Old trees are rare in the landscape, as are many of their associated species. Veteranisation is a method by which attempts are made to create microhabitats, otherwise found only in old trees, in younger trees at an earlier stage than would occur naturally. Here, we analysed the early fungal succession in 6 y-old veteranisation wounds in ca. 100 y old living oak trunks by DNA-barcoding of the wood at eight sites in Sweden and Norway. We hypothesised basidiomycetes would be most abundant, and exposed sapwood and heartwood would select for different communities. We identified 686 fungal taxa, mainly ascomycetes, with a large overlap in species composition and surprisingly similar species richness, i.e. 325 vs. 308–360, between intact and different types of damaged wood, respectively. Endophytes continued to be present and common in damaged wood. The results demonstrate that damage to sapwood and heartwood partly select for different fungi and that 6 y is too early to evaluate if veteranisation can positively favour fungi of conservation interest. The class Archaeorhizomycetes (Taphrinomycotina, Ascomycota) was introduced to accommodate an ancient lineage of soil-inhabiting fungi found in association with plant roots. Based on environmental sequencing data Archaeorhizomycetes may comprise a significant proportion of the total fungal community in soils. Yet the only species described and cultivated in this class is Archaeorhizomyces finlayi . In this paper, we describe a second species from a pure culture, Archaeorhizomyces borealis NS99-600 T (=CBS138755 ExT ) based on morphological, physiological, and multi-locus molecular characterization. Archaeorhizomyces borealis was isolated from a root tip of a Pinus sylvestris seedling grown in a forest nursery in Lithuania. Analysis of Archaeorhizomycete species from environmental samples shows that it has a Eurasian distribution and is the most commonly observed species. Archaeorhizomyces borealis shows slow growth in culture and forms yellowish creamy colonies, characteristics that distinguish A. borealis from its closest relative A. finlayi . Here we also propose a sequence-based taxonomic classification of Archaeorhizomycetes and predict that approximately 500 species in this class remain to be isolated and described. Abstract: The secondary metabolites of the saprotrophic wood-decay basidiomycete fungus Granulobasidium vellereum were studied. Six sesquiterpenes were obtained; 2-hydroxycoprinolone (1), 8-deoxy-4a-hydroxytsugicoline (2), 8-deoxydihydrotsugicoline (3), which were previously not described, radulone A and B, and coprinolone ketodiol. Additionally, base-treatment of 1 yielded the diagnostic degradation products 1a and 1b, whereas radulone A was found to form 4 under mild acidic conditions. The structures were determined by NMR, MS, CD and polarimetry, along with biosynthetic considerations. Radulone A fully inhibited the spore germination of the potentially competing fungi Phlebiopsis gigantea, Coniophora puteana and Heterobasidion occidentale at 10μM, 500μM and 100μM, respectively. None of the other substances tested gave rise to any growth inhibition. [Copyright &y& Elsevier]

Abstract: Fomannoxin is a biologically active benzohydrofuran, which has been suggested to be involved in the pathogenicity of the root rotting fungus Heterobasidion annosum sensu lato. The biosynthesis of fomannoxin was investigated through an isotopic enrichment study utilizing [1-13C]glucose as metabolic tracer. 13C NMR spectroscopic analysis revealed the labeling pattern and showed that the isoprene building block originates from the mevalonic acid pathway, whereas the aromatic motif is formed via the shikimic acid route by elimination of pyruvate from chorismic acid. A natural product, 4-hydroxy-3-(3-methylbut-2-enyl)benzaldehyde (1), was isolated and characterized, and was suggested to be a key intermediate in the biosynthesis of fomannoxin and related secondary metabolites previously identified from the H. annosum fungal species complex. Abstract: Investigation of the production of secondary metabolites of Heterobasidion occidentale led to the isolation and identification of six sesquiterpenes (illudolone A and B, illudolactone A and B, deoxyfomannosin A and B) along with the well-known sesquiterpene fomannosin and the previously described benzohydrofuran fomannoxin. The structures and relative configurations of the compounds were determined by 1D and 2D NMR spectroscopic analysis as well as by HRMS. Their absolute configuration and biosynthesis were suggested and discussed in relation to fomannosin. Four compounds showed growth inhibiting activity against several basidiomycetes, Phlebiopsis gigantea, Phanerochaete chrysosporium and H. occidentale, and toxicity towards the moss Physcomitrella patens. In addition, one compound displayed activity against the bacterium Variovorax paradoxus as well as against the ascomycete Fusarium oxysporum. [Copyright &y& Elsevier]

Abstract: The aim of this work was to clarify taxonomy and examine evolutionary relationships within European Ceriporiopsis species using a combined analysis of the large subunit (nLSU) nuclear rRNA and small subunit (mtSSU) mitochondrial rRNA gene sequences. Data from the ITS region were applied to enhance the view of the phylogenetic relationships among different species. The studied samples grouped into four complex clades, suggesting that the genus Ceriporiopsis is polyphyletic. The generic type Ceriporiopsis gilvescens formed a separate group together with Ceriporiopsis guidella and Phlebia spp. in the phlebioid clade. In this clade, the closely related species Ceriporiopsis resinascens and Ceriporiopsis pseudogilvescens grouped together with Ceriporiopsis aneirina. C. resinascens and C. pseudogilvescens have identical LSU and SSU sequences but differ in ITS. Ceriporiopsis pannocincta also fell in the phlebioid clade, but showed closer proximity to Gloeoporus dichrous than to C. gilvescens or C. aneirina–C. pseudogilvescens–C. resinascens group. Another clade was composed of a Ceriporiopsis balaenae–Ceriporiopsis consobrina group and was found to be closely related to Antrodiella and Frantisekia, with the overall clade highly reminiscent of the residual polyporoid clade. The monotypic genus Pouzaroporia, erected in the past for Ceriporiopsis subrufa due to its remarkable morphological differences, also fell within the residual polyporoid clade. Ceriporiopsis subvermispora held an isolated position from the other species of the genus. Therefore, the previously proposed name Gelatoporia subvermispora has been adopted for this species. Physisporinus rivulosus appeared unrelated to two other European Physisporinus species. Moreover, Ceriporiopsis (=Skeletocutis) jelicii grouped in a separate clade, distinct from Ceriporiopsis species. Finally, the ITS data demonstrated the proximity of some Ceriporiopsis species (Ceriporiopsis portcrosensis and Ceriporiopsis subsphaerospora) to Skeletocutis amorpha. [Copyright &y& Elsevier]

Abstract: The aim of this study was to characterise the genetic variation and molecular relationships of the brown rot polypore, Laetiporus sulphureus s. lat., from Europe, South America, Africa, and Asia, using ITS sequences of the nu-rDNA and by comparing the growth rate in vitro. In a NJ analysis of the sequences of 130 individuals of L. sulphureus s. lat., eight distinct clusters emerged, supported by BS values of 70–100%. Within each cluster, the ITS rDNA sequence variation was below 3%. The sequences were also analysed together with Laetiporus sequences available from GenBank. Results demonstrated the possible presence of L. huroniensis in Europe (invalidly named L. montanus) and L. gilbertsonii in South America, and provided more information on the Pan-American and European distribution of one of the clades, currently known in North America as L. sulphureus. L. conifericola formed a separate distinct clade. Moreover, the analysis revealed two unknown Laetiporus taxa in Korea, one in South Africa, and one in Europe. As L. sulphureus is described from Europe (France), and we show that more than one taxon exist here, it is presently not possible to define L. sulphureus s. str. Abstract: Patterns of colonization of conifer roots by dark septate endopyhtes of the Phialocephala fortinii s. lat. species complex in nurseries in Switzerland and Lithuania were studied. The potential for man-mediated genotype flow was estimated for two Swiss nurseries based on customers'' addresses and the number of delivered plants. Two hundred and forty-nine strains from three Swiss and five Lithuanian nurseries and an afforestation site were characterized using a combination of inter-simple sequence repeat-anchored PCR (ISSR-PCR), single-copy RFLP analysis, and sequence analysis. P. fortinii s. lat. was abundant in nursery seedlings, but the frequency of seedlings colonized varied considerably among and within nurseries. Ten cryptic species (CSP) of P. fortinii s. lat. were identified, including four hitherto undiscovered CSP. P. helvetica was the dominant species in Swiss nurseries, whereas P. fortinii s. str. was the most abundant species in Lithuanian nurseries and the afforestation site. Swiss nurseries deliver plants over distances of more than 200km indicating the high potential for man-mediated genotype flow in P. fortinii s. lat. [Copyright &y& Elsevier]

Hapalopilus croceus is a rare, wood-decay polypore associated with old coarse broadleaf trees, primarily Quercus. It is mainly distributed throughout the temperate zone of the Northern Hemisphere with its main distribution in Europe. Mating systems in fungi influence the probability of in- or outbreeding and may affect the species' fitness. We found the mating system of H. croceus to be bipolar by pairing 18 single spore isolates from one sporocarp. We discuss this finding from the perspective of H. croceus as a threatened species with a very small and fragmented current population associated with a declining habitat and globally red-listed as Vulnerable (VU). Unlike for many other organism groups, conservation translocations of fungi are still rare. Encouraged by recent successful translocations, there is a growing interest in applying this conservation tool to threatened wood-inhabiting fungi. When combined with other conservation or restoration measures, translocation can be an effective measure for preventing further population decline in the short term, and species extinctions in the long term. Translocations can be appropriate for rare and specialist fungal species that occur as small local populations in isolated patches across fragmented landscapes, where there is a low likelihood of successful dispersal between distant host trees that have special qualities and are situated in suitable conditions. As species translocations are a controversial topic, the pros and cons of translocation as a conservation tool for threatened fungi need careful consideration. We highlight the uncertainties and risks that are connected to fungal translocations, and propose ten principles adhering to the precautionary principle. [ABSTRACT FROM AUTHOR]

Effective symbiosis with ectomycorrhizal (ECM) fungi is important for the successful growth and establishment of Pinus radiata. However, the structure of ECM communities varies across soil (edaphic) and environmental conditions, and thus affects the potential range of symbiotic associations. To improve our understanding of these factors, we characterised the range of ECM fungi present across six sites varying in edaphic properties. We also assessed ECM fungi in root tips of P. radiata planted two years earlier at these sites to assess the extent in overlap between root and soil ECM communities. The structure of the ECM community varied substantially with site. Across all sites, correlations were identified between soil pH and metrics describing ECM community structure in both soil and root samples, indicating soil pH was contributing to the differences in the fungal communities between sites. Mineralisable nitrogen, soil carbon, and phosphorus content also varied with site, but were not significantly related to descriptors of the ECM communities. Root tips and surrounding soils shared some taxa but were inhabited by diverse communities of fungi, and a number of ECM P. radiata associations unique to New Zealand were identified. It is possible that the establishment of linkages between metrics could provide an opportunity to begin predicting the response of ECM populations to site modification, and potentially increase the growth of radiata pine and improve forest health through enhanced resilience to disturbance.
