Background: Healthcare-associated norovirus outbreaks place a large burden on healthcare staff. Environmental service workers (ESWs), however, remain understudied despite high contact with potentially contaminated surfaces. Understanding the magnitude of the risk of norovirus infection in healthcare ESWs can protect workers and improve infection control.Aim: This study simulated the risk of norovirus infection for unprotected ESWs after a single fomite contact, assuming no disinfection or protective equipment, in norovirus-positive patient rooms. In addition, the risk of secondary surface transmission from norovirus-exposed ESWs was simulated.Methods: A quantitative microbial risk assessment employing two-dimensional Monte Carlo simulation with parameters extracted from the literature was used to estimate norovirus infection from multiple fomite contact scenarios defined by: norovirus source (patient vomit/diarrhoea), location (bathroom/patient room) and target outcome (ESW/secondary illness).Findings: Unprotected ESWs have a maximum estimated risk of norovirus infection of 33% (1:3) for a single fomite contact in a room where a norovirus-positive patient had a diarrhoeal event. Patient vomit events lead to fomite contact risk estimates that are four orders of magnitude lower than those for diarrhoeal events. The estimated risk of secondary illness from touching a common surface is as high as 25% (1:4) after single fomite exposure following a diarrhoeal event.Conclusions: A single fomite contact may lead to sizable risk of norovirus infection in ESWs if personal protective equipment and disinfection are not used appropriately. ESWs can also transfer virus to secondary surfaces, initiating further infections. There is substantial potential for human exposure to viruses in environmental matrixes. Identification of virally contaminated environmental reservoirs requires assays with sufficient sensitivity to detect low copy numbers of viral targets. However, low detection sensitivity frequently requires sample concentration during which inhibitors to downstream assays co-isolate with desired target. Conventional detection assays (e.g., cell culture, polymerase chain reaction) require a priori selection of appropriate cell lines or primers and probes based on the viruses anticipated to be present in the sample. This can underestimate exposure risks by excluding unidentified or unknown virus. Emerging methods including nonspecific adsorption/elution, filtration, and total nucleic acid sequencing, that are capable of concentrating, purifying, and detecting total virus and/or total virus nucleic acid will aid in estimates of exposure risk, source tracking, intervention efficacy, and evaluation of virus fate and transport. Development and implementation of novel virus detection techniques must integrate quality assurance guidelines to validate results and provide opportunities for interstudy comparison. Abstract: Magnetic ion exchange (MIEX) pretreatment has been increasingly employed by water treatment plants for removal of dissolved organic carbon (DOC). In this study, the effects of MIEX pretreatment on low pressure membrane filtration of natural surface water were investigated under different feedwater qualities, membrane properties, and MIEX dosing conditions. Regardless of feedwater DOC, moderate decrease in the total and hydraulically irreversible fouling was observed for a polyvinylidene fluoride (PVDF) microfiltration membrane and a polyethersulfone ultrafiltration (UF) membrane after MIEX pretreatment, which was coincident with moderate removals of high molecular weight DOC in the feedwaters. Comparatively, the fouling of a PVDF UF membrane did not decrease after MIEX pretreatment, revealing the impact of membrane properties on membrane fouling in the presence of MIEX pretreatment. Reuse of virgin or regenerated MIEX resulted in similar membrane fouling as observed with single use of the virgin MIEX. The level of DOC removal by MIEX was similar to the removal of MS2 bacteriophage spiked in the feedwater, suggesting a potential similarity in the removal of organic and microbial particles. In conclusion, MIEX pretreatment was effective for DOC removal, but less effective in controlling short-term membrane fouling or removing viruses. [Copyright &y& Elsevier]

Cryptosporidiosis, giardiasis and microsporidiosis are serious human diseases of waterborne origin; their etiologic agents and a substantial fecal coliform load can enter surface, drinking and recreational water resources from aquatic birds. The aim of this article is to present interactions between waterfowl and these waters that imply a negative public health impact, reinforcing the need for either better water-quality indicators or for water monitoring specifically for Cryptosporidium, Giardia and microsporidia. Where justifiable, the presence of waterfowl should be supported; however, management of drinking and recreational water resources needs to be improved by incorporating effective protection measures for pathogens linked to these birds. Noroviruses cause significant global health burdens and waterborne transmission is a known exposure pathway. Chlorination is the most common method of disinfection for water and wastewater worldwide. The purpose of this study was to investigate the underlying causes for discrepancies in human norovirus (hNoV) resistance to free chlorine that have been previously published, and to assess hNoV GI and GII persistence during disinfection of municipal secondary wastewater (WW) effluent. Our results reveal that choice of hNoV purification methodology prior to seeding the viruses in an experimental water matrix influences disinfection outcomes in treatment studies. Common hNoV purification processes such as solvent extraction and 0.45-μm filtration were ineffective in removing high levels of organics introduced into water or wastewater samples when seeding norovirus positive stool. These methods resulted in experimental water matrices receiving an additional 190 mg/L as Cl 2 of 15-s chlorine demand and approximately 440 mg/L as Cl 2 of 30-min chlorine demand due to seeding norovirus positive stool at 1% w/v. These high organic loads impact experimental water chemistry and bias estimations of hNoV persistence. Advanced purification of norovirus positive stool using sucrose cushion ultracentrifugation and ultrafiltration reduced 15-s chlorine demands by 99% and TOC by 93% for loose (i.e. unformed diarrhea) stools. Using these methods, hNoV GI and GII persistence was investigated during free chlorination of municipal WW. A suite five of kinetic inactivation models was fit to viral reverse transcription-qPCR reduction data, and model predicted CT values for 1, 2, and 3 log 10 reduction of hNoV GI in municipal WW by free chlorine were 0.3, 2.1, and 7.8 mg-min/L, respectively. Model predicted CT values for reduction of hNoV GII in WW were 0.4, 2.0, and 7.0 mg-min/L, respectively. These results indicate that current WW treatment plant disinfection practices employing free chlorine are likely protective for public health with regards to noroviruses, and will achieve at least 3-log reduction of hNoV GI and GII RNA despite previous reports of high hNoV resistance. Widespread contamination of groundwater with per- and polyfluoroalkyl substances (PFAS) has required drinking water producers to quickly adopt practical and efficacious treatments to limit human exposure and deleterious health outcomes. This pilot-scale study comparatively investigated PFAS adsorption behaviors in granular activated carbon (GAC) and two strong-base gel anion exchange resin (AER) columns operated in parallel over a 441-day period to treat contaminated groundwater dominated by short-chain perfluorocarboxylic acids (PFCA). Highly-resolved breakthrough profiles of homologous series of 2–8 CF 2 PFCA and perfluorosulfonic acids (PFSA), including ultrashort-chain compounds and branched isomers, were measured to elucidate adsorption trends. Sample ports at intermediate bed depths could predict 50% breakthrough of compounds on an accelerated basis, but lower empty bed contact times led to conservative estimates of initial breakthrough. Homologous PFAS series displayed linear (GAC) and log-linear (AER) relationships between chain-length and breakthrough, independent of initial concentration. AERs generally outperformed GAC on a normalized bed volume basis, and this advantage widened with increasing PFAS chain-length. As designed, all treatments would have short full-scale service times (≤142 days for GAC; ≤61 days for AERs) before initial breakthrough of short-chain (2–4 CF 2) PFCA. However, AER displayed far longer breakthrough times for PFSA compared to GAC (>3× treatment time), and breakthrough was not observed for PFSA with >4 CF 2 in AERs. GAC had a finite molar adsorption capacity for total PFAS, leading to a stoichiometric replacement of short-chain PFCA by PFSA and longer-chain PFCA over time. AERs quickly reached a finite adsorption capacity for PFCA, but they showed substantially greater selectivity for PFSA whose capacity was not reached within the duration of the pilot. Breakthrough characteristics of keto- and unsaturated-PFSA, identified in the groundwater by suspect screening, were also evaluated in absence of reference standards. Modified PFAS structures (branched, keto-, unsaturated-) broke through faster than linear and unmodified perfluorinated structures with equal degrees of fluorination, and the effects were more pronounced in GAC compared to AERs. The results highlight that the design of robust PFAS treatment systems should consider facets beyond current PFAS targets including operational complexities and impacts of unregulated and unmonitored co-contaminants. For the fresh-cut produce industry, a critical area of concern is potential pathogen cross-contamination during wash operations when wash water is reused and re-circulated in wash systems continuously imputed with fresh-cut produce. However, little research has focused on the chemical properties of wash water. Organic input from residual soil and vegetable material deteriorates water quality and creates increasing chlorine demand within this wash water. This study evaluated the origins of chlorine demand input and chlorine decay kinetics of fresh-cut produce wash water. Using a model system, vegetable juice released per kg of processed produce for shredded romaine lettuce, shredded iceberg lettuce, shredded carrot and baby spinach was 82.1 mL/kg, 94.5 mL/kg, 158 mL/kg, and 2.26 mL/kg, respectively. Batch water analysis revealed a rapid reaction between constituents in the wash water and chlorine where over a 90 min observation period, 50% of chlorine demand occurred within first 5 min, underscoring the challenge for any water treatment process to reduce chlorine demand once vegetables are deposited into washing systems. Moreover, the results also showed sustained chlorine demand over 90-min periods, indicating an accumulative effect on chlorine consumption with continuous organic input. Additionally, HPLC-SEC analysis showed that the constituents contributing to chlorine demand are predominantly dissolved small molecules (<3400 Da), which will challenge water reuse treatment approaches. This study provides quantitative information of chlorine demand origins and chlorine decay kinetics in wash water and provides baseline data critical for integrating water reuse in the fresh produce processing industry. Recent studies showing an association between fecal indicator organisms (FIOs) in sand and gastrointestinal (GI) illness among beachgoers with sand contact have important public health implications because of the large numbers of people who recreate at beaches and engage in sand contact activities. Yet, factors that influence fecal pollution in beach sand remain unclear. During the 2007 National Epidemiological and Environmental Assessment of Recreational (NEEAR) Water Study, sand samples were collected at three locations (60 m apart) on weekend days (Sat, Sun) and holidays between June and September at two marine beaches — Fairhope Beach, AL and Goddard Beach, RI — with nearby publicly-owned treatment works (POTWs) outfalls. F + coliphage, enterococci, Bacteroidales , fecal Bacteroides spp., and Clostridium spp. were measured in sand using culture and qPCR-based calibrator-cell equivalent methods. Water samples were also collected on the same days, times and transects as the 144 sand samples and were assayed using the same FIO measurements. Weather and environmental data were collected at the time of sample collection. Mean FIO concentrations in sand varied over time, but not space. Enterococci CFU and CCE densities in sand were not correlated, although other FIOs in sand were. The strongest correlation between FIO density in sand and water was fecal Bacteroides CCE, followed by enterococci CFU, Clostridium spp. CCE, and Bacteroidales CCE. Overall, the factors associated with FIO concentrations in sand were related to the sand–water interface (i.e., sand-wetting) and included daily average densities of FIOs in water, rainfall, and wave height. Targeted monitoring that focuses on daily trends of sand FIO variability, combined with information about specific water quality, weather, and environmental factors may inform beach monitoring and management decisions to reduce microbial burdens in beach sand. Abstract: Carbon nanotubes (CNTs) were investigated for their capability and mechanisms to simultaneously remove colloidal natural organic matter (NOM) and humic substances from natural surface water. Static removal testing was conducted via adsorption experiments while dynamic removal was evaluated by layering CNTs onto substrate membranes and filtering natural water through the CNT-layered membranes. Analyses of treated water samples showed that removal of humic substances occurred via adsorption under both static and dynamic conditions. Removal of colloidal NOM occurred at a moderate level of 36–66% in static conditions, independent of the specific surface area (SSA) of CNTs. Dynamic removal of colloidal NOM increased from approximately 15% with the unmodified membrane to 80–100% with the CNT-modified membranes. Depth filtration played an important role in colloidal NOM removal. A comparison of the static and dynamic removal of humic substances showed that equilibrium static removal was higher than dynamic (p < 0.01), but there was also a significant linear relationship between static and dynamic removal (p < 0.05). Accounting for contact time of CNTs with NOM during filtration, it appeared that CNT mat structure was an important determinant of removal efficiencies for colloidal NOM and humic substances during CNT membrane filtration. Abstract: As a result of the widespread use of antibiotics in large-scale U.S. poultry production, a significant proportion of Salmonella strains recovered from conventional poultry farms and retail poultry products express antibiotic resistance. We evaluated whether large-scale poultry farms that transitioned from conventional to organic practices and discontinued antibiotic use were characterized by differences in the prevalence of antibiotic-resistant Salmonella compared to farms that maintained conventional practices. We collected poultry litter, water and feed samples from 10 newly organic and 10 conventional poultry houses. Samples were analyzed for Salmonella using standard enrichment methods. Isolates were confirmed using standard biochemical tests and the Vitek®2 Compact System. Antimicrobial susceptibility testing was performed by Sensititre® microbroth dilution. Data were analyzed using Fisher's exact test and generalized linear mixed models. We detected Salmonella in both conventional and newly organic poultry houses. Salmonella Kentucky was the predominant serovar identified, followed by S. Orion, S. Enteritidis, S. Gostrup and S. Infantis. Among S. Kentucky isolates (n=41), percent resistance was statistically significantly lower among isolates recovered from newly organic versus conventional poultry houses for: amoxicillin–clavulanate (p =0.049), ampicillin (p =0.042), cefoxitin (p =0.042), ceftiofur (p =0.043) and ceftriaxone (p =0.042). Percent multidrug resistance (resistance to ≥3 antimicrobial classes) was also statistically significantly lower among S. Kentucky isolates recovered from newly organic poultry houses (6%) compared to those recovered from conventional houses (44%) (p =0.015). To our knowledge, these are the first U.S. data to show immediate, on-farm changes in the prevalence of antibiotic-resistant Salmonella when antibiotics are voluntarily withdrawn from large-scale poultry facilities in the United States. Abstract: Carbon nanotubes (CNTs) with different physiochemical properties were layered onto low pressure membranes and tested for antifouling properties using a natural surface water with high fouling potential. Membranes modified with the largest diameter pristine multi-walled CNTs (MWCNTs) were most effective in controlling membrane fouling, tripling the time it took for the membrane to become noticeably fouled at a CNT loading of 22 g/m2. The differences in the structure of CNT layers were an important contributing factor for antifouling properties; scanning electron microscopy imaging showed that large diameter MWCNTs formed homogeneous porous layers across the membrane surface, while less effective, small diameter MWCNTs formed heterogeneous layers. Water quality analysis showed that CNT-membranes constructed with larger diameter CNTs were more effective at removing larger organic macromolecules responsible for fouling from feedwater compared to membranes made with smaller diameter CNTs. This reduced the concentration of foulants reaching the PVDF membrane and thus helped reduce membrane fouling. Beneficial for application, increased loadings of CNTs onto the membrane surface increased resistance to fouling while only slightly reducing the clean water permeability of the modified membranes. Overall, CNT layered membranes were shown to highly resist membrane fouling with potential applications in sustainable water treatment. [Copyright &y& Elsevier]

Abstract: Microbial monitoring was conducted over a period of more than 1year at three full-scale riverbank filtration (RBF) facilities, located in the United States along the Ohio, Missouri, and Wabash Rivers. Results of this study demonstrated the potential for RBF to provide substantial reductions in microorganism concentrations relative to the raw water sources. Cryptosporidium and Giardia were detected occasionally in the river waters but never in any of the well waters. Average concentrations and log reductions of Cryptosporidium and Giardia could not be accurately determined due to the low and variable concentrations in the river waters and the lack of detectible concentrations in the well waters. Average concentrations of aerobic and anaerobic spore-forming bacteria, which have both been proposed as potential surrogates for the protozoans, were reduced at the three facilities by 0.8 to >3.1 logs and 0.4 to >4.9 logs, respectively. Average concentrations of male-specific and somatic bacteriophage were reduced by >2.1 logs and ⩾3.2 logs, respectively. Total coliforms were rarely detected in the well waters, with 5.5 and 6.1 log reductions in average concentrations at the two wells at one of the sites relative to the river water. Average turbidity reductions upon RBF at the three sites were between 2.2 and 3.3 logs. Turbidity and microbial concentrations in the river waters generally tracked the river discharge; a similar relationship between the well water concentrations and river discharge was not observed, due to the low, relatively constant well water turbidities and lack of a significant number of detections of microorganisms in the well waters. Further research is needed to better understand the relationships among transport of pathogens (e.g., Cryptosporidium, Giardia, viruses) and potential surrogate parameters (including bacterial spores and bacteriophage) during RBF and the effects of water and sediment characteristics on removal efficiency. • First large-scale peer-reviewed investigation of PFAS in U.S. bottled water. • PFAS were detected in 39/101 uniquely labelled bottled water products. • Ultrashort-chain perfluoropropanoic acid (PFPrA) comprised 42% of detected PFAS mass. • Reverse osmosis-treated water contained less PFAS than other bottled water products. Per- and polyfluoroalkyl substances (PFAS) are compounds of emerging concern due to their persistence in the global water cycle and detection in drinking water sources. However, PFAS have been poorly studied in bottled water, especially in the United States. This study investigated the occurrence of PFAS and related factors in 101 uniquely labelled bottled water products for sale in the U.S. Products were screened for 32 target PFAS by solid phase extraction-liquid chromatography-tandem mass spectrometry (SPE-LC-MS/MS). Fifteen of 32 measured analytes were detected, consisting primarily of C3-C10 perfluorocarboxylic acids (PFCA) and C3-C6 and C8 perfluorosulfonic acids (PFSA). PFAS were detected above method detection limits in 39/101 tested products. The Σ 32 PFAS concentrations detected were 0.17–18.87 ng/L with a median of 0.98 ng/L; 97% of samples were below 5 ng/L. PFCA (83%) and short-chain perfluoroalkyl acids (PFAA) containing 5 or less CF 2 groups (67%) were more prevalent on a mass basis than PFSA and longer-chain PFAA, respectively. Ultrashort-chain PFPrA, measured for the first time in bottled water, accounted for the greatest individual fraction of detected PFAS mass (42%) and was found almost exclusively in products labeled as Spring water. Purified water products contained significantly less PFAS than Spring water products, which was attributed to the use of reverse osmosis (RO) treatment in the majority of Purified waters (25/35) compared to Spring waters (1/45). RO-treated products contained significantly lower Σ 32 PFAS, long-chain, short-chain, and PFPrA concentrations than products without RO. Although no enforceable PFAS regulations exist for bottled water in the U.S., the finding that some products approach levels of concern justify a framework for monitoring PFAS in bottled water production.
