Hearing impairment has frequently been described in β-thalassemia patients with a significant impact on the patients' quality of life. Most studies provided evidence of deferoxamine (DFO) dose-related ototoxicity, however, the data is scarce regarding deferasirox (DFX) as a sole iron chelator. We aimed to assess the prevalence and risk factors of sensorineural hearing loss (SNHL) and vestibular dysfunction in regularly transfused β-thalassemia patients who had been treated with DFX film coated tablets. We conducted a case control study on 57 transfusion dependent β-thalassemia patients with a mean age of 15.3 years who received DFX FCT as monotherapy for at least one consecutive year, and 57 healthy age and sex-matching controls. Comprehensive audiological evaluations using pure tone audiometry (PTA) and transient evoked otoacoustic emission (TEOAE) as well as vestibular evaluation using Video-nystagmography (VNG) were done. SNHL was identified in 12 patients (21.1 %) using PTA and a statistically significant difference was detected between controls and patients at 6 KHz and 12 KHz frequencies. A higher incidence of SNHL was detected using TEOAE, 22 patients (43.1 %) failed to pass TEOAE, with a statistically significant decrease in the signal at frequencies 1, 4 KHz bilaterally and at frequencies 1.5, 2 KHz in the right ear compared to controls. Canal paresis was detected in 21 (36.8 %) of thalassemic children using bithermal caloric test with significantly more unilateral weakness than control children (P = 0.008). We found no significant correlation between audio-vestibular dysfunction and age, sex, serum ferritin, frequency of blood transfusion and dose of DFX FCT in thalassemic children. We conclude that the incidence of SNHL and vestibular dysfunction was high among transfusion dependent β-thalassemia patients. Therefore, we recommend performing pre-treatment baseline audio-vestibular assessment and yearly audio-vestibular monitoring to early detect high risk patients and initiate timely management to prevent permanent damage. Abstract Objective We aimed to describe vestibular/oculomotor function of 7–12-year-old children with CP, Gross Motor Function Classification System (GMFCS) levels (I-III), in comparison to an age-matched control group to understand the effect of the vestibular system on activities and participation of children with CP. Methods Vestibular, oculomotor and balance function were tested in children with CP. Central and peripheral vestibular function was examined using an enclosed rotary chair and infrared video goggles (100 Hz) that measured eye movements. Oculomotor tests included smooth pursuit and optokinetic nystagmus (OKN). Vestibulo-Ocular Reflex (VOR) tests, done in complete darkness, included step rotation (STEP), sinusoidal harmonic acceleration (SHA) test, VOR cancellation and enhancement, and subjective visual vertical and horizontal (SVV/SVH). The integrity of the saccule was tested with the Cervical Vestibular Evoked Myogenic Potential. If able, the participants' balance abilities were examined using the Sensory Organization Test (SOT) to determine ability to maintain standing balance during six conditions that challenged the visual, somatosensory and vestibular systems. Independent t-tests and Mann-Whitney U tests were used to compare results between groups. Results Forty-one children with CP (mean age = 9.44 years, SD = 1.66; 23F/18M; Gross Motor Function Classification System levels: I (n = 19), II (n = 7), III (n = 15) and thirty-three typically developing (TD) children (mean age = 10.16 years, SD = 1.6; 13F/20M) were recruited from the Birmingham, AL community. There was no significant difference between children with CP and TD children in saccular function (i.e. C-VEMP test), and peripheral vestibular end organ (i.e. SHA test and STEP test), VOR enhancement, or OKN gain. Velocity gain for horizontal smooth pursuit was significantly worse in children with CP (p = 0.009), compared to TD children. Poor mediation of central vestibular function were that evident with significantly higher VOR cancellation gain in children with CP (p < 0.0001), compared to TD children and significantly higher SVV variance (p = 0.002), SVH mean (p = 0.001), and SVH variance (p < 0.0001) in children with CP compared to TD children. Compromised balance abilities in children with CP was evident with significantly lower composite scores (p < 0.0001), vestibular ratio (p < 0.0001), and visual ratio (p = 0.021). The somatosensory ratio (p = 0.798) of children with CP was similar to children with TD. Conclusions Although peripheral vestibular function was intact, children with CP had difficulty coupling eye and head movement (VOR cancellation), using the vestibular system for postural control (SOT), demonstrated poor perception of upright (SVV/SVH), and had difficulty following a slow moving target (smooth pursuit eye movement). Abstract The vestibular system enables humans to estimate self-motion, stabilize gaze and maintain posture, but these behaviors are impacted by neural noise at all levels of processing (e.g., sensory, central, motor). Despite its essential importance, the behavioral impact of noise in human vestibular pathways is not completely understood. Here, we characterize the vestibular imprecision that results from neural noise by measuring trial-to-trial vestibulo-ocular reflex (VOR) variability and perceptual just-noticeable differences (JNDs) in the same human subjects as a function of stimulus intensity. We used head-centered yaw rotations about an Earth-vertical axis over a broad range of motion velocities (0–65°/s for VOR variability and 3–90°/s peak velocity for JNDs). We found that VOR variability increased from approximately 0.6°/s at a chair velocity of 1°/s to approximately 3°/s at 65°/s; it exhibited a stimulus-independent range below roughly 1°/s. Perceptual imprecision ("sigma") increased from 0.76°/s at 3°/s to 4.7°/s at 90°/s. Using stimuli that manipulated the relationship between velocity, displacement and acceleration, we found that velocity was the salient cue for VOR variability for our motion stimuli. VOR and perceptual imprecision both increased with stimulus intensity and were broadly similar over a range of stimulus velocities, consistent with a common noise source that affects motor and perceptual pathways. This contrasts with differing perceptual and motor stimulus-dependent imprecision in visual studies. Either stimulus-dependent noise or non-linear signal processing could explain our results, but we argue that afferent non-linearities alone are unlikely to be the source of the observed behavioral stimulus-dependent imprecision. Background The vestibulo-ocular reflex (VOR) maintains stable gaze during head motion. Deficiencies lead to apparent world motion due to incomplete stabilization of eyes in space. VOR measurement requires specialized apparatus, trained operators, and significant setup time. New method We present a system (VON: vestibulo-ocular nulling ) for rapid vestibulo-ocular assessment without measuring eye movements per se . VON uses a head-mounted motion sensor, laptop computer with user input control, and laser target whose position is controlled by the computer. As the head moves, the target is made to move in the same manner with a gain set by the subject. When the subject sets the gain so the target appears stationary in space, it is stationary on the retinas. One can determine from this gain the extent to which the eyes move in space when the head moves, which is the amount by which the VOR is deficient. From this the gain of the compensatory eye movements is derived. Results VON was compared with conventional video-based VOR measures. Both methods track expected changes in gain over 20 min of adaptation to minifying spectacles. VON measures are more consistent across subjects, and pre-adaptation values are closer to compensatory. Comparison with existing method VON is a rapid means to assess vestibulo-ocular performance. As a functional perceptual measure, it accounts for gaze-stabilizing contributions that are not apparent in the standard VOR, such as pursuit and perceptual tolerance. Cisplatin is a chemotherapeutic agent commonly used for the treatment of solid tumors, and its side-effects include vestibulotoxicity. Previous studies have reported cisplatin-induced vestibulotoxicity in various animal models, but no study has investigated in vivo mouse vestibular dysfunction after cisplatin. The aim of this study was to investigate cisplatin-induced vestibulotoxicity in C57BL/6J mice. Vestibular function was assessed by recording the vestibulo-ocular reflex (VOR). This was done during sinusoidal rotations in the horizontal plane at three frequencies (0.5, 1.0 and 2.5 Hz). A high-resolution, high-frequency digital infra-red camera was used with eye-tracking algorithms. Cisplatin at 16 mg/kg, but not 8 mg/kg, decreased the VOR gain at 2.5 Hz compared with the vehicle control. Following 16 mg/kg cisplatin treatment, the animals showed no change in the optokinetic nystagmus response, suggesting that no major changes in visual or oculomotor functions had occurred. Some individuals with noise-induced hearing loss (NIHL) also report balance problems. These accompanying vestibular complaints are not well understood. The present study used a rat model to examine the effects of noise exposure on the vestibular system. Rats were exposed to continuous broadband white noise (0–24 kHz) at an intensity of 116 dB sound pressure level (SPL) via insert ear phones in one ear for three hours under isoflurane anesthesia. Seven days after the exposure, a significant increase in ABR threshold (43.3 ± 1.9 dB) was observed in the noise-exposed ears, indicating hearing loss. Effects of noise exposure on vestibular function were assessed by three approaches. First, fluorescein-conjugated phalloidin staining was used to assess vestibular stereocilia following noise exposure. This analysis revealed substantial sensory stereocilia bundle loss in the saccular and utricular maculae as well as in the anterior and horizontal semicircular canal cristae, but not in the posterior semicircular canal cristae. Second, single unit recording of vestibular afferent activity was performed under pentobarbital anesthesia. A total of 548 afferents were recorded from 10 noise-treated rats and 12 control rats. Noise exposure produced a moderate reduction in baseline firing rates of regular otolith afferents and anterior semicircular canal afferents. Also a moderate change was noted in the gain and phase of the horizontal and anterior semicircular canal afferent's response to sinusoidal head rotation (1 and 2 Hz, 45°/s peak velocity). Third, noise exposure did not result in significant changes in gain or phase of the horizontal rotational and translational vestibulo-ocular reflex (VOR). These results suggest that noise exposure not only causes hearing loss, but also causes substantial damage in the peripheral vestibular system in the absence of immediate clinically measurable vestibular signs. Updating the position of an earth-fixed target during whole-body rotation seems to rely on cognitive processes such as the utilization of external feedback. According to perceptual learning models, improvement in performance can also occur without external feedback. The aim of this study was to assess spatial updating improvement in the absence and in the presence of external feedback. While being rotated counterclockwise (CCW), participants had to predict when their body midline had crossed the position of a memorized target. Four experimental conditions were tested: (1) Pre-test: the target was presented 30° in the CCW direction from participant’s midline. (2) Practice: the target was located 45° in the CCW direction from participant’s midline. One group received external feedback about their spatial accuracy (Mackrous and Simoneau, 2014) while the other group did not. (3) Transfer T 30 CCW : the target was presented 30° in the CCW direction to evaluate whether improvement in performance, during practice, generalized to other target eccentricity. (4) Transfer T 30 CW : the target was presented 30° in the clockwise (CW) direction and participants were rotated CW. This transfer condition evaluated whether improvement in performance generalized to the untrained rotation direction. With practice, performance improved in the absence of external feedback ( p = 0.004). Nonetheless, larger improvement occurred when external feedback was provided ( p s = 0.002). During T 30 CCW , performance remained better for the feedback than the no-feedback group ( p = 0.005). However, no group difference was observed for the untrained direction ( p = 0.22). We demonstrated that spatial updating improved without external feedback but less than when external feedback was given. The central vestibular system plays an important role in higher neural functions such as self-motion perception and spatial orientation. Its ability to store head angular velocity is called velocity storage mechanism (VSM), which has been thoroughly investigated across a wide range of species. However, little is known about the mouse VSM, because the mouse lacks typical ocular responses such as optokinetic after nystagmus or a dominant time constant of vestibulo-ocular reflex for which the VSM is critical. Experiments were conducted to examine the otolith-driven eye movements related to the VSM and verify its characteristics in mice. We used a novel approach to generate a similar rotating vector as a traditional off-vertical axis rotation (OVAR) but with a larger resultant gravito-inertial force (>1 g) by using counter rotation centrifugation. Similar to results previously described in other animals during OVAR, two components of eye movements were induced, i.e. a sinusoidal modulatory eye movement (modulation component) on which a unidirectional nystagmus (bias component) was superimposed. Each response is considered to derive from different mechanisms; modulations arise predominantly through linear vestibulo-ocular reflex, whereas for the bias, the VSM is responsible. Data indicate that the mouse also has a well-developed vestibular system through otoliths inputs, showing its highly conserved nature across mammalian species. On the other hand, to reach a plateau state of bias, a higher frequency rotation or a larger gravito-inertial force was considered to be necessary than other larger animals. Compared with modulation, the bias had a more variable profile, suggesting an inherent complexity of higher-order neural processes in the brain. Abstract: Objective: We compared vibration-induced ocular vestibular evoked myogenic potentials (OVEMPs) with the visual vertical during whole-body roll tilt and eccentric rotation in healthy subjects and patients with unilateral vestibular loss, to determine which test was most sensitive in discriminating impaired utricle function. Methods: OVEMPs and the visual vertical were measured in 11 patients and 11 healthy subjects. Visual vertical was measured during roll tilts between −9.6° and 9.6°, and during rotation at 400°/s with the head upright and the vertical rotation axis located between ±3.5cm from the head center. Results: OVEMPs in patients were strikingly asymmetric, whereas they were approximately symmetric in healthy subjects. Patients showed impaired visual vertical gain during eccentric rotation and increased errors for both roll tilt and eccentric rotation tests. OVEMPs were superior at discriminating between patients and healthy subjects, although eccentric rotation performed nearly as well. Conclusions: OVEMPs provide a powerful test for discriminating between healthy subjects and patients with chronic unilateral vestibular loss, and testing the visual vertical testing during eccentric rotation was superior to testing during whole-body roll tilt. Significance: OVEMPs are easier to administer, less demanding on patients, and in general are more effective at identifying chronic unilateral vestibular loss than visual vertical measurements. Abstract: Since the first description of sound-evoked short-latency myogenic reflexes recorded from neck muscles, vestibular evoked myogenic potentials (VEMPs) have become an important part of the neuro-otological test battery. VEMPs provide a means of assessing otolith function: stimulation of the vestibular system with air-conducted sound activates predominantly saccular afferents, while bone-conducted vibration activates a combination of saccular and utricular afferents. The conventional method for recording the VEMP involves measuring electromyographic (EMG) activity from surface electrodes placed over the tonically-activated sternocleidomastoid (SCM) muscles. The “cervical VEMP” (cVEMP) is thus a manifestation of the vestibulo-collic reflex. However, recent research has shown that VEMPs can also be recorded from the extraocular muscles using surface electrodes placed near the eyes. These “ocular VEMPs” (oVEMPs) are a manifestation of the vestibulo-ocular reflex. Here we describe the historical development and neurophysiological properties of the cVEMP and oVEMP and provide recommendations for recording both reflexes. While the cVEMP has documented diagnostic utility in many disorders affecting vestibular function, relatively little is known as yet about the clinical value of the oVEMP. We therefore outline the known cVEMP and oVEMP characteristics in common central and peripheral disorders encountered in neuro-otology clinics. [Copyright &y& Elsevier]

Abstract: Objective: The n10 component (n10) of the ocular vestibular evoked myogenic potential (oVEMP) to brief bone conducted vibration (BCV) of the forehead at Fz is probably caused by the vibration selectively activating vestibular otolithic receptors. If the n10 is due primarily to utricular activation then diseases which affect only the superior division of the vestibular nerve (SVN) should reduce or eliminate n10. Methods: The n10 component of the oVEMP was measured in 13 patients with unilateral SVN but with inferior vestibular nerve function preserved. Results: We compared the n10 to BCV of these 13 SVN patients to previously published data for healthy subjects and patients after complete unilateral vestibular loss. We found that in 12 out of the 13 patients with SVN, n10 was markedly reduced or absent under the contralesional eye. Conclusion: Since all utricular afferents course in the superior vestibular nerve and in 12/13 of these patients the n10 was reduced we conclude that the n10 component of the oVEMP to BCV is probably mediated by the superior vestibular nerve and probably due to activation of mainly utricular receptors. Abstract: Objective: Synchronous extraocular muscle activity can be recorded from around the eyes at the beginning of a vestibular-evoked eye movement (ocular vestibular evoked myogenic potentials, OVEMPs). As galvanic vestibular stimulation (GVS) evokes the vestibulo-ocular reflex, we wished to investigate GVS-evoked OVEMPs. Methods: We stimulated 10 normals and 6 patients with unilateral vestibular loss (uVL) with bi/unipolar 4mA, 2ms current steps at the mastoid. OVEMPs were recorded from electrodes placed superior and inferior to the eyes. Results: OVEMPs were present beneath both eyes in all normal subjects: an initial positivity ipsilateral to the cathodal electrode (peak latency 9.9ms, amplitude 1.3μV) and an initial negativity contralateral to the cathode (8.8ms, 2.4μV). In the patients, stimulation of the affected side produced little or no response. Stimulation of the intact side produced only contralateral responses. Conclusions: The infra-orbital response is likely produced primarily by the inferior obliques, producing conjugate torsion away from the cathode. The projection to the ipsilateral eye depends upon normal vestibular function on the contralateral side. Significance: OVEMPs can be evoked by GVS. While bilateral effects are obtained with unilateral stimulation in normals, the primary vestibular pathway to the inferior oblique in humans is crossed. [Copyright &y& Elsevier]

Abstract: Recent studies have shown that the vection in depth experienced by stationary observers viewing constant velocity radial flow can be enhanced by adding simulated viewpoint jitter/oscillation. This study examined the effect of manipulating visual–vestibular conflict on the perceived strength and speed of vection in depth. Four conditions were examined: (i) radial flow without viewpoint jitter viewed by stationary observers (consistent visual–vestibular inputs); (ii) radial flow with viewpoint jitter synchronized to lateral head oscillation (consistent inputs); (iii) radial flow with viewpoint jitter viewed by stationary observers (inconsistent inputs); (iv) radial flow without viewpoint jitter viewed during head oscillation (inconsistent inputs). We found that the strength and perceived speed of vection in depth was always greater when simulated viewpoint jitter was introduced. No further vection enhancement was found when this jitter was generated by active head oscillation—even though passive jitter conditions should have generated significant sensory conflicts, whereas active jitter conditions would not. Active head oscillation without display jitter also had little effect, producing similar vection strength/speed ratings to stationary observation of non-jittering optic flow. Horizontal eye tracking suggested that retinal stimulation was similar between comparable active and passive viewing conditions. This stabilization of the retinal image across active and passive conditions appeared to be due to cooperative engagement of the translational vestibuloocular reflex and the visually driven ocular following response. Rather than providing evidence for synergistic integration of self-motion perception, these findings obtained with low-frequency sensory stimuli suggest that self-motion perception is dominated by visual processing centres. Abstract: Mechanoreceptive hair-cells of the vertebrate inner ear have a remarkable sensitivity to displacement, whether excited by sound, whole-body acceleration or substrate-borne vibration. In response to seismic or substrate-borne vibration, thresholds for vestibular afferent fibre activation have been reported in anamniotes (fish and frogs) in the range −120 to −90dB re 1 g. In this article, we demonstrate for the first time that the human vestibular system is also extremely sensitive to low-frequency and infrasound vibrations by making use of a new technique for measuring vestibular activation, via the vestibulo-ocular reflex (VOR). We found a highly tuned response to whole-head vibration in the transmastoid plane with a best frequency of about 100Hz. At the best frequency we obtained VOR responses at intensities of less than −70dB re 1 g, which was 15dB lower than the threshold of hearing for bone-conducted sound in humans at this frequency. Given the likely synaptic attenuation of the VOR pathway, human receptor sensitivity is probably an order of magnitude lower, thus approaching the seismic sensitivity of the frog ear. These results extend our knowledge of vibration-sensitivity of vestibular afferents but also are remarkable as they indicate that the seismic sensitivity of the human vestibular system exceeds that of the cochlea for low-frequencies. Abstract: Objective: Patients with superior canal dehiscence (SCD) typically have enhanced sound-evoked vestibular reflexes, such as vestibulo-collic and vestibulo-ocular reflexes. We wished to investigate whether sound-evoked lower limb EMG responses and postural sway are also enhanced in this condition. Methods: Eight patients with CT confirmed SCD (11 affected ears) and 8 age-matched normal controls participated. Three sound-evoked responses were measured; vestibulo-collic reflexes (i.e. vestibular-evoked myogenic potentials, VEMPs), lower limb vestibulo-spinal reflexes and body sway (centre of pressure in mm). Sound stimuli were 500Hz air-conducted tone bursts of varying lengths (VEMPs: 2ms; vestibulo-spinal: 20ms; sway: 1s and 200ms) set at fixed levels above each subject’s VEMP threshold. Results: SCD patients had very large VEMP and vestibulo-spinal responses following high intensity stimulation, but at the matched intensity of 15dB above threshold amplitudes were similar in both SCD patients and controls. The amplitude of both responses increased linearly with increasing stimulus intensity in both groups. Large (∼20mm), stereotyped sway responses were present in only one (atypical) patient with high intensity stimulation. Small (∼2mm) sway responses were present in the remaining patients, and began immediately following the vestibulo-spinal responses. Conclusions: Despite the presence of large vestibular reflexes, there is usually very little body sway in response to loud sounds in SCD patients. Abstract: We studied ocular asymmetries and orienting responses induced by angular rotation in rabbits with binocular video recordings. Slow phase velocities were significantly larger in the eye moving temporonasally than nasotemporally. The eyes also converged and pitched down during rotation, which increased and refocused binocular overlap in the visual fields. Eye position also shifted into the slow phase direction. Vergence and pitch outlasted the induced nystagmus, suggesting that they were generated by a separate vestibulo-oculomotor subsystem(s). Thus, mechanisms in the rabbit increase compensatory eye velocity in the eye that leads into the direction of rotation and enhance binocular vision. [Copyright &y& Elsevier]

Abstract: The orientations of the semicircular canals determines the response of the canals to head rotations and, in turn, the brain’s ability to interpret those motions. The geometry of chinchillas’ semicircular canals has never been reported. Volumetric representations of three chinchilla skulls were generated using a microCT scanner. The centroids of each semicircular canal lumen were identified as they passed through the image slices and were regressed to a plane. Unit vectors normal to the plane representing canal orientations were used to calculate angles between canal pairs. Pitch and roll maneuvers required to bring any canal into the horizontal plane for physiologic investigation were calculated. The semicircular canals of the chinchilla were found to be relatively planar. The horizontal canal was found to be oriented 55.0° anteriorly upward. Pairs of ipsilateral chinchilla canals were not orthogonal and contralateral synergistic pairs were not parallel. Despite this arrangement, the canal plane unit normal vectors were organized to respond with approximately equal overall sensitivity to rotations in any direction. The non-orthogonal chinchilla labyrinth may provide an opportunity to determine whether the frame of reference used by the central vestibular and oculomotor system is based on directions of afferent maximum sensitivity or prime directions. Abstract: We describe a novel, inexpensive method for real-time measurement of binocular three-dimensional eye position. The method employs consumer-grade digital video cameras (“webcams”) to track an array of three fluorescent non-collinear markers affixed to each eye. The instantaneous position of the marker array relative to a reference position is used to construct a rotation matrix describing the eye rotation. The mathematical computation used to determine the rotation matrix is conceptually simpler and computationally more efficient than methods previously described, allowing generation of binocular three-dimensional eye position in real-time during image acquisition. The fluorescent marker is illuminated using a UV-A light source. The light source and reflective artifacts are filtered out to improve the signal to noise ratio. In addition, we present a method to align the camera with the center of eye rotation. When tested in vitro, the video-oculography (VOG) method had a &#60;2.9% positional error (in each component of 3-D eye position) for eye positions within 20° of center. We directly compared this method of VOG to the search coil technique by measuring three-dimensional eye position simultaneously using search coils and VOG in a chinchilla (C. laniger). The horizontal rotatory vestibulo-ocular reflex (VOR) stabilizes gaze by moving the eyes at an angular velocity proportional to head velocity, and can accomplish this for a broad range of frequencies and amplitudes of head motion. Rotation at 5 Hz and above may be processed differently than lower frequencies by the VOR network. We recorded discharges and calculated spike densities of a small sample of vestibular neurons in alert cats during low-velocity rotation at frequencies up to 8 Hz. At high frequencies, we found both vestibular-only (V-only) and eye-movement-sensitive (EM) cells that generated asymmetric output signals. Asymmetry was primarily of the cutoff type, i.e., changes in spike density were smallest for rotation in the inhibitory direction. Most cells were identified as secondary neurons. The mean spike density was 23 sp/s, which was lower than previously reported in vestibular neurons of monkeys. A few neurons had very high sensitivities, associated with phase-locking, to rotation at high frequencies. In general, vestibular neurons carried a high-pass-filtered version of rotational signals. When synaptic inputs from the vestibular commissure were quantified, we found that the immediate change in probability of firing due to commissural vestibular input was inversely correlated with the degree of high-pass filtering. At high frequencies, increased asymmetry and phase-locking occurred in some neurons. A small number of neurons responded with increased probability of firing to both directions of rotation. Together, these observations suggest that high frequencies of rotation may be encoded differently than low frequencies by central vestibular neurons in alert animals. [Copyright &y& Elsevier]

Despite nearly perfect gaze stability during natural head movements, the amplitude of the vestibulo-ocular reflex during passive head and body translation (TVOR) has been consistently reported to be undercompensatory during near target viewing. Here we have compared the rhesus monkey TVOR during pure head and body translation with the eye movements generated during eccentric yaw rotations, where both semicircular canal and otolith signals are activated. We found a significant increase in both the near target TVOR amplitude and its viewing distance dependence during eccentric rotations, as compared to pure translations. We conclude that the simultaneous activation of the horizontal semicircular canals result in an improvement of the viewing distance-dependence of the rhesus monkey TVOR. [Copyright &y& Elsevier]

This article reviews studies of the organization and development of the vestibulo-ocular reflex arc in the chicken embryo. It summarizes some of the principal features that characterize the development of this circuit, including the gradual clustering of motoneurons in the oculomotor nucleus into functionally identifiable motoneuron pools, the patterning of vestibular projection neurons into coherent clusters with specific axonal trajectories and terminations onto the oculomotor motoneuron pools, the reverse order of synapse formation during development (motoneuron to muscle, then vestibular projection neuron to motoneuron), and the selectivity of initial synaptic termination at both the ultimate and penultimate relays within the reflex arc. Reference to studies in other vertebrate species is made to provide a comparative context, and potential mechanisms are discussed that may contribute to the underlying synaptic specificity in this circuit. Right brain damaged patients affected by left unilateral neglect (N+) typically fail to explore the contralesional space. For the first time, this study investigates the dynamic and spatial features of the horizontal vestibular–ocular response (VOR), the optokinetic response (OKR) and the VOR–OKR interaction in six N+ and in five right brain damaged patients without neglect (N−). No lateral asymmetry of the gain (i.e. eye velocity to head velocity ratio) of VOR slow phases was found in either group. In the VOR, N+ had higher frequency of slow-rightward/fast-leftward phases and higher contralesional shift of the beating field (i.e. orbital position of fast phases). In the VOR–OKR, there was an increase of gain in both lateral directions and in both groups even though in N−, there was a lower phase shift between eye and head velocity. In contrast to the VOR, in the VOR–OKR, N+ had higher frequency of slow-leftward/fast-rightward phases. The VOR–OKR interaction also introduced an ipsilesional shift of the beating field in both N+ and N−. In the OKR, N+ showed a drop in the velocity, amplitude and frequency of slow-rightward/fast-leftward phases. These findings potentially suggest that each hemisphere modulates VOR with contralaterally directed slow phases and OKR with ipsilaterally directed slow phases. This organisation could facilitate maintenance or fast recovery of combined VOR + OKR after unilateral brain damage. The same findings suggest that by inducing slow-leftward phases, vestibular and optokinetic stimulation improve left side neglect through the activation of different hemispheric pathways. No ipsilesional deviation of the subjective “straight ahead” was found in N+. These results show that chronic unilateral neglect can be dissociated both from deficits of ipsilesionally directed VOR and from ipsilesional deviation of the subjective midsagittal plane of the body. The present study investigated vestibulo-ocular reflex (VOR), optokinetic reflex (OKR) and postural function in patients with rheumatoid arthritis (RA). Compared with controls, no differences in gaze-holding, VOR gain or phase, OKR slow phase velocity (SPV) or quick phase amplitude, optokinetic afternystagmus SPV or duration, or latency to the illusion of circularvection, were found. RA patients did exhibit greater sway in the leftward direction (P<0.01), however, this was no greater in the conditions of the Clinical Test of Sensory Interaction and Balance that increase reliance upon vestibular information. Gravity provides an absolute verticality reference for all spatial perception, allowing us to move within and interact effectively with our world. Bayesian inference models explain verticality perception as a combination of online sensory cues with a prior prediction that the head is usually upright. Until now, these Bayesian models have been formulated for judgements of the perceived orientation of visual stimuli. Here, we investigated whether judgements of the verticality of tactile stimuli follow a similar pattern of Bayesian perceptual inference. We also explored whether verticality perception is affected by the postural and balance expertise of dancers. We tested both the subjective visual vertical (SVV) and the subjective tactile vertical (STV) in ballet dancers and non-dancers. A robotic arm traced downward-moving visual or tactile stimuli in separate blocks while participants held their head either upright or tilted 30° to their right. Participants reported whether these stimuli deviated to the left (clockwise) or right (anti-clockwise) of the gravitational vertical. Tilting the head biased the SVV away from the longitudinal head axis (the classical E-effect), consistent with a failure to compensate for the vestibulo-ocular counter-roll reflex. On the contrary, tilting the head biased the STV toward the longitudinal head axis (the classical A-effect), consistent with a strong upright head prior. Critically, tilting the head reduced the precision of verticality perception, particularly for ballet dancers' STV judgements. Head tilt is thought to increase vestibular noise, so ballet dancers seem to be surprisingly susceptible to degradation of vestibular inputs, giving them an inappropriately high weighting in verticality judgements. • Vestibular, proprioceptive, and external cues contribute to verticality perception. • The subjective tactile vertical is biased toward the direction of a head tilt. • The subjective visual vertical is biased away from the direction of a head tilt. • Ballet dancers are particularly susceptible to vestibular noise caused by tilts.
