Abstract: Theranostic polymeric nanocarriers loaded with anticancer drug Taxol and superparamagnetic iron oxide nanocrystals have been developed for possible magnetic resonance imaging (MRI) use and cancer therapy. Multifunctional nanocarriers with a core–shell structure have been prepared by coating superparamagnetic Fe3O4 nanoparticles with block copolymer of poly(ethylene glycol)-b-poly(propylene succinate) with variable molecular weights of the hydrophobic block poly(prolylene succinate). The multifunctional polymer nano-vehicles were prepared using a nanoprecipitation method. Scanning transmission electron microscopy revealed the encapsulation of magnetic nanoparticles inside the polymeric matrix. Energy dispersive X-ray spectroscopy and electron energy loss spectroscopy mapping allowed us to determine the presence of the different material ingredients in a quantitative way. The diameter of the nanoparticles is below 250nm yielding satisfactory encapsulation efficiency. The nanoparticles exhibit a biphasic drug release pattern in vitro over 15 days depending on the molecular weight of the hydrophobic part of the polymer matrix. These new systems where anti-cancer therapeutics like Taxol and iron oxide nanoparticles (IOs) are co-encapsulated into new facile polymeric nanoparticles, could be addressed as potential multifunctional vehicles for simultaneous drug delivery and targeting imaging as well as real time monitoring of therapeutic effects. Abstract: In the present study chitosan grafted copolymers with poly(ethylene glycol) (CS-g-PEG) were prepared and studied using PEG with molecular weights 2000 and 5000g/mol. The materials were characterized using 1H NMR, FTIR and WAXD techniques. These polyelectrolytes were ionically crosslinked with tripolyphosphate (TPP) and poly(glutamic acid) (PGA) at different polymer/crosslinking agent ratios (1:1, 2:1, 3:1 and 4:1, w/w) for the nanoencapsulation of bovine serum albumin (BSA). Prepared nanoparticles are spherical in shape with a mean diameter ranging from 150 to 600nm. The size depends mainly to the molecular weight of the PEG and the crosslinking agent used. The PEG molecular weight also seems to affect the release rate of BSA especially the first burst effect which appears to be high in copolymers containing PEG5000, compared with copolymer prepared with PEG2000, and it is also higher when PGA was used as crosslinking agent, instead of TPP. Abstract: Chitosan (CS) nanoparticles of dorzolamide hydrochloride (Dorzo) and pramipexole hydrochloride (Prami) were prepared by the ionic gelation method and their in vitro properties were studied. The long-term objective is the development of efficient ocular formulations for Dorzo and efficient oral formulations for Prami. The particle size of nanoparticles was affected by the CS/drug ratio whereas it was not affected by the type of drug (Dorzo or Prami). Drug association efficiency to the nanoparticles did not appear to correlate with the drug/CS ratio whereas the loading capacity tended to fall with increasing drug proportion. Based on WAXD data, Dorzo was dispersed in the nanoparticles in crystalline form, probably due to the weak interaction developed between Dorzo and CS/TPP matrix as FT-IR data indicated. In contrast, WAXD and step-scan DSC data indicated that Prami formed a molecular dispersion within the nanoparticles. This was probably due to the potent interactions developed between Prami and CS/TPP matrix, as FT-IR data revealed. The nanoparticles exhibited mucoadhesive properties which diminished with increasing drug content. Sustained in vitro drug release was observed with the Dorzo-loaded CS nanoparticles in PBS (pH 7.4) and with the Prami-loaded CS nanoparticles in simulated intestinal fluid. The results obtained in this study suggest that the Dorzo-loaded CS nanoparticles and the Prami-loaded CS nanoparticles could be further evaluated for the controlled ocular delivery of Dorzo and the controlled oral delivery of Prami, respectively. Abstract: A series of novel poly(ε-caprolactone-co-propylene succinate) P(CL-co-PSu) copolymers having low propylene succinate content and high molecular weight were synthesized following a combinatory scheme of ring opening and polycondensation reactions, in an attempt to obtain copolymers of sufficient performance and increased biodegradation rates. Enzymatic hydrolysis of the copolymers was studied in the presence of mixture of Rhizopus delemar and Pseudomonas cepacia lipases. Much higher hydrolysis rates, comparing to neat PCL, were proved by both mass loss measurements and scanning electron microscopy (SEM) observations of the degraded film surfaces. Thermodynamics of cocrystallization and wide-angle X-ray diffraction (WAXD) patterns were investigated to estimate the extent of comonomer cocrystallization. Results of the study showed that comonomer inclusion may hold, though the molar fraction of the comonomer in the PCL crystals is lower than in the bulk. This means that not only the observed decrease of the degree of crystallinity from about 48% for PCL to about 29% for the P(CL-co-PSu) 75/25 favours enzymatic hydrolysis, but also the enrichment of the amorphous phase in the fast degrading propylene succinate units plays its role. The non-isothermal crystallization rates of the copolymers, like the melting points, decreased substantially when the propylene succinate content exceeded 8mol%. The activation energy of crystallization was calculated using the isoconversional method of Friedman, over the whole range of crystallization temperatures. An increase was found in the activation energy with increasing the comonomer content in the copolymers also proving the reduced symmetry along the copolymer chains due to the presence of comonomer units.
