Abstract

Lacticaseibacillus rhamnosus Lcr35 is a well-known bacterial strain whose efficiency in preventing recurrent vulvovaginal candidiasis has been largely demonstrated in clinical trials. The presence of sodium thiosulfate (STS) has been shown to enhance its ability to inhibit the growth of Candida albicans strains. In this study, we confirmed that Lcr35 has a fungicidal effect not only on the planktonic form of C. albicans but also on other life forms such as hypha and biofilm. Transcriptomic analysis showed that the presence of C. albicans induced a metabolic adaptation of Lcr35 potentially associated with a competitive advantage over yeast cells. However, STS alone had no impact on the global gene expression of Lcr35, which is not in favor of the involvement of an enzymatic transformation of STS. Comparative HPLC and gas chromatography-mass spectrometry analysis of the organic phase from cell-free supernatant (CFS) fractions obtained from Lcr35 cultures performed in the presence and absence of STS identified elemental sulfur (S0) in the samples initially containing STS. In addition, the anti- C andida activity of CFS from STS-containing cultures was shown to be pH-dependent and occurred at acidic pH lower than 5. We next investigated the antifungal activity of lactic acid and acetic acid, the two main organic acids produced by lactobacilli. The two molecules affected the viability of C. albicans but only at pH 3.5 and in a dose-dependent manner, an antifungal effect that was enhanced in samples containing STS in which the thiosulfate was decomposed into S0. In conclusion, the use of STS as an excipient in the manufacturing process of Lcr35 exerted a dual action since the production of organic acids by Lcr35 facilitates the decomposition of thiosulfate into S0, thereby enhancing the bacteria's own anti-fungal effect. Abstract

Abstract: Infection with Shiga-toxin producing Escherichia coli (STEC) may result in the development of the haemolytic–uremic syndrome (HUS), the main cause of acute renal failure in children. While O157:H7 STEC are associated with large outbreaks of HUS, it is difficult to predict whether a non-O157:H7 isolate can be pathogenic for humans. The mucosal innate immune response plays a central role in the pathogenesis of HUS; therefore, we compared the induction of IL-8 and CCL20 in human colon epithelial cells infected with strains belonging to different serotypes, isolated from cattle or from HUS patients. No correlation was observed between strain virulence and chemokine gene expression. Rather, the genetic background of the strains seems to determine the chemokine gene expression profile. Investigating the contribution of different bacterial factors in this process, we show that the type III secretion system of O157:H7 bacteria, but not the intimate adhesion, is required to stimulate the cells. In addition, H7, H10, and H21 flagellins are potent inducers of chemokine gene expression when synthesized in large amount. [Copyright &y& Elsevier]

Abstract

This paper presents results obtained on barium ferrite thick films prepared by electron beam evaporation. First of all, we have looked at the influence of substrate temperature on the films properties. Then, we have fixed the substrate temperature to 700 °C and the effects of films thickness on the different properties has been investigated. Indeed for a substrate temperature of 700 °C we have observed that the layers are crystallized in the BaM phase with in-plane preferential orientation. However, a secondary non magnetic phase (BaFe2O4), which can modify the magnetic properties, appears on some layers. [Copyright &y& Elsevier]

Abstract

The mechanical properties of spaghetti were determined as a function of water content at different temperatures (20, 40, 60, or 80 °C). The mechanical properties of pasta were evaluated by an empirical test and characterized by the apparent strength and apparent relaxation coefficients. The fresh spaghetti (at 45% water db) behave as a visco-plastic and soft product. The dry spaghetti (at 10% water db) behave as an elastic and rigid product. An abrupt mechanical transition separates the fresh state and the dry state. This transition depends either from water content and temperature of pasta. A sigmoid model was used to describe this transition as a function of moisture content and/or temperature. The transition of mechanical properties for spaghetti was nearly superimposed with the glass transition temperature of wheat durum semolina. An apparent “time–temperature–water content” superposition showed that an increase in temperature or in water content induces a plasticizing effect on mechanical properties of spaghetti.
