Solvation characteristics in homogeneous and heterogeneous media have been probed by monitoring the band width of ICT band of 2,6-di-phenyl-4(2,4,6-triphenyl-1-pyridino) phenolate, the indicator solute for ET(30) scale, in pure, mixed binary solvents and aqueous micellar solution. Non-ideal solvation behaviour is observed in all the binary solvent mixtures. Index of preferential solvation has been calculated as a function of solvent composition. Study in micellar media indicates that the dye is located at the micelle–water interface. The effects of variation of micelle concentration, temperature and electrolyte concentration have also been studies. [Copyright &y& Elsevier]

Bladder cancer (BCa) stands as prevalent malignancy of the urinary system globally, especially among men. The clinical classification of BCa into non-muscle invasive bladder cancer (NMIBC) and muscle-invasive bladder cancer (MIBC) is crucial for prognosis and treatment decisions. However, challenges persist in current diagnostic methods like Urine cytopathology that shows poor sensitivity therefore compromising on accurately diagnosing and monitoring BCa. In recent years, research has emphasized the importance of identifying urine and blood-based specific biomarkers for BCa that can enable early and precise diagnosis, effective tumor classification, and monitoring. The convenient proximity of urine with the urinary bladder epithelium makes urine a good source of noninvasive biomarkers, in particular urinary EVs because of the packaged existence of tumor-associated molecules. Therefore, the review assesses the potential of urinary extracellular vesicles (uEVs) as noninvasive biomarkers for BCa. We have elaborately reviewed and discussed the research that delves into the role of urinary EVs in the context of BCa diagnosis and classification. Extensive research has been dedicated to investigating differential microRNA (miRNA) expressions, with the goal of establishing distinct, noninvasive biomarkers for BCa. The identification of such biomarkers has the potential to revolutionize early detection, risk stratification, therapeutic interventions, and ultimately, the long-term prognosis of BCa patients. Despite notable advancements, inconsistencies persist in the biomarkers identified, methodologies employed, and study populations. This review meticulously compiles reported miRNA biomarkers, critically assessing the variability and discrepancies observed in existing research. By synthesizing these findings, the article aims to direct future studies toward a more cohesive and dependable approach in BCa biomarker identification, fostering progress in patient care and management. Several studies on experimental animals indicate that the process of organogenesis crucially depends upon the spatiotemporal dose of certain critical bio-molecules. Tooth development is also not an exception. While most of the knowledge regarding the molecular mechanism of tooth development comes from the studies on mouse model, pathogenic variations identified in human tooth agenesis also provide valuable information on mammalian tooth development. Until now five major candidate genes have been identified for tooth agenesis in human. Among them, PAX9 plays the crucial role in tooth development and in non-syndromic congenital tooth agenesis. In this study, microsatellite and SNP based genotyping identifies a disease specific haplotype block, which includes PAX9 gene, segregates with autosomal dominant tooth agenesis phenotype. Direct sequencing of PAX9 identifies a novel heterozygous G to A transition at the third base (c.3G > A) of initiation codon leading to ATG to ATA shift in all affected individuals which is absent in all unaffected relatives and 200 control chromosomes. Further, in vitro functional analysis creating PAX9 minigene construct did apparently show no effect on the splice-site migration. It is therefore proposed that haploinsufficiency of PAX9 is the causal factor for tooth agenesis in this family. [ABSTRACT FROM AUTHOR]

In this study, water dispersible fluorescent carbon quantum dot (CQD) has been synthesised, having an average size of 8.6 ± 0.4 nm using Cynodon dactylon (CD) following microwave assisted green synthetic one-step method. As-prepared CQD fluoresces strongly at 444 nm having a quantum yield of 1% in water when excited at 350 nm. This fluorescence of CQD is sensitive toward As3+ and Fe3+ metal ions. These CQD are utilized for dual metal ion fluorescence sensing; turn-on fluorescence sensing for As3+ and turn-off fluorescence sensing for Fe3+ ions. Limit of detection for As3+ and Fe3+ ions has been found to be 19 nM and 0.10 μM respectively, which is the lowest value reported for As3+ without any functionalization. The adsorption kinetics of As3+ and Fe3+ ions on CQD have been examined using pseudo-first-order-kinetic model revealing that physical adsorption is dominant over chemical processes in this work. For 0.41 g/L and 1.90 g/L dose of CQD, the equilibrium adsorption capacity was found to be 1.57 × 10−6 mg/g, 2.91 × 10−7 mg/g, and 1.01 × 10−5 mg/g, 1.69 × 10−6 mg/g respectively for As3+ and Fe3+ ions. Despite having low quantum yield in water, as-prepared CQD showed low cytotoxicity and good tolerance against photodegradation of biological cells at concentrations lower than 62.5 μg/mL and when the cells are illuminated up to 12 h. Owing to this, the synthesised CQD have been utilized as fluorescent probes for in itro cell imaging. Loss-of-function mutation in PINK1 is known for causing autosomal recessive early onset Parkinsonism accounting approximately 6.5% of PD cases. Recently, PINK1 has also been shown to cause Parkinson’s disease (PD) in eastern India. Present study is aimed to see its contribution in north-Indian PD patients. A total of 106 PD patients and 60 ethnically matched healthy controls were included in the study. All the patients were screened for mutation in PINK1 by direct DNA sequence analysis of the PCR amplicons covering all exons and exon–intron boundaries. Identified novel variant was reconfirmed by DNA sequencing of 10 randomly selected TA clones containing the variant amplicon. In vitro functional assay of the mutant protein was performed by transfecting COS-7 cell line with wild type and mutant (created by site-directed-mutagenesis) cDNA construct of PINK1 fused to N′ terminal GFP followed by western blot analysis. Two potentially pathogenic, one being novel (p.Q267X) and 6 other apparently non-pathogenic variants were identified. Western blot analysis reveals production of truncated PINK1 fusion protein of ∼55 kDa in p.Q267X mutant instead of 82/93 kDa of wild type PINK1 fusion protein (molecular weight of GFP is ∼27 kDa). Our study concludes that PINK1 variants are prevalent for causing Parkinson’s disease (PD) in India, as revealed by the occurrence of 1.8% (2/106) in PD patients from north Indian population. The novel homozygous variant of PINK1 (c.799C > T) reported here is the plausible cause for disease manifestation in this patient. Future study, however, would be helpful to understand the functional mechanism how this premature PINK1 protein (p.Q267X) responds to cellular stress leading to the PD pathophysiology. The present work deals with the synthesis of efficient water soluble O-carboxymethyl chitosan (O-CMCS) based nanocomposites (NCs) with nanostructured zinc oxide (n-ZnO) by ex-situ grafting method for the delivery of anticancer drug curcumin (Cr). The phase identification, morphology and thermal stability of prepared Cr/O-CMCS/n-ZnO NCs have been investigated by FT-IR, XRD, SEM, TEM and TGA/DTA techniques. The drug entrapment efficiency have found to be 74% and in vitro drug release study performed at 37 °C at pH 4.5 and 7.4 indicated slow and controlled release in the initial phase and sustained in later phase. The MTT assay showed higher and preferential toxicity of the Cr/O-CMCS/n-ZnO NCs against cancer cells (MA104). The cellular uptake study by FACS revealed concentration dependent uptake of the NCs. The utilization of this Cr/O-CMCS/n-ZnO nanoformulation offers an efficient strategy and a novel promising soluble nanomatrix for anticancer therapy and other biomedical applications. A new pyrene-pyridine dithiocarbamate based fluorescent chemodosimeter, potassium (pyren-1-ylmethyl)(pyridin-2-ylmethyl)dithiocarbamate (L1) has been designed and synthesized. The chemodosimeter shows high selectivity and sensitivity (5.2 ppb) for Hg2+ in pure aqueous medium in which emission intensity was quenched by ≈80% due to the formation of new cyclized species, 1. The probe behaves as a chemodosimeter for Hg2+ ions and forms Hg2+ triggered cyclised imidazoline species with approximate detection time of 50 s and exhibits both colorimetric and fluorometric changes on detection of Hg2+ ion. Color of the probe (L1) changed from green to colorless visible to the naked eye and from green to dark blue upon the addition of Hg2+ ions under UV light. The Hg2+ triggered cyclization reaction was confirmed by spectral data analysis and a single crystal structure determination of the cyclised entity 2 obtained from the model compound potassium benzyl(pyridin-2-ylmethyl) dithiocarbamate (L2). L1 finds its application for detection of Hg2+ ions on paper strips, and in BSA (bovine serum albumin) medium. L1 is also applicable for the monitoring of Hg2+ ion in NIH3T3 live cells.
