Abstract: Polyamidoamine (PAMAM) is one of the widely employed non-viral vectors in gene therapy research, and shows excellent biocompatibility and relatively low cytotoxicity. However, it has poor transfection efficiency compared with that of polyethylenimine (PEI, 25kDa). To enhance the gene expression efficiency, we introduced the RRRK peptide from mouse fibroblast growth factor 3 (FGF3) to PAMAM, which is a known nuclear localization signal (NLS). We synthesized PAMAM-KRRR and PAMAM-RRRK to verify the difference of the induced functional status from reversal of the N-terminus. PAMAM containing the FGF3 peptide showed a transfection efficiency corresponding to that of PEI in HEK293, and HeLa cells, and showed much higher gene expression capacity than that of PEI in NIH3T3 cells with relatively decreased cytotoxicity. These results imply that introduction of the FGF 3 peptide has the potential to provide a novel PAMAM-based vector by enhancing its gene expression efficiency. [Copyright &y& Elsevier]

Comparative chemical and structural characterization of various apatite phases from both synthetic and natural sources were carried out by utilizing a multi-sample loading device developed for scanning electron microscopy (SEM) and transmission electron microscopy (TEM) analyses. The device is capable of spraying nanoparticles via ultrasonication, and four separate samples were loaded on a single TEM grid by a masking tool to characterize different apatite phases in consistent instrumental and analytical conditions. SEM energy dispersive spectroscopy (EDS) analysis of the apatite samples revealed distinctive values of O/P and Ca/P intensity ratios except for the fossil apatite due to its high proportion of externally originating phases. Selected area electron diffraction (SAED) pattern analysis presented the characteristic lattice parameters of the apatite phases, especially the bone and fossil apatite which had CO 3 in their chemistry. Analyzing distinctive chemical and structural features among varied apatite phases can be efficiently carried out through electron microscopy with multiple samples on a single TEM grid, and such methods can be employed for characterizing a wider range of materials. We report interesting microscopic features that evolve as a function of electron dose from the starting aluminum trihydroxide to the resulting transition aluminas with layered mesoporous structures. Remarkably, the sizes of pores and crystals in the porous structure were reduced as the electron dose was increased. The properties of the layered structure from the aluminum trihydroxide were maintained through the transition process, and hence the final mesoporous structures (mainly γ-alumina) were of lamella-type microstructure.
