CDK4/6 inhibitors combined with endocrine therapy are standard first- or second-line treatment for patients with HR-positive and HER2-negative advanced breast cancer, however, there is currently no optimal recommendation for therapeutic strategies after progression on CDK4/6i. The aim of this study is to analyze the efficacy and safety of HDAC inhibitor Tucidinostat combined with endocrine therapy in patients after prior CDK4/6 inhibitor progression. The pathological and clinical data of 44 HR-positive and HER2-negative breast cancer patients treated with tucidinostat after progression on CDK4/6i at the Breast Oncology Department of the Fifth Medical Center of the PLA General Hospital from July 2019 to October 2021 were retrospectively analyzed. Observation indexes included progression-free survival (PFS), overall survival (OS), clinical benefit rate (CBR), objective response rate (ORR) and adverse events. At the same time, we attempted to identify potential genomic predictors using available next-generation sequencing (NGS). A total of 44 patients were enrolled in this study. Median follow-up was 10 months (1–26 months) by the data cutoff date (February 2022). The CBR was 6.8% (3/44), the median PFS was 2.0 months (95% CI 1.9–2.1), and the median OS was 14 months (95% CI 6.3–21.7). The mPFS was 4.1 months (95%CI: 0–8.2) in patients with 1 metastatic site, and the mPFS was 4.5 months (95%CI: 4.2–4.8) in patients who received sequential tucidinostat after CDK4/6i failure. Multivariate analysis showed that patients with 1 metastatic site or sequential tucidinostat treatment after failure of CDK4/6i were more likely to benefit from tucidinostat combined with endocrine therapy. Preliminary data showed PIK3CA mutation may be associated with resistance of tucidinostat therapy. No grade 4 adverse events and no treatment-related deaths were recorded in the study. Dose reductions because of adverse events occurred in 4 (9.1%) patients. This study preliminarily shows that tucidinostat combined with endocrine therapy may be an optional sequential strategy for patients with HR+/HER2-advanced breast cancer that has progressed on CDK4/6 inhibitor, especially for these with lower tumor burden and fewer prior palliative treatment. • CDK4/6 inhibitor became standard treatment for HR-positive MBC patients. • There is no optimal treatment strategy after progression on CDK4/6i. • Tcucidinostat was the only HDAC inhibitor approved in solid tumors worldwide. • The mPFS of Tcucidinostat was 4.5 months when used sequentially after prior CDK4/6i. • PIK3CA mutation may be associated with resistance of tucidinostat. National-level marine ranching plays an important role in enhancing the marine ecological environment, fostering social and economic development, and bolstering human well-being. Quantitatively analyzing the level of coordinated development of its "ecological-economic-social" benefits and the factors influencing them is of great theoretical significance and practical value for refining the new round of development planning and guiding its construction and management. Therefore, on the basis of establishing a theoretical framework for the interaction of "ecological-economic-social" benefits, shellfish and algae were selected as research objects. We calculated the levels of ecological, economic, and social benefits, explored the interrelationships among the three using the coupled coordination model, and analyzed the influencing factors by using multivariate linear regression and geographically weighted regression. The results showed that: (1) In 2015 and 2021, the ecological and economic benefits of national-level marine ranching for shellfish and algae were better than the social benefits, in which the ecological and social benefits were stable, while the economic benefits were more variable; (2) The degree of coordination of the coupling between the three benefits in each province was mostly low, with most experiencing severe disorder; (3) The level of openness to the outside world and the standard of living of fishermen contributed positively to the coordination of benefit coupling between shellfish and algae. Conversely, the level of science and technology, innovation capacity, and the level of social development had a slightly negative impact. In conclusion, shellfish and algal aquaculture in China's national-level marine ranching significantly contributes to the ecological, economic, and social systems. However, the merits and demerits of natural, social, and economic factors must be comprehensively considered to promote the sustainable development of marine ranching. • Shellfish and algae, as the main species of China's marine organisms, provide a guarantee for the country's food security. • The assessment of shellfish and algae benefits reflects the sustainable development level of marine ranching. • The relationships of shellfish and algae benefits vary due to factors, leading to discrepancies between theory and practice. • Integrating domestic and international marine ranching developments offers insights for China's marine ranching construction. In recent years, harmful algal blooms（HABs）caused by the eutrophication of water bodies have been occurring, which not only damage the ecological environment, but also threaten human health. To control HABs simply and efficiently, a novel magnetic buoyant-bead flotation method was developed in this study. A magnetic floatable bead of Fe 3 O 4 /CCP was first prepared, and then was characterized by X-ray diffraction, Fourier transform infrared spectroscopy, vibrating sample magnetometer and X-ray photoelectron spectroscopy. Subsequently, this study examined the removal effects of three kinds of floatable bead (closed-cell perlite (CCP), Fe 3 O 4 /CCP and Fe 3 O 4 /CCP +CPAM) on three typical microalgae (Chlorella vulgaris , Microcystis aeruginosa and Anabaena flos-aquae) in water bloom. The results confirmed that the removal rate of Fe 3 O 4 /CCP +CPAM was 27.28%, 45.79% and 42.71% higher than that of Fe 3 O 4 /CCP in the removal of three microalgae, respectively, while it was 32.26%, 50.49% and 46.91% higher than that of CCP, respectively. In addition, CPAM concentration, the amount of Fe 3 O 4 /CCP, stirring time, and stirring speed were found to have significant effects on the removal of three microalgae. The orthogonal experiments further revealed that the CPAM concentration exerted the most significant impact on the removal rate while using the proposed magnetic buoyant-bead flotation method. Under the optimal experimental conditions, the removal rate of C. vulgaris , M. aeruginosa and A. flos-aquae using Fe 3 O 4 /CCP was 98.01%, 97.96% and 97.58%, respectively. Finally, the removal mechanism of microalgae by the combination of Fe 3 O 4 /CCP+CPAM was given. This study can provide a new idea for effectively removing harmful microalgae from water blooms or harvesting the microalgae. [Display omitted] • Buoyant barium alginate bubble beads (BABB) boosted the growth of immobilized microalgae. • BABB reduced the leakage of microalgae by 80% over traditional calcium alginate beads (CAB). • BABB enabled rapid microalgal biomass separation and full freeze−drying in 3.5 h. • Three strategic BABB optimizations were introduced for further enhanced culture. • Optimized BABB in open raceway pond reactor achieved 0.609 mg/bead biomass in 7 days. Microalgae immobilization in alginate beads shows promise for biomass production and water pollution control. However, carrier instability and mass transfer limitations are challenges. This study introduces buoyant barium alginate bubble beads (BABB), which offer exceptional stability and enhance Chlorella vulgaris growth. In just 12 days, compared to traditional calcium alginate beads, BABB achieved a 20 % biomass increase while minimizing cell leakage and simplifying harvesting. BABB optimization involved co-immobilization with BG-11 medium, enrichment of CO 2 in internal bubbles, and the integration of Fe nanoparticles (FeNPs). In the open raceway pond reactor, these optimizations resulted in a 39 % increase in biomass over 7 days compared to the unoptimized setup in closed flasks. Furthermore, enhancements in pigment and organic matter production were observed, along with improved removal of ammonia nitrogen and phosphate.
