Peritoneal carcinomatosis (PC) from colorectal cancer (CRC) is a highly challenging disease to treat. Systemic chimeric antigen receptor (CAR) T cells have shown impressive efficacy in hematologic malignancies but have been less effective in solid tumors. We explored whether intraperitoneal (i.p.) administration of CAR T cells could provide an effective and robust route of treatment for PC from CRC. We generated second-generation carcinoembryonic antigen (CEA)-specific CAR T cells. Various animal models of PC with i.p. and extraperitoneal metastasis were treated by i.p. or intravenous (i.v.) administration of CEA CAR T cells. Intraperitoneally administered CAR T cells exhibited superior anti-tumor activity compared with systemic i.v. cell infusion in an animal model of PC. In addition, i.p. administration conferred a durable effect and protection against tumor recurrence and exerted strong anti-tumor activity in an animal model of PC with metastasis in i.p. or extraperitoneal organs. Moreover, compared with systemic delivery, i.p. transfer of CAR T cells provided increased anti-tumor activity in extraperitoneal tumors without PC. This phenomenon was further confirmed in an animal model of pancreatic carcinoma after i.p. administration of our newly constructed prostate stem cell antigen-directed CAR T cells. Various complications such as posterior capsule opacification (PCO) and endophthalmitis occur after intraocular lenses (IOLs) implantation in cataract surgery. It is urgent to construct antifouling and antibacterial IOLs to lower the incidence of PCO and endophthalmitis. Bionic zwitterionic polymer such as 2-methacryloyloxyethyl phosphorylcholine (MPC) shows excellent performance in resisting nonspecific proteins and bacterial adhesion. In this work, a novel bionic brushes coating containing MPC and N, N, N-trimethyl-2-((4-(2-(4-nonylphenoxy) ethoxy)-4-oxobut-2-enoyl) oxy) ethan-1-aminium chloride (TOEAC), a quaternary ammonium monomer, was prepared onto IOLs using reversible addition–fragmentation chain transfer (RAFT) polymerization method. The P(TOEAC- co -MPC) brushes exhibited excellent antifouling efficiency against bovine serum albumin, Staphylococcus aureus , and human lens epithelial cells. In addition, the P(TOEAC- co -MPC) brushes showed excellent antibacterial and antibiofilm abilities and good biocompatibility. An in vivo study confirmed that the P(TOEAC- co -MPC) brushes effectively prevented PCO and endophthalmitis. Consequently, the P(TOEAC- co -MPC) bionic brushes are promising for IOLs surface modification to resist postoperative complications for long-term implantation. • A novel PU/PDA-Hep/CMCS coating was prepared onto implants mediated by polydopamine. • The PU/PDA-Hep/CMCS coating exhibited excellent improved hemocompatibility. • The PU/PDA-Hep/CMCS coating showed excellent antibacterial and anti-biofilm abilities. • The PU/PDA-Hep/CMCS coating is promising for implants modification for long-term implantation.
