Background This work is development of new molecules of isoniazid derivatives as dealing with potential of antimicrobial activity against clinical pathogens causing infectious disease. Antimicrobial of novel Mannich base derivatives can be achieved via one-pot synthesis in green chemistry approach. This method offers efficient, mild reaction conditions and high yields. In this study, totally 12 compounds (1a–l) was prepared and screened for cytotoxic and antimicrobial activities. Materials and methods Newly synthesised compounds were conformed via FT- IR, 1H, and 13C NMR (Nuclear Magnetic Resonance), and mass spectra analysis. All compounds were checked antibacterial activity against gram-positive bacteria of Enterococcus faecalis, Staphylococcus aureus and gram-negative bacteria of Pseudomonas aeruginosa, Klebsiella pneumoniae and Escherichia coli. All compounds were checked against antifungal activity against Aspergillus fumigatus, Candida albicans, Cryptococcus neoformans, Aspergillus niger, and Microsporum audouinii. All compounds were screened for cytotoxic activity against the MCF-7 (Michigan Cancer Foundation-7) cancer cell line. Result The compound 1g was highly (MIC: 0.25 μg/mL) active against gram-negative bacterial of P. aeruginosa, whereas other compounds 1e and 1h were more active (MIC: 2 μg/mL) in K. pneumoniae and also 1g (MIC: 2 μg/mL) was more active in E. faecalis than standard ciprofloxacin. Antifungal screening, the compound 1b was highly active (MIC: 0.25 μg/mL) against C. albicance, 1g (MIC: 2 μg/mL) and 1h (MIC: 4 μg/mL) was significant of active against A. fumigatus, and the compound 1c (MIC: 4 μg/mL) was extremely active in M. audouinii than clotrimazole. Compound 1g (GI50 = 0.01 μM) exhibited high activity against the MCF-7 cell line, while 1b (GI50 = 0.02 μM) was equipotent active compared with standard doxorubicin. Conclusion A novel set of isoniazid derivatives (1a–l) and 1h were synthesized and screened for antimicrobial and cytotoxic activities. We found some highly active molecules, which are evidencing to be a potential treatment of bacterial and fungal infection candidates.

The shrimp Penaeus monodon was used for the isolation of digestive enzyme producing host-associated probiotic bacteria. Gut was isolated from a healthy animal completely and morphologically different bacterial isolates were screened for the production of hydrolytic enzymes, such as, protease, amylase, lipase and cellulases. Based on their ability to produce enzymes, the potent probiotic bacteria were identified as Bacillus subtilis and B. licheniformis and these two were used for the preparation of probiotic diet for experimental trials. Probiotic diet was prepared by mixing the shrimp feed with 2 g probiotic/100 g artificial diet (F1), 4 g/100 g (F2), 6 g/100 g (F3), 8 g/100 g (F4) and 10 g/100 g (F5). Juvenile shrimp was fed with probiotic and control diet for a period of 7 weeks at 5 and 8% body weight for the first 3 and 7 weeks, respectively. After seven weeks, whole gut was dissected out and protease activity was estimated as 145 ± 12.3 U/g in control animal and increased as 710 ± 15.2 U/ g in F5 feed groups. Amylase activity was 139 ± 10.4 U/g in control and increased as 209 ± 13. 3 U/g in F5 group. Cellulase activities were 171 ± 9.3 in F5 groups and the control group showed only 102 ± 12.4 U/g. Lipase activity was 78 ± 3 U/g in F1 groups and it increased as 85 ± 5 U/g in F3 groups. These findings indicate the potential of host-associated bacteria to enhance the production of enzymes in the gut of juvenile P. monodon.

Background: Environmental exposure to toxicants poses high risk to develop reproductive and developmental chronic toxicity in man. Toluene is one of the commonest industrial agents whose exposure is attributed with potential to induce reproductive and developmental toxicity. Since they contaminate the immediate environment of air and water to which humans are exposed, its containment is of great public health importance. Conventional treatment modalities fail owing to the difficulty to detect these highly volatile agents in environment and human body. The peril of such hazardous exposures is evident only when irreversible structural and functional damages have incurred. In such instances, prevention gains an upper hand when compared to therapeutic interventions. Several natural compounds derived from medicinal herbs possess potential to curb toxicities induced by such xenobiotic agents. Among them Boerhavia diffusa Linn. is a widely distributed and common herb attributed with antitoxic potential and capability for antioxidant defence. A study was performed on the prophylactic efficacy of aqueous extract of B. diffusa in curbing toluene induced developmental toxicity in Drosophila melanogaster. Methods: The study consisted of a preliminary phytochemical screening and HPTLC profiling of B. diffusa aqueous extract (BDAE). LC50 of toluene was assessed and a sublethal dose of 200 ppm was fixed for the study. Four doses of BDAE; 25, 50, 100 and 200 mg/ml designated as Low dose, medium dose 1, medium dose 2 and high dose was used for the study. The parameters used for the study included the determination of larval period, pupal period, percentage of egg hatching, morphometric analysis of egg, larvae, pupae and adults, fertility, fecundity, lifespan and levels of antioxidant enzymes such as catalase, glutathione-S-transferase and superoxide dismutase. Results: The phytochemical and HPTLC characters were as per the pharmacopoeial standards. LC50 of toluene was found to be 430 ppm in this study. BDAE at medium dose 2 and high dose significantly prevented the deterioration of reproductive and developmental toxicity parameters of larval period, pupal period, percentage of egg hatching, morphometric characters of larva, pupa and adult, fertility, fecundity and lifespan in drosophila. Also the drug significantly elevated the levels of antioxidant enzymes. Conclusion: Toluene exposure during lifetime is inevitable. B. diffusa, equipped with its rich active ingredients prevented toluene induced developmental and reproductive toxicity in Drosophila. This medicinal herb provides a ray of hope in preventing environmental toxin induced reproductive and developmental toxicity. A novel aluminium (Al) and its active alloys are extensively been used in nearly all areas owing to their cost-effectiveness. But when it is subjected to an aqueous medium, gets corroded through a chemical response. In this paper, a novel framework was fabricated by copolymer coating over on Al and loaded with zinc via electro polymerization and electrodeposition method ([EDA- OPDA]Al@Zn). The as-fabricated composite has emerged for the sorption of Methylene Blue (MB) aqueous dye and Paracetomal drug (PAR). The as-fabricated composite framework has been categorized via IR spectra, FE-SEM images, and EDX spectra. The sorption progression was optimized for numerous prompting features like pH, contact time and impact of dosage. Based on kinetics data, the growth in QE value by an enhancement in temperature for adsorption and the higher r values shows the adsorption progression is a pseudo-second-order model. The thermodynamic constraints specify that the field of adsorbate is impulsive and typical endothermic process. Instead, the corrosion resistance of a composite in the 3.5% of NaCl. Polycyclic aromatic hydrocarbons (PAHs) pollution occurs in freshwater and marine environment by anthropogenic activities. Moreover, analysis of the PAHs-degradation by the indigenous bacterial strains is limited, compared with other degraders. In this study, naphthalene (NAP) biodegrading bacteria were screened by enrichment culture method. Three bacterial strains were obtained for NAP degradation and identified as Bacillus cereus CK1, Pseudomonas aeruginosa KD4 and Enterobacter aerogenes SR6. The amount of hydrogen, carbon, sulphur and nitrogen of wastewater were analyzed. Total bacterial count increased at increasing incubation time (6-60 days) and moderately decreased at higher NAP concentrations. The bacterial population increased after 48 days at 250 ppm NAP (519 ± 15.3 MPM/mL) concentration and this level increased at 500 ppm NAP concentration (541 ± 12.5 MPM/mL). NAP was degraded by bacterial consortium within 36 h-99% at 30 °C. PAHs degrading bacteria were grown optimally at 4% inoculum concentrations. Bacterial consortium was able to degrade 98% NAP at pH 7.0 after 36 h incubation and degradation potential was improved (100%) after 34 h (pH 8.0). Also at pH 9.0, 100% biodegradation was registered after 36 h incubation. When the agitation speed enhanced from 50 ppm to 150 ppm, increased bacteria growth and increased NAP degradation within 42 h incubation. Among the nutrient sources, beef extract, peptone and glucose supplemented medium supported complete degradation of PAHs within 30 h, whereas peptone supported 94.3% degradation at this time. Glucose supplemented medium showed only 2.8% NAP degradation after 6 h incubation and reached maximum (100%) within 42 h incubation. Bacterial consortium can be used to reduce NAP under optimal process conditions and this method can be used for the removal of various hydrocarbon-compounds.

Objectives: Mycosynthesis offers a benevolent alternative for the synthesis of Silver Nanoparticles (AgNPs) when compared to the conventional physical and chemical methods. Fungi secrete enormous amounts of proteins and enzymes that act as reducing and capping agents in AgNPs synthesis. Methods: AgNPs were synthesized using extracellular fungal extract of Fusarium incarnatum. The reaction mixture was optimized using different parameters to obtain better yield. Anti-melanogenic activity of fungal extract, AgNPs and standard drug (Kojic acid) was evaluated against Human skin melanoma SK-MEL-3 cells. AgNPs were synthesized at 30 °C, pH 5 and 1 mM AgNO3 concentration. Then optimization and the characterization were done by UV–Vis, FTIR, XRD, Zeta potential and H-TEM techniques. Results: The average size of AgNPs was 10 nm and predominantly spherical in shape. Myco-synthesized AgNPs showed significant anti-melanogenic activity against human skin melanoma SK-MEL-3 cells with an IC50 value of 17.70 μg/ml. The concentration of AgNPs required to inhibit SK-MEL-3 cells was lower than the concentrations of F. incarnatum extract and standard Kojic acid. Conclusion: Spherical AgNPs with and average size of 4 nm were synthesized using fungal extract of F. incarnatum. Mycosynthesized AgNPs can be considered as effective anti-melanogenic agents owing to their impressive activity against skin melanoma.

ZnGa2O4 nanocomposites have been widely used for photocatalytic degradation of industrial dyes. In this work, ZnGa2O4 was synthesized from zinc sulphate heptahydrate ZnSO4.10H2O and Gallium (III) oxide (Ga2O3) by hydrothermal method. As prepared, ZnGa2O4 nanocomposites was used as a photocatalyst degradation of three organic dyes rhodamine-B, methylene blue, and methyl orange, under ultraviolet (UV) light irradiation. The ZnGa2O4 nanocomposites structure, morphology, size and optical properties were studied by X-ray diffraction (XRD), Fourier transform Raman spectroscopy (FT-Raman), scanning electron microscopy (SEM), Transmission electron microscopes (TEM) and photoluminescence spectra (PL). Moreover, the results explained the rate-controlling mechanisms of the dye degradation process followed by second-order kinetics. After 100 min of adsorption kinetic models, the decomposition of rhodamine-B (7.2 Ct mg/L, 5.2 Ct mg/L, and 4.1 Ct mg/L), methylene blue (42.8 qt mg/g, 44.8 qt mg/g, and 45.9 qt mg/g), and methyl orange (42.8 qe mg/g, 44.8 qe mg/g, and 45.9 qe mg/g) respectively. This investigation study offers a promising method to design more efficient ZnGa2O4 nanocomposites based photocatalytic degradation of industrial organic dyes.

Objective Abutilon indicum is a medicinal plant belonging to the Malvaceae family. The current study has been developed to detect Abutilon indicum bio-activity to produce an adequate drug design for cancer. Methods The objective of this work is to perform molecular docking and dynamics as well as inhibitors and cancer cell line studies of Abutilon indicum would be essentially effective to use current strong medicines from oncology therapies. Results By Docking best finding binding energy -12.02kcal/mol (ARG310, ASP323, SER291, THR358, GLU293) amino acid has been found to be immersed in the formation of the hydrogen interaction. This finding also indicates that a range of compounds are ADMET positive drug molecules in cancer studies. Network pharmacology showed that the signal rule ERG, PTEN, NKX31, AR, ETV4, STAT3, PTPN11, CBL, KRAS, EREG, STAT3, GRB2, HRAS, and SHC1, and the axis of DRD2. Molecular simulation trajectories show that RMS deviation profiles were relatively stable during the simulation and it indicated the orientations were created by the docking studies. In cell lines MCF-7, the active compound R-N-1'-methoxycarbonyl-2'-phenylethyl-4-hydroxy benzamide has anticancer inhibitory 76.56% at 100μg/mL. The ASP323 interaction of EGFR inhibitors interaction molecules were derived that can be successfully used to explain the cancer activities. Conclusion The results of pharmacodynamic and toxicity for natural organic derived compound and its active results epidermal growth factor receptor for identifying novel drugs for the treatment confirms compound moderate to a good cancer drug.

Public requirements encouraged by the current asset framework drive industry to expand its general effectiveness by enhancing existing procedures or finding new uses for waste. Thus, the aim of this study was the isolation, fabrication, and characterization of pectin derived from jackfruit (Artocarpus heterophyllus) peels and the generation of hybrid of pectin (P)/apatite (HA) (P/HA) bionanocomposites. In this process, the natural pectin polymer derived from the peel of jackfruits was used in different concentrations for the fabrication of HA bionanocomposites. Characterization of the isolated pectin and bionanocomposites samples was performed with 1H NMR and 13C NMR, FTIR, XRD, SEM-EDX, and HR-TEM. Cytocompatibility, ALP, fibroblast stem cells, anti-inflammatory and cell adhesion testing of the fabricated bionanocomposites was showed good biocompatibility. Our results signify that the fabricated bionanocomposites might be applicable as bone graft materials.

Heavy metal ions in aqueous solutions are taken into account as one of the most harmful environmental issues that ominously affect human health. Pb(II) is a common pollutant among heavy metals found in industrial wastewater, and various methods were developed to remove the Pb(II). The adsorption method was more efficient, cheap, and eco-friendly to remove the Pb(II) from aqueous solutions. The removal efficiency depends on the process parameters (initial concentration, the adsorbent dosage of T-Fe3O4 nanocomposites, residence time, and adsorbent pH). The relationship between the process parameters and output is non-linear and complex. The purpose of the present study is to develop an artificial neural networks (ANN) model to estimate and analyze the relationship between Pb(II) removal and adsorption process parameters. The model was trained with the backpropagation algorithm. The model was validated with the unseen datasets. The correlation coefficient adj.R2 values for total datasets is 0.991. The relationship between the parameters and Pb(II) removal was analyzed by sensitivity analysis and creating a virtual adsorption process. The study determined that the ANN modeling was a reliable tool for predicting and optimizing adsorption process parameters for maximum lead removal from aqueous solutions.

Dengue virus (DENV) has emerged as a major economic concern in developing countries, with 2.5 billion people believed to be at risk. Vascular endothelial cells (ECs) lining the circulatory system from heart to end vessels perform crucial functions in the human body, by aiding gas exchange in lungs, gaseous, nutritional and its waste exchange in all tissues, including the blood brain barrier, filtration of fluid in the glomeruli, neutrophil recruitment, hormone trafficking, as well as maintenance of blood vessel tone and hemostasis. These functions can be deregulated during DENV infection. In this study, BALB/c mice infected with DENV serotype 2 were analyzed histologically for changes in major blood vessels in response to DENV infection. In the uninfected mouse model, blood vessels showed normal architecture with intact endothelial monolayer, tunica media, and tunica adventitia. In the infected mouse model, DENV distorted the endothelium lining and disturbed the smooth muscle, elastic laminae and their supporting tissues causing vascular structural disarrangement. This may explain the severe pathological illness in DENV-infected individuals. The overall DENV-induced damages on the endothelial and it's supporting tissues and the dysregulated immune reactions initiated by the host were discussed.

Taxodium is a deciduous conifer belong to family Cupressaceae, commonly known as cypresses. In the present study, two species of Taxodium trees from four locations in Egypt were investigated. Bald cypress and pond cypress as plant genetic resources were surveyed and tested for horticultural and morphological traits using biochemical and molecular genetic techniques. Leaf essential oils (EOs) were analyzed by gas chromatography coupled with mass spectrometry (GC/MS), subjected to bioassay for insecticidal activity against the rice weevil (Sitophilus oryzae L.) and the red flour beetle (Tribolium castaneum (Herbst)), and compared with three monoterpenes (D-limonene, camphor, and carvone). T. distichum var. distichum (bald cypress) growing in the drylands in the Alexandria, El-Beheira, Giza 1, and Qalyubia governorates had lower root hydrogen peroxide (H2O2), proline content, and leaf peroxidase activity than T. distichum var. imbricarium (pond cypress, Giza 2), growing in swamplands of the Giza governorate. Forty random amplified polymorphic DNA (RAPD-DNA) primers and 20 simple sequence repeats (SSRs) indicated 57.41% and 64.50% polymorphism, respectively, between the Taxodium species. The main constituents of leaf EOs from trees growing in Alexandria were germacrene D (14.71%), borneol acetate (6.81%), ledene oxide-(II) (6.13%), and trans(β)-caryophyllene (6.13%); thujopsene (18.27%), 3-carene (14.49%), and cedrol (8.02%) were found in trees from Qalyubia; thujopsene (19.93%), α-caryophyllene oxide (17.32%), and cedrol (10.87%) from trees in Giza 1, α-pinene (63.80%) and thujopsene (10.15%) from trees in Giza 2, and El-Beheira α-pinene (26.66%), thujopsene (22.42%), and cedrol (11.30%) in trees from El-Beheira. S. oryzae was more susceptible than T. castaneum to the EOs from leaves of Taxodium trees from the four localities. Taxodium oil from El-Beheira followed by carvone and EOs from trees grown in Alexandria showed the lowest LC50 against S. oryzae with values of 50 μL/L, respectively. Carvone, Taxodium EOs from El-Beheira and Qalyubia exhibited the lowest LC50 against T. castaneum with values of 53.37, 66.40, and 72.54 μL/L, respectively.

This work was conducted to evaluate the ability of grape molding fungus; Penicillium citrinum to synthesize silver nanoparticles (Ag NPs). The potency of biosynthesized Ag NPs was checked against the aflatoxigenic Aspergillus flavus var. columnaris, isolated from sorghum grains. Biosynthesized Ag NPs were characterized and confirmed in different ways. X ray diffraction (XRD), Energy Dispersive Spectroscopy (EDS), Transmission Electron Microscopy (TEM) and optical absorption measurements confirmed the bio-synthesis of Ag NPs. The in vitro antifungal investigation showed that biosynthesized Ag NPs were capable of inhibiting the growth of aflatoxigenic A. flavus var. columnaris. Utilization of plant pathogenic fungi in the Ag NPs biosynthesis as well as the use of bio-Ag NPs to control fungal plant diseases instead of chemicals is promising. Further work is needed to confirm the efficacy of the bio-Ag NPs against different mycotoxigenic fungi and to determine the potent applicable doses.
