13. 8',9'-Bis-nor-neolignans from Cedrela odorata and their antiangiogenic activity.

Abstract

Two previously undescribed 8',9'-bis- nor -neolignans, 9-acetyltoonin C (1) and threo -guaiacylglycerol-β-methyl vanillate ether (2), were isolated from the stem bark of Cedrela odorata (Meliaceae), together with 4,5-dihydroblumenol A, 2-(3,4-dimethoxyphenyl)ethyl-β- d -glucopyranoside, scopoletin, homovanillic alcohol, and 7-megastigmene-3,6,9-triol. All compounds were characterized by 1D and 2D NMR and HRESIMS data. The new compounds and other lignans obtained from our chemical library, namely erythro -4,7,9,9'-tetrahydroxy-3,3'-dimethoxy-8- O -4'-oxyneolignan (3), urolignoside (4), secoisolariciresinol 9- O -β-D-glucopyranoside (5), icariside E 3 , and erythro -4,7,9,9'-tetrahydroxy-3,3'-dimethoxy-8- O -4'-oxyneolignan-9'- O -β-D-glucopyranoside, were then evaluated for their antiangiogenic activity by the in vivo zebrafish embryos test. Compounds 1 , 2 , and 3 exhibited the best antiangiogenic response, while 4 and 5 showed mild inhibitory effects on EAP release after treatment of zebrafish embryos. In particular, compound 2 induced an inhibition of 42.56 % at 10 μM, similarly to 2-methoxyestradiol used as reference compound. Abstract

We examine the problem of deflagration of a flammable gaseous mixture contained within an elastic spherical shell and ignited at its centre. We predict analytically the pressure wave radiated by an expanding spherical deflagration front and we study its interaction with the elastic shell prior to failure. We predict the waves reflected at the inner wall of the shell and radiated in the space outside the shell, which we assume initially filled with air at atmospheric pressure, and we calculate the magnitude of these waves as a function of density, thickness and elastic modulus of the shell. The findings of this model are used to critically assess laboratory setups used in controlled deflagration experiments, in which a flammable mixture is initially contained by a soft shell. The interpretation of data from these tests currently assumes that the gaseous mixture is effectively unconfined during the deflagration, due to the low mass, low stiffness and early failure of the containing shell. We show that this assumption is not adequate and it leads to errors in the interpretation of the measurements; the proposed model is used to quantify these errors. [ABSTRACT FROM AUTHOR]

Abstract

The threat of accidental hydrocarbon explosions is of major concern to industrial operations; in particular, there is a need for design tools to assess and quantify the effects of potential deflagration events. Here we present a design methodology based on analytical models that allow assessing the loading and structural response of objects exposed to pressure waves generated by deflagration events. The models allow determining: i) the importance of Fluid-Structure Interaction (FSI) effects; ii) the transient pressure histories on box-like or circular cylindrical objects, including the effects of pressure clearing; iii) the dynamic response of structural components that can be idealised as fully clamped beams. We illustrate by three case studies the complete design methodology and validate the analytical models by comparing their predictions to those of detailed CFD and FE simulations. We employ the validated analytical models to perform Monte Carlo analyses to quantify, for box-like structures, how the uncertainty in input design variables propagates through to the expected maximum force and impulse.
