Three undescribed lanostane triterpenoids, ganoellipsic acids A–C (1–3), together with seven known Ganoderma lanostanoids (4 – 10), were isolated from artificially cultivated fruiting bodies of Ganoderma ellipsoideum , strain BCC 16634. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The C-25 absolute configuration of 1 was determined to be 25 S using the phenylglycine methyl ester (PGME) method. A new 3,4- seco -27-norlanostane triterpene, ganoboninketal D ( 1 ), a new lanostane, (24 S )-3-oxo-7α,24,25-trihydroxylanosta-8-ene ( 2 ), together with six known lanostanes ( 3 – 8 ), were isolated from cultivated fruiting bodies of the basidiomycete Ganoderma orbiforme . The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data, and the structures 1 and 2 were further confirmed by chemical correlations to ganoboninketal C and ganodermanondiol, respectively. All isolated compounds were inactive in the antitubercular and antimalarial activity assays against Mycobacterium tuberculosis H37Ra and Plasmodium falciparum K1, respectively, except that compound 8 exhibited weak antitubercular activity (MIC 50 μg/ml). Twelve previously unsubscribed compounds, (9 Z)-9-methoxystrobilurins B (1) and G (2), oudemansin Y (3), 10,14-dehydrooudemansin B (4), favolasin B (5), favolasinones A (6) and B (7), favodehydes G (8) and K (9), 3-methylbenzo[ b ]oxepin-7-carboxylic acid (10), the prenylated primary benzamide 11 , and its hydroxamine derivative 12 , together with sixteen known compounds were isolated from the fungus Favolaschia xtbgensis TBRC-BCC 18687. Compounds in the class of strobilurins and oudemansins, 1 , 2 , and 4 , exhibited strong to moderate antimalarial and cytotoxic activity against NCI–H187 cells. The most potent antimalarial compound was the (9 Z)-9-methoxystrobilurins G (2 , IC 50 0.090 μM), which was comparable to that of its (9 E)-isomer (IC 50 0.061 μM), even though the 3D structures of these two compounds were quite different. Three previously undescribed highly modified lanostane triterpenoids, ganopyrone A, ganocolossusin I, and ganodermalactone Y, were isolated from the artificially cultivated fruiting bodies of the basidiomycete Ganoderma colossus TBRC-BCC 17711. Ganopyrone A possesses an unprecedented polycyclic carbon skeleton with an α -pyrone ring and C-18/C-23 bond. It showed antimalarial activity against Plasmodium falcip arum K1 (multidrug-resistant strain) with an IC 50 value of 7.8 μM (positive control: dihydroartemisinin, IC 50 1.4 nM), while its cytotoxicity (Vero cells) was much weaker (IC 50 103 μM). Twelve aromadendrane sesquiterpenoids, inonotins A–L, and a previously unknown cyclofarnesane, i.e., inonofarnesane, together with two known compounds, were isolated from cultures of the wood-rotting basidiomycete Inonotus sp. BCC 23706. Inonotin I is identical to a previously reported compound with an incorrect structure. Structures of the compounds were elucidated by spectroscopic analysis and X-ray crystallography. Abstract: Seven lanostane triterpenoids, ganorbiformins A–G, together with twelve known compounds, were isolated from cultures of the mushroom fungus Ganoderma orbiforme BCC 22324. Ganorbiformin A is an unusual rearranged analog, whereas the other compounds share the same lanostane skeleton with known ganoderic acids. The C-3 epimer of ganoderic acid T also exhibited significant antimycobacterial activity against Mycobacterium tuberculosis H37Ra (MIC 1.3μM). [Copyright &y& Elsevier]

The absolute configuration at C-25 of ganoderic acid A and its derivatives was determined to be 25 R by application of the phenylglycine methyl ester (PGME) method. Among the isolated compounds, ganoderiol F exhibited the most potent activity against Mycobacterium tuberculosis H37Ra with an MIC value of 0.781 μg/ml. Sixteen previously undescribed lanostane-type triterpenoids (1 – 16), together with fourteen known compounds, were isolated from cultivated fruiting bodies of the basidiomycete Ganoderma casuarinicola , a recently described species. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data.
