We demonstrate an ultra-broadband multi-channel orbital angular momentum (OAM) mode generator using a helical long-period grating (HLPG) pair in a few-mode fiber. The numbers of the channel of the OAM mode generator can be flexibly adjusted by designing the cascaded length of the HLPG pair. When the cascaded length is 50 cm, the maximum channel number of OAM mode generator reaches 46, and the mode convert efficiencies of the channels are more than 90 %, which is, to the best of our knowledge, the highest number of channels among OAM mode converters. By reducing grating length and enhancing the refractive index modulation depth, the 3 dB bandwidth of the HLPG is significantly increased, and thus the wavelength range of the HLPG pair can be expanded. The maximum wavelength ranges of the channels with coupling efficiency greater than 90 % are measured to be 379 nm experimentally. By adjusting the spiral direction of the helical refractive index modulation, we can achieve multi-channel ± 1 order OAM mode conversion for each channel. MicroRNAs (miRNAs) regulate insulin secretion, pancreas development, and beta cell differentiation. In this study, to screen for miRNAs and their targets that function during insulin-producing cells (IPCs) formation, we examined the messenger RNA and microRNA expression profiles of pancreatic progenitor cells (PPCs) and IPCs using microarray and deep sequencing approaches, respectively. Combining our data with that from previous reports, we found that miR-21 and its targets play an important role in the formation of IPCs. However, the function of miR-21 in the formation of IPCs from PPCs is poorly understood. Therefore, we over-expressed or inhibited miR-21 and expressed small interfering RNAs of miR-21 targets in PPCs to investigate their functions in IPCs formation. We found that miR-21 acts as a bidirectional switch in the formation of IPCs by regulating the expression of target and downstream genes ( SOX6 , RPBJ and HES1 ). Small interfering RNAs were used to knock down these genes in PPCs to investigate their effects on IPCs formation. Single expression of si-RBPJ, si-SOX6 and si-HES1 in PPCs showed that si-RBPJ was an inhibitor, and that si-SOX6 and si-HES1 were promoters of IPCs formation, although si-HES1 induced formation of IPCs at higher rates than si-SOX6. These results suggest that endogenous miRNAs involved in the formation of IPCs from PPCs should be considered in the development of an effective cell transplant therapy for diabetes. Abstract: A fibroblast line from ear marginal tissue of Luxi cattle (LXCEM2/2) was successfully established by direct culturing of explants. Biological analysis showed that the population doubling time (PDT) for reviving cells was approximately 24h. Measurement of lactic dehydrogenase (LDH) and malic dehydrogenase (MDH) isoenzymes showed no cross-contamination among the cells. Karyotyping showed that the frequency of cells with chromosome number 2n=60 was 90.7–92.2%. Tests for bacteria, fungi, viruses and mycoplasma were negative. The efficiencies of expression of pEGFP-N3, pEYFP-N1 and pDsRed1-N1 were between 6.3% and 31.6% at 24h, 48h and 72h after transfer; at 24h, fluorescence was well distributed in the cytoplasm and nucleus except for some cryptomeric vesicles. Every index of the Luxi cattle cell line meets the quality control standards of the American Type Culture Collection (ATCC). Not only has the germline of this important cattle breed been preserved at the cell level, but also valuable material had been provided for genome, postgenome and somacloning research. Moreover, the establishment of this technical platform may provide both technical and theoretical support for storing the genetic resources of other animals and poultry at the cell level. • Long-period fiber grating based linearly polarized mode converter operating at a dispersion turning point has been demonstrated. Such mode converter can realize broadband mode conversion. • A broadband and controllable linearly polarized OAM modes generator using a single long-period fiber grating followed by a length of few-mode polarization-maintaining fiber has been demonstrated. • A simple approach was proposed to tunable the linearly polarized OAM mode, the phase difference of the two linearly polarized modes was continually turned by strain, the OAM mode can be smoothly tunable between ±1 order. We demonstrate a broadband and stable linearly polarized mode converter by inscribing long-period grating (LPG) in few-mode polarization-maintaining fibers (FPMFs). The operating wavelength of the mode converter at 1.0- and 1.5- μm waveband has been achieved by using PMF with different cut-off wavelengths. Theoretical and experimental demonstrated broadband linearly polarized mode conversion can be achieved with a maximum 10 dB bandwidth of 140- and 189- nm at 1.0- and 1.5- μm waveband respectively due to the turning point coupling mechanism. Then, we propose and demonstrate a controllable linearly polarized orbital angular momentum (LP-OAM) mode generator using a single LPG-based mode converter and a length of FPMF. The LP-OAM mode is produced by turning axial strain and can be smoothly switched between ± 1 order. Our all-FPMF based grating fabrication offers a new option towards a stable and broadband polarized mode converter and LP-OAM mode generator for application in a few-mode fiber laser and communication system. In this study, a full-length enriched cDNA library was successfully constructed from Siberian tiger, the world's most endangered species. The titers of primary and amplified libraries were 1.28×106 pfu/mL and 1.59×1010 pfu/mL respectively. The proportion of recombinants from unamplified library was 91.3% and the average length of exogenous inserts was 1.06kb. A total of 279 individual ESTs with sizes ranging from 316 to 1258bps were then analyzed. Furthermore, 204 unigenes were successfully annotated and involved in 49 functions of the GO classification, cell (175, 85.5%), cellular process (165, 80.9%), and binding (152, 74.5%) are the dominant terms. 198 unigenes were assigned to 156 KEGG pathways, and the pathways with the most representation are metabolic pathways (18, 9.1%). The proportion pattern of each COG subcategory was similar among Panthera tigris altaica, P. tigris tigris and Homo sapiens, and general function prediction only cluster (44, 15.8%) represents the largest group, followed by translation, ribosomal structure and biogenesis (33, 11.8%), replication, recombination and repair (24, 8.6%), and only 7.2% ESTs classified as novel genes. Moreover, the recombinant plasmid pET32a-TAT-COL6A2 was constructed, coded for the Trx-TAT-COL6A2 fusion protein with two 6× His-tags in N and C-terminal. After BCA assay, the concentration of soluble Trx-TAT-COL6A2 recombinant protein was 2.64±0.18mg/mL. • The pattern of fat deposition in the tail of F2 sheep changed and evolved. • SCD , ESR1 , EMR1 and PHYH genes play a direct role in tail fat deposition. • GPAM , STAT3 and ALDH1A1 genes indirectly regulate the evolution of fat tails in sheep. Fat tail is one of the most important domesticated characteristics in sheep; however its molecular mechanism is poorly understood. Here we took small-tailed F2 hybrid of wild Argali sheep and typical fat-tailed Bashby sheep as research object. First, histological analysis revealed that the mean diameter and area in tail and subcutaneous fat cells, and surface density in tail fat in Bashby sheep were significantly larger than that in F2 sheep, and surface density of fat in subcutaneous fat in Bashby sheep was significantly lower than that in F2 sheep. Second, 873 differentially expressed genes (DEGs) of tail fat between Bashby and F2 sheep were identified by RNA-seq. Third, the tissue expression profile and relative expression difference between Bashby and F2 sheep of 7 of 873 DEGs were analyzed by RT-PCR. SCD, ESR1 , EMR1, PHYH , STAT3 and GPAM genes were highly expressed in fat, muscle and liver, and ALDH1A1 were highly expressed in small intestine. In addition, the expressions of SCD , PHYH and CPAM genes in tail fat of F2 sheep were lower than that of Bashby sheep, while the expression patterns of ESR1 and EMR1 were reversed.
