Experiments and numerical simulations were conducted across a range of contraction ratio to investigate the detonation combustion characteristics of liquid aviation kerosene/air rotating detonation combustor. The research indicates that extremely low and high contraction ratio adversely affect the stable operation of the two-phase rotating detonation combustors. At a contraction ratio that is too low (19.03 % in the RDC discussed in this paper), the atomization and mixing properties of the two-phase fuel are poor, with a lower reaction pressure in the fresh mixture, hindering the initiation of detonation waves. On the other hand, a contraction ratio (85.7 % in the RDC discussed in this paper) that is too high enhances the energy of the detonation wave; it triggers oblique shockwave feedback, preventing the fresh mixture from entering the combustor and thus leading to the quenching of detonation waves. With an increase in the contraction ratio of outlet configurations, there is a noticeable rise in both the peak pressure and combustion efficiency of detonation waves. In contrast, the wave height gradually decreases, and the wave velocity shows a pattern of initially increasing and then decreasing. Along the axial position inside the two-phase rotating detonation combustor, the upstream detonation wave is caused by the reflected shockwaves increasing the pressure of the upstream fresh mixture. In contrast, the downstream detonation wave results from increased fresh mixture pressure and enhanced fuel atomization and mixing due to the increased contraction ratio of outlet configurations and wave feedback. So, the detonation wave exhibits a strong-weak-strong distribution characteristic. • Three-dimensional numerical simulation and experiment in a RDC with separate feeding of kerosene and air is achieved. • Two atomization and mixing structures ("Kerosene" and "Enhanced Kerosene" region) is found. • An optimal contraction ratio size enables the RDC to achieve the best operational characteristics. The TFIIF-like Rpc53/Rpc37 heterodimer of RNA polymerase (pol) III is involved in various stages of transcription. The C-terminal region of Rpc53 dimerizes with Rpc37 to anchor on the lobe domain of the pol III cleft. However, structural and functional features of the Rpc53 N-terminal region had not been characterized previously. Here, we conducted site-directed alanine replacement mutagenesis on the Rpc53 Nterminus, generating yeast strains that exhibited a coldsensitive growth defect and severely compromised pol III transcriptional activity. Circular dichroism and NMR spectroscopy revealed a highly disordered 57-amino acid polypeptide in the Rpc53 N-terminus. This polypeptide is a versatile protein-binding module displaying nanomolar-level binding affinities for Rpc37 and the Tfc4 subunit of the transcription initiation factor TFIIIC. Accordingly, we denote this Rpc53 N-terminus polypeptide as the TFIIIC-binding region or CBR. Alanine replacements in the CBR significantly reduced its binding affinity for Tfc4, highlighting its functional importance to cell growth and transcription in vitro. The production of secondary metabolites is a major mechanism used by beneficial rhizobacteria to antagonize plant pathogens. These bacteria have evolved to coordinate the production of different secondary metabolites due to the heavy metabolic burden imposed by secondary metabolism. However, for most secondary metabolites produced by bacteria, it is not known how their biosynthesis is coordinated. Here, we showed that PhlH from the rhizobacterium Pseudomonas fluorescens is a TetR-family regulator coordinating the expression of enzymes related to the biosynthesis of several secondary metabolites, including 2,4-diacetylphloroglucinol (2,4-DAPG), mupirocin, and pyoverdine. We present structures of PhlH in both its apo form and 2,4-DAPG-bound form and elucidate its ligand-recognizing and allosteric switching mechanisms. Moreover, we found that dissociation of 2,4-DAPG from the ligand-binding domain of PhlH was sufficient to allosterically trigger a pendulum-like movement of the DNA-binding domains within the PhlH dimer, leading to a closed-to-open conformational transition. Finally, molecular dynamics simulations confirmed that two distinct conformational states were stabilized by specific hydrogen bonding interactions and that disruption of these hydrogen bonds had profound effects on the conformational transition. Our findings not only reveal a well-conserved route of allosteric signal transduction in TetR-family regulators but also provide novel mechanistic insights into bacterial metabolic coregulation. Studies found that the Drosha-DGCR8 complex initiates microRNA maturation by precise cleavage of the stem loops, which are embedded in primary transcripts. A model is proposed for this process that is based upon evidence from both computational and biochemical analyses.

The TMC genes encode a set of homologous transmembrane proteins whose functions are not well understood. Biallelic mutations in either TMC6 or TMC8 are detected in more than half of cases of the pre-malignant skin disease epidermodysplasia verruciformis (EV). It is controversial whether EV induced by mutations in TMC6 or TMC8 originates from keratinocyte or lymphocyte defects. Quantification of TMC6 and TMC8 RNA levels in various organs revealed that lymphoid tissues have the highest levels of expression of both genes, and custom antibodies confirmed protein expression in mouse lymphocytes. To study the function of these proteins we generated mice with targeted deletion mutant alleles of Tmc6 or Tmc8. Either TMC6 or TMC8 deficiency induced a reduction in apparent molecular weight and/or amount of the other TMC molecule. Co-immunoprecipitation experiments indicated that TMC6 and TMC8 formed a protein complex in mouse and human T cells. MS and biochemical analysis demonstrated that TMC6 and TMC8 additionally interacted with the CIB1 protein to form TMC6-TMC8-CIB1 trimers. We demonstrated that TMC6 and TMC8 regulated CIB1 levels by protecting CIB1 from ubiquitination and proteasomal degradation. Reciprocally, CIB1 was needed for stabilizing TMC6 and TMC8 levels. These results suggest why inactivating mutations in any of the three human genes leads to similar clinical presentations. We also demonstrated that TMC6 and TMC8 levels are drastically lower and the proteins are less active in regulating CIB1 in keratinocytes than in T cells. Prognosis of hepatocellular carcinoma (HCC) remains poor despite significant recent improvement in therapy. Recent studies have reported that transglutaminase 3 (TGM3) plays an important role in several human cancer types. However, the role of TGM3 in HCC have not been previously elucidated. We evaluated the role of TGM3 in regulating HCC cell proliferation, migration, and invasion. We also investigated the prognostic significance of TGM3 in an HCC cohort. Finally, we explored the signalling pathways that TGM3 regulates in HCC. We identified TGM3 to be overexpressed in HCC compared to normal tissues. Higher expression of TGM3 predicts poor prognosis in HCC patients. TGM3 knockdown led to decreased HCC cell proliferation, invasion, and xenograft tumour growth. TGM3 depletion inhibited AKT, extracellular signal–regulated kinase (ERK), p65, and glycogen synthase kinase 3β (GSK3β)/β-catenin activation, but promoted levels of cleaved caspase 3. Moreover, TGM3 knockdown cells had increased E-cadherin levels and decreased vimentin levels, suggesting that TGM3 contributes to epithelial–mesenchymal transition (EMT) in HCC. Our results suggest that TGM3 controls multiple oncogenic pathways in HCC, thereby contributing to increased cell proliferation and EMT, and TGM3 potentially enhances HCC metastasis. The thermal conductivity of two-dimensional materials like graphene can efficiently be tuned by introducing holes, in which the density and distribution of the holes are the key parameters. Furthermore, the distribution of holes can induce a variation as high as 74% in the thermal conductivity for porous graphene with a given density of holes. Therefore, an existing challenge is to find the optimal distribution of holes that can minimize or maximize the thermal conductivity of porous graphene as the design space expands dramatically with increasing hole density. We therefore apply an inverse design methodology based on machine learning to reveal the relationship between hole distribution and thermal conductivity reduction in monolayer graphene. The methodology reveals that holes that are randomly distributed transverse to the direction of heat flow, but that exhibit some periodicity along the direction of heat flow, represent the optimal distribution to minimizing the thermal conductivity for porous graphene. Lattice dynamics calculations and wave packet simulations reveal that this spatial distribution effectively causes localization of the phonon modes in porous graphene, which reduces the thermal conductivity. This paper studies the effects of shear inhomogeneities on the structure and stability characteristics of turbulent piloted premixed flames. The shear is introduced by splitting the fuel-air mixture in two stream and feeding each stream at different velocities through two annular concentric tubes residing within the pilot annulus. The shear is varied by varying the proportion of fluid injected through the streams and also by changing the recess distance of the inner tube with respect to the jet exit plane. It is noted that with increasing rates of shear, the Bunsen-like flames become shorter with the flame tip opening downstream of the pilot stream. Measurements of flow fields reveal useful information about the level and location of the shear layers. Joint imaging of Laser-Induced Fluorescence (LIF) of OH and CH show that, with increasing rates of shear, the degree of flame front fragmentation increases implying the occurrence of local extinction even in regions extremely close to the pilot. The flame surface area, marked by LIF-CH is taken as an indicator of consumption rates of reactants. For all flames examined here, the peak flame surface area increases with increasing shear and subsequently decreases as the flame shortens and approaches blow-off. The peak flame surface density decreases with increasing shear. Regime diagrams show that the flames investigated here bridge the regions of broadened preheat thin reactions as well as the distributed reaction zone. While the thickness of the CH layer remain somewhat uniform, the number of flame fragments, as indicated by breaks in the CH profiles, increases with increasing shear before stabilizing at some peak value. The observed mechanism of local extinction in highly sheared Bunsen-like, turbulent piloted premixed flames complements existing results for bluff-body stabilised flames. [ABSTRACT FROM AUTHOR]

Fullerene-based sandwiches have excellent controllable physical properties such as cross-plane thermal conductivity and charge transfer, which provide new possibilities for advancing the field of thermoelectricity. Because the pattern of the fullerene cluster impacts the physical properties of the sandwiches, it is important to understand the mechanisms for their structural transformations. In the present work, we find that the graphene/fullerene/graphene sandwich structure transforms among three configurations in an ideal environment, depending on the fullerene to graphene area ratio. Molecular dynamics simulations show that there are two critical values for the area ratio. The fullerene pattern transforms from circular to rectangular at the first critical area ratio of 1 π. The critical value of 1 π is successfully derived by comparing the geometrical perimeter of the circular and rectangular shapes for the fullerene cluster without any physical parameters. At the second critical area ratio, the graphene layers surrounding the fullerene cluster become separated due to the competition between the bending energy and the cohesive energy. The vacuum space is predicted to be 34 Å based on the analytic model. These findings provide fundamental insights into the mechanisms driving structural transformation of fullerene-based sandwiches, which will enable the selection of appropriate structures and materials to guide the design of future electronic and energy storage applications. The thermal expansion coefficient of van der Waals layered structures plays an important role in governing the thermal stability of high-performance electronic devices. Meanwhile, bubbles are frequently observed in the van der Waals layered structures due to the weak inter-layer interaction and the extremely small bending stiffness of the layered structure, but the effect of bubbles on the thermal expansion coefficient for layered structures remains unclear. We derive an analytic formula for the bubble effect on the length variation of the MoS 2 layer. In the analytic derivation, the MoS 2 layer is described by three mechanics models, including the elastic membrane theory, the nonlinear plate theory, and the linear plate theory. The gas inside the bubble is described by two types of state equations, including the ideal gas law and the generalized van der Waals equation. It is found that the nonlinear plate theory with the generalized van der Waals equation gives the most accurate description for the bubble effect, since MoS 2 has a moderate bending stiffness. The analytic formula shows that the bubble can cause strong thermal contraction for few-layer MoS 2 with increasing temperature. The analytic predictions are verified by comprehensive molecular dynamic simulations for few-layer MoS 2 with different layer numbers and different gas molecule numbers. Graphene has been expected to revolutionize the electronics, but suffered from its zero bandgap that will lead to an unacceptable leakage current in graphene-based transistors. Although a bandgap engineered graphene structure may have a bandgap, such bandgap is sensitively dependent on the geometry of the structure. Finding new two-dimensional (2D) semiconducting carbon allotropes is therefore extremely important for the new emerging all-carbon electronics. Based on first principles calculations, here we predict a new 2D semiconducting carbon allotrope, referred to as twin graphene. The allotrope has an intrinsic direct bandgap, very close to that of silicon and tunable by in-plane strain, indicating its great potential in nanoelectronics. The stability of twin graphene is carefully verified by both the first principles calculations and the molecular dynamics simulations. We determine also the mechanical properties of twin graphene, including the in-plane stiffness, Poisson’s ratio, shear stiffness and bending stiffness. Its excellent electronic and mechanical properties suggest that twin graphene may have a significant impact on many disciplines and fields related to nanotechnology. [ABSTRACT FROM AUTHOR]

Edge effects play an important role for many properties of graphene. While most works have focused on the effects from isolated free edges, we present a novel knotting phenomenon induced by the interactions between a pair of free edges in graphene, and investigate its effect on the buckling of monolayer graphene. Upon compression, the buckling of graphene starts gradually in the form of two buckling waves from the warped edges. The collision of these two buckling waves results in the creation of a knot structure in graphene. The knot structure enables the buckled graphene to exhibit two unique post-buckling characteristics. First, it induces a five-fold increase in graphene’s mechanical stiffness during the buckling process. Second, the knotted structure enables graphene to exhibit a mechanically stable post-buckling regime over a large (3%) compressive strain regime, which is significantly larger than the critical buckling strain of about 0.5%. The combination of these two effects enables graphene to exhibit an unexpected post-buckling stability that has previously not been reported. We predict that numerical simulations or experiments should observe two distinct stress strain relations for the buckling of identical graphene samples, due to the characteristic randomness in the formation process of the knot structure. Purpose To study the genetics and functional alteration of a family with X-linked lissencephaly and subcortical band heterotopia. Methods Five affected patients (one male with lissencephaly, four female with subcortical band heterotopia) and their relatives were studied. Sanger sequencing of DCX gene, allele specific PCR and molecular inversion probe technique were performed. Mutant and wild type of the gene products, namely doublecortin, were expressed in cells followed by immunostaining to explore the localization of doublecortin and microtubules in cells. In vitro microtubule-binding protein spin-down assay was performed to quantify the binding ability of doublecortin to microtubules. Key findings We identified a novel DCX mutation c.785A > G, p.Asp262Gly that segregated with the affected members of the family. Allele specific PCR and molecular inversion probe technique demonstrated that the asymptomatic female carrier had an 8% mutant allele fraction in DNA derived from peripheral leukocytes. This mother had 7 children, 4 of whom were affected and all four affected siblings carried the mutation. Functional study showed that the mutant doublecortin protein had a significant reduction of its ability to bind microtubules. Significance Low level mosaicism could be a cause of inherited risk from asymptomatic parents for DCX related lissencephaly-subcortical band heterotopia spectrum. This is particularly important in terms of genetic counselling for recurrent risk of future pregnancies. Thiazole orange is a commonly used cyanine dye for binding to nucleic acids. Recently, it has been used for the detection of G-quadruplexes. However, thiazole orange is non-selective for G-quadruplex and other nucleic acids, thus hampering its further application. Herein, we designed and synthesized new fluorescent probes by incorporating hydrocarbon rings into the chromophore of thiazole orange. This simple modification dramatically improved selective binding to certain G-quadruplexes. The most promising probe, the cyclopentane fused analogue, exhibited significant fluorescence enhancement when treated with G-quadruplexes but retained weak fluorescence in the presence of double-stranded and single-stranded DNA. The cyclopentane fused probe also displayed considerable selectivity for parallel G-quadruplexes. These modifications reduced the quantum yield of thiazole orange. Further study of the mechanism revealed that the introduction of a hydrocarbon ring altered the planarity of the chromophore as well as the binding affinities for G-quadruplexes, and therefore, influenced the ability to detect G-quadruplexes.
