Abstract

This study aimed to: (i) understand how women perceived their recently developed patellofemoral pain (PFP) regarding its cause, prognosis, and willingness to seek treatment; (ii) investigate self-reported function, knee-related quality of life (QoL), fear of movement, and physical activity level at the onset of PFP. Mixed-methods longitudinal study. University. Sixty-eight pain-free women were followed up over one year. Those who developed PFP were interviewed within one month of the development of symptoms. Self-reported function, kinesiophobia, knee-related QoL, and physical activity were obtained at baseline and follow-up assessments. Twenty-one women developed PFP. Most participants reported believing the increase in physical activity and/or sitting time was associated with the onset of PFP. Many reported believing symptoms would improve over time without any treatment. Only a small number of participants intended to seek care. Quantitatively, decreases in self-reported function and QoL, as well as increases in the physical activity level were observed after PFP development. Although decreases in self-reported function and QoL were observed, women reported believing their PFP is self-limiting and do not need treatment. Strategies to accurately disseminate knowledge about PFP are needed to help stimulating early care. • Overuse and sitting time are commonly associated with patellofemoral pain onset. • Women with acute patellofemoral pain believe that their pain is self-limiting. • Seeking early care is not a priority for women with acute patellofemoral pain. • Knowledge dissemination about patellofemoral pain can help stimulating early care. Abstract

• Uninduced and neuronally-induced DPSCs demonstrate potential for Parkinson's disease therapy. • Uninduced and neuronally-induced DPSCs promote significant recovery in dopaminergic neurons and locomotion. • Neuronally-induced DPSCs prevent GFAP elevation and alter DARPP-32 phosphorylation states. • Uninduced DPSCs reduce JUN levels and mitigate glycosylated DAT elevation. In recent years, dental pulp stromal cells (DPSCs) have emerged as a promising therapeutic approach for Parkinson's disease (PD), owing to their inherent neurogenic potential and the lack of neuroprotective treatments for this condition. However, uncertainties persist regarding the efficacy of these cells in an undifferentiated state versus a neuronally-induced state. This study aims to delineate the distinct therapeutic potential of uninduced and neuronally-induced DPSCs in a rodent model of PD induced by 6-Hydroxydopamine (6-OHDA). DPSCs were isolated from human teeth, characterized as mesenchymal stromal cells, and induced to neuronal differentiation. Neuronal markers were assessed before and after induction. DPSCs were transplanted into the substantia nigra pars compacta (SNpc) of rats 7 days following the 6-OHDA lesion. In vivo tracking of the cells, evaluation of locomotor behavior, dopaminergic neuron survival, and the expression of essential proteins within the dopaminergic system were conducted 7 days postgrafting. Isolated DPSCs exhibited typical characteristics of mesenchymal stromal cells and maintained a normal karyotype. DPSCs consistently expressed neuronal markers, exhibiting elevated expression of βIII-tubulin following neuronal induction. Results from the animal model showed that both DPSC types promoted substantial recovery in dopaminergic neurons, correlating with enhanced locomotion. Additionally, neuronally-induced DPSCs prevented GFAP elevation, while altering DARPP-32 phosphorylation states. Conversely, uninduced DPSCs reduced JUN levels. Both DPSC types mitigated the elevation of glycosylated DAT. Our results suggested that uninduced DPSCs and neuronally-induced DPSCs exhibit potential in reducing dopaminergic neuron loss and improving locomotor behavior, but their underlying mechanisms differ. Abstract

Background: The coronavirus disease 2019 (COVID-19) presents an urgent threat to global health. Prediction models that accurately estimate mortality risk in hospitalized patients could assist medical staff in treatment and allocating limited resources. Aims: To externally validate two promising previously published risk scores that predict in-hospital mortality among hospitalized COVID-19 patients. Methods: Two prospective cohorts were available; a cohort of 1028 patients admitted to one of nine hospitals in Lombardy, Italy (the Lombardy cohort) and a cohort of 432 patients admitted to a hospital in Leiden, the Netherlands (the Leiden cohort). The endpoint was in-hospital mortality. All patients were adult and tested COVID-19 PCR-positive. Model discrimination and calibration were assessed. Results: The C-statistic of the 4C mortality score was good in the Lombardy cohort (0.85, 95CI: 0.82−0.89) and in the Leiden cohort (0.87, 95CI: 0.80−0.94). Model calibration was acceptable in the Lombardy cohort but poor in the Leiden cohort due to the model systematically overpredicting the mortality risk for all patients. The C-statistic of the CURB-65 score was good in the Lombardy cohort (0.80, 95CI: 0.75−0.85) and in the Leiden cohort (0.82, 95CI: 0.76−0.88). The mortality rate in the CURB-65 development cohort was much lower than the mortality rate in the Lombardy cohort. A similar but less pronounced trend was found for patients in the Leiden cohort. Conclusion: Although performances did not differ greatly, the 4C mortality score showed the best performance. However, because of quickly changing circumstances, model recalibration may be necessary before using the 4C mortality score.

Abstract

Background: The prevention of healthcare-associated infections requires continuous effort. In order to achieve better practical results, the control of environmental microbial biofilms with effective disinfection strategies should be addressed.Aim: To test the efficacy of different time cycles of nebulized hydrogen peroxide (H2O2) against bacterial and yeast dry biofilms.Methods: The efficacies of a standard cycle (SC) and a fast cycle (FC) of nebulized H2O2 were compared. Microbial biofilms were grown on different material coupons. The metabolic activity of biofilms was determined by XTT assay, and the total biomass of biofilms was determined by crystal violet assay.Findings: Regarding the efficacy of nebulized H2O2 against biofilms, the mean reduction in metabolic activity for the SC was 55.2% [standard deviation (SD) 19.4%], compared with 50.4% (SD 17.7%) for the FC. The mean reduction in total biomass for the SC was 45.5% (SD 22.7%), compared with 46.7% (SD 21.7%) for the FC. No significant differences were found between the tested cycles and materials.Conclusion: H2O2 nebulization was found to exhibit good efficacy against healthcare-associated microbial dry biofilms.
