When nanomaterials with antibacterial properties were sent to the infected area, it was predicted that infection and related complications could be prevented. The nanoparticles can be designed to possess magnetic and luminescence (magneto-luminescent) properties to be effectively targeted and localized at the infection foci without dispersing into the body. Simultaneously, the magneto-luminescent characteristic of particles allows visualization and confirmation of localized particles at the desired area. In this regard, there are no studies on the use of antibacterial magneto-luminescent bioactive glass for orthopedic applications and the treatment of orthopedic device-related infections. In this study, antibacterial magneto-luminescent 58S bioactive glasses were synthesized by the modified Stöber using coupled with a layer-by-layer assembly approach to possess core/shell particle morphology. SPION/Bioactive glass nanoparticles had an average size of 50 nm and displayed superparamagnetic behavior. While the saturation magnetization value (σ s) of the undoped 58S sample was 25.32 emu/g, that of the co-doped sample (2% Eu, 2% Zn) was 21.74 emu/g; this showed that the doping slightly reduced the magnetization value. Europium (Eu) doping of SPION/Bioactive glass nanoparticles induced characteristic red emission originating from Eu emissions belonging to 5D 0 –7F J (J = 1–4) transitions and the strongest peak was at 612 nm (electric-dipole transition, 5D 0 –7F 2). Color chromaticity coordinates confirmed emission in the red region. XPS spectrum revealed the existence of Eu and Zn dopant elements in 58S bioactive glass. After soaking characteristic peaks at 31.74° and 45.43° belonging to the hexagonal hydroxyapatite phase were detected in the XRD data, confirming the SEM images. 2% Eu doped SPION/Bioactive glass nanoparticles had the highest osteoblast viability up to 7 days in vitro , while doping the samples with 2% zinc did not yield bone cell viability as high as the Eu doped ones. Importantly, Eu doped SPION/Bioactive glass nanoparticles inhibited gram-positive Staphylococcus aureus (S. aureus) and gram-negative Escherichia coli (E. coli) growth up to 48 h in vitro. The results showed that Eu doping of SPION/Bioactive glass nanoparticles increased osteoblast viability and inhibited bacterial growth, while possessing superparamagnetic properties and exhibiting red luminescence. A straightforward wet chemical method has been applied for the fabrication of Yb 2 O 3 nanoparticles (NPs) from ytterbium nitrate solution by using ammonium carbonate as precipitation agent. Effects of precursor molarity (0.1, 0.15 and 0.2 M), calcination temperature (800, 900 and 1000 °C) and time (2, 4 and 6 h) on average crystallite size (CS) of the NPs were statistically investigated by using Box-Behnken design. A simple and effective quadratic model was proposed for controlling the CS. The CS values were calculated by using Williamson Hall method (W-H) from X-Ray Diffraction (XRD) broadening data and found to be ranging between 13 and 26 nm. Agglomerated NPs morphologies and particle sizes were revealed by Field Emission Gun-Scanning Electron Microscopy (FEG-SEM). Fourier Transform Infrared (FTIR) spectrophotometry confirmed that as-received Yb 2 (CO 3) 3 xH 2 O powders were successfully transformed into Yb 2 O 3 NPs with calcination. High-Resolution Transmission Electron Microscopy (HRTEM) results verified the average CS values. ANOVA analyses revealed that linear and squared terms of the production parameters were significantly related to the CS whereas interaction terms were insignificant with the confidence level of 95% (R2 = 92.67%, R2-adj = 87.17%). The calcination temperature had the highest impact on the average CS followed by the time and precursor molarity. Increasing calcination parameters resulted in bigger crystallites whereas increasing precursor molarity exhibited a critical supersaturation value (0.15 M) from which the average CS was decreased. In this study, Y 2 O 3 :Eu3+ phosphorescent powders with different dopant rates were synthesized by sol-gel method. Statistical experimental design methods were used to investigate the effects of calcination temperature (800, 900 and 1200 °C), time (5 and 7 h) and dopant rate (5 and 7% at.) on the colour of the photoluminescence emission. The XRD results showed that as-prepared powders were amorphous. After calcination, the crystal structures became cubic bixbyite-type. XRD and Raman spectroscopy results confirmed the existence of Eu3+ in the Y 2 O 3 crystal. Crystallite sizes and lattice parameters of the powders were calculated by using Williamson-Hall and Cohen-Wagner methods, respectively. Photoluminescence emission spectra of the samples were measured at room temperature by using an optical emission spectroscope with 380 nm monochromatic excitation source and the emission spectra were transformed to CIELab colour space coordinates. The emission spectra exhibited a strong characteristic peak at 611 nm and a smaller peak at 614 nm. Depending on the dopant rate and the calcination parameters, other smaller peaks appeared at 587, 592, 606, 623 and 631 nm. The CIELab colour space transformation results showed that the powders possessed orange-red emission. The dopant rate and the calcination conditions slightly changed the emission colour. According to the statistical analysis, the dopant rate had the strongest effect on a* colour space value (redness) whereas all parameters had equally significant effect on b* colour space value (yellowness). The ANOVA analysis revealed that the interaction between the temperature and the dopant rate was also statistically significant with the confidence level of 95%. This interaction was attributed to the relation between lattice strain and calcination temperature, which confirms the strong symmetry dependence of the electron-dipole (ED) transitions of the Y 2 O 3 :Eu3+ phosphors. The incidence of chronic kidney disease (CKD) is increasing rapidly. Diabetes mellitus (DM) is the most important cause of CKD. We studied the possible role of DM in CKD patients with respect to DNA damage, as assessed by the comet assay in 60 CKD patients (with or without DM) undergoing hemodialysis and in 26 controls. Effects of other factors, such as age, sex, hypertension, duration of hemodialysis, body mass index (BMI), and levels of hemoglobin (HB), intact parathormone (iPTH), and ferritin (FER), were also examined. Primary DNA damage measured by the comet assay was significantly higher in CKD patients than in controls. Among CKD patients, the following correlations were observed. (1) There was no difference in comet tail length or tail intensity between diabetic and non-diabetic individuals. (2) Age, sex, hemoglobin, hypertension, duration of hemodialysis, and ferritin levels affected neither tail length nor intensity. (3) BMI values above 25 kg/m 2 and iPTH levels above 300 pg/ml were associated with significantly greater comet tail length. Our results indicate that primary DNA damage is increased in CKD patients undergoing hemodialysis, compared to controls; however, DM had no additional effect. [ABSTRACT FROM AUTHOR]

Due to high operating temperature (800–1000 °C) of SOFC's, limited number of alloys can be employed for the purpose of current collection. The electrical conductivity of these alloys usually decreases during the operation of SOFC because of highly corrosive cathode (air side) environment. Cr evaporation from such alloys is also an important problem of performance degradation. A spinel Mn 1.5 Co 1.5 O 4 and perovskite La 0.60 Sr 0.40 FeO 3 mixture is coated on Crofer alloy for corrosion and chromium protection in the cathode side of a SOFC. In study firstly, the performance of the fuel cell with the Crofer 22 APU as interconnect material was measured up to 100 h without any coating. Secondly, using a screen printing method, bare Crofer 22 APU was coated with Mn 1.5 Co 1.5 O 4 and finally Mn 1.5 Co 1.5 O 4 + La 0.60 Sr 0.40 FeO 3 ceramic powder slurries were employed as the coating material. Then, the performances of a SOFC short stack with interconnects coated by two methods were compared with uncoated interconnect. Results show that uncoated interconnect experienced a significant performance lost from 4.5 W to 3.8 W. On the other hand the cells with screen printing coated with a mixture Mn 1.5 Co 1.5 O 4 and La 0.60 Sr 0.40 FeO 3 exhibited decreases only from 5.78 W to 5.42 W in the short-term performance tests. Endemic Muscari muscarimi Medikus is the most fragrant plant among Muscari species and has a high ornamental potential. The natural populations of M. muscarimi , are severely affected by increased environmental pollution and urbanization. There is a need to develop a micropropagation method that should serve effectively for commercial propagation and conservation. Therefore, the study targeted to set up a strategy for efficient in vitro bulblet regeneration system of M. muscarimi using twin scale bulb explants on 1.0 × MS medium containing 4.44, 8.88, 17.76 μM BAP (6-Benzylaminopurine) plus 2.685, 5.37, 10.74 μM NAA (α-Naphthalene acetic acid). Maximum number of 19 daughter axillary bulblets and 16 daughter adventitious bulblets per twin bulb scale explant was regenerated on 1.0 × MS medium containing 17.76 μM BAP plus 10.74 μM NAA and 17.76 μM BAP plus 2.685 μM NAA respectively. The daughter bulblets regenerated on twin bulb scales on 8 out of 9 regeneration treatment could be easily rooted on 1.0 × MS medium containing 4.9 μM IBA (Indole-3-butyric acid). The daughter bulblets regenerated on 9th treatment (1.0 × MS medium containing 17.76 μM BAP plus 10.74 μM NAA) were transferred to 1.0 × MS medium containing 30 g/l sucrose to break negative carry over effect of this dose of BAP–NAA, where they grew 2–3 roots of variable length. Daughter bulblet diameter was increased by culturing them on 1.0 × MS medium containing 4.44 μM BAP plus 5.37 μM NAA. The results verified that both age and the source of explants had significant effect on regeneration. In another set of experiments, twin scales were obtained from in vitro regenerated daughter bulblets, although they induced bulblets, yet their bulblet regeneration percentage, mean number of bulblets per explant and their diameter were significantly reduced. In vitro regenerated bulblets were acclimatized in growth chamber under ambient conditions of temperature and humidity on peat moss, where they flowered. The study provides important information about selection of suitable micropropagation medium, strategies to improve bulblet diameter and rooting of M. muscarimi which offers a scope for commercial propagation. Cynarin is an artichoke phytochemical that possesses a variety of pharmacological features including free-radical scavenging and antioxidant activity. The origin of artichoke species appears to be Mediterranean region. Two of these species, globe artichoke (Cynara cardunculus var. scolymus L.) and cardoon (Cynara cardunculus var. altilis DC), are widely cultivated and consumed. This vegetable, as the basis of the mediterranean diet, has been used as herbal medicine for its therapeutic effects since ancient times. Therefore, this study was performed to determine genotoxic and antigenotoxic effects of cynarin against MMC (mitomycin C) and H 2 O 2 (hydrogen peroxide) induced genomic instability using chromosome aberrations (CAs), sister chromatid exchanges (SCEs), micronucleus (MN), and comet assays in human lymphocytes. Lymphocytes obtained from two healthy volunteers (1 male and 1 female) were exposed to different concentrations of cynarin (12–194 μM) alone and the combination of cynarin and MMC (0.60 μM) or cynarin and H 2 O 2 (100 μM, only for comet assay). Cynarin alone did not induce significant genotoxic effect in the CA, SCE (except 194 μM), MN, and comet assays. The combination of some concentrations of cynarin and MMC decreased the frequency of CAs, SCEs and MN induced by MMC. Furthermore, the combination of cynarin and H 2 O 2 reduced all comet parameters at all the concentrations compared to H 2 O 2 alone. While the highest concentrations of cynarin significantly decreased mitotic index (MI), the combination of cynarin and MMC increased the reduction of MI induced by MMC alone. All the results obtained in this study demonstrated that cynarin exhibited antigenotoxic effects rather than genotoxic effects. Sitagliptin is an active ingredient of antidiabetic drug used in the treatment of type 2 diabetes mellitus (T2DM). In this study, the genotoxic effects of sitagliptin were determined in human lymphocytes by using chromosome aberrations (CAs), sister chromatid exchanges (SCEs), micronucleus (MN) and comet assays. 31.25–1000 μg/mL concentrations of sitagliptin were used. Sitagliptin significantly increased the frequency of CAs and SCEs at the highest concentration at 24 h treatment and all concentrations (except 250 μg/mL for CA, except 31.25 and 62.50 μg/mL for SCE) at 48 h treatment compared with solvent control (DMSO). This compound increased the MN at only the highest concentration compared with the solvent control. Mitotic index (MI) significantly decreased at the three highest concentrations of sitagliptin at 48 h treatment. However, replication (RI) and nuclear division (NDI) indices were not affected at all the treatments. Comet assay results indicated that sitagliptin significantly increased mean comet tail intensity and tail moment at only two concentrations (62.50 and 1000 μg/mL for intensity, 125 and 1000 μg/mL for tail moment), and tail length at all concentrations (except 125 and 500 μg/mL). Food additives are approved chemicals used for various purposes in foods; to provide nutritional safety, increase flavor, extend shelf life, reduce nutrient losses etc. In this study, the in vitro genotoxic effects of flavor enhancers, Monopotassium glutamate (MPG) and Magnesium diglutamate (MDG) were investigated in human peripheral blood lymphocytes by using chromosome aberrations (CAs), sister chromatid exchanges (SCEs), cytokinesis-block micronucleus cytome (CBMN-Cyt), and comet assays. Four concentrations of MPG (125, 250, 500, and 1000 μg/mL) and MDG (93.75, 187.5, 375, and 750 μg/mL) were used. Both food additives significantly reduced mitotic index and increased the frequency of CAs at high concentrations. MPG and MDG (except 93.75 μg/mL) significantly increased SCEs/Cell in concentration-dependent manner. In the CBMN-Cyt test, both MPG and MDG increased the formation of micronucleus, nuclear buds, and nucleoplasmic bridges compared to control in a concentration-dependent manner. However, these increases were statistically significant at higher concentrations. MPG (at 500 and 1000 μg/mL) and MDG (except 93.75 μg/mL) significantly increased DNA damages observed by comet assay. Chronic kidney disease (CKD) is a common health problem. The primary etiology of CKD is diabetes mellitus (DM). The aim of our study is to determine the possible role of DM and also effects of other factors such as hypertension, duration of hemodialysis (HD), age, sex, body mass index (BMI), and levels of hemoglobin (HB), intact parathormone (iPTH), and ferritin on genetic alterations in maintenance HD patients using chromosomal aberrations (CAs), sister chromatid exchanges (SCEs), and micronucleus (MN) tests. According to the results, the frequency of CAs (p = 0.001), SCEs (p < 0.001) and MN (p < 0.001) statistically increased in HD patients compared to controls. However, there was no significant effect of diabetes as well as other factors on CA, SCE (except at factor of age), and MN in HD patients compared to controls. The mitotic (MI), replication (RI) and nuclear division indices (NDI) significantly decreased in HD patients compared to controls (p < 0.001). In addition, RI (p < 0.001) and NDI (p = 0.047) were significantly decreased in diabetic HD patients than the non-diabetic HD patients. There was no relation between the frequency of CA, SCE and MN and duration of HD treatment with correlation analysis. According to univariate regression analyses, only having CKD was significantly associated with the values of CA, SCE and MN. However, in multivariate analyses, only having CKD remained as significantly associated with CA, SCE and MN values. Hyperoside is a flavonol glycoside isolated from various plant genera such as Hypericum and Crataegus. It has an important place in the human diet and is used medically to relieve pain and ameliorate cardiovascular functions. However, a comprehensive profile of the genotoxic and antigenotoxic effects of hyperoside is not known. The current study aimed to investigate the genotoxic and antigenotoxic effects of hyperoside against genetic damages induced by two genotoxins (MMC and H 2 O 2) using chromosomal aberrations (CAs), sister chromatid exchanges (SCEs), and micronucleus (MN) assays in human peripheral blood lymphocytes in vitro. Blood lymphocytes were incubated with 7.8–62.5 μg/mL concentrations of hyperoside alone and simultaneously with 0.20 μg/mL Mitomycin C (MMC) or 100 μM Hydrogen peroxide (H 2 O 2). Hyperoside did not exhibit genotoxic potential in the CA, SCE, and MN assays. Moreover, it did not cause a decrease in mitotic index (MI) which is an indicator of cytotoxicity. On the other hand, hyperoside significantly decreased CA, SCE, and MN (except for MMC treatment) frequencies induced by MMC and H 2 O 2. Hyperoside, increased mitotic index against both mutagenic agents at 24-h treatment when compared to positive control. Our results demonstrate that hyperoside exhibited antigenotoxic effects rather than genotoxic in vitro human lymphocytes. Therefore, hyperoside may be a potential preventive agent in inhibiting chromosomal and oxidative damage induced by genotoxic chemicals. Exopolysaccharide isolated from Lactobacillus salivarius (new genus name Ligilactobacillus) KC27L strain (EPS KC27L) exhibits antioxidant properties with 1,1-diphenyl-2-picrylhydrazase (DPPH) radical and superoxide anion radical (O 2 -.) scavenging effect and iron ion (Fe2+) chelating activity. This study aimed to investigate the in vitro genotoxic effects of EPS KC27L alone (12.50, 25.00, 50.00, and 100.00 μg/mL) and its antigenotoxic activity against DNA damage induced by mitomycin-C (MMC; 0.20 μg/mL), methyl methanesulfonate (MMS; 5.00 μg/mL), and hydrogen peroxide (H 2 O 2 ; 100 μM). For this purpose, chromosome aberration (CA), sister chromatid exchange (SCE), micronucleus (MN), and comet assays were performed in human peripheral lymphocytes. In addition, the structure of EPS KC27L was investigated in the scanning electron microscope (SEM). EPS KC27L alone did not cause a significant genotoxic effect in CA, SCE, MN, and comet tests. EPS KC27L significantly decreased the frequency of CA, SCE, and MN induced by MMC and MMS. EPS KC27L also significantly reduced DNA damage induced by H 2 O 2. This study showed that the EPS KC27L alone has no genotoxic risk at these concentrations and shows antigenotoxic activity against MMC, MMS, and H 2 O 2. • Pullulan is not genotoxic in human lymphocytes. • It revealed antigenotoxic effect against Mitomycin C and Hydrogen Peroxide. • It induces cytotoxic effect in human breast cancer (MCF-7) cells. Pullulan is a biocompatible and water-soluble exo-polysaccharide produced by primary strains of the fungus Aureobasidium pullulans. It is frequently used in the pharmaceutical and food industries. In this study, possible cytotoxic effect of pullulan was assessed using the MTT assay in the human breast cancer (MCF-7) cell line. Micronucleus (MN), micronucleus-FISH (MN-FISH), random amplified polymorphic DNA (RAPD-PCR), and comet assays were used to investigate genotoxic and antigenotoxic effects of pullulan against mitomycin C (MMC) (at MN assay) and hydrogen peroxide (at comet assay) in human lymphocytes. Antigenotoxicity was determined using two different applications: 1 h pretreatment and simultaneous treatment. In the MTT assay, pullulan significantly reduced the cell viability at 15.6−2000 μg/mL compared to the control. No significant alterations in MN rates were found in human lymphocytes treated with different concentrations of pullulan compared to the control. In contrast, co-treatment of pullulan and MMC decreased the frequency of MN in almost all the treatment concentrations and durations compared to the MMC. No significant change was observed in the frequency of the centromere-positive C + or negative C- MNi compared to the positive control. In comet assay, pullulan did not affect comet tail intensity compared to the negative control. On the contrary, pullulan in combination with H 2 O 2 significantly decreased tail intensity at almost all the concentrations compared to the positive control. The changes occurring in RAPD-PCR profiles following pullulan treatments included an increase or decrease in band intensity and gain or loss of bands. These results indicate that exopolysaccharide Pullulan is not genotoxic; moreover, it possesses a protective effect against MMC and H 2 O 2 induced genotoxicity.
