The objective of this study was to quantify the farm gate nitrogen (N) offset potential of perennial ryegrass (Lolium perenne L.; PRG) white clover (Trifolium repens L.; WC) swards by comparing the herbage and milk production from dairy farmlets that were simulations of full farming systems. A study was established where 120 cows were randomly assigned to 4 farmlets of 10.9 ha (stocking rate: 2.75 cow/ha), composed of 20 paddocks each. Cows were fed 526 kg of DM of concentrate on average each year. The 4 grazing treatments were PRG-only at 150 or 250 kg of N/ha and PRG-WC at 150 or 250 kg of N/ha. Cows remained in their treatment group for an entire grazing season and were re-randomized as they calved across treatments each year. As cows calved in the spring as standard practice in Ireland, they were rotationally grazed from early February both day and night (weather permitting) to mid-November, to a target postgrazing sward height of 4.0 cm. Mean sward WC content was 18.1% and 15.4% for the 150 and 250 kg of N/ha PRG-WC treatments, respectively over the 3-yr period. When WC was included, lowering the N rate did not reduce pregrazing yield, pregrazing height, or herbage removed, but those factors decreased significantly when WC was absent. Total annual herbage DM production was 13,771, 15,242, 14,721, and 15,667 kg of DM/ha for PRG-only swards receiving 150 or 250 kg of N/ha and PRG-WC swards receiving 150 or 250 kg of N/ha, respectively. In addition, when WC was present, compressed postgrazing sward heights were lower (4.10 vs. 4.21 cm) and herbage allowance (approximately 17 kg/cow feed allocation per cow per day) higher than the high-N control (+ 0.7 kg of DM/cow per day). There was a significant increase in milk production, both per cow and per hectare, when WC was included in PRG swards. Over the 3-yr study, cows grazing PRG-WC had greater milk (+304 kg) and milk solids (+31 kg of fat + protein) yields than cows grazing PRG-only swards. This significant increase in milk production suggests that the inclusion of WC in grazing systems can be effectively used to increase milk production per cow and per hectare and help offset nitrogen use. This result shows the potential to increase farm gate N use efficiency and reduce the N surplus compared with PRG-dominant sward grazing systems receiving 250 kg of N/ha, without negatively affecting milk solids yield or herbage production, thus increasing farm profit by €478/ha. This study investigated the effects of acacia (extract of Acacia mearnsii) and sainfoin (Onobrychis viciifolia) as condensed tannin (CT)-rich sources on ruminal and manure methane (CH 4) emissions in comparison with non-CT silages characterized by different contents of the cell wall and water-soluble carbohydrates. In a 3 × 6 incomplete Latin square design, 30 Holstein cows (63 ± 23 d in milk; mean ± SD; 33.8 ± 7.6 kg of milk per day, body weight 642 ± 81 kg) were provided with ad libitum access to 1 of 6 total mixed rations comprising 790 g of silage and 210 g of concentrate per kilogram of dry matter (DM). The silages were either rich in sainfoin [neutral detergent fiber (NDF): 349 g/kg of DM], perennial ryegrass (NDF: 420 g/kg of DM), or red clover (NDF: 357 g/kg of DM). Each silage was supplemented with 20 g/kg (of total diet DM) of acacia or straw meal. Feed intake and milk yield were recorded daily. Milk composition and ruminal fluid characteristics and microbiota were analyzed. The individual ruminal CH 4 production was determined using the GreenFeed system, and CH 4 emissions from the manure of cows fed the same diets were measured in a parallel experiment over 30 d at 25°C using a dynamic flux chamber. The CT sources did not reduce CH 4 yield or emission intensity. Acacia reduced milk production (from 26.3 to 23.2 kg/d) and DM intake (from 19.7 to 16.7 kg/d) when supplemented with ryegrass, and both CT sources reduced the milk protein content and yield. Acacia supplementation and ryegrass silage reduced the ruminal acetate:propionate ratio. Furthermore, during acacia treatment, the abundance of Methanobrevibacter archaea tended to be lower and that of Thermoplasmata was higher. Acacia reduced the CH 4 emissions from manure for the ryegrass group by 17% but not for the sainfoin and clover groups. Feeding sainfoin silage resulted in the lowest manure-derived CH 4 emissions (−47% compared with ryegrass). In conclusion, acacia reduced ruminal CH 4 production by 10%, but not emission intensity, and the mitigation effect of sainfoin depended on the silage to which it was compared. Because mitigation was partially associated with animal productivity losses, careful evaluation is required before the implementation of tanniferous feeds in farm practice. This study aims to investigate the ability of an imidazolium biobased Zwitterionic Ionic Liquids (ZILs) in enhancing the phytoavailability of copper from garden (G) and vineyard (V) soils using the model plant ryegrass. Uncontaminated and artificially contaminated CuSO 4 soils, unamended and ZIL-amended soil modalities were designed. The copper/ZIL molar ratio (1/4) introduced was rationally established based on molecular modeling and on the maximal copper concentration in artificially contaminated soil. Higher accumulation of copper in the shoots was detected for the uncontaminated and copper contaminated ZIL amended V soils (18.9 and 23.3 mg/kg, respectively) contrary to G soils together with a ZIL concentration of around 3% (W/W) detected by LC-MS analyses. These data evidenced a Cu-accumulation improvement of 38% and 66% compared to non-amended V soils (13.6 and 13.9 mg/kg respectively). ZIL would be mainly present under Cu(II)-ZIL 4 complexes in the shoots. The impact on the chemical composition of shoot was also studied. The results show that depending on the soils modalitity, the presence of free copper and/or ZIL led to different chemical compositions in lignin and monomeric sugar contents. In the biorefinery context, performances of enzymatic hydrolysis of shoots were also related to the presence of both ZIL and copper under free or complex forms. Ecotoxicity assessment of the vineyard soil samples indicated that the quantity of copper and ZIL remaining in the soils had no significant toxicity. The aim of the study was to investigate the effects of substituting silage of chopped grass with pulp silage of grass fractionated once or twice in a biorefinery using a screw press on fiber kinetics, protein value, and production of CH 4 in dairy cows. Six lactating multiparous Holstein cows in mid-lactation (176 ± 93 d in milk), cannulated in the rumen, duodenum, and ileum, were used in an incomplete 6 × 4 Latin square design with a 2 × 3 factorial arrangement of treatments. Perennial ryegrass was harvested in third regrowth from the same field at early and late developmental stage (35 and 44 d of regrowth, respectively) and subjected to 1 of 3 types of processing within each developmental stage. Grass was either harvested for normal silage making (mowed, wilted, chopped, and ensiled), or harvested fresh and fractionated using a screw press. Half of the pulp from the first fractionation was ensiled, whereas the other half of the pulp was rehydrated, fractionated a second time, and pulp hereof was ensiled. The grass and pulp silages were used with concentrates (65:35 forage to concentrate ratio) to make total mixed rations (TMR) based on either silage of chopped grass (GS), pulp silage of grass fractionated once (1×P), or pulp silage of grass fractionated twice (2×P), harvested either at early (E) or late (L) developmental stage resulting in 6 different TMR treatments (EGS, E1×P, E2×P, LGS, L1×P, L2×P). The TMR were fed for ad libitum intake and samples of intestinal digesta and feces were collected for determination of digestibility. The effect of processing on ash-free neutral detergent fiber (aNDFom) concentration in silages depended on developmental stage, but showed that within each developmental stage, pulp silage of grass fractionated twice had higher aNDFom concentration than pulp silage of grass fractionated once and silage of chopped grass. The 2×P resulted in lower (14.9 ± 0.55 vs. 17.5 ± 0.54 kg/d) dry matter intake (DMI) compared with GS. The effects of processing and developmental stage interacted such that apparent total-tract aNDFom digestibility was higher (784 ± 13 vs. 715 ± 13 g/kg) for L2×P compared with LGS, whereas no difference was found between E2×P and EGS. Moreover, the protein value was higher (106 ± 5 vs. 92 ± 5 g AA digested in the small intestine/kg of DMI) for 2×P compared with GS. Unexpectedly, processing had no effect on fractional rate of digestion of digestible aNDFom or CH 4 yield (L/kg of DMI), whereas feeding forages harvested at early compared with late developmental stage resulted in lower CH 4 yield. Feeding pulp silage of grass fractionated once generally yielded results intermediate to cows fed silage of chopped grass and pulp silage of grass fractionated twice. This study showed that pulp silage of fractionated grass could serve as feed for dairy cows because the fiber digestibility and protein value improved, but further research investigating effects of physical processing of forage on fiber kinetics is required. α-Ketoglutarate (AKG) is a key intermediate metabolite in the tricarboxylic acid cycle of respiration and a precursor for glutamate, playing important roles in regulating plant growth and stress tolerance. The objectives of this study were to examine effects of AKG on heat tolerance characterized by leaf senescence in a cool-season grass species by foliar application and to determine major metabolites and associated metabolic pathways regulated by AKG for its effects on heat tolerance. Perennial ryegrass (Lolium perenne L.) plants were exposed to heat stress (35/30 °C, day/night) or optimal temperature (25/20 °C, day/night, non-stress control) in controlled-environment growth chambers. The solution containing AKG (5 mM) was applied to leaves by spraying 7 d prior to the initiation of heat stress and every 7 d during the heat stress period. Exogenous application of AKG enhanced heat tolerance in perennial ryegrass, as manifested by significant increases in leaf chlorophyll content, photochemical efficiency, and membrane stability, as well as activities of antioxidant enzymes for H 2 O 2 scavenging in AKG-treated plants relative to untreated control plants exposed to heat stress. Metabolic profiling and pathway analysis demonstrated that exogenous AKG application enhanced metabolite accumulation in four major metabolic pathways, including antioxidant metabolism, amino acid metabolism, glycolysis and tricarboxylic acid cycle of respiration, and pyrimidine metabolism, contributing to AKG-improved heat tolerance in perennial ryegrass. • α-Ketoglutarate (AKG) enhanced heat tolerance in perennial ryegrass. • AKG suppressed heat-induced leaf senescence. • AKG regulated the metabolism of antioxidant, amino acids, pyrimidine, glycolysis, and TCA cycle of respiration. Plantain (Plantago lanceolata) is an herb used to reduce the forage deficit of ryegrass-based pastures during the summer. This herb is being promoted for its reduced environmental impact in terms of nitrogen emissions, particularly reducing urinary nitrogen. However, the effect of plantain on emissions of enteric CH 4 , the main greenhouse gas produced from ruminant-based production systems, is not known. The aim of the present trial was to determine CH 4 emissions and rumen fermentation characteristics of nonlactating dairy cows fed 100% plantain (PLT) or 100% perennial ryegrass (RG; Lolium perenne) in 2 experiments (E1 and E2). The forages were in a vegetative growth stage in E1 and were in a reproductive growth stage in E2. Methane emissions from 16 cows in each experiment were measured in respiration chambers for 2 d. Methane emissions per unit of dry matter intake (CH 4 yield) were 15 and 28% less for cows fed PLT than those fed RG in E1 and E2, respectively. Dry matter digestibility of PLT was 7 and 27% less than that of RG in E1 and E2, respectively, and CH 4 per unit of dry matter digested was similar for PLT and RG in both experiments. There were only minor (but some significant) differences in rumen fermentation characteristics between cows fed PLT and RG in both experiments. In conclusion, CH 4 yield was lower for cows fed PLT compared with those fed RG in both experiments and this reduction was largely explained by the lesser dry matter digestibility of PLT. Protein extracts from green leaves, consisting of mainly RuBisCO, have potential applications for human consumption. However, their properties are highly influenced by the extraction process applied. This work assessed the impact of the extraction method on the solubility profile and emulsifying properties of two protein concentrates from perennial ryegrass, obtained using two different approaches. These two methods differed in the application (or absence) of a heating step (50 °C for 30 min) to improve the removal of chloroplast material. The functionality of both protein concentrates was highly influenced by the isoelectric point of the proteins. The extract containing less residual chloroplast material showed greater solubility, up to 40 % more, and superior emulsifying capacity and stability, at non-acidic pH, similar to WPI. At pH above 6, the protein extract which contained residual chloroplast material was significantly less soluble, which limited its performance as emulsifier. Therefore, the extraction process critically impacted the final product's functionality. This work demonstrated that effectively removing chloroplast material is essential to achieve targeted techno-functional properties of an ingredient and the food product in which it is used. While plant toxicity reduction remains the primary metric for judging the success of metal immobilization in soil, the suitability of microorganisms as universal indicators of its effectiveness in various contaminated soils remains a point of contention. This study assessed the sensitivity of microbial bioindicators in monitoring metal immobilization success in smelter-impacted soils. It compared plants and microorganisms as indicators of the efficiency of natural Fe–Mn nodules from the Gulf of Finland in immobilizing metals in soils contaminated by a Ni/Cu smelter, on the Kola Peninsula, Murmansk region, Russia. Perennial ryegrass (Lolium perenne) was grown on nodule-amended and control soils. Plant responses in the smelter-impacted soils proved to be sensitive and robust indicators of successful metal immobilization. However, microbial responses exhibited a more complex story. Despite the observed reductions in soluble metal concentrations, shoot metal contents in ryegrass, and significant improvements in plant growth, certain microbial bioindicators were unresponsive to metal immobilization success brought about by the addition of Fe–Mn nodules. Among microbial bioindicators studied, community-level physiological profiling, microbial biomass carbon, and basal respiration were sensitive indicators of metal immobilization success, whereas the number of saprotrophic, oligotrophic, and Fe-oxidizing bacteria and fungi, the biomass of bacteria and fungi, and enzymatic activity were less robust indicators. Interestingly, the correlations between different microbial responses measured were weak or even negative. Some microbial responses also exhibited negative correlations with plant biomass. These findings underscore the need for further research on comparative evaluations of plants and microorganisms as reliable indicators of metal immobilization efficacy in polluted environments. Perennial ryegrass (Lolium perenne) is a fructan-accumulating plant constituting one of the most important grassland species with high herbage production, nutritive value and digestibility for grazing cattle. Although fructans were reported to be involved in plant defense acting as antioxidants or stress signals, their contribution in root protection is still to be explored. In roots, atypical defense is provided by the "Root Extracellular Trap" or "RET" at the root-soil interface. The molecular composition and structural organization of the RET are essential to provide root defense against pathogen attacks and abiotic stresses. The RET was reported to be mainly composed of polysaccharides (homogalacturonan, xylogalacturonan, xyloglucan) and proteoglycans such as arabinogalactan proteins (AGPs). Our aim is to characterize the RET composition of L. perenne using cell imaging techniques and a wide range of monoclonal antibodies directed against epitopes from cell wall glycomolecules and to investigate the potential presence of fructans. Interestingly, we found that both mucilage and cell wall surface of border cells were enriched in AGP epitopes. An increased amount of the AGP-containing mucilage was produced by L. perenne root tip in response to both elicitor and osmotic stress. Fructan epitopes were also detected in root cap cells and appeared to be released in the RET under stress conditions. Australia has tropical, summer dominant rainfall in the north transitioning to temperate, winter dominant rainfall in the south. Temperate pasture species with a cool season growth pattern are commonly grown in the south, but tropical pasture species that are highly productive and responsive to summer rainfall, may also have a role in southern pasture systems with climate change. Determine the value of replacing temperate pastures with tropical pastures; and assess the emergence success of tropical grasses in response to historic climate change in eastern Australia. The AusFarm model was used to test replacing lucerne (Medicago sativa L.) (L); annual ryegrass (Lolium rigidum L.)/subterranean clover (Trifolium subterraneum L.) (RC); or lucerne/annual ryegrass/subterranean clover (LRC) with tropical pasture at varying rates of inclusion at five locations across the subtropical-temperate transition zone. The emergence success of tropical grasses with a bare fallow, short fallow and increased ground cover management was also tested. The modelling covered two periods, the Far Climate (1960–1990) and the Near Climate (1990–2020). Tropical pastures had higher plant growth in late summer-early autumn when temperate pastures are less active. Pasture growth decreased from the Far to the Near Climate period across all sites. The pastures with L lost the most production over this period, with minimal change in other pasture types. Supplementary feeding was reduced at all sites with the inclusion of tropical pastures. The cost of supplementary feeding from the Far to the Near Climate increased by $20/ha with increased variability between years, which in-turn decreased gross margin by an average of $26/ha. Tropical pastures improved profitability on average by $24/ha (11%), but with different responses between environments. At the low rainfall sites (Condobolin and Leeton) there was a substantial benefit when tropical grass-clover (TC) pasture replaced L, but at the higher rainfall sites (Cowra and Wagga Wagga) the benefit was replacing RC with TC. There was a gradient of successful emergence from very high (>90%) in the north, to reduced opportunities in the south (<45%), and marginal influence of sowing management. Adding tropical pastures to grazing systems with temperate pastures increased summer-autumn growth, the animal requirements met by pasture, and gross margins. Profitability of livestock systems decreased with climate change, but this can be overcome by optimising pasture composition. Complex rhizoremediation is the main mechanism of phytoremediation in organic-contaminated soil. Low molecular weight organic acids (LMWOAs) in root exudates have been shown to increase the bioavailability of contaminants and are essential for promoting the dissipation of contaminants. The effects of root exudates on the dissipation of organophosphate esters (OPEs) in soil are unclear. Consequently, we studied the combined effects of root exudates, soil enzymes and microorganisms on OPEs (tri (1-chloro-2-propyl) phosphate (TCPP) and triphenyl phosphate (TPP)) dissipation through pot experiments. Oxalic acid (OA) was confirmed to be the main component of LMWOAs in root exudates of ryegrass. The existence of OA increased the dissipation rate of OPEs by 6.04%–25.50%. Catalase and dehydrogenase activities were firstly activated and then inhibited in soil. While, urease activity was activated and alkaline phosphatase activity was inhibited during the exposure period. More bacteria enrichment (e.g., Sphingomonas , Pseudomonas , Flavisolibacter , Pontibacter , Methylophilus and Massilia) improved the biodegradation of OPEs. In addition, the transformation paths of OPEs hydrolysis and methylation under the action of root exudates were observed. This study provided theoretical insights into reducing the pollution risk of OPEs in the soil. Cadmium (Cd) and arsenic (As) are two highly toxic heavy metals and metalloids that coexist in many situations posing severe threats to plants. Our investigation was conducted to explore the different regulatory mechanisms of ryegrass (Lolium perenne L.) responding to individual and combined Cd and As stresses in hydroponics. Results showed that the ryegrass well-growth phenotype was not affected by Cd stress of 10 mg·L-1. However, As of 10 mg·L-1 caused rapid water loss, proline surge, and chlorosis in shoots, suggesting that ryegrass was highly sensitive to As. Transcriptomic analysis revealed that the transcription factor LpIRO2 mediated the upregulation of ZIP1 and YSL6 that played an important role in Cd tolerance. We found that the presence of As caused the overexpression of LpSWT12 , a process potentially regulated by bHLH14, to mitigate hyperosmolarity. Indoleacetic acid (IAA) and abscisic acid (ABA) contents and expression of their signaling-related genes were significantly affected by As stress rather than Cd. We predict a regulatory network to illustrate the interaction between transporters, transcription factors, and signaling transduction, and explain the antagonism of Cd and As toxicity. This present work provides a research basis for plant protection from Cd and As pollution. In this study, municipal waste pyrolytic char (PEWC) was prepared by pyrolysis from municipal solid waste extracted in landfills, and Fe-based modified pyrolytic char (Fe-PEWC) was prepared by modification. Focusing on the evaluation of the stabilization capacity of Fe-PEWC for vanadium (V) and chromium (Cr) in soils, the effects of PEWC addition on soil properties, bioavailability and morphological distribution of V and Cr, ryegrass growth, and V and Cr accumulation were thoroughly investigated. The results of pot experiment showed that the application of PEWC and Fe-PEWC significantly (P < 0.05) improved soil properties (such as pH, EC, total nitrogen, available phosphorus, available potassium, and organic matter). After 42 days of cultivation, Fe-PEWC has a better fixation effect on heavy metals, and the bioavailable V and Cr of 3% Fe-PEWC decreased by 14.96% and 19.48%, respectively. The exchangeable state and reducible state decreased, while the oxidizable state and residual state increased to varying degrees. The Fe-PEWC can effectively reduce the accumulation of V and Cr in ryegrass by 71.25% and 76.43%, respectively, thereby reducing their toxicity to plants. In summary, modified pyrolytic char can effectively solidify heavy metals in soil, improve soil ecology and reduce the toxicity to plants. The use of excavated waste as a raw material for the preparation of soil heavy metal curing agent has the significance of resource recycling, low price, and practical application. In this study, a straw-based degradable mulch film was prepared with a similar heat and moisture retention effect to polyethylene mulch film and good magnetically controlled-release performance for glyphosate to promote growth of crop and inhibit grass simultaneously. To solve three main problems in agriculture (plastic mulch pollution, difficult straw recycling, and low utilization efficiency of pesticides), a multifunctional mulch film (MF) was fabricated. In this study, corn straw was mixed with xanthan gum, glyphosate (Gly), Fe 3 O 4 , and MIL-100(Fe) to obtain a MF named as SMFG with magnetically controlled-release Gly. To improve the hydrophobicity and stability of SMFG, the surface was coated by amino silicone oil (ASO) to get SMFG@ASO. SMFG@ASO had well insulation and moisture retention properties close to polyethylene MF. Meanwhile, the weight loss ratio of SMFG@ASO increased with time and could reach 59% underground in 120 d, indicating the good biodegradation behavior of SMFG@ASO. Importantly, pot experiment showed that SMFG@ASO could significantly promote growth of crop and inhibit grass. Besides, earthworm experiment illustrated that SMFG@ASO possessed a high biosafety and could increase the relative abundance of beneficial bacteria (Flavobacterium) from 0.08 to 0.21 and soil quality. Epichloë endophytes in grass associations express a myriad of secondary metabolites which can affect the health of grazing animals and reduce the impact of insect pests on pasture. The ideal endophyte-grass association must have a favourable chemical profile such that the impact on animal health is minimised while the beneficial, deterrent effect on insect pests is maximised. A number of endophyte-perennial ryegrass associations have been successfully commercialised but research is on-going to further improve production in farming systems. Secondary metabolites expressed by endophyte-infected tall fescue include lolines, an animal-safe class of compound which imparts a potent effect on insects. Since endophyte-infected perennial ryegrass does not express lolines, a tall fescue endophyte, AR501, was inoculated into perennial ryegrass in an attempt to improve the insect resistance of this pasture type. In addition to animal safety, it is imperative that consideration is given to the safety of humans consuming animal products derived from livestock grazing the novel pasture. Although pure loline alkaloids have previously been tested on mice it is essential that the entire AR501 endophyte-infected perennial ryegrass matrix is tested since this will result in the exposure of both known and unknown secondary metabolites to mice. Three treatment groups each containing 6 male and 6 female mice were fed diets containing AR501 endophyte-infected perennial ryegrass seed (30%), perennial ryegrass seed containing no endophyte (30%) or a diet without seed (control) for 3 weeks. Mice fed control diet ate more than either of the treatment groups fed a diet containing seed. Male mice fed diet containing Nil endophyte seed ate more than those eating AR501 endophyte-infected perennial ryegrass seed although there was no difference observed in the food intake of female mice. While a few statistically significant differences were observed in the haematology and serum biochemical data, in every instance the difference was restricted to only one gender so is considered unlikely to be of toxicological significance. Mice fed AR501 endophyte-infected perennial ryegrass seed remained healthy throughout the experimental period despite consuming 62,000 mg/kg lolines and 4600 mg/kg peramine per day as well as the wide array of other unknown secondary metabolites expressed by this endophyte. Our aim was to evaluate whether increasing soluble carbohydrates in the herbage by changing the time of the grazing session or including Acacia mearnsii tannin in the diet would affect intake, digestion, N partitioning, and productive performance of dairy cows fed a diet combining ryegrass herbage with partial total mixed ration (PMR). We hypothesized that both strategies could reduce the concentration of NH 3 -N in the rumen, reducing urinary N excretion. Nine Holstein cows were used in a triplicate 3 × 3 Latin square experiment with 3 experimental periods of 22 d. The cows were fed a fixed amount of PMR [60% of the predicted individual dry matter intake (DMI)], and an unrestricted amount of herbage in 1 grazing session of 5 h/d. The treatments were (1) morning grazing session and afternoon PMR meal (AM); (2) morning PMR meal and afternoon grazing session (PM); and (3) morning grazing session and afternoon PMR meal supplemented with 15.0 g of tannins/kg of PMR dry matter (TAN). Milk production was not affected by treatments. Although the protein concentration was lower for TAN than for PM, no differences were detected for the yield of any component between treatments. The concentration of individual or grouped fatty acids in milk fat was not affected by treatments, except for 16:1 cis -9 and Δ9-desaturase ratios 14:1/14:0 and 16:1/16:0, which were lower for TAN. Treatments did not affect total DMI, but PM tended to increase herbage DMI and reduce dry matter and crude protein digestibilities. Treatments did not affect cow eating and ruminating behavior except for the proportion of time spent eating PMR, which was higher for PM and TAN. Although no relevant effects of treatments on ruminal fermentation, purine derivatives excretion in urine, or N excretion in milk were detected, both PM and TAN decreased the total N excreted in urine by an average of 8% compared with AM. In conclusion, changing the grazing session from the morning to the afternoon and including tannins in the diet were effective in decreasing the excretion of urinary N but did not change the productive performance of dairy cows fed PMR and ryegrass herbage. Environmental degradation has been attributed to inefficient nitrogen utilization from pastoral dairy production systems. This degradation has especially been associated with the urine patch, which has been identified as a key component of nitrate leaching to waterways. However, a lack of information exists regarding the pattern of urination events and individual urination characteristics across the day, which would help inform strategic management decisions. The aim of this study was therefore to evaluate and report the patterns and characteristics of fecal and urination events throughout the day for cows divergent for milk urea nitrogen breeding values (MUNBV) on either a plantain [ Plantago lanceolata L. (PL)] or ryegrass [ Lolium perenne L. (RG)] diet as ways to reduce environmental impact. Sixteen multiparous lactating Holstein Friesian × Jersey cows divergent for MUNBV were housed in metabolism crates for 72 h, with all excretion events captured and analyzed. Cows selected as low for MUNBV consistently had a 65.2-kg lower urinary urea nitrogen (UUN) load (kg/ha) than high MUNBV cows for all hours of the day when consuming RG. The association between lower urinary urea loading rates and less N leaching implies a reduced environmental impact from low MUNBV cows consuming RG. When cows consumed PL, regardless of MUNBV, they had on average a 137.5-kg (UUN/ha) lower loading rate compared with high MUNBV cows on RG and a 72.2-kg (UUN/ha) lower loading rate compared with low MUNBV cows consuming RG across the day. Cows on PL also exhibited a different diel pattern of UUN load compared with cows consuming RG. Differences in the diel pattern of N excreted in feces were also detected based on MUNBV and by diet, with low MUNBV cows excreting on average 3.06 g more N in feces per event for the majority of the day compared with high MUNBV cows when consuming RG. Lower UUN loading rates and more N excreted in feces indicate a potentially lower environmental impact from low MUNBV cows when consuming RG compared with high MUNBV cows. The use of the PL diet also resulted in lower UUN loading rates and greater levels of N excreted in feces compared with RG, therefore also indicating its ability to reduce environmental impact compared with RG. In sheep production systems, diet type is an important factor affecting the eating quality of the meat produced. The objective of this study was to assess the effect of binary swards on the slaughter performance, descriptive sensory profile, consumer liking and instrumental texture of lamb meat. Five dietary treatments were investigated, perennial ryegrass (Lolium perenne L.; PRG), PRG plus white clover (Trifolium repens L.; PRG+WC), PRG plus red clover (Trifolium pratense L.; PRG+RC), PRG plus chicory (Chicorium intybus L.; PRG+Chic) and PRG plus plantain (Plantago lanceolata L.; PRG+Plan). Post – weaning sixty Texel sired crossbred lambs (n = 12 per dietary treatment) were selected from the main cohort of lambs born in 2022 and further balanced for sex and reared litter size. The M. longissimus thoracis et lumborum (LTL) was obtained from each lamb post-slaughter. Meat samples were analysed by a trained sensory panel, a consumer panel as well as Warner-Bratzler shear force (WBSF) measurements. All data were analysed using a linear mixed model PROC MIXED in SAS. Mean slaughter weights of 46.0 ± 0.36 kg and cold carcass weights of 22.2 ± 0.51 kg were achieved with no differences observed across dietary treatments (P > 0.05). On average, lambs were 217 d of age at slaughter. Lambs finished on PRG and PRG+Plan had similar growth rates, however binary mixtures containing PRG plus any one of WC, RC (P < 0.01) or Chic (P < 0.05) increased average daily gain (ADG) and reduced the number of days required for lambs to reach slaughter (P < 0.05), relative to lambs grazing the PRG monoculture. Meat from PRG+Plan lambs was scored lower by the trained sensory panel for initial tenderness (P < 0.001) and overall tenderness (P < 0.001) when compared to meat from any other dietary treatment. Consumers scored meat from PRG or PRG+WC lambs higher for tenderness (P < 0.05) and flavour (P < 0.01) in comparison to meat produced from PRG+Plan lambs. Relative to meat from PRG+Plan lambs, PRG, PRG+WC and PRG+Chic lamb meat scored higher for overall liking (P < 0.01) when assessed by the consumer panel. Results show that companion forage inclusion increased lamb growth performance in the post-weaning period thus reducing age at slaughter. Multipurpose cropping is promoted to provide biomass to meet the growing demand for biorefining, but excessive biomass removal may decrease soil C stocks and ecosystem multifunctionality. Here, soil C stocks across 0–200 cm depth and soil ecosystem multifunctionality at 0–40 cm were evaluated based on a 19–year field experiment, including three multipurpose cropping systems as continuous cotton (CC), ryegrass–sweet sorghum double cropping (RS) and perennial switchgrass (SG). Compared to CC, RS and SG increased the soil C stocks by 19% and 28%, and N stocks by 19% and 23% at 0–200 cm, respectively. The soil C stocks were positively correlated with belowground C input and negatively associated with C mineralization. RS and SG increased soil ecosystem multifunctionality by 16.8 and 15.7 folds at 0–20 cm and 0.34 and 0.61 folds at 20–40 cm relative to CC, respectively. The benefit of soil C stocks and ecosystem multifunctionality was more pronounced in SG than in RS due to the deeper root system. In conclusion, relative to continuous monoculture, rotational and perennial cropping can enhance soil C sequestration, maintain soil function, and provide feedstock for bioenergy. Green manure application is a feasible agricultural practice to effectively improve soil fertility. However, our understanding of the effects and mechanisms of different green manure residue amounts on soil fertility improvement remains insufficient. Regarding a new type of Italian ryegrass (Lolium multiflorum L.)-rice (Oryza sativa L.) rotation (IRR) system in Southern China, the effects of six Italian ryegrass residue amounts (IRR0: 15,700 kg·ha−1, IRR1: 30,700 kg·ha−1, IRR2: 35,400 kg·ha−1, IRR3: 40,100 kg·ha−1, IRR4: 44,800 kg·ha−1, IRR5: 49,500 kg·ha−1) on the soil nutrient contents, soil enzyme activities, and bacterial communities were evaluated. Compared to the CK, appropriate amounts of Italian ryegrass residue application increased total nitrogen (36.45–37.38 %), soil organic carbon (15.34–26.55 %), microbial biomass carbon (14.07–33.93 %) and microbial biomass nitrogen (30.20–47.65 %) (P < 0.05). The activities of soil polyphenol oxidase, urease, β-glucosidase, peroxidase, cellulase, and total enzyme were also improved significantly in comparison with the CK (P < 0.05). Bacterial community structures in response to IRR were different from those in the CK. The application of Italian ryegrass residue significantly enhanced the relative abundance of beneficial taxa, including Acinetobacter , Pseudomonas , and Arenimonas (P < 0.05). The 30,700–35,400 kg·ha−1 Italian ryegrass residue exhibited the most practical amounts for enhancing soil fertility, soil enzyme activities, and altering soil microbial communities, suggesting that the improvement effects of green manure application on soil were dependent on the residue returning amounts. Perennial ryegrass (Lolium perenne) is a widely used cool season turfgrass with outstanding turf quality and grazing tolerance. High temperature is the key factor restricting the distribution of perennial ryegrass in temperate and sub-tropic regions. In this study, we found that one HEAT SHCOK TRANSCRIPTION FACOTR (HSF) class A gene from perennial ryegrass, LpHSFA3 , was highly induced by heat stress. LpHSFA3 is localized in nucleus and functions as a transcription factor. Ectopic overexpression of LpHSFA3 in Arabidopsis improved thermotolerance and rescued heat sensitive deficiency of athsfa3 mutant. Overexpression of LpHSFA3 in perennial ryegrass enhanced heat tolerance and increased survival rate in summer season as evidenced by decreased EL and MDA, increased number of green leaves and total chlorophyll content. LpHSFA3 binds to the HSE region in LpHSFA2a promoter to constitutively activate the expression of LpHSFA2a and downstream heat stress responsive genes. Ectopic overexpression of LpHSFA2a consequently rescued thermal sensitivity of athsfa3 mutant and enhanced thermotolerance of athsfa2 mutant. Perennial ryegrass protoplasts with overexpression of LpHSFA3 and LpHSFA2a exhibited induction of similar subsets of heat responsive genes. Forestry and fruit industry is a unique and advantageous industry in Xinjiang, China, and it is an important channel for farmers to increase income. With limited land resources and a shortage of forage in southern Xinjiang, farmers began to establish a fruit-grass interplanting model to improve the comprehensive economic benefits of orchards. The objective of this study was to explore the effects of apple/ryegrass intercropping system on root spatial distribution and competition, and to evaluate the performance of field microclimate, soil physical properties and productivity under intercropping. In the study, three different planting patterns were designed: sole-cropped apple (The spacing of the rows of apple trees was 4.0 m, and the plant spacing was 1.0 m), sole-cropped ryegrass (Strip sowing, row spacing was 0.25 m), and apple/ryegrass intercropping (The distance from apple row to ryegrass row was 1.0 m). Two-year field experiments were conducted to measure the root length density (RLD), specific root length (SRL), interspecific competitiveness index, soil moisture content, organic matter content, air temperature, relative humidity, wind speed, yield, land equivalent ratio (LER) and aggressivity. Compared with sole cropping, intercropping reduced the RLD of apple and ryegrass and increased the SRL of ryegrass. The competition space between apple and ryegrass was concentrated in the soil area of 100 cm horizontally and 0–20 cm vertically, and apple was more competitive than ryegrass. Intercropping increased the content of soil moisture and organic matter and improved the microclimate of farmland, decreased the air temperature, increased the relative humidity of an orchard, and decreased wind speed. Although intercropping reduced ryegrass and apple yield, land equivalent ratio (LER) was 1.27–1.30, indicating that intercropping improved land use efficiency. Root competition led to an uneven distribution of root length density (RLD) and specific root length (SRL). The plasticity of root morphology and root spatial distribution was key to adapting to competition and maximizing the absorption of water and nutrients in apple/ryegrass agroforestry systems. Apple/ryegrass intercropping system maximizes resource utilization and reduces interspecies resource competition. Crop nitrogen (N) fertilization diagnoses via the N nutrition index (NNI)-yield relationship have been tested for several crop species, but a cross-species comparison of that relationship has not been performed yet. This study aimed to perform a cross-species comparison of the relationship between NNI and yield with emphasis on the yield sensitivity to N deficiency, slope of the models. Additionally, we conducted an evaluation to determine the best NNI sampling moment to predict relative yield, with focus on major grain crops. Based on a recently published global dataset to parametrize critical dilution curves , we calculated integrated NNI, instantaneous NNI, relative yield, and relative shoot biomass for annual ryegrass, tall fescue, maize, potato, rice, and wheat. We obtained 238 observations to fit integrated NNI-relative yield linear mixed-effects models and 1606 observations to fit instantaneous NNI-relative yield models. Subsequently, we performed a sensitivity analysis to determine the best NNI sampling moment to predict relative yield, with focus on major grain crops (maize, rice, and wheat). Our results show that there was low inter-species variation of sensitivity to N deficiency, i.e., the slope of the relationship between relative yield and integrated NNI. For grain crops, instantaneous NNI around anthesis demonstrated a better predictive capability for relative yield, outperforming other vegetative stages. This finding contributed to improving the understanding of the association between relative yield and NNI with implications for breeding programs, nutrient management practices, and crop modelling. In order to improve the soil remediation efficiency, we used the joint remediation method of magnetically modified biochar and ryegrass to remediate the soil polluted by compound heavy metals (chromium, nickel, copper, zinc, arsenic and cadmium), and evaluate the effect on the process of organic carbon mineralization in polluted soils. It was found that magnetic biochar and ryegrass together decreased the concentrations of Cr, Ni, Cu, Zn, As, and Cd in soils by 24.12 %, 23.30 %, 22.01 %, 9.98 %, 14.83 %, and 15.08 %, respectively, and reduced the available fractions. Ryegrass roots were the main accumulation part of heavy metals, and the order of enrichment effect was ranked as Zn > As > Cr > Cu > Ni > Cd. In addition, magnetic biochar can maintained the stability of the organic carbon pool, and inhibited the emission of volatile organic compounds from ryegrass. Overall, this study indicates that magnetic biochar spheres combined with ryegrass is an effective method for heavy metals co-contaminated soils, and has the excellent remediation ability for actual co-contaminated soils. The temporal profile of methane (CH 4) emissions from ruminants is affected by feed composition and forage type. This study aimed to 1) describe and compare the post-feeding pattern of CH 4 emissions from lambs fed fresh ryegrass substituted with graded levels of forage rape, and 2) to evaluate the association between the magnitude of the variation in CH 4 emission patterns and organic fermentation products quantified in pre- and- post- feeding rumen content samples. Methane emissions were measured at approximately 6 min intervals in respiration chambers from 70 lambs (n = 14 per treatment) fed ryegrass only (FR0) or as a proportion, 0.25 forage rape + 0.75 ryegrass (FR25), 0.50 forage rape + 0.50 ryegrass (FR50), 0.75 forage rape + 0.25 ryegrass (FR75), and forage rape only (FR100). The magnitude of the variability in CH 4 emissions was defined as the ratio of maximum to minimum CH 4 emissions in a 24 h period. This ratio was correlated (P < 0.05) with the proportions of major short chain fatty acids, and calculated hydrogen (H 2) available per unit of glucose fermented (H 2 /GEF), quantified in rumen content samples collected pre- and- post- feeding. The CH 4 emission profile after each feeding followed an asymmetrical positively skewed shape for FR0 to FR75, but not in lambs fed FR100. The lowest CH 4 emission rates were observed before morning feeding which was followed by a CH 4 peak within the 2 h after feeding, and then the CH 4 emissions decreased until the next feeding. In FR100 lambs, the CH 4 emission rate was, unexpectedly, relatively constant after feeding, without any clear CH 4 peaks. The magnitude of the variability in the CH 4 emission rate increased when the pH of rumen contents and H 2 /GEF measured in pre-feeding samples decreased. Including forage rape at 0.75, or greater proportion, in the diet of lambs drastically changed the CH 4 emissions pattern, likely due to a continuous low pH in the rumen which is detrimental for methanogenesis. In conclusion, the daily variation in CH 4 emissions decreased with increasing forage rape inclusion in the diet of sheep and this was associated with a decreasing acetate proportion in the rumen liquid. • Lambs fed 100% forage rape had relatively constant CH 4 emissions throughout the day. • Adding increasing pasture to the forage rape increased the variability of CH 4. • H 2 available per unit of glucose fermented was correlated to the variability of CH 4. There is an increasing demand for developing new sources of proteins, and plants are considered the preferred alternative to animal proteins. In particular, green leaves are a source of RuBisCO (ribulose-1,5-bisphosphate carboxylase/oxygenase). This work assessed perennial ryegrass as a source of soluble proteins using mild treatments to preserve their functional properties. Two different extraction methods were compared, differing in the presence or abscence of a heating step (at 50 °C for 30 min) aimed to improve the removal of chloroplast material. The method that included the heating step resulted in a protein concentrate with high solubility (>90% at pH 7), and higher concentration of phenolic compounds. On the other hand, avoiding the heating step resulted in a higher extraction yield, but the residual chloroplast material affected the solubility of the final powder (>60% at pH 7). The protein extracts obtained by both methods consisted mainly of RuBisCO and presented a balanced amino acid profile covering the FAO (2011) recommendations for all the essential amino acids. Therefore, perennial ryegrass showed potential to become an alternative source of proteins for food applications with good nutritional value, whose phenolic compound, chlorophyll content and solubility was strongly dependent on the extraction process. Policy measures have been taken to improve water quality in the Netherlands. These measures include the abolishment of derogation, which allowed dairy farmers to go beyond the maximum application of 170 kg nitrogen (N) from organic fertiliser per hectare, and additional measures of the 7th Nitrates Action program. Grass-clover swards, known for their symbiotic N fixation, could be a strategy to deal with stricter N policies and can potentially improve the environmental sustainability and economic viability of Dutch dairy farms. The aim was to assess the effects of stricter N policies on the farm structure, farm income and environmental performance of a representative Dutch dairy farm on a sandy soil, and to assess the effect of incorporating perennial ryegrass-red white clover (GC rw) swards and perennial ryegrass-white clover (GC w) swards into the grassland management of this farm using a model. A whole-dairy farm linear programming model was used with the objective function to maximize farm income. The model was combined with a farm nutrient balance and life-cycle assessment to determine the impact on nutrient surpluses and greenhouse gas emissions. We modelled a representative Dutch dairy farm with perennial ryegrass (PRG) before and after implementing stricter N policies. Thereafter, the implications of implementing GC rw and GC w swards was assessed. Including the policy measures increased the share of maize land (+12%), decreased the number of dairy cows (-9 cows), reduced farm income (€-18,858 yr-1), led to similar greenhouse gas emissions (∼800 kg carbon dioxide equivalents (CO 2 -eq) per tonne (t) of fat and protein corrected milk (FPCM)), and resulted in a lower N surplus (-65 kg ha-1 yr-1) and phosphate surplus (-4.4 kg ha-1 yr-1) for a scenario with only PRG. The use of GC rw and GC w swards could partly compensate for the reduction in farm income (€+9,255 up to +14,706 yr-1). A combination of PRG, GC rw and GC w resulted in the highest farm income. The use of grass-clover swards only had the most positive effect on greenhouse gas emissions (767 kg CO 2 -eq t-1 FPCM) and N surplus (113 kg ha-1 yr-1). Sensitivity analysis showed the importance of yields and feed characteristics on the obtained farm income. Grass-clover swards might partly compensate for the negative economic consequences of stricter N policies for Dutch dairy farms. As the subject has not been previously examined, the aim of this study was to investigate the role of salicylic acid (SA) and calcium (Ca) as signaling molecules under combined drought and salt stresses on two different carbon-fixative turfgrasses, Lolium perenne L. (perennial ryegrass; C3) and Cynodon dactylon (L.) Pers. (Bermuda grass; C4). Twenty-one days after germination, PEG6000 (5%) and NaCl (150 mM) were applied simultaneously as a combined drought and salinity (DS) treatment. Before starting the treatment, half of the seedlings were treated with SA (0.5 mM) solution for 24 h; Ca (5 mM) was also applied simultaneously to the SA-treated groups. Combined drought and salt stresses caused substantial decreases in leaf growth parameters, relative water content, and osmotic potential in both cultivars, whereas lipid peroxidation (TBARS), hydrogen peroxide (H 2 O 2), and electrolyte leakage increased. However, the deleterious effect of the combined stress was much more pronounced in Bermuda grass. On the other hand, activities of superoxide dismutase (SOD), peroxidase (POX), ascorbate peroxidase (APX), and glutathione reductase (GR) in perennial ryegrass and only GR in Bermuda grass were reduced by DS. Moreover, Na accumulation was greater in perennial ryegrass (33.9-fold) than in Bermuda grass (11.1-fold). Under stress, simultaneous SA+Ca application was more effective on the recovery potential of Bermuda grass than on that of perennial ryegrass, with enhanced growth as well as increased proline, K, Mg, and Ca contents, and retained RWC. However, the SA+Ca treatment for the perennial ryegrass under DS significantly enhanced the antioxidant defense activities of SOD, POX, APX, and GR as well as increasing the TBARS and H 2 O 2 content, compared with plants subjected to DS alone. Taken together, these results indicated that simultaneous application of SA+Ca could play a positive regulatory role, which in turn could improve the double-stress tolerance of turfgrasses. Dairy heifers developed in certified organic programs, especially those utilizing pasture-based management schemes, have lower rates of gain than heifers raised in nonorganic confinement production systems in temperate climates, such as in the Intermountain West region of the United States. This study investigates the effects that different forages in a rotational grazing system have on development of organically raised Jersey heifers. Over 3 years, 210 yearling Jersey heifers were randomly assigned to one of 9 treatments, including a conventional confinement control where animals were fed a total mixed ration or one of 8 pasture treatments: Cache Meadow bromegrass (Bromus riparius Rehmann), QuickDraw orchard grass (Dactylis glomerata L.), Amazon perennial ryegrass (Lolium perenne L.), or Fawn tall fescue (Schendonorus arundinaceus [Schreb.] Dumort) and each individual grass interseeded with birdsfoot trefoil (Lotus corniculatus L., BFT). Each treatment had 3 blocks/yr over the 3-yr period, with each block having a 0.4 ha pasture of each treatment. Every 35 d, over a 105-d period, heifers were weighed and measured for hip height, and blood samples were collected to determine serum insulin-like growth factor-1 and blood urea nitrogen concentrations. Fecal egg counts were also assessed. Heifer body weight (BW), blood urea nitrogen, and insulin-like growth factor-1 concentrations were affected by treatment when analyzed over time. Heifers on grass-BFT pastures had increased BW compared with heifers on monoculture grass pastures. Heifers receiving a total mixed ration or perennial ryegrass+BFT had increased BW gain over the 105-d period compared with heifers grazing tall fescue+BFT, orchard grass, perennial ryegrass, meadow bromegrass, or tall fescue. Individually for all grass species, heifers grazing +BFT pastures had greater ending BW and weight gain than heifers grazing the respective grass monocultures. Furthermore, weight gain for heifers on perennial ryegrass+BFT, meadow bromegrass+BFT, and orchard grass+BFT were not different from those on a total mixed ration. Heifers grazing grass-BFT pastures had increased blood urea nitrogen compared with heifers grazing monoculture grass pastures. Heifer hip height and fecal egg counts were not affected by treatment. These results show that the addition of BFT to organic pasture improves growth of grazing replacement heifers. Economic analyses also demonstrate that interseeding grass pastures with BFT results in an increased economic return compared with grazing monoculture grass pastures. Grazing efficiency has been shown to differ between perennial ryegrass varieties. Such differences affect the utilization of grass within grazing systems, influencing the profitability of grass-based ruminant production systems. The Pasture Profit Index (PPI) is an economic merit grass variety selection tool developed to identify varieties with the greatest economic potential for grass-based dairy production systems. A new grass utilization subindex was developed and incorporated into the PPI to identify varieties with superior grazing efficiency. The subindex rewards varieties with superior grazing efficiency, measured as Residual grazed height, as these varieties allow increased amounts of herbage dry matter to be used by grazing animals. The economic values of all other traits within the PPI were reviewed and updated to ensure that the index was reflective of the current economic scenarios with appropriate assumptions included in the models, thus ensuring that varieties excelling in the agronomic traits with the greatest effect on profitability were recognized. The difference between the highest and lowest performing varieties for the grass utilization trait ranged from €23 to −€24. A range of €211 to €43 was recorded between the highest and lowest ranked varieties within the updated PPI. Spearman's rank correlation between the updated and original PPI lists was 0.96. The introduction of the utilization subindex will allow farmers to make informed variety selection decisions when reseeding pasture, particularly on their grazing platforms and it will allow a demand-based communication process between the farmer and the grass merchant or breeder, ultimately affecting trait selection for future breeding strategies. Low dietary energy and decreased intake of herbage have been attributed to the reduced performance of grazing dairy cattle. We hypothesized that grasses with inherently greater energy would interact in a complementary way with condensed tannins (CT) in birdsfoot trefoil to increase herbage intake by grazing dairy heifers. Eight pasture treatments comprising high-sugar perennial ryegrass (Lolium perenne L.), orchardgrass (Dactylis glomerata L.), meadow bromegrass (Bromus riparius Rehmann), and tall fescue [ Schendonorus arundinaceus (Schreb.) Dumort] were established in Lewiston, Utah as monocultures and binary mixtures with birdsfoot trefoil (Lotus corniculatus L.; BFT). Pasture treatments were rotationally stocked by Jersey heifers for 105 d in 2017 and 2018, and herbage samples were collected pre- and postgrazing for each 7-d grazing period and analyzed for herbage mass, nutritive value, and apparent herbage intake. We observed differences among pasture treatments in herbage quantity and nutritive value, as well as differences in herbage intake by grazing Jersey heifers. On average, grass-BFT mixtures had greater herbage intake than grass monocultures, and every grass-BFT treatment individually had greater herbage intake than their respective grass monocultures. Using multivariate analyses, we determined that approximately 50% of the variation in herbage intake was due to nutritive and physical herbage characteristics, with the most explanatory being characteristics related to fiber and energy, followed by those related to the percent of BFT in the herbage. Grass monocultures exhibited a range of inherent dietary energy, but there was indication that an imbalance of energy to crude protein (e.g., protein deficient) reduced intake of grass monocultures. Moreover, there was some evidence of a complementary effect between increased dietary energy and CT; however, low CT levels made it impossible to determine the effect of CT on herbage intake per se. This study confirmed that chemical and physical characteristics inherent to different pasture species have a large effect on herbage intake by grazing cattle. Several studies have been conducted to improve grazing management and supplementation in pasture-based systems. However, it is necessary to develop tools that integrate the available information linking the representation of biological processes with animal performance for use in decision making. The objective of this study was to evaluate the precision and accuracy of the Molly cow model predictions of ruminal fermentation, nutrient digestion, and animal performance by cows consuming pasture-based diets to identify model strengths and weaknesses, and to derive new digestive parameters when relevant. Model modifications for adipose tissue, protein synthesis in lean body mass and viscera representation were included. Data used for model evaluations were collected from 25 publications containing 115 treatment means sourced from studies conducted with lactating dairy cattle. The inclusion criteria were that diets contained ≥45% perennial ryegrass (Lolium perenne L.), and that dry matter intake, dietary ingredient composition, and nutrient digestion observations were reported. Animal performance and N excretion variables were also included if they were reported. Model performance was assessed before and after model reparameterization of selected digestive parameters, global sensitivity analysis was conducted after reparameterization, and a 5-fold cross evaluation was performed. Although rumen fermentation predictions were not significantly improved, rumen volatile fatty acids absorption rates were recalculated, which improved the concordance correlation coefficient (CCC) for rumen propionate and ammonia concentration predictions but decreased CCC for acetate predictions. Similar degradation rates of crude protein were observed for grass and total mixed ration diets, but rumen-undegradable protein predictions seemed to be affected by the solubility of the protein source as was the intestinal digestibility coefficient. Ruminal fiber degradation was greater after reparameterization, driven primarily by hemicellulose degradation. Predictions of ruminal and fecal outflow of neutral detergent fiber and acid detergent fiber, as well as total fecal output predictions, improved significantly after reparameterization. Blood urea N and urinary N excretion predictions resulted in similar accuracy using both sets of model parameters, whereas fecal N excretion predictions were significantly improved after reparameterization. Body weight and body condition score predictions were greatly improved after model modifications and reparameterization. Before reparameterization, yield predictions for daily milk, milk fat, milk protein, and milk lactose were greatly overestimated (mean bias of 61.0, 58.7, 73.7, and 64.6% of mean squared error, respectively). Although this problem was partially addressed by model modifications and reparameterization (mean bias of 3.2, 1.1, 1.7, and 0.4% of mean squared error, respectively), CCC values were still small. The ability of the model to predict grass digestion and animal performance in dairy cows consuming pasture-based diets was improved, demonstrating the applicability of this model to these productive systems. However, the failure to predict grass digestion based on standard model inputs without reparameterization indicates there are still fundamental challenges in characterizing feeds for this model. The silicon (Si) uptake system of two ryegrass (Lolium perenne L.) cultivars was characterised by assessing the concentration- and time-dependent kinetics. Additionally, a Si transporter gene was isolated from ryegrass and their expression pattern was analysed. The concentration-dependent kinetics was examined in Jumbo and Nui cultivars supplied with 0, 0.5, 1.0, 2.0, and 4.0 mM Si and harvested at 24 h and 21 d. The time-dependent kinetics was evaluated at 0, 0.5, or 2 mM Si doses after 0, 3, 6, 9, 12, and 24 h. RACE-PCR was performed to isolate a full-length sequence codifying for a Si transporter, and semi-quantitative and quantitative RT-PCR was used to analyse its expression pattern. Differential Si uptake between ryegrass cultivars was found. Moreover, Lineweaver-Burk linearization showed similar V max values between cultivars; however, different K m suggested that Jumbo and Nui may have different affinities for silicic acid. The dissimilarities in K m between cultivars might involve either the differential contribution of known proteins responsible for Si uptake and transport or the involvement of undiscovered Si transporters. We identified a putative Si transporter from ryegrass Nui (LpLsi1), which was only expressed in roots and down-regulated by Si supply. The predicted amino acid sequence of LpLsi1 did not only show a high similarity and close phylogenetic relationship with monocot Si influx transporters but also indicated that it is a membrane protein possessing a high conservation of domains essential for silicic acid selectivity. Our findings provide evidence of LpLsi1 in ryegrass, which supports its high Si accumulation capacity. • Silicon concentration and Si uptake ability is higher in cv. Jumbo than in cv. The objective of this study was to investigate the effect of perennial ryegrass (Lolium perenne L.; PRG) ploidy and white clover (Trifolium repens L.) inclusion on milk production, dry matter intake (DMI), and milk production efficiencies. Four separate grazing treatments were evaluated: tetraploid PRG only, diploid PRG only, tetraploid PRG with white clover, and diploid PRG with white clover. Individual DMI was estimated 8 times during the study (3 times in 2015, 2 times in 2016, and 3 times in 2017) using the n -alkane technique. Cows were, on average, 64, 110, and 189 d in milk during the DMI measurement period, corresponding to spring, summer, and autumn, respectively. Measures of milk production efficiency were total DMI/100 kg of body weight (BW), milk solids (kg of fat + protein; MSo)/100 kg of BW, solids-corrected milk/100 kg of BW, and MSo/kg of total DMI. Perennial ryegrass ploidy had no effect on DMI; however, a significant increase in DMI (+0.5 kg/cow per day) was observed from cows grazing PRG-white clover swards compared with PRG-only swards. Sward white clover content influenced DMI as there was no increase in DMI in spring (9% sward white cover content), whereas DMI was greater in summer and autumn for cows grazing PRG-white clover swards (+0.8 kg/cow per day) compared with PRG-only swards (14 and 23% sward white clover content, respectively). The greater DMI of cows grazing PRG-white clover swards led to increased milk (+1.3 kg/cow per day) and MSo (+0.10 kg/cow per day) yields. Cows grazing PRG-white clover swards were also more efficient for total DMI/100 kg of BW, solids-corrected milk/100 kg of BW, and MSo/100 kg of BW compared with cows grazing PRG-only swards due to their similar BW but higher milk and MSo yields. To reveal the structural characteristics and physicochemical properties of perennial ryegrass lignin, sequential alkali extractions or double ball-milling and enzymatic hydrolysis on the basis of ultrasonic and hydrothermal pretreatments were proposed in this study. Results revealed that sequential alkali extractions released 89.4% of original lignin from the ryegrass cell walls and 0.75–4.16% of associated carbohydrates as compared to the double ball-milling and enzymatic hydrolysis (96.0% and 18.39%). It was observed that the two types of lignin prepared were SGH-type and had different amounts of p -coumarates and ferulates, and primarily consisted of β-O-4 ′ linkages combined with minor amounts of β-β ′ and β-5 ′ linkages. Besides, alkali-soluble lignins exhibited relatively fewer β-O-4 ′ linkages, higher S/G ratios and H-type units, and abundant phenolic OH groups as compared to the double enzymatic lignin. Haloxylon ammodendron , a typical xerophyte, tolerates various abiotic stresses and is widely distributed in desert areas. Two PGPR strains, Bacillus sp. WM13-24 and Pseudomonas sp. M30-35, were previously isolated from the rhizosphere of H. ammodendron in Tengger Desert, Gansu province, northwest China. The aim of this study was to investigate the role of M30-35 and WM13-24 in drought stress alleviation of ryegrass (Lolium perenne L.). Under normal condition, both M30-35 and WM13-24 increased shoot fresh and dry weight, chlorophyll content, total nitrogen and phosphorus contents and altered phytohormone distribution compared to control. Moreover, after 7 days of drought stress, WM13–24 and M30-35 enhanced photosynthetic capacity, relative water content, the activities of catalase (CAT) and peroxidase (POD) and proline content, resulted in decreased malondialdehyde (MDA) content, relative membrane permeability (RMP) and H 2 O 2 accumulation; interestingly, the two strains decreased ABA content in leaves. This study demonstrated that the two PGPR strains promoted ryegrass growth and root development via regulating plant hormone distribution and enhanced drought tolerance of ryegrass through improving the activities of antioxidant enzymes, regulating ABA signaling and maintaining plant growth. Our results indicated that PGPR strains from rhizosphere of the desert plant species could be considered as promising bioinoculants for grass plants. • Both strains Bacillus sp. The objective of this study was to evaluate the effect of rolled barley supplementation on microbial composition and omasal flows of bacterial, protozoal, and nonmicrobial AA in cows fed fresh perennial ryegrass (Lolium perenne L.; PRG). Ten ruminally cannulated multiparous Holstein cows averaging (mean ± standard deviation) 49 ± 23 d in milk and 513 ± 36 kg of body weight were assigned to 1 of 2 treatments in a switchback design. The treatment diets were PRG only or PRG plus 3.5 kg of dry matter rolled barley (G+RB). The study consisted of three 29-d periods where each period consisted of 21 d of diet adaptation and 8 d of data and sample collection. A double-marker system was used to quantify nutrient flow entering the omasal canal along with 15N-ammonium sulfate to label and measure the microbial and nonmicrobial omasal flow of AA. Overall, rolled barley supplementation had no effect on the AA composition of the omasal liquid-associated and particle-associated bacteria. Rolled barley supplementation affected the AA concentrations of omasal protozoa; however, the differences were nutritionally minor. Particle-associated bacteria AA flow was increased for all AA, except for Trp and Pro, in cows fed the G+RB diet. Rolled barley supplementation had no effect on protozoal AA flow. On average, protozoa accounted for 23% of the microbial essential AA flow, which ranged from 17 to 28% for Trp and Lys, respectively. The flow of all AA in omasal true digesta increased in cows fed the G+RB diet compared with the PRG-only diet, resulting in a 228 g/d increase in total AA flow in cows fed the G+RB diet. This increase in total AA flow in cows fed the G+RB diet was due to an increase in microbial AA flow. Rolled barley supplementation had no effect on nonmicrobial AA flow. The nonmicrobial AA flow modestly contributed to total AA flow, accounting for 15.6% on average. These results indicated that extensive ruminal degradation of PRG AA occurred (83.5%), and we demonstrated that cows consuming PRG-based diets exhibit a large dependence on microbial AA to support metabolizable AA supply. Rolled barley supplementation can increase the omasal flow of microbial AA in cows consuming PRG-based diets. The current study highlights the synthesis of Zn-enriched biosourced catalysts (BIOCATs) obtained from ryegrass (Lolium perenne L.) cultivated on contaminated soils located in the North of France (Noyelles-Godault and Auby, Hauts-de-France). These cultures were located on brownfield land, next to two former industrial sites where the zinc and lead metallurgists Metaleurop and Nyrstar were installed. Physical and morphological properties of BIOCATs were compared to heterogeneous catalysts (CATs) prepared from commercial zinc chloride and montmorillonite K10. The catalysts were thermally activated (120 °C, 280 °C or 500 °C). BIOCATs have been further compared to CATs and different commercial metal chlorides in terms of efficiency in the synthesis of anti-inflammatory agents, Piroxicam and Meloxicam, both of them being involved in the inhibition of cyclooxygenases (COX) and being the main ingredients of the Feldene® and Mobic® drugs, respectively. BIOCAT-assisted synthesis resulted in the best reaction yields (84% and 78%, respectively) among all the tested catalysts. The results indicated that ryegrass provides appropriate quantity of biomass and can be considered as a bio-ore resource to improve green metrics in an environmental context involving clean and green technologies. Furthermore, the Zn/Cd and Zn/Pb ratios obtained in the ashes of ryegrass shoots were in general comprised between 153 and 202. This result demonstrated the excellent selectivity of ryegrass for zinc at the expense of the more problematic Cd and Pb, which are carcinogenic, mutagenic and reprotoxic. The indiscriminate use of sulfadiazine has caused severe harm to the environment, and biodegradation is a viable method for the removal of sulfadiazine. However, there are few studies that consider sulfadiazine biodegradation mechanisms. To comprehensively investigate the process of sulfadiazine biodegradation by plants in a soil system, a potted system that included ryegrass and soil was constructed in this study. The removal of sulfadiazine from the system was found to be greater than 95% by determining the sulfadiazine residue. During the sulfadiazine removal process, a significant decrease in ryegrass growth and a significant increase in antioxidant enzyme activity were observed, which indicates the toxic response and detoxification mechanism of sulfadiazine on ryegrass. The ryegrass transcriptome and soil bacterial communities were further investigated. These results revealed that most of the differentially expressed genes (DEGs) were enriched in the CYP450 enzyme family and phenylpropanoid biosynthesis pathway after sulfadiazine exposure. The expression of these genes was significantly upregulated. Sulfadiazine significantly increased the abundance of Vicinamibacteraceae , RB41 , Ramlibacter, and Microvirga in the soil. These key genes and bacteria play an important role in sulfadiazine biodegradation. Through network analysis of the relationship between the DEGs and soil bacteria, it was found that many soil bacteria promote the expression of plant metabolic genes. This mutual promotion enhanced the sulfadiazine biodegradation in the soil system. This study demonstrated that this pot system could substantially remove sulfadiazine and elucidated the biodegradation mechanism through changes in plants and soil bacteria. Knowledge of the environmental compatibility of pervious concrete with recycled coarse aggregate (RA) is significant, especially when used in riparian buffer areas. In this study, investigations on the growth performance of two grass combinations (I: Festuca and Ryegrass; II: Cosmos bipinnata, Festuca and Ryegrass) and water purification capacity of the pervious concrete with the RA were carried out to evaluate the environmental compatibility; the former was performed by measuring the maximum height and germination rate during 28 days of growth; the latter was measured using a water immersion test with measurements of the nutrient content (NO 3 --N, NO 2 --N, NH 4 +-N, T-N, PO 4 3--P, and T-P) and organic matter (COD and DOC) at certain intervals. Prior to that, the effects of RA grading (5–15 mm, 15–20 mm, 20–25 mm) and water-cement ratio (W/C) (0.4, 0.45, 0.5) on strength and porosity were determined, along with suitable concrete being selected for the test of environmental compatibility. The results showed that a maximum RA grading of 20–15 mm was the best for plant growth, manifesting the greatest maximum height of 9.8 cm and germination rate of 42.42%. Pervious concrete with vegetation can efficiently purify the surrounding water, with a removal efficiency of more than 98% for NH 4 +-N, T-P, PO 4 3--P, and COD. The removal efficiency increased over time, except for the DOC. W/C had no distinct effect on plant growth and matter removal, except for NO 2 --N and NO 3 --N, which increased as W/C increased. The primary aim of this study was to assess the impact of liquid (S-LAB) and lyophilized (L-LAB) probiotics sourced from Rye-Grass Lactic Acid Bacteria on broilers experiencing heat stress. The study involved 240 broiler chicks divided into six groups. These groups included a negative control (Control) with broilers raised at a normal temperature (24 °C) on a basal diet, and positive control groups (S-LAB and L-LAB) with broilers under normal temperature receiving a lactic acid bacteria supplement (0.5 mL/L) from rye-grass in their drinking water. The heat stress group (HS) comprised broilers exposed to cyclic heat stress (5–7 h per day at 34–36 °C) on a basal diet, while the heat stress and probiotic groups (S-LAB/HS and L-LAB/HS) consisted of broilers under heat stress supplemented with the rye-grass-derived lactic acid bacteria. Results indicated that heat stress without supplementation (HS) led to reduced body weight gain, T3 levels, citrulline, and growth hormone levels, along with an increased feed conversion ratio, serum corticosterone, HSP70, ALT, AST, and leptin levels. Heat stress also negatively impacted cecal microbiota, decreasing lactic acid bacteria (LABC) while increasing E. coli and coliform bacteria (CBC) counts. Probiotic supplements (S-LAB/HS and L-LAB/HS) mitigated these effects by enhancing broilers' resilience to heat stress. In conclusion, rye grass-derived S-LAB and L-LAB probiotics can effectively support broiler chickens under heat stress, promoting growth, liver function, hormonal balance, gut health, and cecal microbiome ecology. [Display omitted] • The first-time volatiles of Rathybacter toxicus are identified with cultivating bacteria with a novel approach. • Associated endophytic bacteria were explored for their antibacterial potential against R. toxicus. • Pantoea agglomerans and Pseudomonas cedrina could provide a new avenue for developing bio-control strategies against R. toxicus. The study involved the isolation of bacteria from the surface of Rathayibacter toxicus infested ryegrass seeds, followed by an analysis of volatile organic compounds (VOCs) to identify biomarkers associated with each bacterial species. Among the four isolated bacteria (a species of Pantoea , two species of Pseudomonas and R. toxicus), three were evaluated for their antibiosis potential against R. toxicus. The main objective of this research was to characterize the volatile compounds produced by R. toxicus and to assess the capability of the other three bacteria to serve as biological control agents. The volatile chemicals were identified by using headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry (GC–MS) in the aforementioned bacterial species. The identified compounds such as 1-nonanol, geraniol, decanoic acid and 2-decanone may serve as biomarkers for R. toxicus. Additionally, obtained information on VOCs may facilitate chemical communication between bacteria and the nematode which is responsible to carry it to the ryegrass plant. During the antibiosis assay, Pantoea agglomerans and Pseudomonas cedrina , among the endophytic/epiphytic bacterial species, produced compounds that significantly inhibited R. toxicus growth in vitro, with capacity values 22.7 ± 2.3 and 20.0 ± 2.0, respectively. In conclusion, this study's findings have significant implications for the management of annual ryegrass toxicity (ARGT), as they highlight the promising potential of P. agglomerans and Ps. cedrina as viable candidates for biological control strategies in ARGT management. The dominant ammonium bacteria established in different crop rotations are directly related to the conversion and utilization of nitrogen (N) in paddy soil. This study investigated the ammonia oxidation process in the rice rhizosphere of Italian ryegrass (Lolium multiflorum L.)-rice (IRR), celery (Apium graveolens)-rice (CLR) and winter fallow-rice (WFR) rotations in South China through the amoA functional gene analysis based on Miseq high-throughput sequencing platform and qPCR methods. The abundance of amoA gene in IRR was increased by 116.67 % (P < 0.05) and 42.76 % (P > 0.05) compared with CLR and WFR, respectively. Nitrosomonas became the dominant genus in the rhizosphere of IRR, with the highest relative abundance (59.30 %). The contents of soil organic matter, total N and nitrate nitrogen (NO 3 −-N) in the rice rhizosphere soil in IRR were significantly higher than other two treatments (P < 0.05) by 12.47–14.31 %, 16.88–25.00 %, and 97.48–159.60 %, respectively. Although lower microbial diversity was observed in IRR, the soil fluorescein diacetate hydrolysis enzyme activities and ammonia monooxygenase activities in the rice rhizosphere were higher in IRR than those in CLR and WFR (P < 0.05). Also, the soluble sugar and crude protein of milled rice were highest in IRR among the three rotation treatments (P < 0.05). The ammonia oxidation efficiency of ammonium-oxidizing bacteria was largely dependent on key taxa, such as Nitrosomonas (dominant genus in IRR), rather than the dynamics of AOB community structure. The key amoA- nitrifier taxa of the AOB community developed in IRR are capable of promoting rice production by improving soil N cycle efficiency and reducing the loss of N. Phosphorus (P) is critical to plants in metal-contaminated soils because it participates in various biochemical reactions during plant growth. However, the mechanisms of P in mitigating the toxicity of heavy metals to ryegrass root is still veiled. In this study, the physiological and biochemical dynamics of the ryegrass root under various cadmium (Cd) and P conditions were investigated in a hydroponic system. Cd stress decreased the length of the ryegrass root, but P application enhanced the root elongation to reduce the Cd concentration in the root. Both Cd and P dosages were positively correlated with hemicellulose 1 content, pectin content, and PME activity, while having a negative effect on cellulose content. Moreover, the addition of 80 mg L−1 P increased the contents of pectin and hemicellulose 1 by 2.5 and 5.8% even with 4 mg L−1 Cd. In addition, P supply increased pectin methylesterbase activity under Cd stress, which further changed the extra-cytoplasmic structures and cell wall composition. Thus, exogenous P promoted the immobilization of Cd onto the cell wall and protected protoplast primarily through indirectly regulating the binding capacity of the root cell wall for Cd. The type-B response regulator (ARR) family members represent the elements of two-component regulatory systems and play crucial roles in the cytokinin signaling, plant growth, and stress responses. The objective of this study was to characterize type-B ARR genes from perennial ryegrass, and to investigate their roles in regulating to salt tolerance. Twelve type-B ARRs , namely LpARR1 - 12 , were identified in perennial ryegrass. They were clustered into three subgroups and were unevenly distributed on chromosomes. The stress responsive elements (SREs) and hormone responsive elements (HREs) were identified in LpARRs promoters and the transcription of LpARRs genes were responsive to various abiotic stress (heat, salt, and osmotic) and hormonal (cytokinin, ABA, and ethylene) treatments. LpARR10 was strong induced by salt stress, therefore it was selected for function analysis. Overexpressing LpARR10 in Arabidopsis led to a significant improvement in salt tolerance. The LpARR10 -overexpressing (LpARR10 -OE) lines exhibited lower levels of Na+ content compared to WT plants, and the expression levels of SOS1 and SOS3 were significantly higher in the LpARR10 -OE lines. Results of yeast auto-transactivation and plant cell transcriptional activity assays shows LpARR10 is a nucleus localized transcriptional activator and directly bind and transactivated the promoter of SOS1 and SOS3. Together, LpARR10 enhances salt tolerance may via trans-activating SOS1 / SOS3 in Arabidopsis. Silicon (Si) has the potential to promote plant yield and improve their insect resistance, but the direct defense mechanisms associated with the effects of Si on forage grasses and its regulation are not known. In this study, the conditions of silica fertilization nutrition (1 and 3 mmol L−1 recorded as low concentration (LC) and high concentration (HC) groups, respectively) were changed during the seed germination period, which did not significantly affect the seed germination of ryegrass. However, the shoot biomass and insect resistance of ryegrass were significantly increased in the HC group. Comparison of the transcriptional results from RNA-seq assays revealed that the accumulation of ryegrass biomass may be closely related to the silicon intervention in photosynthesis, and the metabolic synthesis of lignin, carbohydrates, and hemicellulose. There was an additive effect of silicon concentration on the regulatory effects of ryegrass at the transcriptional level. Meanwhile, 98 and 1291 differentially expressed genes were assigned to the metabolic process in the LC and HC groups, respectively. Metabolomic results combined with LC-MS analysis showed that Si initiated a direct defense network in ryegrass with Phenylpropanoid biosynthesis as the backbone, and Flavonoid, Flavone and flavonol, and Isoflavonoid biosynthesis as important branches. The relative content of phenolics was significantly up-regulated, especially in the HC group, and most of these phenolics were potent SOS substances, closely related to the physiological disturbances suffered by Spodoptera frugiperda , with inhibitory effects on larval body weight. The above results confirm that silicon promotes an increase in ryegrass yield while initiating its direct defense network against insects, and that silicon is an effective supplement in improving ryegrass yield and insect resistance. Genotype-by-environment interaction (G×E) is commonly observed in perennial ryegrass species and significantly impacts dry matter yield (DMY) performance across environments. Variation in DMY of perennial ryegrass cultivars and their distinct responses to different environments directly influence the profitability of the Australian dairy industry. In this study, we implemented a separate two-way factor analytic strategy to fit multi-environment-multi-harvest data collected from 18 trials spanning 14 years to account for the G×E effects across complex pasture environments in south-eastern Australia to achieve accurate DMY predictions. Three mega-environments were identified, and the seasonal and environmental DMYs of 126 cultivars or breeding lines (defined by a combination of varieties and endophytes) were predicted. Statistical differences in DMY performance and crossover G×E effects in cultivar rankings were observed among the three mega-environments. Several high-yielding cultivars specific to each mega-environment were also identified. The analysis demonstrated the importance of accounting for G×E effects when predicting the DMY of perennial ryegrass and highlighted the potential for identifying high-yielding cultivars specific to mega-environments. Grazing birds like geese can have a big impact on agricultural land, potentially causing damage to crops. Their exploitation of agricultural land varies within and between seasons, and these temporal grazing patterns can be influenced by management. Furthermore, management practices using scaring and accommodation aim to influence local grazing pressures. To achieve efficient goose management we need a good understanding of the effects of timing and intensity of grazing on grass development and subsequent yield loss. We performed an exclosure study on twelve fields in Friesland (Fryslân), the Netherlands to study the effect of grazing on grass development and potential yield loss. Grazing was prevented either from November or from early April until farmers anticipated their first harvest. Every two weeks we measured grass height and calculated growth in exclosures and in ungrazed plots, and performed dropping counts. The results show that the difference in grass height between grazed and ungrazed plots increases with grazing pressure, but less so for higher grazing pressures. We also find that both winter and spring grazing can result in a larger difference in grass height between grazed and ungrazed plots, and grazing into the growing season delays the start of grass growth. This appears due to the relationship between grass height and growth rate. Because the optimal height for growth increases over the season, spring grazing has a much larger impact on grassland yields than winter grazing. Overall these results show that the amount of yield loss depends on different aspects of grazing, most prominently the recovery time, duration and grazing intensity. We hypothesize that barnacle geese may select fields with denser swards and may stimulate sward density by frequent grazing throughout winter and early spring as well as across consecutive years. Future studies should look into the effect of harvest delays and sward density on the size and quality of yield across the season and especially at the last cut. Plant roots exude nitrogen (N)-containing compounds, which are assimilated by the rhizobiome to maintain stoichiometric homeostasis. Various 15N-tracing methods exist to estimate the rhizobiome-N derived from root exudation but have never been validated with a full factorial experiment. We exposed ryegrass (Lolium multiflorum) to 15N solutions through different plant organs (stem or leaf feeding), with different 15N-tracers (urea or NH 4 NO 3) and concentrations (64.5, 129 or 193 mmol 15N L−1). Stem feeding with either 15N-source at the highest concentration was the only effective way to quantify root exudate-N assimilation by the rhizobiome, and we recommend this 15N-labeling method for the reliable assessment of microbiome-N dynamics in the rhizosphere. • Stem feeding quantifies root exudate-N transfer to ryegrass rhizobiome. • Nitrogen concentration determines rhizobiome 15N enrichment. • Root exudates are a N source to the ryegrass rhizobiome in N-rich soil. Sheep farmers can increase production levels as a means to improve economic outcomes. However, this intensification can potentially result in more nitrogen (N) leaching which is at odds with the increasing pressure on farmers to reduce the quantity of N lost into the environment. Therefore, there is increasing interest in the use of alternative forages, such as Italian ryegrass and plantain, which could potentially allow farmers to increase production while decreasing N leaching. This study quantified and compared the quantity of N leaching under sheep grazing four different forages: perennial ryegrass/white clover (RGWC), Italian ryegrass/WC (IRWC), plantain/WC (PWC) and a winter brassica. Five replicates of each forage were established on plots of 800 m2 (40 m x 20 m). Each plot had an independent mole-pipe drain system with tipping-bucket monitoring equipment at the end of the pipe drain. Drainage volume and N concentrations (nitrate-N: NO 3 ⁻-N; ammonium-N; total-N) in drainage water were measured to estimate N losses. The forages were grazed according to the best management practice for each forage type over a two-and-a-half-year period (Year 1: July-December 2019; Year 2: January-December 2020; and Year 3: January-December 2021). In the first two years of this study, NO 3 ⁻-N leaching losses under IRWC and PWC were very small (ranging from 0.4 to 0.5 kg N/ha), and they were not different (P > 0.05) from those under RGWC. However, in Year 3, the leaching losses under IRWC and PWC treatments were greater (P < 0.05) than those under RGWC. This increased leaching was probably due to the need to re-establish these pastures. As part of renewing these swards, the IRWC and PWC plots were fallow during the key late summer-autumn period. In contrast, NO 3 ⁻-N leaching losses under brassica forages (3.5–6.4 kg N/ha) were greater (P < 0.05) than under RGWC (0.5–1.5 kg N/ha). Overall, the leaching losses measured under sheep grazing the forages were much smaller than those commonly reported by dairy studies.
