Abstract

The tunnel engineering has become increasingly common with the needs of urban development. However, excavation sequence has a significant effect on the stability of the tunnel perimeter rock, which can lead to more unpredictable perimeter rock hazards and more difficulty in maintaining perimeter rock stability. In this study, to investigate the effect of different excavation sequences on tunnel deformation and creep, biaxial compression experiments were conducted on muddy siltstone specimens by using the acoustic emission (AE) and digital image correlation (DIC) techniques. The results indicated that, as the distance from the tunnel portal increases, the amount of stress and strain change decreases, with the largest values of stress changes occurring in the tunnel vault and middle area of twin tunnels. The specimen undergoes two stages in the creep process: deceleration creep and steady-state creep. The steady-state creep rate of the measurement points depends on the degree of excavation disturbance impact it experiences. The AE events during excavation can be divided into two phases: the quiet period when the specimen is not excavated and the active period when the specimen is excavated, and the cumulative counting curve shows a "stepwise" increase with the number of excavations. There is a sudden drop in the b-value during excavation and a decrease in the b-value during the deceleration creep phase. The results of this study can reflect the creep patterns of the surrounding rock under different excavation sequences, which can help to choose a more suitable excavation plan in terms of rock creep stability. Abstract

Our objective was to determine the feasibility of extending our previously reported PET imaging study of pancreatic cancer (PnCa) with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 to radioimmunotherapy (RIT) by exploiting the β-particle and Auger electron emissions of 64Cu (PET theranostic concept). To enhance the effectiveness of 64Cu]Cu-NOTA-panitumumab F(ab′) 2 , we further combined RIT with radiosensitizing gemcitabine (GEM) and the poly(ADP)ribose polymerase inhibitor (PARPi), rucaparib. Normal tissue toxicity was assessed in non-tumor-bearing NOD-scid mice injected i.v. with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (1.85–9.25 MBq; 10 μg) or 64Cu]Cu-NOTA-anti-mouse EGFR Ab30 F(ab′) 2 (12.95 MBq). Body weight was monitored, and hematopoietic (CBC), liver (ALT) and kidney [creatinine (SCr)] toxicity were assessed. RIT studies were performed in NOD-scid mice with s.c. OCIP23 human PnCa patient-derived xenografts (PDX) administered 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (3.7 MBq; 10 μg), unlabeled panitumumab F(ab′) 2 (10 μg) or normal saline every two weeks. Subsequent studies evaluated RIT with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (12.95 MBq; 10 μg) administered alone or combined with GEM and the PARPi, rucaparib administered on a 14-day treatment cycle for up to 6 cycles in NOD-scid mice with s.c. PANC-1 human PnCa xenografts. The radiation absorbed dose in PANC-1 tumors and normal organs in mice after a single i.v. injection of 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (12.95 MBq; 10 μg) was estimated based on previously reported biodistribution studies of 64Cu]Cu-NOTA-panitumumab F(ab′) 2. No normal tissue toxicity was observed in non-tumor-bearing NOD-scid mice administered up to 3.7 MBq (10 μg) of 64Cu]Cu-NOTA-panitumumab F(ab′) 2 but slightly increased ALT was noted at 9.25 MBq. Administration of 64Cu]Cu-NOTA-anti-mouse EGFR Ab30 F(ab′) 2 (12.95 MBq; 10 μg) caused some hematopoietic toxicity but no increase in ALT or SCr or decreased body weight. A slight tumor growth delay and increased survival was noted in NOD-scid mice with s.c. OCIP23 PDX treated with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (3.7 MBq; 10 μg) or unlabeled panitumumab F(ab′) 2 (10 μg) compared to normal saline treated mice. RIT with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (12.95 MBq; 10 μg) combined with GEM + PARPi for up to 6 cycles was most effective for the treatment of PANC-1 tumors. Tumor doubling time increased to 13.3 ± 0.9 days vs. 7.8 ± 3.7 days for RIT alone and 9.3 ± 2.2 days for normal saline treatment. Median survival was significantly longer (P < 0.05) than in mice treated with normal saline (35 days) for RIT + GEM + PARPi (71 days), GEM + PARPi (44 days) and RIT + GEM (43 days) but not for RIT alone (25 days). RIT + GEM + PARPi provided a longer median survival than RIT (P < 0.01), GEM + PARPi (P = 0.01) but not RIT + GEM (P = 0.23). Nonetheless, PANC-1 tumors grew exponentially in all treatment groups. The absorbed dose in PANC-1 tumors after a single i.v. injection of 64Cu]Cu-NOTA-panitumumab F(ab′) 2 (12.85 MBq; 10 μg) was 0.8 Gy, while the dose in normal organs ranged from 0.6–1.2 Gy. We conclude that RIT with 64Cu]Cu-NOTA-panitumumab F(ab′) 2 did not cause significant normal tissue toxicity but was not effective when administered alone for treatment of PnCa xenografts in NOD-scid mice. Combining RIT with GEM and the PARPi, rucaparib enhanced its effectiveness but tumors continued to grow exponentially. Our results suggest that 64Cu is not feasible for RIT of PnCa due to low tumor absorbed doses. 177Lu which has a higher abundance of moderate energy β-particle emissions may be more effective than 64Cu. The hematopoietic toxicity of 64Cu]Cu-NOTA-anti-mouse EGFR Ab30 F(ab′) 2 may be mediated by binding to mouse EGFR expressed on some hematopoietic stem cells. Direct extension of PET with 64Cu(Cu)-NOTA-panitumumab F(ab′) 2 to RIT exploiting the β-particle and Auger electron emissions of 64Cu is not feasible. Abstract

Can single-nucleotide polymorphisms (SNP) of genes related to progesterone synthesis predict the risk of premature serum progesterone elevation in women undergoing gonadotrophin-releasing hormone agonist protocols for ovarian stimulation? A total of 765 women were divided into high progesterone and normal progesterone groups according to progesterone concentration on the day of human chorionic gonadotrophin (HCG) administration, with the 75th percentile as the threshold between the group. Associations were analysed of genetic information from whole exome sequencing and the clinical characteristics of the two groups to identify the relationship between SNP, haplotypes and serum progesterone elevation. Among 40 common SNP of eight genes (FSHR, LHCGR, ESR1, ESR2, PGR, HSD3B1, CYP11A1 and CYP17A1), no statistical significance between the high and normal progesterone groups was identified in the distribution of genotypes and allele frequencies after multiple test correction to adjust the false discovery rate (P FDR > 0.05). When compared with the most common haplotypes of each gene, haplotype GAAG in CYP17A1 was associated with a 1.44-fold increased risk of progesterone elevation (95% confidence interval [CI] 1.22–1.69, P FDR < 0.001), while haplotypes of the following genes showed a decreased risk of progesterone elevation: haplotype CC in FSHR and LHCGR (0.66-fold, P FDR = 0.020, and 0.64-fold, P FDR < 0.001, respectively), CA in ESR1 (0.90-fold, P FDR < 0.001), TCTGG in ESR2 (0.92-fold, P FDR = 0.007) and GAACC in HSD3B1 (0.42-fold, P FDR < 0.001). In this paper, a new formulation of three-dimensional spherical discontinuous deformation analysis (DDA) based on second-order cone programming has been proposed. Artificial springs with open-close iteration used in classic DDA have been removed, given that improper stiffness parameters might cause numerical problems. Furthermore, to account for irregular granular shapes, a rolling resistance model is incorporated in the variational formulation. The proposed formulation can be cast into a standard second-order cone programming program, which can be solved using efficient off-the-shelf optimisation solvers. Abstract In this paper, an innovative method to replace steel reinforcement cage by steel fiber to form a steel shape and steel fiber reinforced concrete has been proposed to solve the problems of steel reinforced concrete structure during construction, such as position conflict between shape steel and steel bars, or difficulty of concrete placement, etc. Eighteen beams were tested under a four-point bending configuration to study the effectiveness of steel fibers on shear and flexural strengths, failure mechanisms, crack control, and ductility. The major factors considered were shear span depth ratio (λ = 2.5, 3.5), with or without longitudinal reinforcement and web reinforcement (none, stirrups and/or steel fibers). The response of Steel Fiber Steel Reinforcement Concrete (SFSRC) beams were assessed depending on the results of crack patterns, load at yielding, failure and ultimate bearing capacity, failure modes, ductility and displacement increment of plastic development, The experimental results showed that the addition of steel fibers improves the mechanical response, specifically speaking, flexural and shear strengths and ductility of the tested specimens. The longitudinal reinforcement can greatly enhance the bearing capacity, and the stirrups and steel fibers can make the reinforcement effect more prominent. The stitching effect of steel fiber between cracks leads to stable bearing capacity of the specimens always stable after the peak load. Increasing the content of steel fiber contributes to delay or even avoid bond failure. Steel fibers can be an alternative to shear reinforcement. A formula was proposed to predict the bearing capacity of SFSRC specimens, and the comparison between predictions and experiment results proved the reasonable of the formula. Abstract 5-Aminolevulinic acid photodynamic therapy(ALA-PDT) has been widely used for cervical cancer treatment, but the mechanisms are still not fully delineated. Here we showed that ALA-PDT significantly upregulated HMGB1 while downregulated miR-34a expression levels in cervical cancer tissues, and the percentages of mature DCs(mDCs) were increased in ALA-PDT treated patients' peripheral blood. After treating HPV-positive Hela, SiHa, Caski and HPV-negative C33 A cervical cancer cell lines with ALA-PDT, HPV-positive cells' proliferative ability was significantly inhibited and apoptosis rates were elevated, while no significant changes were found in HPV-negative C33 A cell line. Most importantly, in HPV-positive cells, we found that miR-34a were downregulated in cytoplasm, and both cytoplasm and exosome HMGB1 were significantly elevated comparing to cancer cells without ALA-PDT treatment, and it could be reversed by miR-34a mimic transfection, which indicated that HPV infection and miR-34a downregulation might be vital for ALA-PDT treatment. Based on the HMGB1 is the potential target of miR-34a and an inverse correlation between miR-34a and HMGB1 in ALA-PDT treated cancer tissues, we verified that HMGB1 could be targeted and downregulated by miR-34a mimic, and ALA-PDT promotes HMGB1 secretion by inhibiting miR-34a expression. By co-culturing cervical cancer cell lines with immature DCs(imDCs) in the Transwell systems, we found that ALA-PDT induced HMGB1 exosomes could promote DCs maturation, which could be reversed by silencing HMGB1 in HPV-positive cervical cancer cells. In vivo animal experiments also proved that ALA-PDT inhibited tumor growth in tumor bearing mice, which was reversed by co-transfected with miR-34a mimic or silencing HMGB1 in HPV-positive cells. Hence we concluded that ALA-PDT treatment specifically inhibited HPV-positive cervical cancer cells' proliferative ability, promoted cell apoptosis and modulated DCs maturation by regulating miR-34a mediated HMGB1 exosomes secretion.
