious disease dynamics on dairy farms. Comprehensive understanding of these infections may lead to better farm management practices and pathogen reduction programs to control and reduce the on-farm contamination of these pathogens and to prevent their further entry into the food-chain. Abstract: Acidithiobacillus ferrooxidans biofilm grown on uranium ore substrate was analyzed by a micro-Raman spectrometer and an atomic force microscope (AFM). The bacterium employed for this study, A. ferrooxidans BM1, was isolated from a uranium mine (Jaduguda, India). Micro-Raman analysis revealed the different constituents of molecular fragments present in microbial cells and in secreted extracellular polymeric substances (EPSs). AFM images clearly revealed bacterial cells surrounded by EPS. From Raman spectral data, the composition of EPS from A. ferrooxidans BM1 appeared to be similar to that of EPS secreted in a different Pseudomonas bacterium. [Copyright &y& Elsevier]

Abstract: The polycrystalline samples of La-modified Bi(Sc1/2Fe1/2)O3 (i.e., (Bi1−x La x )(Sc1/2Fe1/2)O3) were prepared using a solid-state reaction technique. The formation of single-phase compound was checked with an X-ray diffraction (XRD) technique. A small amount of La-substitution (0≤ x ≤0.15) at the Bi site does not change the crystal structure of Bi(Sc1/2Fe1/2)O3. Detailed studies of dielectric properties (i.e., dielectric constant, loss tangent and ac conductivity) showed significant effect of La doping on these properties. High dielectric constant with relatively low loss tangent and electrical conductivity of the mixed-type (i.e., space charge and oxygen ion vacancies) are some of the special features of the La-modified Bi(Sc1/2Fe1/2)O3 compounds. The increasing demand of ready-to-eat poultry products has led to serious concerns over product safety, and more emphasis has been placed on thorough cooking of products. In this study, processing conditions and thermal inactivation of Listeria innocua in chicken breast meats were evaluated during convection cooking in a pilot-plant scale air-steam impingement oven. A predictive model was developed by integrating heat and mass transfer models with a pathogen kinetics model to predict temperature, water content, product yield, and bacterial inactivation during air-steam impingement cooking. Skinless boneless chicken breasts were cooked at oven air temperatures of 177 and 200°C for 2 to 10 min at a humidity of 70 to 75% (moisture by volume) and an air velocity of 1 m/s at the exit of the nozzles. The reduction in Listeria in chicken breasts after 2 to 5 min of cooking was from 0.3 to 1.4 log CFU/g and from 0.8 to 1.8 log CFU/g at 177 and 200°C, respectively. After cooking for 10 min at both temperatures, no survivors were detected in any of the cooked chicken breasts from an initial bacterial concentration of 106 CFU/g. The standard errors of prediction for the endpoint center temperatures after 2 to 10 min of cooking were 2.8 and 3.0°C for air temperatures of 177 and 200°C, respectively. At 177 and 200°C, the median relative errors of prediction for water content were 2.5 and 3.7% and those for product yield were 5.4 and 8.4%, respectively. Contamination and penetration of salmonellae into hatching eggs may comprise an important link in the transmission of these bacteria to growing birds, processed carcasses, and eventually to the consumer. In this study, a predictive model for Salmonella typhimurium as a function of initial cell number and storage or incubation time at a nearly constant temperature and humidity was developed and evaluated to compute the bacterial load after 1 d (holding), 10 d (candling), 17 d (incubation), and 21 d (chick processing). Experiments were conducted for S. typhimurium with both high initial bacterial load (HIBL) and low initial bacterial load (LIBL) of 6.0 and 3.5 log cfu/egg, respectively. Eggs with HIBL experienced 2.0 log reduction in the bacterial load after holding at 4°C for 24 h and 3.0 log increase in the bacterial load during incubation and hatch at approximately 37°C between 17 d and 21 d. Experimental data showed that bacterial load of S. typhimurium from holding to chick processing changed from 3.7 to 6.6 log cfu/egg and from 3.7 to 2.7 log cfu/egg in HIBL and LIBL eggs, respectively. The developed model was able to predict bacterial load of S. typhimurium from 3.6 to 6.6 log cfu/egg in HIBL eggs and from 3.4 to 2.7 log cfu/egg in LIBL eggs from holding to chick processing. Root mean square errors and plot of predicted compared with observed bacterial load of S. typhimurium in contaminated eggs yielded a good fit and prediction. The predicted and experimental results indicated that incubated broiler eggs have an increase in internal bacterial loads between incubation and hatch. In addition to vitamin A (1000 μg retinol equivalents), the intervention groups received either folic acid (FA; 400 μg), FA + iron (60 mg), FA + iron + zinc (30 mg), or multiple micronutrients (MNs; the foregoing plus 10 μg vitamin D, 10 mg vitamin E, 1.6 mg thiamine, 1.8 mg riboflavin, 2.2 mg vitamin B-6 2.6 μg vitamin B-12, 100 mg vitamin C, 64 μg vitamin K, 20 mg niacin, 2 mg Cu, and 100 mg Mg). The control group received vitamin A only. Results: None of the supplements reduced fetal loss. Compared with control infants, infants whose mothers received FA alone or with iron or iron + zinc had a consistent pattern of 15-20% lower 3-mo mortality; this pattern was not observed with MNs. The effect on mortality was restricted to preterm infants, among whom the relative risks (RRs) were 0.36 (95% CI: 0.18, 0.75) for FA, 0.53 (0.30, 0.92) for FA + iron, 0.77 (0.45, 1.32) for FA + iron + zinc, and 0.70 (0.41, 1.17) for MNs. Among term infants, the RR for mortality was close to 1 for all supplements except MNs (RR: 1.74; 95% CI: 1.00, 3.04). Conclusions: Maternal micronutrient supplementation failed to reduce overall fetal loss or early infant mortality. Among preterm infants, FA alone or with iron reduced mortality in the first 3 mo of life. MNs may increase mortality risk among term infants, but this effect needs further evaluation.
