Abstract The honeycomb worm Sabellaria alveolata is capable of building extensive bioconstructions, including what are currently considered Europe's largest biogenic reefs. The size and volume of these bioconstructions, however, vary greatly, such that not all habitats engineered by S. alveolata may be easily identified as reefs. Given that European environmental legislation protects marine habitats that are classified as "reefs", it is important to identity a clear set of definition criteria. Furthermore, quantifiable and unequivocal criteria are also needed to evaluate the ecological (health) state of these reefs, in order to best monitor and protect them. Here we propose new terminology to describe the physical appearance of these bioconstructions and attempt to link these physical criteria to the physiological state of the tube-building polychaete. We tested whether a bioconstruction displaying outward signs of growth is built by "healthy" worms devoid of physiological stress by analysing three macromolecules (carbohydrates, proteins, lipids), four polar lipid fatty acids, six neutral lipid fatty acid markers and three metabolic enzymes (citrate synthase, catalase and superoxide dismutase). The worms were sampled in bioconstructions of different "Type" (veneer vs. hummock), "Phase" (progradation vs. retrogradation), and "Shore Level" (high shore vs. low shore) at Champeaux in Mont-Saint-Michel Bay, France. Our results show that worms sampled in retrograding reefs (i.e. displaying signs of erosion and colonisation by epibionts such as oysters or mussels), were less physiologically stressed than worms sampled in prograding bioconstructions, possibly due to lower intraspecific competition and hence greater food availability. We therefore suggest management measures should encompass the whole mosaic of biogenic construction Types and Phases. We propose the inclusion of the polar lipid fatty acid arachidonic acid, in combination with the activity of two metabolic enzymes, citrate synthase and superoxide dismutase, as the three key biochemical markers to consider for quantitative information on the physiological state of this particular ecosystem engineer. Our results also revealed the influence of both sex and size on fatty acid and enzyme levels, highlighting the importance of taking into account both these variables when sampling and subsequently pooling individuals by sex and size category for laboratory analyses. Once seasonal and site variation have been addressed, these biochemical indicators could be examined in parallel with S. alveolata bioconstruction physical criteria as part of a European-wide protocol for monitoring ecological status in this potential reef habitat. Multifactorial studies assessing the cumulative effects of natural and anthropogenic stressors on individual stress response are crucial to understand how organisms and populations cope with environmental change. We tested direct and indirect causal pathways through which environmental stressors affect the stress response of wild gilthead seabream in Mediterranean costal lagoons using an integrative PLS-PM approach. We integrated information on 10 environmental variables and 36 physiological variables into seven latent variables reflecting lagoons features and fish health. These variables concerned fish lipid reserves, somatic structure, inorganic contaminant loads, and individual trophic and stress response levels. This modelling approach allowed explaining 30 % of the variance within these 46 variables considered. More importantly, 54 % of fish stress response was explained by the dependent lagoon features, fish age, fish diet, fish reserve, fish structure and fish contaminant load latent variables included in our model. Abstract: Pacific oyster Crassostrea gigas suffers from chronic or sporadic mortality outbreaks worldwide, resulting from infectious diseases and/or physiological disorders triggered by environmental factors. Since 2008, ostreid herpesvirus OsHV-1 μVar has been identified as the main agent responsible for mass mortality of juvenile oysters in Europe. Previous studies of genome-wide expression profiling have provided candidate genes that potentially contribute to genetically-based resistance to summer mortality. To assess their value in determining resistance to the juvenile mass mortality that has occurred in France since 2008, we analyzed the expression of 17 candidate genes in an experimental infection by OsHV-1 μVar, and in an in vivo field experiment. Individual quantification of mRNA levels of 10 out of the 17 targeted genes revealed significant variation, of which 7 genes were showed differences between conditions that created significant differences in mortality, and 6 depended on the number of OsHV-1 genome copies individually quantified in mantle tissue. Complex SOD metalloenzymes known to be part of the antioxidant defense strategies may at least partly determine susceptibility or resistance to OsHV-1-associated mortality. Furthermore, inhibitor 2 of NF-κB, termed CgIκB2, exhibited highly significant variation of mRNA levels depending on OsHV-1 load in both experiments, suggesting its implication in the antiviral immune response of C. gigas. Our results suggest that CgIκB2 expression would make a good starting point for further functional research and that it could be used in marker-assisted selection. [Copyright &y& Elsevier]

Abstract: Carbon stable isotopes and fatty acids were measured in the suspended particulate organic matter (POM) of the Thau lagoon to study its qualitative temporal changes in relation to environmental factors and to identify the food sources of bivalves over a one-yr-cycle in relation to their growth. Reciprocally, the impact of shellfish farming on POM was also studied. Oysters and mussels were sampled and measured for biometry, stable isotopes and fatty acid composition. Water samples were collected at two sites, both inside and outside of the shellfish farming area, to determine concentrations in POM, chlorophyll a (Chl a) and stable isotopes. Carbon isotopes and fatty acids in bivalves reflected seasonal changes in food sources, which varied consistently with the environment. Seasonal changes in δ13C and fatty acids in the bivalves suggested that dietary phytoplankton contribution varied according to season. Terrestrial organic matter and bacteria can contribute to the diet of bivalves during non-bloom periods. Mussels seemed to rely more on diatoms and less on terrestrial organic matter and bacteria than oysters did, particularly when phytoplankton biomass was low during the summer. Although one- and two-yr-old oysters showed similar δ13C, their fatty acid dynamics differed slightly. Periods of high growth rate in bivalves were mainly fuelled by diatoms, thus highlighting the importance of seasonal blooms of microphytoplankton during the critical period of bivalve growth and gamete production. Although there was no significant effect of shellfish farms on Chl a and POM δ13C, consistent differences indicate that stable isotopes could be used successfully to investigate the effects of bivalve aquaculture. Abstract: A temperature decrease usually induces an ordering effect in membrane phospholipids that can lead to membrane dysfunction. Ectotherms typically counteract this temperature effect by remodeling membrane lipids as stipulated in the homeoviscous adaptation theory (HVA). Previous studies mostly focused on the remodeling of membrane lipids during long-term acclimatization or acclimation at constant temperature regimes, whereas in nature, many organisms experience variations in temperature on a daily basis and must react to this changing thermal environment. The objective of this study was to examine the composition of membrane lipids in oysters subjected to long-term acclimation at constant temperatures (12 or 25 °C) or to environmentally realistic daily fluctuations in temperature between 12 and 25 °C for 7 d. The lipid composition of gill in oysters subjected to long-term acclimation at a constant temperature or to daily temperature fluctuations varied in a way consistent with HVA: oysters adjusted their phospholipid to sterol ratio in response to long-term acclimation to a constant temperature but not to daily temperature fluctuations. In contrast, the unsaturation index of polar lipids in oysters varied in response to both long-term acclimation to a constant temperature and to daily temperature fluctuations, mainly due to changes in 22:6n-3 and 20:5n-3. The 20:4n-6 levels in oyster gills increased as temperature rose, suggesting an increasing availability of this fatty acid for eicosanoid biosynthesis during stress responses. Abstract: Quantification of mRNA of genes related to metabolism, immunity and cellular stress was examined in relation to a massive mortality event during the culture of American oyster larvae, Crassostrea virginica which was probably, in regard to previous microbiological analysis, induced by Vibrio infection. To document molecular changes associated with the mortality event, mRNA levels were compared to biochemical and physiological data, previously described in a companion paper. Among the 18 genes studied, comparatively to the antibiotic control, 10 showed a lower relative gene expression when the massive mortality occurred. Six of them are presumed to be related to metabolism, corroborating the metabolic depression associated with the mortality event suggested by biochemical and physiological analyses. Relationships between the regulation of antioxidant enzyme activities, lipid peroxidation, and the mRNA abundance of genes linked to oxidative stress, cytoprotection, and immune response are also discussed. Finally, we observed an increase in the transcript abundance of two genes involved in apoptosis and cell regulation simultaneously with mortality, suggesting that these processes might be linked. In intertidal zones, species such as sessile shellfish exhibit extended phenotypic plasticity to face rapid environmental changes, but whether frequent exposure to intertidal limits of the distribution range impose physiological costs for the animal remains elusive. Here, we explored how phenotypic plasticity varied along foreshore range at multiple organization levels, from molecular to cellular and whole organism acclimatization, in the Pacific oyster (Crassostrea gigas). We exposed 7-month-old individuals for up to 16 months to three foreshore levels covering the vertical range for this species, representing 20, 50 and 80% of the time spent submerged monthly. Individuals at the upper range limit produced energy more efficiently, as seen by steeper metabolic reactive norms and unaltered ATP levels despite reduced mitochondrial density. By spending most of their time emerged, oysters mounted an antioxidant shielding concomitant with lower levels of pro-oxidant proteins and postponed age-related telomere attrition. Instead, individuals exposed at the lower limit range near subtidal conditions showed lower energy efficiencies, greater oxidative stress and shorter telomere length. These results unraveled the extended acclimatization strategies and the physiological costs of living too fast in subtidal conditions for an intertidal species. We investigated the effect of brown ring disease (BRD) development and algal diet on energy reserves and activity of enzymes related to energy metabolism, antioxidant system and immunity in Manila clam, Ruditapes philippinarum . We found that algal diet did not impact the metabolic response of clams exposed to Vibrio tapetis . At two days post-injection (dpi), activities of superoxide dismutase and glutathione peroxidase (GPx) decreased whereas activities of nitric oxide synthase (iNOS) and catalase increased in infected clams, although no clinical signs were visible (BRD−). At 7 dpi, activities of several antioxidant and immune-related enzymes were markedly increased in BRD-likely indicating an efficient reactive oxygen species (ROS) scavenging compared to animals which developed clinical signs of BRD (BRD+). Therefore, resistance to BRD clinical signs appearance was associated with higher detoxification of ROS and enhancement of immune response. This study provides new biochemical indicators of disease resistance and a more comprehensive view of the global antioxidant response of clam to BRD development. [ABSTRACT FROM AUTHOR]

The Pacific oyster Crassostrea gigas is established in the marine intertidal zone, experiencing rapid and highly dynamic environmental changes throughout the tidal cycle. Depending on the bathymetry, oysters face oxygen deprivation, lack of nutrients, and high changes in temperature during alternation of the cycles of emersion/immersion. Here we showed that intertidal oysters at a bathymetry level of 3 and 5 m delayed by ten days the onset of mortality associated with Pacific Oyster Mortality Syndrome (POMS) as compared to subtidal oysters. Intertidal oysters presented a lower growth but similar energetic reserves to subtidal oysters but induced proteomic changes indicative of a boost in metabolism, inflammation, and innate immunity that may have improved their resistance during infection with the Ostreid herpes virus. Our work highlights that intertidal harsh environmental conditions modify host-pathogen interaction and improve oyster health. This study opens new perspectives on oyster farming for mitigation strategies based on tidal height.
