This study aims to estimate the prevalence and factors of depression in primary caregivers of people with dementia in China, based on a biopsychosocial medical model. A sample of 285 caregiver-patient dyads was recruited from a tertiary psychiatric hospital in Nanjing, between December 2018 and November 2019. The prevalence of depression among primary caregivers of people with dementia was 42.8%. Binary logistic regression analyses revealed that caregivers' gender (OR=4.692), social support (OR=0.131), health condition (OR=12.994), extraversion (OR=0.102) and neuroticism (OR=2.978) were predictive of depression in those caregivers. Of the above, health condition was the major factor associated with caregiver's depression. The Box-Tidwell method was used to show a linear relationship between continuous independent variables and dependent variable logit conversion values (p = 0.0045). Highlights • Deacetylation removed 66% of acetyl groups in corn stover and sugarcane bagasse. • Glucose recovery rate was 92.2% from deacetylated and pretreated stover. • Residual xylose reduced 61.3% by blending a C6 yeast with engineered C5/C6 yeast. Abstract Corn stover and sugarcane bagasse are the most widely available agriculture processing biomass and could serve as feedstocks for production of biofuel. In this study, three different technologies are combined to develop a more efficient conversion process for each of these feedstocks. The three technologies are diluted alkaline deacetylation process, combined thermochemical and mechanical shear pretreatment, and fermentation using a combined inoculum of two commercial Saccharomyces yeast strains. The two yeast strains used were a non-GMO and GMO strain engineered for xylose fermentation. The final ethanol concentrations obtained were 35.7 g/L from deacetylated corn stover and 32.9 g/L from sugarcane bagasse. Blending the two yeast reduced residual xylose content from 1.24 g/L to 0.48 g/L and increased ethanol production by 6.5% compared to solely using the C5/C6 yeast. In this study, liquid hot water pretreatment was combined with disk milling for pretreatment of sugarcane bagasse. Sugarcane bagasse was pretreated using liquid hot water (LHW) at 140–180 °C for 10 min (20% w/w solids content) and then disk milled. Disk milling improved glucose release 41–177% and ethanol production from glucose/xylose cofermentation by 80% compared to only using LHW pretreatment. The highest ethanol conversion efficiency achieved was 94%, which was observed when bagasse was treated at 180 °C with LHW and disk milled. However, a small amount of residual xylose (3 g/L) was indicative that further improvement could be achieved to increase ethanol production. [ABSTRACT FROM AUTHOR]

Biodiesel production in the U.S. from vegetable oils has increased substantially during the past decade. However, its further increase is limited by the low amounts of oil produced per hectare from temperate oilseed crops. Recently novel transgenic sugarcane has been developed to accumulate both sugars and lipids in stems, making it a promising dual-purpose feedstock to produce both ethanol and biodiesel. In this study, two lines of the transgenic lipid producing sugarcane (lipid-cane) and the non-transformed sugarcane were characterized and processed. The total lipid concentrations were 0.7%, 0.9% and 1.3% for the non-transformed sugarcane and lipid-cane lines19B and 25 C, respectively. Lipid composition analysis showed that about 31–33% of the total lipids were triacylglycerols, main feedstock for biodiesel production, for the lipid-cane samples, while this value was only 5% for the non-transformed sugarcane. By processing the sugarcane stems with a juicer, about 90% of the sugars and 60% of the lipids were extracted with juice. The extracted sugars in juice were fermented to ethanol and the lipids were later recovered from the fermented juice using organic solvents. The recovered lipids from the fermented juice were 0.3, 0.5 and 0.8 g/100 g dry stem for the non-transformed sugarcane and lipid-cane lines 19B and 25 C, respectively. This study proved the concept of the lipid and sugar coproduction from the novel lipid-cane, which have a potential to make a large-scale replacement of fossil derived fuel without unrealistic demands on land area. • Mutations conferring resistance to gatifloxacin (GAT) remain inadequately characterized. • 123 GAT-resistant mutants were isolated and subjected to whole-genome sequencing. • Sixty-eight of 123 mutants had gyrA gene mutations (G280A, A281G, G280T and G262T). • The remaining mutants harboured gyrB gene mutations (A1495G, C1497A, C1497G and A1503C). • Mutations in the gyrA and gyrB genes are the main mechanisms of resistance to GAT. Gatifloxacin (GAT), a fourth-generation fluoroquinolone (FQ), is used to treat drug-resistant tuberculosis. Although DNA gyrase mutations are the leading cause of FQ resistance, mutations conferring resistance to GAT remain inadequately characterized. GAT-resistant mutants were selected from 7H10 agar plates containing 0.5 mg/L GAT (critical concentration). Mutations involved in GAT resistance were identified through whole-genome sequencing. In total, 123 isolates demonstrated resistance to GAT. Among these isolates, 55.3% (68/123) had gyrA gene mutations [G280A (D94N), A281G (D94G), G280T (D94Y) and G262T (G88C)]. The remainder (44.7%, 55/123) harboured gyrB gene mutations [A1495G (N499D), C1497A (N499K), C1497G (N499K) and A1503C (E501D)]. Mutations in the gyrA and gyrB genes are the main mechanisms of GAT resistance. • The first trial to explore all-oral short regimens for MDR-TB in China. • Two optimized regimens, designed according to local conditions. • Similar high culture conversion in the early stage in both regimens. • High treatment success rate for patients who completed the treatment. • Good adherence in the pandemic of COVID-19. Multidrug-resistant tuberculosis (MDR-TB) patients have been suffering long, ineffective, and toxic treatment until short-course injectable-free regimens emerged. However, the new WHO-recommended regimens might be less feasible in the real-world setting. Here, we evaluated two optimized all-oral short-course regimens in China. From April 2019 to August 2020, we conducted a prospective nonrandomized controlled trial and consecutively included 103 MDR-TB patients diagnosed with pulmonary MDR-TB in Shenzhen, China. A 4-5 drug regimen of 9-12 months was tailored to the strain's resistance patterns, patients' affordability, and tolerance to drugs. This was an interim analysis, focusing on the early treatment period. 53.4% (55/103) of patients were prescribed linezolid, fluoroquinolone (FQ), clofazimine, cycloserine, and pyrazinamide, followed by a regimen in which clofazimine was replaced by bedaquiline (35/103, 34.0%). The culture conversion rate was 83.1% and 94.4% at two and four months, respectively, with no significant difference between bedaquiline-free and bedaquiline-containing cases and between FQ-susceptible and FQ-resistant cases. Among 41 patients who completed treatment, 40 (97.6%) patients had a favorable outcome and no relapse was observed. Peripheral neuropathy and arthralgia/myalgia were the most frequent AEs (56.3%, 58/103). 18 AEs caused permanent discontinuation of drugs, mostly due to pyrazinamide and linezolid. Mycobacterium tuberculosis (Mtb), the pathogenic agent of tuberculosis, remains a primary inducement of morbidity and mortality globally. Mtb have evolved mechanisms to recognize diverse signals, such as acidic pH within phagolysosomes and therefore to reprogram multiple physiological and metabolic processes to adapt to intracellular survival. Moreover, lysine malonylation has been suggested to participate in regulation of enzymes in carbon metabolism. However, lysine malonylation in Mtb and its association with acidic pH associated metabolism adaptation remain unknown. Here, we systematically characterized the comparative malonylome of Mtb H37Rv grown in normal (7H9-Tyloxapol (Ty)-7.4) and acidic (7H9-Ty-4.5) medium mimicking lysosome pH. In total, 2467 lysine malonylation sites within 1026 proteins were identified, which related to diverse biological processes, particularly accumulated in metabolic process. 1090 lysine malonylation sites from 562 proteins were quantified, among which 391 lysine malonylation sites in 273 protein were down-regulated while 40 lysine malonylation sites from 36 proteins were up-regulated in acidic medium, indicating that malonylation may participate in acidic pH associated metabolism. Accordingly, the enzyme activity of GlcB was reduced under acidic stress corresponding to decreased malonylation of GlcB compared with that of normal condition and this was further demonstrated by site-specific mutations. We further found that Mtb-CobB, a sirtuin-like deacetylase and desuccinylase, involved in demalonylase activity. Together, the Mtb malonylome not only indicates the critical role of malonylation in metabolism regulation, but may provide new insights of malonylation on metabolism adaptation to acidic micro-environment in vivo.
