Accurate description of visual effect of colored moiré pattern caused by layered optical structures consisting of gratings and Fresnel lens is proposed in this work. The colored moiré arising from the periodic and quasi-periodic structures is numerically simulated and experimentally verified. It is found that the visibility of moiré pattern generated by refractive optical elements is related to not only the spatial structures of gratings but also the viewing angles. To effectively control the moiré visibility, two constituting gratings are slightly separated. Such scheme is proved to be effective to globally eliminate moiré pattern for displays containing refractive optical films with quasi-periodic structures. [ABSTRACT FROM AUTHOR]

Solenaia oleivora , a freshwater shellfish endemic to China, is becoming one of the most threatened freshwater mussels owing to water pollution, habitat fragmentation, and overfishing. Hence, exploring its response to different environmental factors is important for its conservation. In this work, we investigated the physiological and transcriptomic response of S. oleivora to increased salinity. We found that increased salinity caused the death of S. oleivora. High salinity caused shrinking and deformation of gill filaments, reduced gill cilia, and induced cell apoptosis in gills. The activities of superoxide dismutase (SOD), catalase (CAT), acid phosphatase (ACP), alkaline phosphatase (AKP), as well as glutathione (GSH) content were increased at the beginning of salinity stress (3−12 h), while SOD and ACP activities decreased at 48 h. Transcriptome data revealed that high salinity stress (48 h) induced 766 differentially expressed genes (DEGs). Among these DEGs, the majority of the stress response and ion transport-related genes were up-regulated, while most of the immune-related genes were down-regulated. In conclusion, these findings suggest that the antioxidant and immune functions of S. oleivora can be inhibited by high salinity, which may be one of the main reasons for its low survival rate under conditions of increasing salinity. Vinorine synthase (EC 2.3.1.160) catalyses the acetyl-CoA- or CoA-dependent reversible formation of the alkaloids vinorine (or 11-methoxy-vinorine) and 16-epi-vellosimine (or gardneral). The forward reaction leads to vinorine, which is a direct biosynthetic precursor along the complex pathway to the monoterpenoid indole alkaloid ajmaline, an antiarrhythmic drug from the Indian medicinal plant Rauvolfia serpentina. Based on partial peptide sequences a cDNA clone was isolated and functionally expressed in Escherichia coli. The Km values of the native enzyme for gardneral and acetyl-CoA were determined to be 7.5 and 57 μM. The amino acid sequence of vinorine synthase has highest level of identity (28–31%) to that of Papaver salutaridinol acetyltransferase, Fragaria alcohol acyltransferase, and Catharanthus deacetylvindoline acetyltransferase involved in morphine, flavor, and vindoline biosynthesis, respectively. Vinorine synthase is a novel member of the BAHD superfamily of acyltransferases. Site-directed mutagenesis of 13 amino acid residues provided clear evidence that both, His160 and Asp164 of the consensus sequence HxxxD belong to the catalytic center. The mutations also showed that an amino acid triad is not characteristic of vinorine synthase. The experiments demonstrated the importance of the conserved motif SxL/I/VD near the N-terminus and the consensus sequence DFGWG near the C-terminal. [Copyright &y& Elsevier]

The acetyl-CoA-dependent enzyme vinorine synthase was isolated from hybrid cell suspension cultures of Rauvolfia serpentina and Rhazya stricta. The sarpagan-type alkaloid gardneral was used as a substrate of the enzyme leading to the ajmalan-type 10-methoxyvinorine. An HPLC-based assay was developed to monitor vinorine synthase activity, which allowed establishing a five step purification procedure combining anion exchange, hydrophobic interaction, hydroxyapatite and gel filtration. Purification resulted in a yield of 0.2% and an approximately 991-fold enrichment of the acetyltransfer activity. SDS-PAGE analysis showed a Mr for the enzyme of ∼50 kDa. The four peptide fragments generated by proteolysis of the pure enzyme with endoproteinase LysC and the N-terminal part of the enzyme were sequenced. The enzyme preparation (>875-fold enrichment) delivering the N-terminal sequence was isolated from R. serpentina cell suspensions. Sequence alignment of the five peptides showed highest homologies in a range of 30–71% to acetyltransferases from other higher plants involved in natural plant product biosynthesis. Solenaia oleivora is a valuable freshwater mussel endemic to China with a high content of high-quality proteins, but the lack of structural information and limited functionality of Solenaia oleivora proteins constrained their application in the food industry. This study investigates the changes in structural characteristics and functionality of Solenaia oleivora protein under ultrasound processing at power from 200 to 600 W. The ultrasound treatment caused increased contents of β-turn and α-helix, and the exposure of interior hydrophobic groups, resulting in the increased hydrophobicity by around 3 folds. The ultrasound treatment could significantly decrease particle size and increase surface charges of Solenaia oleivora proteins, facilitating the increase of hydrosolubility from 10.2% to 81.7%. These structural changes and increased hydrosolubility contributed to the enhancement of emulsifying and foaming properties, and in vitro digestibility. The results suggested that the ultrasound-treated Solenaia oleivora proteins possessed the potential as an alternative protein in food applications. Abstract Biotransformation of refractory organics by ammonia-oxidizing microorganisms in nitrifying sludge have been widely reported, while the contribution of heterotrophic bacteria in nitrifying sludge in the biotransformation and degradation process might be overlooked. Here, we provide metagenomic and metatranscriptomic evidences showing that heterotrophic bacteria in a highly enriched autotrophic nitrifying sludge could significantly contribute to the aromatic biotransformation and biodegradation. Diverse genes encoding enzymes for aromatic degradation were observed in an enriched autotrophic nitrifying sludge. These genes are involved in the degradation of at least 15 complex aromatics. Genome binning results showed that these genes were mainly carried by species in Bacteroidetes (Flavobacteriaceae and Sphingobacteriales), Alphaproteobacteria (Rhodobacter) and Betaproteobacteria (Bordetella , Acidovorax , Ramlibacter and Pusillimonas). According to the metatranscriptomic analysis, the overall expression of the potential aromatic-degrading genes was significantly upregulated, and almost all genes involved in phenol degradation were over expressed after the nitrifying sludge was exposed to phenol. Overall, our results suggest that certain heterotrophs in nitrifying sludge are involved aromatic biotransformation and biodegradation and advance our knowledge of the underlying properties and metabolic mechanisms of the nitrifying sludge. Abstract Previous studies have demonstrated that transcription factor-miRNA-gene feed-forward loops (FFLs) played important roles in tumorigenesis. However, the lncRNA-involved FFLs have not been explored very well. Understanding the characteristics of lncRNA-involved FFLs in breast cancer subtypes may be a key question with clinical implications. In this study, we firstly constructed an integrated background regulatory network. Then, based on mRNA, miRNA, and lncRNA differential expression, we identified 147, 140, 284, 1031 dysregulated FFLs for luminal A, luminal B, HER2+ and basal-like subtype of breast cancer, respectively. Importantly, the known breast cancer-associated lncRNAs and miRNAs were enriched in the identified dysregulated FFLs. Through merging the dysregulated FFLs, we constructed the regulatory sub-network for each subtype. We found that all sub-networks were enriched in the well-known cancer-related pathways, such as ce ll cycle , pathways in cancer. Next, we also identified potential prognostic FFLs for subtypes of breast cancer, such as the hsa-miR-182-5p_JUN_XIST in basal-like subtype. Finally, we also discussed the potential application of inferring the candidate drugs for breast cancer treatment through modulating the lncRNA expression in the dysregulated FFLs. Collectively, this study elucidated the roles of lncRNA-involved FFLs in breast cancer subtypes, which could contribute to understanding breast cancer pathogenesis and improving the treatment. Anammox bacteria are being increasingly investigated as part of an emerging nitrogen removal technology. However, due to the difficulty in culturing, current understanding of their behavior is limited. In this study, anaerobic microfluidic chips were used to study anammox bacteria, showing great advantages over reactors. On-chip fluorescence in situ hybridization (FISH) showed the relative abundance of free form anammox bacteria increased by 56.1 % after one week's culture, an increase that is three times higher than that of bioreactor (17.1 %). For granular form cultures, the nitrogen removal load reached 2.34 ∼ 2.51 kg-N/(m3·d), which was also substantially higher than the bioreactor (∼1.22 kg-N/(m3·d)). Furthermore, studying the kinetics of nitrite inhibition of granular sludge with different particle sizes (100–900 μm) showed that the maximum ammonia load and the nitrite semi-saturation coefficient noticeably decreased for smaller particle sizes. • Aromatics may significantly impact the diversity and abundance of bacterial ARGs. • Wastewater containing high levels of PAP leads to more abundant ARGs in bioreactors. • Ozone pretreatment of wastewater containing aromatics can reduce ARGs in bioreactors. • Bacteria harboring both ARGs and ADGs play major roles in degrading aromatics. Aromatic matters are widely present in wastewater, especially industrial wastewater, and may lead to a high abundance of antibiotic resistance genes (ARGs) in wastewater treatment bioreactors and stimulate horizontal transfers of ARGs. Here, we investigated a practical approach that applying ozone pretreatment to mitigate ARGs in bioreactors treating wastewater containing a typical aromatic pollutant, p -aminophenol (PAP). The results showed that ozone pretreatment could effectively reduce the aromaticity of wastewater, and the relative abundance of ARGs in the bioreactor fed with ozone treated wastewater decreased by over 70% compared to the control reactor. Multidrug, quinolone, mupirocin, polymyxin, aminoglycoside, glycopeptide, beta-lactam, and trimethoprim resistance genes were all reduced in the bioreactors receiving wastewater pretreated by ozone. Metagenomic analysis suggested that the reduction of ARGs could be attributed to the co-occurrence of ARGs and aromatic degradation genes in bacteria. Furthermore, we expanded our analysis to investigate 71 metagenomes from different environments, and the results indicated that the impact of aromatics on ARG abundance widely occurs in various ecosystems and confirmed that high levels of aromatics could lead to high abundance of ARGs. Taken together, our work confirmed that the aromatics played critical roles in selecting ARGs and proposed a feasible approach to reduce ARGs in wastewater treatment bioreactors. Titanium xerogel coagulant (TXC) worked better than titanium tetrachloride (TC) and polytitanium chloride (PTC) in a wider pH/dose range for the removal of turbidity. However, the underlying mechanisms were not comprehensively understood. In this work, the better coagulation performance of TXC than TC and PTC was systematically elucidated from the following aspects: the physicochemical properties of the three coagulants, the removal of turbidity and organic matter, and the complexation reactions in coagulation. The results demonstrate that the merits of TXC were attributable to the following characteristics: (1) the higher surface charge density/total surface site concentration/isoelectric point of TXC hydrolysates, (2) the formation of TXC hydrolysates with a net-work structure, and (3) the strong binding affinity of TXC hydrolysates to organic matter caused by the bonded acetylacetone in the TXC framework. In short, the hydrolysis behavior of TXC significantly differed from both its precursor, TC, and the prehydrolyzed PTC. The difference in the hydrolysis of TXC was derived from the gelation process, which led to the polymerization of Ti in a way different from prehydrolyzation. The elucidation of the hydrolysis mechanisms is useful for the better application of Ti-based coagulants and may shed light on the preparation of other metal salts. [ABSTRACT FROM AUTHOR]

• The combinatorial optimization algorithm is applied to estimate the transformation matrix in hand eye calibration. • The influence of spherical mean square error and radius deviation on the calibration results is considered comprehensively. • It effectively reduces the random error, and has practical value for the hand eye calibration of industrial robots. In order to improve the measurement accuracy of laser profiler and industrial robot measurement system and reduce the influence of random error in calibration process, a hand eye calibration method based on combinatorial optimization is proposed. Firstly, the laser profiler is positioned by the robot to scan a standard sphere from different postures. Then, combinations from all the measurement data are selected to calculate several transformation matrices and the corresponding profile datasets in base coordinate system can be derived consequently. Since all the profile data come from the same stationary standard sphere, sphere fitting is applied to each dataset and the optimal hand eye transformation matrix can be derived which corresponds to the minimum fitting error. Experimental results show that the novel method can effectively improve the calibration accuracy and sphere loss which is weighted sum of sphere fitting error and radius error is reduced from 1.976 to 0.539 after combinatorial optimization.
