Active magnetic bearings are used in a number of applications but their disadvantage is the high asynchronous error due to sensor noise amplification. In this paper a new radial position sensor for active magnetic bearings (AMB) based on linear optical encoders is presented. A commercial encoder scanning head faces a round scale with concentric, coplanar lines on its face. Because such a scale is not readily available, it is made by high precision micro machining and different options are compared. In experiments a measurement noise of 3.5 nm at 10 kHz bandwidth is achieved. In addition, a magnetic bearing is built to demonstrate the sensor in closed-loop. [ABSTRACT FROM AUTHOR]

All measurements performed in science and industry are based on the International System of Units, the SI. It has been proposed to revise the SI following an approach which was implemented for the redefinition of the unit of length, the metre, namely to define the SI units by fixing the numerical values of so-called defining constants, including c , h , e , k and N A . We will discuss the reasoning behind the revision, which will likely be put into force in 2018. Abstract: This paper presents a review of the latest research activities and gives an overview of the state of the art in understanding changes in machine tool performance due to changes in thermal conditions (thermal errors of machine tools). The topics are focused on metal cutting machine tools, especially on turning and milling machines as well as machining centres. The topics of the paper thermal issues in machine tools include measurement of temperatures and displacements, especially displacements at the tool centre point, computations of thermal errors of machine tools, and reduction of thermal errors. Computing the thermal errors of machine tools include both, temperature distribution and displacements. Shortly addressed is also to avoid thermal errors with temperature control, the influence of fluids and a short link to energy efficiency of machine tools. The paper presents the summary of research work in the past and current. Research challenges in order to achieve a thermal stable machine tool are discussed. The paper apprehend itself as an update and not a substitution of two published keynote papers of Bryan et al. in 1990 and Weck et al. in 1995. [Copyright &y& Elsevier]

Summary: The Wilms'' tumor 1 protein WT1 is a transcriptional regulator that is involved in cell growth and differentiation. The transcriptional corepressor BASP1 interacts with WT1 and converts WT1 from a transcriptional activator to a repressor. Here, we demonstrate that the N-terminal myristoylation of BASP1 is required in order to elicit transcriptional repression at WT1 target genes. We show that myristoylated BASP1 binds to nuclear PIP2, which leads to the recruitment of PIP2 to the promoter regions of WT1-dependent target genes. BASP1''s myristoylation and association with PIP2 are required for the interaction of BASP1 with HDAC1, which mediates the recruitment of HDAC1 to the promoter and elicits transcriptional repression. Our findings uncover a role for myristoylation in transcription, as well as a critical function for PIP2 in gene-specific transcriptional repression through the recruitment of histone deacetylase. Targeted deletion of the Runx2 gene in mice has demonstrated that Runx2 is a master regulator of osteoblast differentiation. Runx2 has therefore largely been regarded as a bone-specific transcription factor. Runx2[sup -/-] mice die shortly after birth and therefore the role of Runx2 in later developing tissues remains unclear. Here we show that the Runx2 protein is expressed in several mammary epithelial cell lines and in primary mammary epithelial cells. In addition, we have also found that it has a functionally important role in gene regulation. Osteopontin (OPN) is expressed in mammary epithelial cells during pregnancy and lactation and has been shown to have a role in mammary gland differentiation. Here we show that a Runx2 site in the OPN promoter is required for activation of the promoter in mammary epithelial cells. Moreover, dominant-negative Runx proteins can inhibit both activation of an OPN promoter reporter in transient transfections and expression of the endogenous OPN gene in mammary epithelial cells. Our data suggest, for the first time, that the osteoblast transcription factor Runx2 has a role in the normal regulation of gene expression in mammary epithelial cells. [ABSTRACT FROM AUTHOR]

Specificity protein 1 (Sp1) is an important transcription factor implicated in numerous cellular processes. However, whether Sp1 is involved in the regulation of RNA polymerase III (Pol III)- directed gene transcription in human cells remains unknown. Here, we first show that filamin A (FLNA) represses Sp1 expression as well as expression of TFIIB-related factor 1 (BRF1) and general transcription factor III C subunit 2 (GTF3C2) in HeLa, 293T, and SaOS2 cell lines stably expressing FLNA-silencing shRNAs. Both BRF1 promoter 4 (BRF1P4) and GTF3C2 promoter 2 (GTF3C2P2) contain putative Sp1-binding sites, suggesting that Sp1 affects Pol III gene transcription by regulating BRF1 and GTF3C2 expression. We demonstrate that Sp1 knockdown inhibits Pol III gene transcription, BRF1 and GTF3C2 expression, and the proliferation of 293T and HeLa cells, whereas Sp1 overexpression enhances these activities. We obtained a comparable result in a cell line in which both FLNA and Sp1 were depleted. These results indicate that Sp1 is involved in the regulation of Pol III gene transcription independently of FLNA expression. Reporter gene assays showed that alteration of Sp1 expression affects BRF1P4 and GTF3C2P2 activation, suggesting that Sp1 modulates Pol III-mediated gene transcription by controlling BRF1 and GTF3C2 gene expression. Further analysis revealed that Sp1 interacts with and thereby promotes the occupancies of TATA box-binding protein, TFIIAα, and p300 at both BRF1P4 and GTF3C2P2. RNA polymerase II (pol II) is required for the transcription of all protein-coding genes and as such represents a major enzyme whose activity is tightly regulated. Transcriptional initiation therefore requires numerous general transcriptional factors and cofactors that associate with pol II at the core promoter to form a pre-initiation complex. Transcription factor IIA (TFIIA) is a general cofactor that binds TFIID and stabilizes the TFIID-DNA complex during transcription initiation. Previous studies showed that TFIIA can make contact with the DNA sequence upstream or downstream of the TATA box, and that the region bound by TFIIA could overlap with the elements recognized by another factor, TFIIB, at adenovirus major late core promoter. Whether core promoters contain a DNA motif recognized by TFIIA remains unknown. Here we have identified a core promoter element upstream of the TATA box that is recognized by TFIIA. A search of the human promoter database revealed that many natural promoters contain a TFIIA recognition element (IIARE). We show that the IIARE enhances TFIIA-promoter binding and enhances the activity of TATA-containing promoters, but represses or activates promoters that lack a TATA box. Chromatin immunoprecipitation assays revealed that the IIARE activates transcription by increasing the recruitment of pol II, TFIIA, TAF4, and P300 at TATA-dependent promoters. These findings extend our understanding of the role of TFIIA in transcription, and provide new insights into the regulatory mechanism of core promoter elements in gene transcription by pol II. This paper presents a novel concept, a virtual metrology frame, for enhancing the dynamic performance of a machine tool with a flexible structural frame. The dynamic properties of a machine are directly affected by the stiffness of its frame, and its reference system; thus, by having an unstressed metrology frame, superior dynamic capabilities can be achieved. The developed concept does not require physical components associated with metrology frame; hence it is ideal for machine tools with requirements for small footprint and ultra-precision performance. The concept relies on an accelerometer based dynamic displacement feedback technique, where the accelerometer is used as a precision frame displacement sensor. The concept does not require a complex controller, and was realized in an off-the-shelf CNC controller. The concept was demonstrated on a linear motion system, a simplified version of a compact size CNC machine, and its servo bandwidth and dynamic stiffness were improved by 36% and 70% respectively, which are the key parameters for improving the machining accuracy. Cytoskeletal filamin A (FLNA) is an important protein involved in multiple cellular processes. Previous studies have shown that FLNA can promote or inhibit cancer growth and development; however, the mechanisms underlying these events are not fully understood. Here we show that, in both 293T and SaOS2 cells, knockdown of FLNA significantly enhanced transcription of RNA polymerase (pol) III-transcribed genes except for a subset of tRNA genes. In contrast, re-expression of FLNA in an FLNA-deficient melanoma cell line (A7) repressed transcription of all pol III-transcribed genes, suggesting that FLNA inhibits pol III transcription in a cell type-specific manner. Chromatin immunoprecipitation assays revealed that the repression of pol III gene transcription by FLNA correlates with the decreased occupancy of the RNA pol III transcription machinery at promoters. Immunofluorescence microscopy and coimmunoprecipitation assays revealed that FLNA can associate with the RNA pol III transcription machinery through its actin-binding domain within nuclei. Mechanistic analysis revealed that FLNA suppresses pol III gene transcription by confining the recruitment of the RNA pol III transcription machinery at the promoters of the genes that are sensitive to the alteration of FLNA expression. This paper presents a method for optimizing the performance of a real-time, long term, and accurate accelerometer based displacement measurement technique, with no physical reference point. The technique was applied in a system for measuring machine frame displacement. The optimizer has three objectives with the aim to minimize phase delay, gain error and sensor noise. A multi-objective genetic algorithm was used to find Pareto optimal estimator parameters. The estimator is a combination of a high pass filter and a double integrator. In order to reduce the gain and phase errors two approaches have been used: zero placement and pole-zero placement. These approaches were analysed based on noise measurement at 0g-motion and compared. Only the pole-zero placement approach met the requirements for phase delay, gain error, and sensor noise. Two validation experiments were carried out with a Pareto optimal estimator. First, long term measurements at 0g-motion with the experimental setup were carried out, which showed displacement error of 27.6 ± 2.3 nm. Second, comparisons between the estimated and laser interferometer displacement measurements of the vibrating frame were conducted.
