Traba (b) Abstract: This study presents an assessment, resulting from consultation with experts in steppe-bird conservation (scientists, officers and conservationists) using the Delphi method, of a broad range of agri-environmental measures (AEMs) which have been applied in agricultural areas in Spain, with particular reference to four threatened steppe bird species. The measures which experts have valued most highly relate to the maintenance of fallow land, the prohibition of agrochemicals and the suspension of certain agricultural practices when the species are nesting. Other AEMs which have frequently been mentioned as beneficial for steppe birds, including the maintenance of straw-mulched fallows and the abandonment of farmland, were rejected by the experts. The assessment showed a high degree of consensus between experts, although differences between the four studied species were detected. Delphi assessment indicated that different birds need different AEMs. In addition, expert evaluation showed that different AEMs can cause the same effect on the target species, which could generate an over-implementation of measures. Finally, we evaluated the financial implementation of the AEMs selected by the Delphi using a Special Protection Area for birds (SPA) in the Madrid region as a case study. All the hypothetical scenarios used yielded assumable costs, oscillating between 1 and 2 times the current AEMs expenditure. In conclusion, in extensive agrarian systems with already high conservation merits, the implementation of AEMS could be improved using species-specific assessments, thus avoiding over-implementation and improving the fit between costs and benefits for conservation. Twenty-four hours of fasting is known to blunt activation of the human NLRP3 inflammasome. This effect might be mediated by SIRT3 activation, controlling mitochondrial reactive oxygen species. To characterize the molecular underpinnings of this fasting effect, we comparatively analyzed the NLRP3 inflammasome response to nutrient deprivation in wild-type and SIRT3 knock-out mice. Consistent with previous findings for human NLRP3, prolonged fasting blunted the inflammasome in wild-type mice but not in SIRT3 knock-out mice. In SIRT3 knock-out bone marrow-derived macrophages, NLRP3 activation promoted excess cytosolic extrusion of mitochondrial DNA along with increased reactive oxygen species and reduced superoxide dismutase 2 (SOD2) activity. Interestingly, the negative regulatory effect of SIRT3 on NLRP3 was not due to transcriptional control or priming of canonical inflammasome components but, rather, occurred via SIRT3-mediated deacetylation of mitochondrial SOD2, leading to SOD2 activation. We also found that siRNA knockdown of SIRT3 or SOD2 increased NLRP3 supercomplex formation and activation. Moreover, overexpression of wild-type and constitutively active SOD2 similarly blunted inflammasome assembly and activation, effects that were abrogated by acetylation mimic-modified SOD2. Finally, in vivo administration of lipopolysaccharide increased liver injury and the levels of peritoneal macrophage cytokines, including IL-1β, in SIRT3 KO mice. These results support the emerging concept that enhancing mitochondrial resilience against damage-associated molecular patternsmayplay a pivotal role in preventing inflammation and that the anti-inflammatory effect of fasting-mimetic diets may be mediated, in part, through SIRT3-directed blunting of NLRP3 inflammasome assembly and activation. Background: Most studies reveal that the mechanism of action of propolis against bacteria is functional rather than structural and is attributed to a synergism between the compounds in the extracts.Hypothesis/purpose: Propolis is said to inhibit bacterial adherence, division, inhibition of water-insoluble glucan formation, and protein synthesis. However, it has been shown that the mechanism of action of Russian propolis ethanol extracts is structural rather than functional and may be attributed to the metals found in propolis. If the metals found in propolis are removed, cell lysis still occurs and these modified extracts may be used in the prevention of medical and biomedical implant contaminations.Study Design: The antibacterial activity of metal-free Russian propolis ethanol extracts (MFRPEE) on two biofilm forming bacteria: penicillin-resistant Staphylococcus aureus and Escherichia coli was evaluated using MTT and a Live/Dead staining technique. Toxicity studies were conducted on mouse osteoblast (MC-3T3) cells using the same viability assays.Methods: In the MTT assay, biofilms were incubated with MTT at 37°C for 30min. After washing, the purple formazan formed inside the bacterial cells was dissolved by SDS and then measured using a microplate reader by setting the detecting and reference wavelengths at 570nm and 630nm, respectively. Live and dead distributions of cells were studied by confocal laser scanning microscopy.Results: Complete biofilm inactivation was observed when biofilms were treated for 40h with 2µg/ml of MFRPEE. Results indicate that the metals present in propolis possess antibacterial activity, but do not have an essential role in the antibacterial mechanism of action. Additionally, the same concentration of metals found in propolis samples, were toxic to tissue cells. Comparable to samples with metals, metal free samples caused damage to the cell membrane structures of both bacterial species, resulting in cell lysis.Conclusion: Results suggest that the structural mechanism of action of Russian propolis ethanol extracts stem predominate from the organic compounds. Further studies revealed drastically reduced toxicity to mammalian cells when metals were removed from Russian propolis ethanol extracts, suggesting a potential for medical and biomedical applications. • Passive acoustic monitoring (PAM) can be used for monitoring human-wildlife conflicts. • One of such conflicts is that occurring between apiarists and bee-eaters. • Vocal Activity Rate (VAR) is an useful indicator of bee-eater pressure at beehives. • Bee-eater pressure was higher at early morning and evening and during bird migration. • Honeybees activity was negatively associated with VAR but varied between hours. In human-wildlife conflicts, it is crucial to develop accurate protocols for the reliable verification of the causative species and its relationship with potential damage claims. One of such conflicts is that occurring between apiarists and bee-eaters. In this work, we aim to assess the utility of passive acoustic monitoring (PAM) as an efficient methodology to measure European bee-eater (Merops apiaster) predation pressure at beehives and its impact on honeybees foraging activity. Using Autonomous Recording Units (ARUs) in apiaries, coupled to automated recognition methods for bee-eater calls identification, we found a positive relationship between Vocal Activity Rate (VAR) index and number of bee-eaters hunting attempts on honeybees. We also found that VAR varied over time, showing a lower predation pressure during midday hours and higher during the post-breeding migratory period. Honeybees flying activity was negatively associated with VAR and this relationship was conditioned by the hour of the day. Our study offers a new application of PAM and acoustic derived indices for the evaluation of potential damages caused by wildlife. We focused on the interaction between honeybees and the European bee-eater, but we expect PAM might be useful also to remotely monitor impacts to human activities produced by other vocally active species. Wind energy has experienced a notable development during the last decades, driving new challenges for animal communities. Although bird collisions with wind turbines and spatial displacement due to disturbance have been widely described in the literature, other potential impacts remain unclear. In this study, we addressed the effect of turbine noise on the vocal behaviour of a threatened shrub-steppe passerine highly dependent on acoustic communication, the Dupont's lark Chersophilus duponti. Based on directional recordings of 49 calling and singing males exposed to a gradient of turbine noise level (from 15 up to 51 dBA), we tested for differences in signal diversity, redundancy, and complexity, as well as temporal and spectral characteristics of their vocalizations (particularly the characteristic whistle). Our results unveiled that Dupont's lark males varied the vocal structure when subject to turbine noise, by increasing the probability of emitting more complex whistles (with increased number of notes) and shifting the dominant note (emphasizing the longest and higher-pitched note). In addition, males increased duration and minimum frequency of specific notes of the whistle, while repertoire size and signal redundancy remain constant. To our knowledge, this is the first study reporting multiple and complex responses on the vocal repertoire of animals exposed to turbine noise and unveiling a shift of the dominant note in response to anthropogenic noise in general. These findings suggest that the Dupont's lark exhibits some level of phenotypic plasticity, which might enable the species to cope with noisy environments, although the vocal adjustments observed might have associated costs or alter the functionality of the signal. Future wind energy projects must include fine-scale noise assessments to quantify the consequences of chronic noise exposure. It has been known for a long time that mitochondria isolated from hepatocytes treated with glucagon or Ca2+-mobilizing agents such as phenylephrine show an increase in their adenine nucleotide (AdN) content, respiratory activity, and calcium retention capacity (CRC). Here, we have studied the role of SCaMC-3/slc25a23, the mitochondrial ATP-Mg/Pi carrier present in adult mouse liver, in the control of mitochondrial AdN levels and respiration in response to Ca2+ signals as a candidate target of glucagon actions. With the use of SCaMC-3 knock-out (KO) mice, we have found that the carrier is responsible for the accumulation of AdNs in liver mitochondria in a strictly Ca2+-dependent way with an S0.5 for Ca2+ activation of 3.3 ± 0.9 μM. Accumulation of matrix AdNs allows a SCaMC-3-dependent increase in CRC. In addition, SCaMC-3-dependent accumulation of AdNs is required to acquire a fully active state 3 respiration in AdN-depleted liver mitochondria, although further accumulation of AdNs is not followed by increases in respiration. Moreover, glucagon addition to isolated hepatocytes increases oligomycinsensitive oxygen consumption and maximal respiratory rates in cells derived from wild type, but not SCaMC-3-KO mice and glucagon administration in vivo results in an increase in AdN content, state 3 respiration and CRC in liver mitochondria in wild type but not in SCaMC-3-KO mice. These results show that SCaMC-3 is required for the increase in oxidative phosphorylation observed in liver mitochondria in response to glucagon and Ca2+-mobilizing agents, possibly by allowing a Ca2+-dependent accumulation of mitochondrial AdNs and matrix Ca2+, events permissive for other glucagon actions. Abstract: We examined sexual differences in patterns of vegetation structure selection in the sexually dimorphic little bustard. Differences in vegetation structure between male, female and non-used locations during reproduction were examined and used to build a presence/absence model for each sex. Ten variables were measured in each location, extracting two PCA factors (PC1: a visibility-shelter gradient; PC2: a gradient in food availability) used as response variables in GLM explanatory models. Both factors significantly differed between female, male and control locations. Neither study site nor phenology was significant. Logistic regression was used to model male and female presence/absence. Female presence was positively associated to cover of ground by vegetation litter, as well as overall vegetation cover, and negatively to vegetation density over 30cm above ground. Male presence was positively related to litter cover and short vegetation and negatively to vegetation density over 30cm above ground. Models showed good global performance and robustness. Female microhabitat selection and distribution seems to be related to the balance between shelter and visibility for surveillance. Male microhabitat selection would be related mainly to the need of conspicuousness for courtship. Accessibility to food resources seems to be equally important for both sexes. Differences suggest ecological sexual segregation resulting from different ecological constraints. These are the first detailed results on vegetation structure selection in both male and female little bustards, and are useful in designing management measures addressing vegetation structure irrespective of landscape composition. Assessment using entrustable professional activities (EPAs) is currently under study as a joint effort of the American Board of Pediatrics and program director community. However, limited evidence exists for the feasibility of EPA-based assessment in pediatrics. Among pediatric residency programs that implemented EPA-based assessment over 3 academic years (2015-16, 2016-17, 2017-18), determine how often programs were "unable to assess" a resident on a given EPA. 23 programs reported clinical competency committee (CCC) determined EPA supervision level assignments for all residents in their program for a subset of 5-6 of the 17 general pediatrics EPAs biannually (Fall and Spring), except interns who were only assessed at the end of the academic year (Spring). They indicated "unable to assess" whenever applicable. Across the 5 data collection cycles, there were 29,643 opportunities to assign an EPA supervision level to 1987 residents from all three post-graduate years. Of these, 4140 assignments (14%) were deemed "unable to assess." Across all EPAs, this determination was made less often in mid-year reports than year-end reports (p=.005). Post-graduate year 3 (PGY3) residents and PGY2 residents had significantly lower frequencies of "unable to assess" assignments when compared with PGY1 residents (Figure). EPAs with the lowest rates of "unable to assess" included: EPA 4 (patients with acute, common dx) at 0.4%, EPAs 10 (initial resuscitation/stabilization) and 16 (handovers) at 0.9%, EPA 11 (manage information) at 4%, and EPA 13 (practice management) at 5%. EPAs with the highest rates of "unable to assess" included EPA 8 (transition to adult care) at 43%, EPA 17 (procedures) at 27%, EPA 14 (quality improvement) at 20%, and EPAs 12 (make referrals) and 9 (behavioral and mental health) at 14%. "Unable to assess" assignments are related to the given EPA, training year, and timing of assessment within the academic year. CCCs may be less likely to assign EPA supervision levels at timepoints viewed as higher stakes, such as transition from one training year to the next. Abstract: Seeds of the Mediterranean Hackberry Celtis australis are often encountered in fox faeces. In order to evaluate the effect of gut transit on the size of seeds selected, the rates and speed of germination and on the survival of the seedlings, Mediterranean Hackberry seeds from fox faeces were germinated in a greenhouse. The results were compared with those of seeds taken from ripe, uneaten fruits. Fox-dispersed seeds were smaller and lighter than the control ones and had higher (74% vs. 57%) and more rapid germination (74.5 days vs. 99.2 days). Seedlings from fox-dispersed seeds showed significantly greater survival by the end of the study period (74.1% vs. 43.6%) than the control ones. Survival in seedlings from fox-dispersed seeds was related to germination date, late seedlings showing poorer survival. This relationship was not observed away in the control seedlings. Seed mass did not affect seedling survival. Seedling arising from fox-dispersed seeds grew faster than control ones. These results suggest that fox can play a relevant role as seed disperser of Mediterranean Hackberry. [Copyright &y& Elsevier]

The eyelid movements are mediated mainly by the orbicularis oculi (OO) and the levator palpebrae superioris (LPS) muscles. Dissociated upper lid functions exhibit different counterbalanced action of these muscles, and in blinking they show a strictly reciprocal innervation. The disturbance of this close LPS–OO relationship likely leads to many of the central lid movement disorders. Three groups of supranuclear motor impairment of lid movements are considered: the disorders of the lid-eye movements’ coordination, the disturbances of blinking and lid “postural” maintenance, and the alteration of voluntary lid movements. Nuclei of the posterior commissure control the inhibitory modulation of LPS motor-neuronal activity and they are involved in the lid-eye coordination disorders such as lid retraction, which is observed in the Parinaud’s syndrome and also in parkinsonism and progressive supranuclear palsy. Spontaneous (SB) and reflex blinking consist of two components: the inhibition of the basal tonic LPS activity, which keeps the eyes open, and the concurrent activation of the OO muscles. LPS inhibition precedes and outlasts the OO activation. This normal configuration is impaired in parkinsonism and blepharospasm (BSP). SB shows a highly interindividual rate variation (among 10–20 per minute in adults) and abnormal blink rates occur in neurological diseases related to dopaminergic transmission impairments. Lid postural abnormalities include involuntary eyelid closure, which is usually associated with inability to open the eyes. Two major disorders share these two aspects: BSP and blepharocolysis (BCO). BSP consists of an involuntary overactivity of the OO, with LPS co-contraction activity, and is expressed as frequent and prolonged blinks, clonic bursts, prolonged tonic contraction or a blend of all of them. BCO (commonly named “so-called lid opening apraxia”) is an overinhibition of the LPS with no evidence of ongoing OO activity. BSP and BCO occur in many instances of idiopathic dystonias and basal ganglia diseases and, less frequently, in rostral brainstem lesions. Both may coincide in the same patient. Voluntary lid movement disorders comprise the impairment of Bell’s phenomenon, the voluntary eyelid closure palsy and the so-called cerebral ptosis, all related to lesions of frontal cortical areas and/or the corticospinal system. Increasing food security and preventing further loss of biodiversity are two of humanity's most pressing challenges. Yet, efforts to address these challenges often lead to situations of conflict between the interests of agricultural production and those of biodiversity conservation. Here, we focus on conflicts between livestock production and the conservation of wild herbivores, which have received little attention in the scientific literature. We identify four key socio-ecological challenges underlying such conflicts, which we illustrate using a range of case studies. We argue that addressing these challenges will require the implementation of co-management approaches that promote the participation of relevant stakeholders in processes of ecological monitoring, impact assessment, decision-making, and active knowledge sharing. Conflicts between the interests of livestock production and those of wild herbivore conservation are an increasing global challenge. Addressing these conflicts is hindered by a poor understanding of key underlying social and ecological drivers. In particular, there is a need to reconcile the real and perceived costs–benefits of livestock–wild herbivore interactions. This includes better understanding how these interactions vary across migration ranges and are influenced by trophic network structure. In many cases, these conflicts are perpetuated by a disconnect between livestock husbandry practices and scientific research on sustainable management. Addressing these conflicts will require the development of reliable protocols for impact assessment and the implementation of participatory processes that bring together relevant stakeholders.
