Hydrogels are widely used in nerve tissue repair and show good histocompatibility. There remain, however, challenges with hydrogels for applications related to neural signal recording, which requires a tissue-like biomechanical property, high optical transmission, and low impedance. Here, we describe a transparent hydrogel that is highly biocompatible and has a low Young's modulus (0.15 MPa). Additionally, it functions well as an implantable electrode, as it conformably adheres to brain tissue, results in minimal inflammation and has a low impedance of 150 Ω at 1 kHz. Its high transmittance, corresponding to 93.35% at a wavelength of 300 nm to 1100 nm, supports its application in two-photon imaging. Consistent with these properties, this flexible multimodal transparent electrophysiological hydrogel (MTEHy) electrode was able to record neuronal Ca2+ activity using miniature two-photon microscopy. It also used to monitor electrocorticogram (ECoG) activity in real time in freely moving mice. Moreover, its compatibility with magnetic resonance imaging (MRI), indicates that MTEHy is a new tool for studying activity in the cerebral cortex. Future brain science research requires better-performing implantable electrodes to detect neuronal signaling in the brain. In this study, we developed a new hydrogel material, MTEHy-3, that shows high biocompatibility, high optical transmittance (93.35%) and a low Young's modulus (0.15 MPa). Using as high-biocompatible metal-free hydrogel electrode, MTEHy-3 can be implanted for a long time to study the cerebral cortex, and synchronously record the Ca2+ signaling activity of individual neurons and monitor electrocorticogram activity through ionic conduction in freely moving mice. At the same time, non-metallic MTEHy-3 is also suitable for magnetic resonance imaging. • Luteolin inhibits proliferation and invasion of glioma stem cells. • The function of glioma stem cells to promote macrophage polarization was inhibited by luteolin. • Luteolin weakened the support effect of TAMs on glioma stem cells and reshaped the immune microenvironment. Glioblastoma is the most malignant and prevalent primary human brain tumor, and the immunological microenvironment controlled by glioma stem cells is one of the essential elements contributing to its malignancy. The use of medications to ameliorate the tumor microenvironment may give a new approach for glioma treatment. Glioma stem cells were separated from clinical patient-derived glioma samples for molecular research. Other studies, including CCK8, EdU, Transwell, and others, supported luteolin's ability to treat glioma progenitor cells. Network pharmacology and molecular docking models were used to study the drug target, and qRT-PCR, WB, and IF were used to evaluate the molecular mechanism. Intracranial xenografts were examined using HE and IHC, while macrophage polarization was examined using FC. We originally discovered that luteolin inhibits glioma stem cells. IL6 released by glioma stem cells is blocked during medication action and inhibits glioma stem cell proliferation and invasion via the IL6/STAT3 signaling pathway. Additionally, luteolin inhibits the secretion of TGFβ1, affects the polarization function of macrophages in the microenvironment, inhibits the polarization of M2 macrophages in TAM, and further inhibits various functions of glioma stem cells by affecting the IL6/STAT3 signaling pathway, luteolin crosstalk TGFβ1/SMAD3 signaling pathway, and so on. Through the suppression of the immunological microenvironment and inhibition of the IL6/STAT3 signaling pathway, our study determined the inhibitory effect of luteolin on glioma stem cells. This medication's dual inhibitory action, which has a significant negative impact on the glioma stem cells' malignant process, makes it both a viable anti-glioma medication and a candidate for targeted glioma microenvironment therapy. The utilization of Chinese medicine as an adjunctive therapy for cancer has recently gained significant attention. Ferroptosis, a newly regulated cell death process depending on the ferrous ions, has been proved to be participated in glioma stem cells inactivation. We aim to study whether ginsenoside Rg5 exerted inhibitory effects on crucial aspects of glioma stem cells, including cell viability, tumor initiation, invasion, self-renewal ability, neurosphere formation, and stemness. Through comprehensive sequencing analysis, we identified a compelling association between ginsenoside Rg5 and the ferroptosis pathway, which was further validated through subsequent experiments demonstrating its ability to activate this pathway. To elucidate the precise molecular targets affected by ginsenoside Rg5 in gliomas, we conducted an intersection analysis between differentially expressed genes obtained from sequencing and a database-predicted list of transcription factors and potential targets of ginsenoside Rg5. This rigorous approach led us to unequivocally confirm NR3C1 (Nuclear Receptor Subfamily 3 Group C Member 1) as a direct target of ginsenoside Rg5, a finding consistently supported by subsequent experimental investigations. Moreover, we uncovered NR3C1′s capacity to transcriptionally regulate ferroptosis -related genes HSPB1 and NCOA4. Strikingly, ginsenoside Rg5 induced notable alterations in the expression levels of both HSPB1 (Heat Shock Protein Family B Member 1) and NCOA4 (Nuclear Receptor Coactivator 4). Finally, our intracranial xenograft assays served to reaffirm the inhibitory effect of ginsenoside Rg5 on the malignant progression of glioblastoma. These collective findings strongly suggest that ginsenoside Rg5 hampers glioblastoma progression by activating ferroptosis through NR3C1, which subsequently modulates HSPB1 and NCOA4. Herpes zoster (HZ) is a common medical condition accompanied by several distressing symptoms, including acute pain. Pien Tze Huang (PZH) is a well-known traditional Chinese medicine (TCM) with numerous pharmacological effects, including antiviral properties, neuroprotection, and immunity regulation. To investigate the efficacy and safety of PZH capsules in patients with HZ. A multicenter, double-blinded, randomized, and placebo-controlled trial from 8 hospitals in 5 cities of China. Eligible participants were randomly assigned to the PZH capsule and placebo group at a 1:1 ratio. Treatment was conducted for 14 days with a window period of no more than 2 days. For the first 7 days, participants received antiviral drugs combined with PZH capsules (0.6 g/time, 3 times a day) or placebos. For the remaining 7 days, they were only treated with PZH capsules (0.6 g/time, 3 times a day) or placebos. We included 222 patients in the full analysis set (FAS), and 187 patients in the per protocol set (PPS). The change of numeric rating scale pain scores from baseline to the seventh day (±1 day) after treatment in the PZH capsule group was statistically superior to the placebo group (FAS: 2.33 vs. 1.71, 97.5% CI : 0.03 ∼ 1.19; PPS: 2.29 vs. 1.51, 97.5% CI : 0.18 ∼ 1.38). In the PPS, there was a significant difference in the time (days) of pain relief between the placebo group and the PZH capsule group (Mean (SD): 5.71 (3.76) vs. 4.69 (3.57), p = 0.046). On the seventh day (±1 day) after treatment, the level of CD8+ cells in the PZH capsule group were higher than those of the placebo group (FAS: Mean (SD): 24.08 (6.81) vs. 21.93 (8.19), p = 0.007; PPS: Mean (SD): 24.26 (6.93) vs. 22.15 (8.51), p = 0.012). The level of cytotoxic lymphocyte cells found similar results on the seventh day (±1 day) (FAS: Mean (SD): 12.17 (4.65) vs. 10.55 (4.15), p = 0.018; PPS: Mean (SD): 12.25 (4.65) vs. 10.11 (3.93), p = 0.002). No serious adverse events were noted and PZH capsules were well tolerated. PZH capsules confer therapeutic effects on HZ with the TCM symptom of stagnated heat of liver channel by substantially reducing the pain intensity, shortening the time of pain relief as well as regulating the immune function. Flowering means that the plant enters the reproductive growth stage, which is a crucial stage in the plant life cycle. Delaying flowering time to prolong vegetative growth is an important method to increase biomass yield and saccharification efficiency in switchgrass, It is of great significance to study the molecular mechanism of plant flowering and regulate the process of plant flowering in the process of biomass production. In this study, we identified 55 serine/threonine-protein kinase genes related to flower development from the switchgrass transcriptome database. Simultaneously, we cloned one of them, PvSTK1 , whose expression level and differential fold were significantly higher than other members. PvSTK1 is located on chromosome 8N and its protein was in the cell membrane, cytoplasm, and nucleus. The spatio-temporal expression analysis of the PvSTK1 in switchgrass displayed that the PvSTK1 is crucial in vegetative period, however, not in the transition to reproductive period. Overexpression of PvSTK1 in Arabidopsis resulted in down-regulation of flower-promoting genes and up-regulation of flower-suppressing genes, thereby delaying flowering. In addition, PvSTK1 caused atrophy of the ovules of the florets at the base of the inflorescence, leading to sterility of the florets. The function of PvSTK1 is to inhibit the development of floral organs, and its overexpression can prolong its vegetative period. Angiogenesis is recognized as a sign of cancer and facilitates cancer progression and metastasis. Suppression of angiogenesis is a desirable strategy for gastric cancer (GC) management. In this study, we showed a novel role of gastrin in angiogenesis of GC. We observed that treatment with gastrin 17 (G17) increased the proliferation of AGS cells and enhanced tube formation during normoxia and hypoxia. The expression level of VEGF were increased by G17 treatment as well. Experiments on the mechanism showed that G17 promoted HIF-1α expression, which subsequently enhanced β-catenin nuclear localization and activation of TCF3 and LEF1 and finally resulted in angiogenesis by upregulating VEGF. An in vivo experiment confirmed that G17 enhanced GC cell proliferation and angiogenesis in the resultant tumor. Guajadial, one of natural dialdehyde meroterpenoids, demonstrated significant antineoplasmic activity. The present research was to investigate the inhibitory effects of guajadial by using two NSCLC cells (A549 and H1650) proliferation and migration. Western blotting was employed to explore the underlying mechanisms of VEGF receptor (VEGFR)2-mediated. This research indicated that guajadial not only inhibited endothelial cell proliferation and migration but also suppress tumor growth in human NSCLC xenograft mouse models. Abstract Objective The aim of this meta-analysis was to analysis the expression of livin in human osteosarcoma. Methods We searched the Pubmed, Science Direct, Embase and Web of Science, CNKI, Wanfang and VIP for relevant original studies. Statistical analysis was performed by Stata 11.0 software. Results Our study indicated that livin expressed in the osteosarcoma tissue was significantly higher than the control group (OR = 18.814, P c = 0.000, 95% CI: 10.973–32.257), and the positive expression of livin was correlated with the size of osteosarcoma tumor and Enncking staging (OR = 4.832, 95% CI: 2.198–10.621; OR = 4.851, 95% CI: 3.053–7.709, respectively). Conclusion Livin was highly expressed in osteosarcoma, and osteosarcoma Enncking staging and tumor size were positively correlated, both may be involved in the occurrence and development of osteosarcoma, and be closely related to the prognosis of osteosarcoma patients. High Intensity D–T fusion Neutron Generator (HINEG), developed by Institute of Nuclear Energy Safety Technology (INEST), Chinese Academy of Sciences (CAS)•FDS Team, is an essential platform for advanced nuclear energy systems, which can be used for researches on nuclear technology and safety including the validation of neutronic method and software, validation and measurement of nuclear data, radiation shielding and protection, material irradiation damage and activation, neutronic performance of components, etc. In addition, the application of HINEG can be extended to nuclear medicine, radiotherapy, neutron imaging and other nuclear technology applications. A full-digital multidimensional redundancy control for HINEG system has been implemented by full-digital hardware of the Server/Client mode, and software infrastructure of data acquisition and process control designed by modular and multidimensional redundancy method. Either local users or remote users can execute all authorized control function by unified interface under the condition of radiation, ultra-high voltage and complex electromagnetic environment. With the application of HINEG Control System (HINEG CS), it is the first time for the neutron generator control system to realize full-digital and multidimensional redundancy control which can improve the reliability of neutron generator. Tetrabromobisphenol A (TBBPA) is the world's most widely used brominated flame retardant but there is growing concern about its fate and toxicity in terrestrial organisms. In this study, two ecologically different earthworms, Metaphire guillelmi and Eisenia fetida , were exposed to soil spiked with 14 C-labeled TBBPA for 21 days. M. guillelmi accumulated more TBBPA than E. fetida , evidenced by a 2.7-fold higher 14 C-uptake rate and a 1.3-fold higher biota-soil accumulation factor. Considerable amounts of bound residues (up to 40% for M. guillelmi and 18% for E. fetida ) formed rapidly in the bodies of both earthworms. 14 C accumulated mostly in the gut of M. guillemi and in the skin of E. fetida , suggesting that its uptake by M. guillelmi was mainly via gut processes whereas in E. fetida epidermal adsorption predominated. The TBBPA transformation potential was greater in M. guillelmi than in E. fetida , since only 5% vs. 34% of extractable 14 C remained as the parent compound after 21 days of exposure. Besides polar metabolites, the major metabolites in both earthworms were TBBPA mono- and dimethyl ethers ( O -methylation products of TBBPA). Acute toxicity assessments using filter paper and natural soil tests showed that the methylation metabolites were much less toxic than the parent TBBPA to both earthworms. It indicated that earthworms used O -methylation to detoxify TBBPA, and M. guillelmi exhibited the higher detoxification ability than E. fetida . These results imply that if only the free parent compound TBBPA is measured, not only bioaccumulation may be underestimated but also its difference between earthworm species may be misestimated. The species-dependent fate of TBBPA may provide a better indicator of the differing sensitivities of earthworms to this environmental contaminant. [ABSTRACT FROM AUTHOR]

The fate of organic pollutants in the environment, especially the formation and stability of non-extractable (i.e., bound) residues (NERs) determines their environmental risk. Using 14C-tracers, we studied the fate of the carcinogen phenanthrene in active or sterilized oxic loamy soil in the absence and presence of the geophagous earthworm Metaphire guillelmi and characterized the NERs derived from phenanthrene. After incubation of 14C-phenanthrene in active soil for 28 days, 40 ± 3.1% of the initial amount was mineralized and 70.1 ± 1.9% was converted to NERs. Most of the NERs (>92%) were bound to soil humin. Silylation of the humin-bound residues released 45.3 ± 5.3% of these residues, which indicated that they were physically entrapped, whereas the remainder of the residues were chemically bound or biogenic. By contrast, in sterilized soil, only 43.4 ± 12.6% of the phenanthrene was converted to NERs and all of these residues were completely released upon silylation, which underlines the essential role of microbial activity in NER formation. The presence of M. guillelmi in active soil significantly inhibited phenanthrene mineralization (24.4 ± 2.6% mineralized), but NER formation was not significantly affected. Only a small amount of phenanthrene-derived residues (1.9-5.3% of the initial amount) accumulated in the earthworm body. When humin-bound residues were mixed with fresh soil, 33.9% (humin recovered from active soils) and 12.4% (humin recovered from sterilized soils) of the residues were mineralized after 75 days of incubation, respectively, which indicated a high bioavailability of NERs, albeit lower than the initial addition of phenanthrene. Our results indicated that many phenanthrene-derived NERs, especially those physically entrapped, are still bioavailable and may pose a toxic threat to soil organisms. [ABSTRACT FROM AUTHOR]

Water evaporation-driven hydrovoltaic generators (HGs) with the property of sustainable electricity generation offer the promise of clean and renewable power. Despite recent efforts to advance HGs by enhancing solid-liquid interface interactions and nanopore structures design, extremely scarce usable natural water source and slow heat replenishment remain obstacles. Here, we demonstrated a solar thermal conversion boosted hydrovoltaic power generation system (HPGS) by rationally integrating environmental water harvesting patterned coating, solar steam generator and hydrovoltaic functional coating with solar thermal conversion capability. In the system, the water collected or readily available in the environment with high ion concentration (conductivity of 505.95 μS cm−1) is purified by a solar steam generator into a low conductivity pure water (~ 6.32 μS cm−1) to supply for HGs, realizing the solution to the water source limitation for HGs. The Al 2 O 3 /CB nanoparticles constructed hydrovoltaic functional coating exhibits remarkable solar thermal conversion ability to raise device temperatures by over 18 °C, resulting an increase of V oc by 130.7% from 2.54 V to 5.86 V at 50% RH under 1 standard sun radiation. Such HPGS based on solar thermal conversion makes it possible for HGs to directly use the environmental available water and break through the shackles of restricted ambient temperature and slow heat replenishment. Continental subduction and its interaction with overlying mantle wedge are recognized as fundamental solid earth processes, yet the dynamics of this system remains ambiguous. In order to get an insight into crust–mantle interaction triggered by partial melting of subudcted continental crust during its exhumation, we carried out a combined study of the Shidao alkaline complex from the Sulu ultrahigh pressure (UHP) terrane. The alkaline complex is composed of shoshonitic to ultrapotassic (K 2 O: 3.4–9.3 wt.%) gabbro, pyroxene syenite, amphibole syenite, quartz syenite, and granite. Field studies suggest that the mafic rocks are earlier than the felsic ones in sequence. LA-ICPMS zircon U–Pb dating on them gives Late Triassic ages of 214 ± 2 to 200 ± 3 Ma from mafic to felsic rocks. These ages are slightly younger than the Late Triassic ages (225–210 Ma) of the felsic melts from partial melting of the Sulu UHP terrane during exhumation. The alkaline rocks have wide ranges of SiO 2 (49.7–76.7 wt.%), MgO (8.25–0.03 wt.%), Ni (126.0–0.07 ppm), and Cr (182.0–0.45 ppm) contents. The contents of MgO, total Fe 2 O 3 , CaO, TiO 2 and P 2 O 5 decrease with increasing SiO 2 contents. The contents of Na 2 O, K 2 O, and Al 2 O 3 increase from gabbro to amphibole syenite, and decrease from amphibole syenite to granite, respectively. The alkaline rocks have characteristics of an arc-like pattern in trace element distribution, e.g., enrichment of LREE, LILE (Rb and Ba), Th and U, depletion of HFSE (Nb, Ta, P and Ti), and positive Pb anomalies. From the mafic rocks to the felsic rocks, the (La/Yb) N ratios and the contents of the total REE, Sr and Ba decrease but the Rb contents increase. The alkaline rocks with high SiO 2 contents also display features of an A2-type granitoids, e.g., high contents of total alkalis, Zr and Nb and high ratios of Fe 2 O 3 T /MgO, Ga/Al, Yb/Ta and Y/Nb, suggesting a post-collisional magmatism during exhumation of the Sulu UHP terrane. The alkaline rocks have homogeneous initial 87 Sr/ 86 Sr ratios (0.7058–0.7093) and negative ε Nd (t) values (− 18.6 to − 15.0) for whole-rock. The Sr–Nd isotopic data remain almost unchanged with varying SiO 2 and MgO contents, suggesting a fractional crystallization (FC) process from the same parental magma. Our studies suggest a crust–mantle interaction in continental subduction interface as follows: (1) hydrous felsic melts from partial melting of subducted continental crust during its exhumation metasomatized the overlying mantle wedge to form a K-rich and amphibole-bearing mantle; (2) partial melting of the enriched lithospheric mantle generated the Late Triassic alkaline complex under a post-collisional setting; and (3) the alkaline magma experienced subsequent fractionational crystallization mainly dominated by olivine, clinopyroxene, plagioclase and alkali feldspar. Tetrabromobisphenol A (TBBPA) is the brominated flame retardant with the highest production volume and its bioaccumulation in environment has caused both human health and environmental concerns, however the fate and metabolism of TBBPA in plants is unknown. We studied the fate, metabolites, and transformation of 14 C-labeled TBBPA in rice cell suspension culture. During the incubation for 14 days, TBBPA degradation occurred continuously in the culture, accompanied by formation of one anisolic metabolite [2,6-dibromo-4-(2-(2-hydroxy)-propyl)-anisole] (DBHPA) (50% of the degraded TBBPA) and cellular debris-bound residues (46.4%) as well as mineralization (3.6%). The cells continuously accumulated TBBPA in the cytoplasm, while a small amount of DBHPA (2.1% of the initially applied TBBPA) was detectable inside the cells only at the end of incubation. The majority of the accumulated residues in the cells was attributed to the cellular debris-bound residues, accounting for 70–79% of the accumulation after the first incubation day. About 5.4% of the accumulation was associated with cell organelles, which contributed 7.5% to the cellular debris-bound residues. Based on the fate and metabolism of TBBPA in the rice cell suspension culture, a type II ipso -substitution pathway was proposed to describe the initial step for TBBPA degradation in the culture and balance the fate of TBBPA in the cells. To the best of our knowledge, our study provides for the first time the insights into the fate and metabolism of TBBPA in plants and points out the potential role of type II ipso -hydroxylation substitution in degradation of alkylphenols in plants. Further studies are required to reveal the mechanisms for the bound-residue formation (e.g., binding of residues to specific cell wall components), nature of the binding, and toxicological effects of the bound residues and DBHPA.
