Abstract: Cell wall components, such as lignin, cellulose, and hemicelluloses, play an important role in the conversion efficiency of corn stover into ethanol. Understanding the molecular basis of cell wall formation is fundamental for marker assisted selection to develop lines more suitable for ethanol production. In this study, we evaluated a set of 40 European forage maize lines for cellulose, lignin, total hemicellulose, glucuronoarabinoxylan (GAX), and monosaccharides, such as arabinose (ara), xylose (xyl), and glucuronic acid (GlcA). The most significant correlations were observed between hemicelluloses and GAX (0.9), and hemicelluloses and cellulose (−0.81). Cell wall digestibility (CWD, estimated by digestible neutral detergent fiber, DNDF) was negatively correlated with Xyl (−0.34). The association analysis between the evaluated traits and polymorphisms within ten “lignin” genes revealed significant associations between polymorphisms within CCoAOMT1, CCoAOMT2, 4CL2 and C4H, and cellulose/xyl, cellulose, cellulose, and GclA, respectively. Abstract Thermochemical processing is a promising method for the rapid depolymerization of biomass. This study investigated switchgrass, corn stover, red oak, hybrid poplar, and loblolly pine in terms of heteropolymer and elemental composition, and the distribution and composition of the fast pyrolysis products. Corn stover differed from other biomass types in that less of the biomass was recovered as sugar or phenolic oil (PO) and more of the biomass was recovered as bio-char and bio-gas. The sugar-rich aqueous stream recovered from the bio-oil heavy fraction was characterized in terms of sugar content and distribution, inhibitor content, and ability to support production of ethanol by Escherichia coli KO11 + lgk as a model biorenewable product. Levoglucosan was the most abundant sugar from each type of biomass, followed by either xylose or cellobiosan. For hybrid poplar, cellobiosan accounted for 30 wt% of the total sugar pool. Each of the sugar streams also contained a variety of inhibitors, particularly 5-hydroxymethylfurfural (5-HMF) and methylcyclopentenolone. Methylcyclopentenolone, maple lactone, was found to decrease the specific growth rate of E. coli by 50% when present at 0.72 wt%, indicating that it is less toxic than furfural, acetic acid and guaiacol. Sugars produced from switchgrass contained 4-fold less contaminants on a per-sugar basis than those from poplar and pine. All of the sugar streams contained too many inhibitors to be used at an industrially feasible concentration without additional detoxification. The poplar-derived pyrolytic sugar syrup was particularly inhibitory, possibly due to the high abundance of aromatic hydrocarbons, such as xylenes, and anisoles. Durum wheat is naturally more susceptible to Fusarium graminerum infection in comparison to common wheat. The improvement of durum wheat resistance against F. graminearum is a challenge due to the lack of resistance sources in its gene pool. FHB-resistance factors were introduced in durum wheat by generating recombinant inbred lines (RILs), obtained by crossing the hexaploid resistant accession 02-5B-318 with the susceptible durum wheat cv. Saragolla. In this work we explored the possible contribution of cell wall (CW) in RILs with improved FHB resistance. We thoroughly studied CW components, mycotoxins content and the expression of related genes in different RILs selected for their extremely high and low resistance to FHB. Differences were found in resistant and susceptible lines in the degree of pectin methylesterification and in deoxynivalenol (DON) accumulation after fungal infection. Genes involved in biochemical modification of CW structure ( WheatPme-1, Glu-1 ) and mycotoxins accumulation ( ns-LTP-1 ) were analyzed as putative candidates for FHB resistance. Xyloglucan is the most abundant hemicellulose in the primary cell wall of dicotyledonous plants. In Arabidopsis, three xyloglucan xylosyltransferases, XXT1, XXT2, and XXT5, participate in xylosylation of the xyloglucan backbone. Despite the importance of these enzymes, there is a lack of information on their structure and the critical residues required for substrate binding and transferase activity. In this study, the roles of different domains of XX2 in protein expression and catalytic activity were investigated by constructing a series of N- and C-terminal truncations. XXT2 with an N-terminal truncation of 31 amino acids after the predicted transmembrane domain showed the highest protein expression, but truncations of more than 31 residues decreased protein expression and catalytic activity. XXT2 constructs with C-terminal truncations showed increased protein expression but decreased activity, particularly for truncations of 44 or more amino acids. Site-directed mutagenesis was also used to investigate six positively charged residues near the C-terminus and found that four of the mutants showed decreased enzymatic activity. We conclude that the N- and C-termini of XXT2 have important roles in protein folding and enzymatic activity: the stem region (particularly the N-terminus of the catalytic domain) is critical for protein folding and the C-terminus is essential for enzymatic activity but not for protein folding. Abstract: In this paper, we continue our studies of the previously discovered [O.A. Zabotina, D.A. Ayupova, O.N. Larskaya, O.N. Nikolaeva, G.I. Petrovicheva, A.I. Zabotin, Physiologically active oligosaccharides, accumulating in the roots of winter wheat during adaptation to low temperature, Russian Journal of Plant Physiology 45 (1998) 262] oligosaccharin (physiologically active oligosaccharide) GXAG, which stimulates the acquisition of freezing tolerance in winter varieties of Triticum aestivum L. The transient accumulation of GXAG in the tissues of winter wheat correlates with the temporal activation of cell wall glycosidases during the first hours of cold acclimation (2 °C). This finding suggests that the oligosaccharin is liberated as a result of the intensification of hemicellulose turnover. At low concentrations, GXAG initiates the acquisition of freezing tolerance in winter plants, in a manner similar to ABA, even at room temperature. The resultant effect of ABA and GXAG on the freezing tolerance of winter wheat depends on the sequence of pre-treatments with these two factors. When seedlings are pre-treated with GXAG a few hours before treatment with ABA, the effect is synergistic, and its impact depends on the duration of pre-treatment with GXAG. When ABA is applied first, the resultant effect on freezing tolerance is additive. The results obtained here lead to the conclusion that oligosaccharin, accumulating during the first hours of cold acclimation, functions as a partner of ABA during the initiation of freezing tolerance acquisition in winter plants.
