Little is known about longitudinal associations between added sugar consumption (solid and liquid sources) and glucose-insulin homeostasis among youth. Caucasian children (8-10 y) with at least one obese biological parent were recruited in the QUébec Adipose and Lifestyle InvesTigation in Youth (QUALITY) cohort (n = 630) and followed-up 2 y later (n = 564). Added sugars were assessed by 3 24-h dietary recalls at baseline. Two-year changes were examined in multivariate linear regression models, adjusting for baseline level, age, sex, Tanner stage, energy intake, fat mass (dual-energy X-ray absorptiometry), and physical activity (7 d accelerometer). Added sugar intake in either liquid or solid sources was not related to changes in adiposity measures (fat mass, body mass index, or waist circumference). However, a higher consumption (10 g/d) of added sugars from liquid sources was associated with 0.04 mmol/L higher fasting glucose, 2.3 pmol/L higher fasting insulin, 0.1 unit higher homeostasis model assessment of insulin resistance (HOMA-IR), and 0.4 unit lower Matsuda-insulin sensitivity index (Matsuda-ISI) in all participants (P < 0.01). No associations were observed with consumption of added sugars from solid sources. Overweight/obese children at baseline had greater increases in adiposity indicators, fasting insulin, and HOMA-IR and decreases in Matsuda-ISI during those 2 y than normal-weight children. Consumption of added sugars from liquid or solid sources was not associated with changes in adiposity, but liquid added sugars were a risk factor for the development of impaired glucose homeostasis and insulin resistance over 2 y among youth at risk of obesity. The Wnt signaling plays pivotal roles in embryogenesis and cancer, and the three DIX domain-containing proteins, Dvl, Axin, and Ccd1, play distinct roles in the initiation and regulation of canonical Wnt signaling. Overexpressed Dvl has a tendency to form large polymers in a cytoplasmic punctate pattern, whereas the biologically active Dvl in fact forms low molecular weight oligomers. The molecular basis for how the polymeric sizes of Dvl proteins are controlled upon Wnt signaling remains unclear. Here we show that Ccd1 up-regulates canonical Wnt signaling via acting synergistically with Dvl. We determined the crystal structures of wild type Ccd1-DIX and mutant Dvl1-DIX(Y17D), which pack into "head-to-tail" helical filaments. Structural analyses reveal two sites crucial for intra-filament homo- and hetero-interaction and a third site for inter-filament homo-assembly. Systematic mutagenesis studies identified critical residues from all three sites required for Dvl homo-oligomerization, puncta formation, and stimulation of Wnt signaling. Remarkably, Ccd1 forms a hetero-complex with Dvl through the "head" of Dvl-DIX and the "tail" of Ccd1-DIX, depolymerizes Dvl homo-assembly, and thereby controls the size of Dvl polymer. These data together suggest a molecular mechanism for Ccd1-mediated Wnt activation in that Ccd1 converts latent polymeric Dvl to a biologically active oligomer(s). [ABSTRACT FROM AUTHOR]

In this study, the mercury adsorption characteristics of HBr-modified fly ash in an entrained-flow reactor were investigated through thermal decomposition methods. The results show that the mercury adsorption performance of the HBr-modified fly ash was enhanced significantly. The mercury species adsorbed by unmodified fly ash were HgCl2, HgS and HgO. The mercury adsorbed by HBr-modified fly ash, in the entrained-flow reactor, existed in two forms, HgBr2 and HgO, and the HBr was the dominant factor promoting oxidation of elemental mercury in the entrained-flow reactor. In the current study, the concentration of HgBr2 and HgO in ash from the fine ash vessel was 4.6 times greater than for ash from the coarse ash vessel. The fine ash had better mercury adsorption performance than coarse ash, which is most likely due to the higher specific surface area and longer residence time. [ABSTRACT FROM AUTHOR]

The adsorption of carbon dioxide on hydrotalcite-like compounds was investigated. Two different powdered hydrotalcites were used containing the cations nickel and iron. The powdered materials were screened for carbon dioxide adsorption using a thermogravimetric method and it was found that NiMgAl (Sample 1) hydrotalcite has the largest capacity for CO2, adsorbing 1.58 mmol g-1 at 20 °C, and highest rate of adsorption of up to 0.17 mmol g-1 min-1. This represented an increase of 53% in adsorption capacity, compared with NiMgAlFe (Sample 2). In order to improve the rheological behaviour of hydrotalcite paste for extrusion, hydrotalcite powders were combined with boehmite alumina (70:30 and 50:50 ratios of hydrotalcite:boehmite) before extrusion into pellets suitable for use in a fixed bed adsorber. These pellets were then re-crushed and further tested by thermogravimetric methods. The effects of temperature, composition and pre-treatment of the hydrotalcites on the adsorption of carbon dioxide and nitrogen are reported. At 20 °C, the amount of carbon dioxide adsorbed was between 2.0 and 2.5 mmol g-1 for all the hydrotalcite/alumina samples in this study, although this decayed rapidly with increasing temperature. The results are compared with silica gel as a common sorbent reference, and with literature values. Hydrotalcite/alumina samples have thermal stability and a high adsorption capacity for carbon dioxide over a wide range of temperatures. The composition of the hydrotalcite/alumina pellets investigated in this study has less effect upon the adsorption behaviour compared with the non-calcined hydrotalcite powder, thus allowing a wide choice of pellet compositions to be used. Abscisic acid (ABA) is one of the most important phytohormones in plant. PYL proteins were identified to be ABA receptors in Arabidopsis thaliana. Despite the remarkably high degree of sequence similarity, PYL1 and PYL2 exhibit distinct responses toward pyrabactin, an ABA agonist. PYL1 inhibits protein phosphatase type 2C upon binding of pyrabactin. In contrast, PYL2 appears relatively insensitive to this compound. The crystal structure of pyrabactin-bound PYL1 revealed that most of the PYL1 residues involved in pyrabactin binding are conserved, hence failing to explain the selectivity of pyrabactin for PYL1 over PYL2. To understand the molecular basis of pyrabactin selectivity, we determined the crystal structure of PYL2 in complex with pyrabactin at 1.64 Å resolution. Structural comparison and biochemical analyses demonstrated that one single amino acid alteration between a corresponding valine and isoleucine determines the distinct pyrabactin selectivity by PYL1 and PYL2. Pyrabactin is a synthetic abscisic acid (ABA) agonist that selectively inhibits seed germination. The use of pyrabactin was pivotal in the identification of the PYR1/PYL/RCAR family (PYL) of proteins as the ABA receptor. Although they both act through PYL proteins, pyrabactin and ABA share no apparent chemical or structural similarity. It remains unclear how pyrabactin functions as an ABA agonist. Here, we report the crystal structure of pyrabactin in complex with PYL1 at 2.4 Å resolution. Structural and biochemical analyses revealed that recognition of pyrabactin by the pocket residues precedes the closure of switch loop CL2. Structural comparison between pyrabactin- and ABA-bound PYL1 reveals a general principle in the arrangements of function groups of the two distinct ligands. MecA is an adaptor protein that regulates the assembly and activity of the ATP-dependent ClpCP protease in Bacillus subtilis. MecA contains two domains. Although the amino-terminal domain of MecA recruits substrate proteins such as ComK and Com'S, the carboxyl-terminal domain (residues 121-218) has dual roles in the regulation and function of ClpCP protease. MecA-(121-218) facilitates the assembly of ClpCP oligomer, which is required for the protease activity of C1pCP. This domain was identified to be a non-recycling degradation tag that targets heterologous fusion proteins to the C1pCP protease for degradation. To elucidate the mechanism of MecA, we determined the crystal structure of MecA-(121-218) at 2.2 Å resolution, which reveals a previously uncharacterized α/β fold. Structure-guided mutagenesis allows identification of surface residues that are essential for the function of MecA. We also solved the structure of a carboxyl-terminal domain of YpbH, a paralogue of MecA in B. subtilis, at 2.4 Å resolution. Despite low sequence identity, the two structures share essentially the same fold. Comparing with traditional methods, high speed centrifugal spinning has the advantages of simple fabrication principle and high preparation efficiency. In this study, the flow law of spinning solution in the nozzle and the effect of spinning parameters on the morphology of nanofibers were explored. A motion model of spinning solution in the nozzle was established to study the velocity distribution of spinning solution in the nozzle, moreover, the relationship between the critical rotational speed required to form the jet and the spinning parameters was studied. It was found that the current velocity of spinning solution was mainly concentrated at the axis of the nozzle and shifted in the direction of Coriolis acceleration. The relationship between spinning parameters and the critical rotational speed was verified. The critical rotational speed for forming the spinning jet was directly proportional to the spinning solution concentration and inversely proportional to the nozzle diameter. Polyoxyethylene (PEO) nanofibers were fabricated by selectively adjusting spinning parameters and characterized by using scanning electron microscopy (SEM). The results showed that the diameter distribution of nanofibers could be adjusted by changing the spinning parameters, such as spinning solution concentration, rotational speed, nozzle length, nozzle diameter and collection distance. The high speed centrifugal spinning has the potential to produce single continuous nanofibers efficiently.
