Epilepsy is a neurodegenerative condition characterized by uncontrollable convulsions caused by a misalignment of the central nervous system's inhibitory and excitatory branches. Vateria indica is a medicinal herb with anti-inflammatory, anthelmintic, antiulcer, antitumor, and anticancer properties. To investigate the antiepileptic activity of Vateria indica using maximal electrical shock (MES), pentylenetetrazole (PTZ), and isoniazid (INH) induced experimental animal models. Vateria indica bark was subjected to Soxhlet extraction using ethanol and quantitative and qualitative analysis was performed. The antiepileptic activity of Vateria indica bark extract (VIE) was investigated using different animal models in mice. GABA levels in the brain and antioxidant capacity in vitro were estimated. Treatment of mice with VIE significantly reversed the MES-induced convulsions, which was reflected by the decrease in the duration (sec) of all the phases of MES-induced convulsions, with an increment in the GABA levels. In the PTZ and INH models, pretreatment with VIE delayed the latency to clonic convulsions (p 0.001), reduced the intensity and duration of clonic convulsions, and reduced the mortality rate in the treatment groups in a dose-dependent manner. VIE intervention dose-dependently restored brain GABA levels. VIE also exhibited significant in-vitro antioxidant activity. Overall, the findings imply that Vateria indica has substantial antiepileptic activities, mediated by positive GABAergic neurotransmission and antioxidant capabilities. To summarize, Vateria indica may provide adequate protection against epileptic seizures, suggesting that it could be used to treat petitmal and grandmal epilepsy. We plan to provide pure lead compounds derived from Vateria indica in the future in order to better understand the role it could play in the development of natural anticonvulsant drugs. The adsorption properties of 2,4-Dichlorophenoxyacetic acid (2,4-D) onto biochar, obtained through HCl-assisted hydrothermal carbonization process of Vateria indica fruits (VI-BC), were extensively studied using traditional and statistical physics approaches. The traditional adsorption investigations encompassed kinetics, equilibrium, and thermodynamics studies. Subsequently, the Hill statistical physics model was employed to interpret the mechanism. Also, artificial neural network (ANN) and adaptive neuro-fuzzy inference system (ANFIS) machine learning tools were successfully employed to model the adsorption data wherein both models had high prediction potential (R2 > 0.99). The outcomes demonstrated that the produced VI-BC exhibited remarkable adsorptive traits, having a considerable specific surface area (111.54 m2/g), pore size (5.89 nm), a variety of functional groups, and appropriate attributes for efficiently adsorbing 2,4-D. For 10 mg/L 2,4-D, at pH 2.0 and with 0.3 g/L dose, an impressive 91.67% adsorption efficiency was achieved within a 120-min. Pseudo-second-order model aptly depicted the kinetic behavior of 2,4-D adsorption, while the Freundlich model provided a more accurate representation of the isotherms. 2,4-D maximum adsorption capacity stood at 131.39 mg/g at 303 K. The Hill statistical physics model elucidated that the adsorption primarily occurred via physisorption mechanisms, involving electrostatic attractions, π-π conjugation, and pore filling. This conclusion was further substantiated by post-adsorption characterization of the VI-BC. Thermodynamic analysis indicated that the interactions between VI-BC and 2,4-D were favorable, spontaneous, and exothermic. The calculated low energy of adsorption (1.255 kJ/mol) and ΔH° value (−20.49 kJ/mol) further supported physisorption as the dominant mechanism. In summary, this study underscores the significant potential of the newly developed biochar as a promising alternative material for efficiently removing the 2,4-D herbicide from polluted environments. [Display omitted] Human triple-negative breast cancer (TNBC) being an aggressive cancer type accounts for about 15–20% of global breast cancer cases. In the present study, the cytotoxicity of pure silver (AgVI) and silver/zinc oxide (Ag/ZnOVI) nanostructures was evaluated against the TNBC cells. The nanostructures synthesized from a green route using Vateria indica (L.) fruit extract were characterized to scrutinize their formation, crystal phase, size, shape, and surface properties via FTIR, PXRD, FE-SEM coupled with EDS spectroscopy, and BET analysis. The results of the studies have unveiled the formation of 26.43 nm and 20.97 nm sized AgVI and Ag/ZnOVI nanostructures in their purest form. The in-vitro anticancer study performed on human TNBC cells [MDA-MB468] revealed the enhancement in the antiproliferative potentiality of bimetallic Ag/ZnOVI nanostructures from 66.99 ± 0.13 to 79.73 ± 0.23 in comparison to pure AgVI nanostructures. In addition to this, the greenish yellow-fluorescence observed in the TNBC nuclei during the AO-EB staining study manifested the early apoptosis. Furthermore, the anti-inflammatory and cytotoxicity study performed on the human RBC and normal NIH3T3 murine fibroblasts cells proved the biocompatibility and non-toxic nature of the synthesized nanostructures with membrane stabilization percentage up to 94.5 ± 0.001. Additionally, the antioxidant and antidiabetic studies carried out have corroborated the radical scavenging and α-amylase inhibition capability up to 85.87 ± 0.001 and 89.60 ± 0.002 % respectively. Thus the overall results of the study substantiate the superlative antioxidant, antidiabetic, and antiproliferative property of green synthesized AgVI and Ag/ZnOVI nanostructures with excellent biocompatibility. Two new stilbenoids, vateriaphenols A (1) and B (2), were isolated from the stem bark of Vateria indica along with known 10 stilbenoids (3–12) and bergenin (13). The structures of isolates were established based on spectroscopic analysis. The structures of vateriaphenols A and B were characterized as an octamer and a tetramer of resveratrol, respectively. The spectral properties of the highly condensed vateriaphenol A were also discussed. [Copyright &y& Elsevier]
