Membrane fouling is one of the most pressing obstacles during the operation of filtration membranes. Its different physical stages of concentration polarization, fouling layer deposition and reorganisation are difficult to access experimentally. Normally, the phenomenon is observed during membrane filtration when the flux turns time variant or the flux increase does not follow the applied pressure difference linearly. Flux or pressure stepping methods are state of the art to detect the onset of fouling, however they give only limited information on the properties of the deposition layer in the fluid phase adjacent to the membrane surface. Here we present a new hydraulic spectroscopy method allowing to fingerprint the properties of the deposition layer. At different flux levels, we vary the flux around an average value in a sinusoidal fashion. From the measured pressure drop values and the time delay as compared to the imposed flux, we can calculate the so-called hydraulic impedance. Varying the frequency of the flux at a set amplitude, we can explore the frequency dependent hydraulic impedance and the different single contributions to its overall magnitude. We have applied this method to a variety of different solutions and filtration configurations showcasing its sensitivity. Abstract: Optimal cell interaction with biomaterial scaffolds is one of the important requirements for the development of successful in vitro tissue-engineered tissues. Fast, efficient and spatially uniform cell adhesion can improve the clinical potential of engineered tissue. Three-dimensional (3-D) solid free form fabrication is one widely used scaffold fabrication technique today. By means of deposition of polymer fibers, scaffolds with various porosity, 3-D architecture and mechanical properties can be prepared. These scaffolds consist mostly of solid round fibers. In this study, it was hypothesized that a corrugated fiber morphology enhances cell adhesion and proliferation and therefore leads to the development of successful in vitro tissue-engineered constructs. Corrugated round fibers were prepared and characterized by extruding poly(ethylene oxide terephthalate)-co-poly(butylene terephthalate) (300PEOT55PBT45) block co-polymer through specially designed silicon wafer inserts. Corrugated round fibers with 6 and 10 grooves on the fiber surface were compared with solid round fibers of various diameters. The culture of mouse pre-myoblast (C2C12) cells on all fibers was studied under static and dynamic conditions by means of scanning electron microscopy, cell staining and DNA quantification. After 7days of culturing under static conditions, the DNA content on the corrugated round fibers was approximately twice as high as that on the solid round fibers. Moreover, under dynamic culture conditions, the cells on the corrugated round fibers seemed to experience lower mechanical forces and therefore adhered better than on the solid round fibers. The results of this study show that the surface architecture of fibers in a tissue engineering scaffold can be used as a tool to improve the performance of the scaffold in terms of cell adhesion and proliferation. Reverse osmosis (RO) membrane systems are widely used in the desalination of water. However, flux decline due to fouling phenomena in RO remains a challenge. To minimize fouling, a reliable index is necessary to predict the fouling potential of the RO feed water. The ASTM introduced the silt density index (SDI) as a standard fouling index to measure the fouling potential due to colloidal and suspended particles. For decades, the SDI is worldwide accepted and applied. There are growing doubts about the predictive value of this parameter. In addition there are several deficiencies observed, affecting the accuracy and reproducibility e.g. no correction factor for temperature, nor for variations in membrane resistance, and no linear correlation with the concentration of colloidal/suspended particles. This paper gives an overview of our work on limitations, improvements and alternatives for the SDI. Firstly, the influence of the applied 0.45 lm test membrane on the SDI will be investigated. Variations in SDI values can be attributed to differences in properties of these membranes. In order to quantify the influence of pressure, temperature and membrane resistance on the SDI a mathematical relation was developed between the SDI and the MFI0.45, assuming cake filtration. In addition, also other fouling mechanisms were incorporated in the model using the wellknown blocking laws. Based on a cake filtration fouling mechanism and assuming 100% particle retention, the models were used to normalize the experimental SDI values for temperature, pressure and membrane resistance to the SDI+. By applying this normalization, the results of SDI tests carried out under different conditions and/or with different membranes can be compared easily as was proven experimentally in the lab and at a seawater desalination plant. Finally, an alternative filtration index will be introduced, the volume-based SDI_v. The SDI_v compares the initial flow rate to the flow rate after filtering a standard volume of feed water using MF membranes with an average pore size of 0.45 lm. Our experimental results show that SDI_v is independent of the membrane resistance. In that way, it eliminates most of the disadvantages of the SDI and has great potential to replace the SDI in the field. [ABSTRACT FROM AUTHOR]

The ASTM considers the silt density index (SDI) test as a standard test for fouling potential of RO and NF feed waters. Up to date, the SDI is used at many full- and pilot-scale installations. The design and choice of the applied RO pretreatment is to a large extent based on the SDI test on the raw feed water. Comparing and monitoring UF/MF membrane performance is another SDI application. From a practical point of view, the SDI of RO feed water preferably should be lower than 3. The SDI has several disadvantages making it an unreliable test. The SDI has a non linear relationship with the colloidal concentration in the water and it is not corrected for the feed water temperature. Besides that, the SDI is not based on any filtration model. SDI is trying to simulate the RO fouling using dead-end MF membrane. The modified fouling index (MFI) is another fouling index. The MFI is based on the cake filtration model, can be corrected for pressure and temperature and is therefore used as a promising alternative for the SDI. Nevertheless, the procedure of measuring a MFI is more difficult and not directly suitable for carrying out "in the field". The objective of this study therefore is to determine a theoretical relationship between SDI and MFI, and to validate this with experimental results. This relationship can be used to investigate the influence of membrane and testing parameters on SDI under cake filtration conditions, implying this model is valid for cake filtration mechanism and a particle rejection of 100%. In order to calculate the SDI, the times t1 and t2 for collecting the first and second sample are predicted using the measured MFI value and the MFI definition. In this research, the influence of several parameters (such as temperature, membrane resistance, etc.) on the SDI will be shown. The experimental results show a good agreement with the theoretical work, but only if the cake filtration start builds up directly at the beginning of the experiment. In general, this work clearly demonstrates that SDI currently is not reliable test for RO fouling. Either corrections for the SDI are necessary to give a more reliable index, or a new index has to be developed. [ABSTRACT FROM AUTHOR]

Abstract: Sufficient nutrient and oxygen transport is a potent modulator of cell proliferation in in vitro tissue-engineered constructs. The lack of oxygen and culture medium can create a potentially lethal environment and limit cellular metabolic activity and growth. Diffusion through scaffold and multi-cellular tissue typically limits transport in vitro, leading to potential hypoxic regions and reduction in the viable tissue thickness. For the in vitro generation of clinically relevant tissue-engineered grafts, current nutrient diffusion limitations should be addressed. Major approaches to overcoming these include culture with bioreactors, scaffolds with artificial microvasculature, oxygen carriers and pre-vascularization of the engineered tissues. This study focuses on the development and utilization of a new perfusion culture system to provide adequate nutrient delivery to cells within large three-dimensional (3D) scaffolds. Perfusion of oxygenated culture medium through porous hollow fiber (HF) integrated within 3D free form fabricated (FFF) scaffolds is proposed. Mouse pre-myoblast (C2C12) cells cultured on scaffolds of poly(ethylene-oxide-terephthalate)–poly(butylene-terephthalate) block copolymer (300PEOT55PBT45) integrated with porous HF membranes of modified poly(ether-sulfone) (mPES, Gambro GmbH) is used as a model system. Various parameters such as fiber transport properties, fiber spacing within a scaffold and medium flow conditions are optimized. The results show that four HF membranes integrated with the scaffold significantly improve the cell density and cell distribution. This study provides a basis for the development of a new HF perfusion culture methodology to overcome the limitations of nutrient diffusion in the culture of large 3D tissue constructs. Abstract: Hollow fiber membranes with microstructured inner surfaces were fabricated from a PES/PVP blend using a spinneret with a microstructured needle. The effect of spinning parameters such as polymer dope flow rate, bore liquid flowrate, air gap and take-up speed on the microstructure and shape of the bore and its deformation was investigated. It was found that when a high bore liquid flowrate was used, the microstructure in the bore surface was destroyed. The bores were deformed to an oval shape when the fiber walls were thick. This was attributed to buckling of the fiber shell as a result of the coagulation and shrinkage of the outer surface. Fibers were also fabricated with a round-needled spinneret for comparison. The intrinsic pure water permeabilities (based on the actual bore surface areas) of fibers with structured and round bores were found to be similar. On the other hand, the structured fibers have larger pores in the skin layer. Smaller pores on the round fibers are considered to form when the inner surface coagulates and the skin layer is pulled inwards due to the shrinkage caused by phase separation. When the bore is structured, the wavy shape can damp this contraction effect resulting in larger pores. The skin layer thickness of the fibers was investigated using a colloidal filtration method. It was shown that fibers with microstructured bores which have mostly uniform skin layer thickness and reasonably narrow pore size distribution can be fabricated. [Copyright &y& Elsevier]
