Purpose: To investigate the safety and effectiveness of emergent transjugular intrahepatic portosystemic shunt (TIPS) as a first-line therapy in advanced cirrhotic patients with acute variceal bleeding (AVB).Materials and Methods: From July 2016 to June 2019, 76 patients with advanced cirrhosis and AVB were included in this retrospective study. All patients underwent emergent TIPS as first-line therapy within 24 hours. Gastroesophageal varices (GVs) in patients with cirrhosis were diagnosed with contrast-enhanced CT due to the fact that emergent endoscopy in this center has not been routinely performed. The primary outcomes were the control rate of bleeding and the rate of rebleeding. The secondary outcomes were the technical success rate of procedure, transplantation-free survival, the mean hospitalization time, the time of stay in the intensive care unit (ICU) and adverse events.Results: All the patients underwent TIPS creation successfully and were transferred to the general wards. The median follow-up time was 21.7 months (interquartile range 12.6 to 28.1 months). The control rate of bleeding (≤5 days) was 100%. The rates of early (>5 days to 6 weeks) and late (>6 weeks to 2 years) rebleeding were 6.6% and 1.3%, respectively. The 6-week, 1-year and 2-year transplantation-free survival rates were 94.7%, 93.4% and 84.6%, respectively. The incidence of acute liver failure, hepatic encephalopathy and shunt dysfunction were 5.3%, 25% and 5.3%, respectively.Conclusion: Emergent TIPS as a first-line therapy for advanced cirrhotic patients with AVB is safe and effective. Understanding the mechanisms driving parasite distributions is not only important for understanding ecosystem functioning, but also crucial for disease control. Previous studies have documented the important roles of host sex, host body size, host behavioral trait (such as boldness and trappability), and seasonality in shaping parasite load. However, few studies have simultaneously assessed the roles of these factors, as well as their interactions. In spring and summer of 2021, we conducted live trapping in Hohhot, China, to collect ectoparasites on Daurian ground squirrel (Spermophilus dauricus), a small rodent widely distributed in East Asian grassland. We then used generalized linear models to explore the effects of several biological factors (sex, body weight, trappability, and reproductive status) and seasonality on the abundance of ticks and fleas in S. dauricus. Significant but inconsistent seasonal effects were observed: tick load was significantly greater in summer than in spring, while flea load was greater in spring than in summer. Seasons also significantly interacted with host trappability and body weight to affect tick abundance. Our results highlight the importance of considering seasonal changes in parasitism, as well as interactions between season and host biological traits in shaping parasite distributions. In this article, the updated status of interventional radiology (IR) in China is reported and compared vs that a decade ago based on a poll carried out in 2017 in Jiangsu Province, where the economy and overall health level are among the best of the 31 provinces in China. All 98 polled centers responded, and 56 IR departments (57%) had become independent departments separate from the radiology department; 74 (76%) had inpatient wards. In 2017, there were 538 interventional radiologists performing IR procedures in Jiangsu Province, with a total of 69,277 procedures performed, with interventional oncologic procedures accounting for the largest proportion (58%). Corticosteroid insensitivity is a feature of airway inflammation in chronic obstructive pulmonary disease (COPD). Erythromycin exhibits anti-inflammatory activity in COPD, but the concrete mechanism is still unclear. This study aimed to investigate the effects of erythromycin on corticosteroid sensitivity in peripheral blood mononuclear cells (PBMCs) and U937 cells (a human monocytic cell line). PBMCs were collected from non-smokers, healthy smoker volunteers, and COPD subjects. U937 cells were incubated with or without erythromycin and stimulated with TNF-α in the presence or absence of cigarette smoke extract (CSE). The dexamethasone (Dex) concentration required to achieve 50% inhibition of TNF-α-induced interleukin (IL)-8 production was determined and the mitogen-activated protein kinase (MAPK)/Activator protein-1 (AP-1) pathway was also evaluated. Erythromycin improved corticosteroid sensitivity in PBMCs obtained from COPD patients and CSE-treated U937 cells. This improvement in corticosteroid sensitivity was associated with reduced c-Jun expression, which resulted from the inhibition of P38 Mitogen-activated protein kinase (P38MAPK), extracellular signal-regulated protein kinase (ERK)1/2, and c-Jun N-terminal kinase (JNK) phosphorylation. Erythromycin had no effects on the phosphorylated and total protein expression levels of P38MAPK and ERK; however, it induced inhibition of the phosphorylated and total protein expression levels of JNK. This study provides evidence that erythromycin restores corticosteroid sensitivity in PBMCs and U937 cells. JNK inhibition by erythromycin restores corticosteroid sensitivity via the inhibition of c-Jun expression. Thus, JNK/c-Jun is a potential novel therapeutic target for COPD. Abstract Endocrine disrupting chemicals (EDCs) include a wide array of pollutants, such as some metals and other toxic elements, which may cause changes in hormonal homeostasis. In addition to affecting physiology of individuals directly, EDCs may alter the transfer of maternal hormones to offspring, i.e. causing transgenerational endocrine disruption. However, such effects have been rarely studied, especially in wild populations. We studied the associations between environmental elemental pollution (As, Cd, Cu, Ni, Pb) and maternally-derived egg thyroid hormones (THs) as well as nestling THs in great tits (Parus major) using extensive sampling of four pairs of polluted and reference populations across Europe (Finland, Belgium, Hungary, Portugal). Previous studies in these populations showed that breeding success, nestling growth and adult and nestling physiology were altered in polluted zones compared to reference zones. We sampled non-incubated eggs to measure maternally-derived egg THs, measured nestling plasma THs and used nestling faeces for assessing local elemental exposure. We also studied whether the effect of elemental pollution on endocrine traits is dependent on calcium (Ca) availability (faecal Ca as a proxy) as low Ca increases toxicity of some elements. Birds in the polluted zones were exposed to markedly higher levels of toxic elements than in reference zones at the populations in Finland, Belgium and Hungary. In contrast to our predictions, we did not find any associations between overall elemental pollution, or individual element concentrations and egg TH and nestling plasma TH levels. However, we found some indication that the effect of metals (Cd and Cu) on egg THs is dependent on Ca availability. In summary, our results suggest that elemental pollution at the studied populations is unlikely to cause overall TH disruption and affect breeding via altered egg or nestling TH levels with the current elemental pollution loads. Purpose: To construct the albumin-bilirubin (ALBI) grade and the Child-Turcotte-Pugh (CTP) score based on nomograms, as well as to develop an artificial neural network (ANN) to compare the prognostic performance of the 2 scores for hepatocellular carcinoma (HCC) that has undergone transarterial chemoembolization.Materials and Methods: This multicentric retrospective study included patients with HCC who underwent transarterial chemoembolization monotherapy as an initial treatment at 4 institutions between January 2008 and December 2016. In the training cohort, significant risk factors associated with overall survival (OS) were identified by univariate and multivariate analyses. The prognostic nomograms and ANN were established and then validated in 2 validation cohorts.Results: A total of 838 patients (548, 115, and 175 in the training cohort and validation cohorts 1 and 2, respectively) were included. The median OS was 10.4, 15.7, and 9.2 months in the training cohort and validation cohorts 1 and 2, respectively. In the training cohort, both ALBI grade and CTP score were identified as significant risk factors. The ALBI grade and CTP score based on nomograms were established separately and showed similar prognostic performance when assessed externally in validation cohorts (C-index in validation cohort 1: 0.823 vs 0.802, P = .417; in validation cohort 2: 0.716 vs 0.729, P = .793). ANN showed that ALBI grade had higher importance on survival prediction than CTP score.Conclusions: ALBI grade performs at least no worse than CTP score regarding survival prediction for HCC receiving transarterial chemoembolization. To analyze the dynamic performance of the vehicle-FST system accurately, a vehicle-FST dynamic interaction model considering the flexibility of wheelsets is established. A method for flexible wheelset-rail relationship is described in detail. In addition, a virtual degrees of freedom method is adopted to improve the computational efficiency. To solve the stiffness nonlinearity of the rubber spring, an incremental numerical integration and matrices updating method is adopted. The numerical validation shows the reliable of the incremental numerical integration. Finally, the influence of the rubber spring parameters, stiffness nonlinearity of the rubber spring and wheelset flexibility on the dynamic performance of the system is analyzed in detail from the perspective of the frequency-domain and time-domain. Purpose: To develop a modified assessment for retreatment with transarterial chemoembolization (mART) score that may be more suitable for Chinese patients with hepatocellular carcinoma (HCC).Materials and Methods: Chinese patients with HCC who were treated with transarterial chemoembolization in four hospitals were included. A univariate analysis and a multivariate forward Cox regression analysis were used to identify significant prognostic factors of overall survival (OS). A point scoring model was subsequently developed from the training cohort, and the validation process was performed in the validation cohort.Results: The study included 259 patients (124 patients in the training cohort and 135 patients in the validation cohort). Increase in Child-Pugh scores relative to the baseline (P < .001), Barcelona Clinic Liver Cancer (BCLC) stage B before first transarterial chemoembolization (P = .001), and absence of radiologic tumor response (P < .001) were identified as negative prognostic factors for OS and were used to create the mART scores. BCLC staging was substituted for aspartate aminotransferase increase in the mART scores. The mART scores differentiated two groups with distinct prognosis by a cutoff score of 2.5 points (22.9 mo [95% confidence interval (CI), 17.4-28.4] vs 8.9 mo [95% CI, 7.5-10.3] in median survival; P < .001). In the validation cohort, the C index in assessment for retreatment with transarterial chemoembolization (ART) criteria was 0.64, whereas it was 0.82 in mART criteria.Conclusions: In Chinese patients with HCC, mART score of > 2.5 before second transarterial chemoembolization was associated with poor prognosis. Surface-enhanced Raman scattering (SERS) spectra of P-Aminobenzoic Acid (PABA) adsorbed on the silver nano-particles were studied, respectively, in the silver colloidal solution and on the dried silver-coated filter paper. To further study the adsorption behaviors of PABA on the different substrates, we analyze the problem by means of theoretical calculation. Five models of PABA adsorbed on the surfaces of silver nano-particles were established. The Raman spectra of these five models using DFT-B3PW91 with lanl2dz were calculated. By comparing the theoretical values with the experimental values, we found that the calculated Raman frequencies of the models were in good agreement with experimental values, respectively. So we can believe that the simplified models are probably reasonable to describe some surface- enhanced Raman experiments. [ABSTRACT FROM AUTHOR]

Abstract: Th17 cells are highly proinflammatory and involved in the immunopathogenesis of severe autoimmune diseases. Selective phosphodiesterase 4 (PDE4) inhibitors, which elevate intracellular cAMP by inhibiting the hydrolysis of cAMP, have been demonstrated to be an effective anti-inflammatory agent in airway inflammatory diseases. In the present study, we assessed the effect of a selective PDE4 inhibitor Zl-n-91 on IL-17 production by PBMCs and by purified CD4+ T cells following stimulation. The results for the first time demonstrated that the addition of Zl-n-91 into cell cultures of PBMCs and purified CD4+ T cells could result in the suppression of IL-17 production at the protein and mRNA levels. Further analysis indicated that Zl-n-91 had a direct inhibitory effect on the IL-17 production by memory Th17 cells via the suppression of activation, proliferation and division of CD4+ T cells. Our data suggested that Zl-n-91 might have beneficial effects in the treatment of IL-17-related autoimmune diseases. [Copyright &y& Elsevier]

Muscle creatine kinase (CK) is a crucial enzyme in energy metabolism, and it exists in two forms, the reduced form (R-CK) and the oxidized form (O-CK). In contrast with R-CK, O-CK contained an intrachain disulfide bond in each subunit. Here we explored the properties of O-CK and its regulatory role on muscle CK. The intrachain disulfide bond in O-CK was demonstrated to be formed between Cys74 and Cys146 by site-directed mutagenesis. Biophysical analysis indicated that O-CK showed decreased catalytic activity and that it might be structurally unstable. Further assays through guanidine hydrochloride denaturation and proteolysis by trypsin and protease K revealed that the tertiary structure of O-CK was more easily disturbed than that of R-CK. Surprisingly, O-CK, unlike R-CK, cannot interact with the M-line protein myomesin through biosensor assay, indicating that O-CK might have no role in muscle contraction. Through in vitro ubiquitination assay, CK was demonstrated to be a specific substrate of muscle ring finger protein 1 (MURF-1). O-CK can be rapidly ubiquitinated by MURF-1, while R-CK can hardly be ubiquitinated, implying that CK might be degraded by the ATP-ubiquitin-proteasome pathway through the generation of O-CK. The results above were further confirmed by molecular modeling of the structure of O-CK. Therefore, it can be concluded that the generation of O-CK was a negative regulation of R-CK and that O-CK might play essential roles in the molecular turnover of MM-CK. [ABSTRACT FROM AUTHOR]

Protecting proteins from aggregation is one of the most important issues in both protein science and protein engineering. In this research, the mechanism of enhancing the refolding of guanidine hydrochloride-denatured carbonic anhydrase B by polyvinylpyrrolidone 40 (PVP40) was studied by both kinetic and equilibrium refolding experiments. The reactivation and refolding kinetics indicated that the rate constant of refolding the first refolding intermediate (I1) to the second one (I2) is promoted by the addition of PVP. Fluorescence quenching studies further indicated that PVP could bind to the aggregation-prone species l1, resulting in the protection of the exposed hydrophobic surface, a minimization of the protein surface, and more importantly, an increase of the refolding rate of I1. These properties were quite different from those of poly(ethylene glycol) (PEG), which has been shown to have a strong and stoichiometric binding to I1 and does not interfere with the refolding pathway. Unlike PEG, the binding of PVP to I1 does not block the aggregation pathway directly but decreases the energy barrier for I1 to refold to I2 and thus reduces the accumulation of I1. These results suggested that PVP works by a quite different mechanism from those well established ones in chaperones and chemical promoters. PVP is more like a folding catalyst rather than a chemical chaperone. The distinct mechanism of enhancing protein aggregation by PVP is expected to facilitate the attempt to develop new chemical compounds as well as new strategies to protect proteins from aggregation. Hematopoietic growth factors mediate the survival and proliferation of blood-forming cells, but the mechanisms through which these proteins produce their effects are incompletely known. Recent studies have identified the pim family of kinases as mediators of cytokinedependent survival signals. Several studies have identified substrates for the pim-1 kinase, but little is known about the other family members, pim-2 and pim-3. We have investigated potential functions for the pim-2 kinase in factor-dependent murine hematopoietic cells. We find that pim-2 mRNA and protein expression are regulated by cytokines similarly to pim-1. Three PIM-2 protein isoforms are produced in cytokinetreated cells. All three forms are active kinases, and the short (PIM-2(34 kDa)) form is the most active at enhancing survival of FDCP1 cells after cytokine withdrawal. This pro-survival function involves inhibition of apoptosis and caspase activation. Enforced expression of PIM2(34 kDa) kinase does not appear to regulate expression of BCL-2, BCL-xL, BIM, or BAX proteins. However, the kinase can phosphorylate the pro-apoptotic protein BAD on serine 112, which accounts in part for its ability to reverse Bad-induced cell death. Our results indicate that pim-2 functions similarly to pim-1 as a pro-survival kinase and suggest that BAD is a legitimate PIM-2 substrate. [ABSTRACT FROM AUTHOR]

The potential of the Polar Silk Road (PSR) to accelerate the shipping trade is gaining importance because sea ice is currently melting in the Arctic region. Economic growth generates negative environmental and social impacts (TBL aspects); however, only a few studies have discussed the opportunities and threats in balancing TBL aspects. To fill this knowledge gap, this study addresses the opportunities and threats presented by the PSR. Nevertheless, information on the PSR is spread across diverse databases and is incomplete. This study utilizes a data-driven approach to handle these diverse databases and incomplete information and thereby to provide visual two- and three-dimensional impact analyses of opportunities and threats. This study contributes to the literature by (1) bridging TBL aspects of the PSR, (2) using a proposed hybrid method to address the aspect-balancing problem, and (3) providing insights on the opportunities and threats of the PSR. The results reveal negative effects of the PSR in terms of the balance between the economic and environmental aspects and that between the economic and social aspects. This study confirms that the scale and speed of development of the PSR pose a challenge to the environmental and social standards upheld by the surrounding countries and regions. The ability to maintain a (relatively) stable body temperature in a wide range of thermal environments by use of endogenous heat production is a unique feature of endotherms such as birds. Endothermy is acquired and regulated via various endocrine and molecular pathways, and ultimately allows wide aerial, aquatic, and terrestrial distribution in variable environments. However, due to our changing climate, birds are faced with potential new challenges for thermoregulation, such as more frequent extreme weather events, lower predictability of climate, and increasing mean temperature. We provide an overview on thermoregulation in birds and its endocrine and molecular mechanisms, pinpointing gaps in current knowledge and recent developments, focusing especially on non-model species to understand the generality of, and variation in, mechanisms. We highlight plasticity of thermoregulation and underlying endocrine regulation, because thorough understanding of plasticity is key to predicting responses to changing environmental conditions. To this end, we discuss how changing climate is likely to affect avian thermoregulation and associated endocrine traits, and how the interplay between these physiological processes may play a role in facilitating or constraining adaptation to a changing climate. We conclude that while the general patterns of endocrine regulation of thermogenesis are quite well understood, at least in poultry, the molecular and endocrine mechanisms that regulate, e.g. mitochondrial function and plasticity of thermoregulation over different time scales (from transgenerational to daily variation), need to be unveiled. Plasticity may ameliorate climate change effects on thermoregulation to some extent, but the increased frequency of extreme weather events, and associated changes in resource availability, may be beyond the scope and/or speed for plastic responses. Cryptogenic-organizing pneumonia (COP) is sensitive to corticosteroid therapy, whereas idiopathic pulmonary fibrosis (IPF) is not. Glucocorticoid receptor-α (GR-α) and histone deacetylase 2 (HDAC2) play critical roles in the sensitivity to corticosteroid therapy; however, it is unclear whether HDAC2 and/or GR-α are expressed in the lung tissues of patients with COP and/or IPF. Possible aberrant expressions of HDAC2 and GR-α in IPF and COP were investigated in the current study. Lung tissue samples were obtained from patients with COP (n = 9), IPF (n = 8), pulmonary abscesses (n = 7), or pulmonary inflammatory pseudotumors (n = 6) before corticosteroid treatment, as well as from control subjects (n = 10). The expression of GR-α, HDAC2, PI3K- δ , and NF-κBp65 in the samples was assessed by immunohistochemistry. GR-α expression was the same in lung tissues from COP patients and control subjects, but was significantly lower in lung tissue from IPF. In addition, HDAC2 was significantly higher in lung tissues of COP patients compared to both IPF and control subjects. Furthermore, the transcription factor NF-κBp65 was significantly lower in lung tissues from both COP and control compared to IPF subjects, whereas there was no difference in NF-κBp65 when comparing tissues from COP patients to controls. HDAC2 and GR-α were negatively correlated with NF-κBp65 in COP lung tissue. HDAC2 and GR-α expression in lung tissues are potential biomarkers for predicting corticosteroid sensitivity when initially treating COP and IPF, as well as other forms of ILD. Purpose: To establish a nomogram for predicting the occurrence of early biliary infection (EBI) after percutaneous transhepatic biliary stent (PTBS) placement in malignant biliary obstruction (MBO).Materials and Methods: In this multicenter study, patients treated with PTBS for MBO were allocated to a training cohort or a validation cohort. The independent risk factors for EBI selected by multivariate analyses in the training cohort were used to develop a predictive nomogram. An artificial neural network was applied to assess the importance of these factors in predicting EBI. The predictive accuracy of this nomogram was determined by concordance index (c-index) and a calibration plot, both internally and externally.Results: A total of 243 patients (training cohort: n = 182; validation cohort: n = 61) were included in this study. The independent risk factors were length of obstruction (odds ratio [OR], 1.061; 95% confidence interval [CI], 1.013-1.111; P = .012), diabetes (OR, 5.070; 95% CI, 1.917-13.412; P = .001), location of obstruction (OR, 2.283; 95% CI, 1.012-5.149; P = .047), and previous surgical or endoscopic intervention (OR, 3.968; 95% CI, 1.709-9.217; P = .001), which were selected into the nomogram. The c-index values showed good predictive performance in the training and validation cohorts (0.792 and 0.802, respectively). The optimum cutoff value of risk was 0.25.Conclusions: The nomogram can facilitate the early and accurate prediction of EBI in patients with MBO who underwent PTBS. We found that after stimulation for a few hours, memory but not naive CD4+ T cells produced a large amount of IFN- γ; however, the mechanism of rapid response of memory CD4+ T cells remains undefined. We compared the expression of transcription factors in resting or activated naive and memory CD4+ T cells and found that T-bet, but not pSTAT-1 or pSTAT-4, was highly expressed in resting memory CD4+ T cells and that phenotypic characteristics of T-bet+CD4+ T cells were CD45RAlowCD62Llow CCR7low. After short-term stimulation, purified memory CD4+ T cells rapidly produced effector cytokines that were closely associated with the pre-existence of T-bet. By contrast, resting naive CD4+ T cells did not express T-bet, and they produced cytokines only after sustained stimulation. Our further studies indicated that T-bet was expressed in the nuclei of resting memory CD4+ T cells, which might have important implications for rapid IFN-γ production. Our results indicate that the pre-existence and nuclear mobilization of T-bet in resting memory CD4+ T cells might be a possible transcriptional mechanism for rapid production of cytokines by human memory CD4+ T cells. Most proinflammatory actions of C-reactive protein (CRP) are only expressed following dissociation of its native pentameric assembly into monomeric form (mCRP). However, little is known about what underlies the greatly enhanced activities of mCRP. Here we show that a single sequence motif, i.e. cholesterol binding sequence (CBS; a.a. 35-47), is responsible for mediating the interactions of mCRP with diverse ligands. The binding of mCRP to lipoprotein component ApoB, to complement component C1q, to extracellular matrix components fibronectin and collagen, to blood coagulation component fibrinogen, and to membrane lipid component cholesterol, are all found to be markedly inhibited by the synthetic CBS peptide but not by other CRP sequences tested. Likewise, mutating CBS in mCRP also greatly impairs these interactions. Functional experiments further reveal that CBS peptide significantly reduces the effects of mCRP on activation of endothelial cells in vitro and on acute induction of IL-6 in mice. The potency and specificity of CBS are critically determined by the N-terminal residues Cys-36, Leu-37, and His-38; while the versatility of CBS appears to originate from its intrinsically disordered conformation polymorphism. The increase in atmospheric oxygen during the late Neoproterozoic Era (ca. 800-542 Ma) may have stimulated the oxygenation of the deep oceans and the evolution of macroscopic multicellular organisms. However, the mechanism and magnitude of Neoproterozoic oxygenation remain uncertain. We present Fe isotopes, Fe species and other geochemical data for two sections of the Doushantuo Formation (ca. 635-551 Ma) deposited after the Nantuo glacial episode in the Yangtze Gorge area, South China. It is highlighted that highly positive δ56Fe values reflect a lower oxidation rate of Fe(II)aq under ferruginous conditions, and in turn near zero δ56Fe values indicate oxidizing conditions. Our study suggests that during the deposition of the bottom of Member II of the Doushantuo Formation the shallow seawater was oxic, but the deep water was characterized by ferruginous conditions, which is consistent with a redox chemical stratification model. Subsequent anoxic conditions under shallow seawater, represented by positive δ56Fe and negative δ13Ccarb excursions, should be ascribed to the upwelling of Fe(II)aq and dissolved organic carbon (DOC)-rich anoxic deep seawater. The oxidation of Fe (II)aq and DOC-rich anoxic deep seawater upon mixing with oxic shallow water provides an innovative explanation for the well-known negative δ13Ccarb excursions (ENC2) and positive δ56Fe excursions in the middle of Doushantuo Formation. Meanwhile, the upwelling Fe (II)aq- and DOC-rich anoxic deep seawater could increase photosynthetic activity. Cell fate specification is typically initiated by a master regulator, which is relayed by tissue-specific regulatory proteins (usually transcription factors) for further enforcement of cell identities, but how the factors are coordinated among each other to "finish up" the specification remains poorly understood. Caenorhabditis elegans epidermis specification is initiated by a master regulator, ELT-1, that activates its targets, NHR-25 and ELT-3, two epidermis-specific transcription factors that are important for development but not for initial specification of epidermis, thus providing a unique paradigm for illustrating how the tissue-specific regulatory proteins work together to enforce cell fate specification. Here we addressed the question through contrasting genome-wide in vivo binding targets between NHR-25 and ELT-3. We demonstrate that the two factors bind discrete but conserved DNA motifs, most of which remain in proximity, suggesting formation of a complex between the two. In agreement with this, gene ontology analysis of putative target genes suggested differential regulation of metabolism but coordinated control of epidermal development between the two factors, which is supported by quantitative analysis of expression of their specific or common targets in the presence or absence of either protein. Functional validation of a subset of the target genes showed both activating and inhibitory roles of NHR-25 and ELT-3 in regulating their targets. We further demonstrated differential control of specification of AB and C lineage-derived epidermis. The results allow us to assemble a comprehensive gene network underlying C. elegans epidermis development that is likely to be widely used across species and provides insights into how tissue-specific transcription factors coordinate with one another to enforce cell fate specification initiated by its master regulator. Abstract: We report here the ontogenic changes in mRNA expression of chicken ghrelin (cGhrelin) and its receptor (cGHS-R1a) and the effects of fasting and refeeding on cGhrelin and cGHS-R1a mRNAs expression in 30-day-old broiler chickens. The level of cGhrelin mRNA in the proventriculus was low from embryo—day 15 (E15) to E19, but dramatically increased at post-hatching-day 2 (P2), then remained constant until P30 and followed by a significant decrease at P44 when there was a diet transition at P31 and thereafter. The decreased level was reversed at P58. Hypothalamic cGhrelin mRNA and proventriculus and hepatic cGHS-R1a mRNA were significantly increased at P30. The cGhrelin mRNA level in the proventriculus significantly increased in response to either 12-h or 36-h fasting but did not decrease after subsequent 12-h refeeding. The level of cGHS-R1a mRNA in the proventriculus was significantly upregulated in response to a 12-h fast but not to a 36-h fast and returned to the control level upon 12-h refeeding. Interestingly, it was apparent that the mRNA levels of both cGhrelin and cGHS-R1a in the liver were upregulated in response to fasting in a time-dependent manner and returned to the control level with subsequent refeeding. These results suggest that the expression pattern of ghrelin and its receptor mRNAs distinctly change in tissues depending on ontogenic stages and feeding states in poultry. [Copyright &y& Elsevier]

Abstract: We sought to determine the changes in brain interleukin-1β (IL-1β) following the coadministration of norfloxacin (25 mg/kg, i.p.) with biphenylacetic acid (100 mg/kg, p.o.) in rats. Norfloxacin provoked clonic convulsions in rats treated concomitantly with biphenylacetic acid, a major metabolite of the nonsteroidal anti-inflammatory drug fenbufen. Seizure activity was analyzed by EEG monitoring. Behavioral changes were also monitored. IL-1β expressions in the prefrontal cortex and hippocampus at different time intervals were studied by reverse transcriptase–polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The epileptiform discharges appeared in all the rats, accompanied with limb twitching and clonic–tonic seizures after administration of norfloxacin coadministered with biphenylacetic acid. Norfloxacin plus biphenylacetic acid-induced convulsions rapidly and transiently enhanced IL-1β mRNA in the prefrontal cortex and hippocampus. IL-1β mRNA expression in the prefrontal cortex and hippocampus was detected as soon as 30 min after norfloxacin injection, and decayed to control levels by 6 h. ELISA analysis revealed significant increase of the IL-1β protein in the prefrontal cortex and hippocampus at 2 h and 6 h. Administration of either norfloxacin or biphenylacetic acid alone did not elicit convulsions and increase in IL-1β mRNA and protein expressions. The results suggest that the increased IL-1β expressions in the prefrontal cortex and hippocampus induced by norfloxacin with biphenylacetic acid relate to seizure activities, and that these brain regions play pivotal roles in norfloxacin-induced convulsions. DREB1/C-repeat binding factor (CBF) is a plant-specific family of transcription factors and plays a crucial role in freeze tolerance. In the present work, two groups of drought-responsive element binding factor (DREB)-like genes were isolated from Brassica napus, named Group I and Group II. The two groups of genes were both induced by low temperature, but the expression of Group I preceded that of Group II. The Group I DREBs could specifically bind with the DRE cis-acting element and activate the expression of downstream genes, but Group II factors were trans-inactive although they still had the ability to bind with DRE, which was confirmed by electrophoretic mobility shift assay. Fluorescence quenching assays indicated that the DRE binding ability of the two groups was similar. Co-expression of Group II could depress the trans-activation activity of Group I DREB in a concentration-dependent manner. These results strongly suggested that the trans-active Group I DREBs were expressed at the early stage of cold stress to open the DRE-mediated signaling pathway in cold stress, whereas the trans-inactive Group II DREBs were expressed at the later stage to close the signal pathway in a competitive manner. Protein disulfide isomerase (PDI) functions as an isomerase to catalyze thiol:disulfide exchange, as a chaperone to assist protein folding, and as a subunit of prolyl-4-hydroxylase and microsomal triglyceride transfer protein. At a lower concentration of 0.2 µM, PDI facilitated the aggregation of unfolded rabbit muscle creatine kinase (CK) and exhibited anti-chaperone activity, which was shown to be mainly due to the hydrophobic interactions between PDI and CK and was independent of the cross-linking of disulfide bonds. At concentrations above 1 µM, PDI acted as a protector against aggregation but an inhibitor of reactivation during CK refolding. The inhibition effect of PDI on CK reactivation was fur. ther characterized as due to the formation of PDI-CK complexes through intermolecular disulfide bonds, a process involving Cys-36 and Cys-295 of PDI. Two disulfide-linked complexes containing both PDI and CK were obtained, and the large, soluble aggregates around 400 kDa were composed of 1 molecule of tetrameric PDI and 2 molecules of inactive intermediate dimeric CK, whereas the smaller one, around 200 kDa, was formed by 1 dimeric PDI and 1 dimeric CK. To our knowledge this is the first study revealing that PDI could switch its conformation from dimer to tetramer in its functions as a foldase. According to the observations in this research and our previous study of the folding pathways of CK, a working model was proposed for the molecular mechanism of CK refolding catalyzed by PDI. The effects of four fluoroquinolones (sparfloxacin, fleroxacin, ofloxacin and levofloxacin) on K+ currents were investigated in pyramidal neurons acutely isolated from rat hippocampus, to evaluate their relative potencies for inhibiting these channels. Using patch-clamp electrophysiological techniques, we found that all four compounds inhibited the delayed rectifier K+ current (IK), but with different potencies. Sparfloxacin was the most potent compound, displaying an IC50 value of 6.44×10−4 M, followed by fleroxacin, ofloxacin and levofloxacin, their IC50 values being 7.09×10−3, 8.42×10−3 and 1.10×10−2 M, respectively. In contrast, the fast transient K+ current (IA) was blocked only by sparfloxacin (IC50=2.86×10−3 M) and fleroxacin (IC50=4.38×10−3 M), but not by ofloxacin and levofloxacin even at concentrations up to 1 mM. The K+ current inhibition was reversible after washout of the compounds. Further study is needed to clarify the possible involvement of this novel action in the adverse effects of fluoroquinolones in the central nervous system (CNS). [Copyright &y& Elsevier]

Hierarchical porous activated carbon (AC) was obtained by the carbonization of dried spinach leaves followed by activation with KOH. The product was characterized by nitrogen adsorption, FT-IR, FE-SEM and electrochemical tests. The AC has a dominant number of micropores, a medium number of mesopores and a few macropores with a specific surface area up to 2616m²/g and a large number of oxygen-containing functional groups. The AC electrode exhibits a good double-layer capacitive behavior with a specific capacitance of 238F/g in a 2mol/L KOH electrolyte. A supercapacitor made of the AC has a high energy density of 10.1Wh/kg at a current density of 0.5A/g and an excellent cyclic stability for 2000 cycles over a potential range from 0 to 1.2V.
