33. Crystal structure of constitutive endothelial nitric oxide synthase: a paradigm for pterin function involving a novel metal center

Abstract

A high-resolution crystal structure of a constitutive endothelial nitric oxide synthase (eNOS) heme domain, alone and in combination with tetrahydrobiopterin (H4B), is presented. A catalytic role for H4B is evidenced by the similarity of the pterin-binding site in NOS with other pterin-using enzymes. Structural evidence for the ability of the substrate, L-arginine, to bind at the site of H4B is also presented. Based on the substrate's cofactory mimicry, a mechanism that involves the involvement of a pterin radical cation in the catalytic cycle leading to nitric oxide biosynthesis is identified.

Abstract

Six water-soluble macrocyclic photosensitizers, the members of two groups of expanded porphyrins (metallotexaphyrins and free-base sapphyrins) containing hydrophilic substituents and meso-tetra(4-sulfonatophenyl)-porphyrin, were tested by UV–Vis absorption and resonance Raman spectroscopy in the in vitro binding experiments with intact mitochondria isolated from swine liver. Studied macrocycles showed markedly different affinity to mitochondria. The highest uptake was observed for sapphyrin–sugar conjugate and metallotexaphyrins. Sapphyrin–polyamine conjugates exhibit something less affinity to mitochondria, while the porphyrin of anionic character showed very low mitochondrial uptake. Obtained spectroscopic results confirm that the binding process altered the self-aggregation degree of expanded porphyrins. [Copyright &y& Elsevier]

Abstract

Varied therapeutic peptides and proteins represent a rapidly growing part of marketed drugs and have an undisputed place alongside other established therapies. Nevertheless, such biodrugs have several drawbacks that hinder their therapeutic application. These are undesirable physicochemical properties, such as variable solubility, low bioavailability and limited stability. These issues can be overcome by addition of stabilizing agents and directed injectable administration, which can however result in low patient compliance. Hence, there is a drive in the biotechnology industry to produce needle-free and more user-friendly drugs, and this has led to the growth of nano-enabled drug delivery systems in the last decade. As discussed here, nanobiotechnology is becoming a commercially feasible and promising opportunity for oral, pulmonary and transdermal administration routes. [Copyright &y& Elsevier]
