This article reports on the dataset gathered following the census of 83 present-day Infralittoral Prograding Wedges (IPWs), surveyed on the inner continental shelf of the Central-Eastern Tyrrhenian Sea. The purpose of the census was to explore their bathymetric range and assess the observational laws governing this variability. The ensued dataset (Campania Region IPW Dataset, CRID) includes geographic, topographic and morpho-bathymetric indices, descriptive of each IPW and more, the exposure of each IPW to wave forcing (Geographical fetch, Effective fetch and extreme significant wave height, HS). In this work, histograms contribute to describe all the variables and highlight the dominant features of each IPW. Location maps univocally links the geographic position of each IPW to the appropriate attribute record in the dataset. Further, thematic maps illustrate eight wave fields obtained by offshore-to-nearshore transformation by as many sea states scenarios with 200-year return period. Such wave fields are used as sources for significant wave height representing wave conditions over each IPW. This dataset could be implemented with new measures at a broader scale, by following analogue procedures for measurements, to enlarge the observational scale on IPWs and improve the numerical models which might eventually derive by the analysis of this dataset.

With the aim to develop alternative therapeutic tools for the treatment of resistant cancers, here we propose targeted Pluronic(®) P123/F127 mixed micelles (PMM) delivering niclosamide (NCL) as a repositioning strategy to treat multidrug resistant non-small lung cancer cell lines. To build multifunctional PMM for targeting and imaging, Pluronic(®) F127 was conjugated with biotin, while Pluronic(®) P123 was fluorescently tagged with rhodamine B, in both cases at one of the two hydroxyl end groups. This design intended to avoid any interference of rhodamine B on biotin exposition on PMM surface, which is a key fundamental for cell trafficking studies. Biotin-decorated PMM were internalized more efficiently than non-targeted PMM in A549 lung cancer cells, while very low internalization was found in NHI3T3 normal fibroblasts. Biotin-decorated PMM entrapped NCL with good efficiency, displayed sustained drug release in protein-rich media and improved cytotoxicity in A549 cells as compared to free NCL (P

The input flow of groundwater from the seabed to the coastal ocean, known as Submarine Groundwater Discharge (SGD), has been only recently recognized as an important component of continental margin systems. It potentially impacts physical, chemical and biological marine dynamics. Independently of its specific nature (seepage, submarine springs, etc.) or fluid chemical composition, a SGD is generally characterized by low flow rates, hence making its detection and quantification very difficult, and explaining why it has been somewhat neglected by the scientific community for a long time. Along with the growing interest for SGDs emerged the need for in-situ observations in order to characterize in details how these SGDs behave. In this work, we describe the morphology of a pockmark field, detected in the Southern Tyrrhenian Sea (Mediterranean Sea), and provide observational evidences of the presence of active submarine springs over the coastal shelf area. We describe the effect of the fluid seeps on the water column stratification close to the main plumes and in the neighbouring areas, providing quantitative estimates of the intensity of the turbulent mixing and discussing their potential impact on the seabed morphology and pockmark formation in the context of turbulent buoyant plumes analytical modelling.

The introduction of total mesorectal excision (TME) and preoperative multimodality treatment have substantially improved the management of rectal cancer reducing local recurrence and increasing sphincter-saving surgery; distant metastases however remain a clinical challenge. Besides, although surgery remains the mainstay for cure of rectal cancer with the multimodality approach (chemotherapy, radiotherapy and surgery) being the standard of care for the majority of rectal cancer patients, there is a need of individualized risk-adapted treatment schemes based on clinico-pathological features because of treatment-induced morbidity and quality of life deterioration. This short viewpoint describes the emerging strategies addressing all these issues.

Background/Aims : Studies on non-alcoholic fatty liver disease (NAFLD) have included chronic liver damage attributed to various causes. Our investigation was held to observe the main clinical, histological, and pathophysiological aspects of NAFLD in patients not exposed to any known cause of chronic liver disease. Methods : We evaluated, in 84 in-patients (male/female, 66/18; median age, 36 years), the clinical and biochemical characteristics of NAFLD, and particularly its association with diabetes, dyslipidemia, hyperinsulinemia and/or with the increase of parameters of oxidative stress (blood levels of malonyldialdehyde, 4-hydroxynonenal and total plasma antioxidant capacity). Results : Ninety percent of patients had an increased body mass index (BMI), 35% had dyslipidemia, 40% had sub-clinical diabetes (only 3% had overt diabetes), 60% had hyperinsulinemia, and more than 90% had enhanced levels of lipid peroxidation markers. In 48 patients who had consented to liver biopsy, we found: 14 with simple steatosis, 32 with steatohepatitis, and two with cirrhosis. Conclusions : Our data indicate that in our country, NAFLD may occur in young males with an increased BMI, with or without hyperinsulinemia, dyslipidemia and diabetes, generally associated with disorders of redox status, and that it may be differentiated from steatosis to steatohepatitis or cirrhosis only with a liver biopsy.

The sandy deposits from dredging can have negative effects on the environment such as increase in suspended solids in the water column and their consequent transport. An experimental study was conducted to characterize water masses, dynamics, and sedimentation rates on the Ligurian continental shelf (Italy), where both a sand deposit, that could be used for beach nourishment, and a nearby Posidonia oceanica meadow coexist. The environmental plan provides a mathematical simulation of the sediment-dispersion to evaluate the possible impact on the meadow. It has been calculated that the dredging could double the concentration of suspended particles, but its scheduling will preclude a sediment accumulation. All the information obtained from this work will be used to study the environmental feasibility of the sand deposit exploitation and as starting point for drawing up the monitoring plan in case of dredging.

We investigated the ability of the histone deacetylase (HDAC) inhibitor trichostatin A (TSA) to interact with gemcitabine (GEM) in inducing pancreatic cancer cell death. The combined treatment with TSA and GEM synergistically inhibited growth of four pancreatic adenocarcinoma cell lines and induced apoptosis. This effect was associated with the induction of reactive oxygen species (ROS) by GEM, increased expression of the pro-apoptotic BIM gene by both TSA and GEM and downregulation of the 5′-nucleotidase UMPH type II gene by TSA. The expression of other genes critical for GEM resistance (nucleoside transporters, deoxycytidine kinase, cytidine deaminase, and ribonucleotide reductase genes) was not affected by TSA. The functional role of ROS in cell growth inhibition by GEM was supported by (i) a significantly reduced GEM-associated growth inhibition by the free radical scavenger N-acetyl-l-cysteine, and (ii) a positive correlation between the basal level of ROS and sensitivity to GEM in 10 pancreatic cancer cell lines. The functional role of both Bim and 5′-nucleotidase UMPH type II in cell growth inhibition by TSA and GEM was assessed by RNA interference assays. In vivo studies on xenografts of pancreatic adenocarcinoma cells in nude mice showed that the association of TSA and GEM reduced to 50% the tumour mass and did not cause any apparent form of toxicity, while treatments with TSA or GEM alone were ineffective. In conclusion, the present study demonstrates a potent anti-tumour activity of TSA/GEM combination against pancreatic cancer cells in vitro and in vivo, strongly supporting the use of GEM in combination with an HDAC inhibitor for pancreatic cancer therapy.

Ice surface temperature (IST) imagery from the thermal infrared bands of the satellite-borne Moderate Resolution Imaging Spectroradiometer (MODIS) has been used to analyze the activity of the Terra Nova Bay polynya from 2005 to 2010. In clear sky conditions, the shape of the open water area of the polynya (i.e. the portion of the ocean free of sea-ice) is revealed by the skin sea-ice temperatures close to the water freezing point provided by the MODIS-derived IST product. The swaths of the MODIS sensors onboard Terra and Aqua satellites over the Terra Nova Bay consist of about 5500 scenes per year, useful to estimate the cloud cover too. The open water area was extracted from clear sky scenes and from scenes affected by fog but providing continuity of the shape of the polynya. Using about 40% of the available swaths, the polynya was detected for more than 80% of the sea-ice period with a revisit time below 24 h and for more than 50% with a revisit time below 3 h. The open water area was extracted using thresholds of IST based on the IST values detected within the open water area and within the band of sea-ice surrounding the polynya, obtained scene by scene. An ice free area larger than 100 km2 was observed near the Nansen Ice Shelf for more than 70% of time (larger than 500 km2 for 50% of time). The mean annual open water area, around 900 km2, was close to the lower limit of the reported range except during 2006, around 600 km2, due to the limited activity in the second half of the cold season. The largest extents, between 6000 and 8500 km2, occurred in different periods of the Antarctic winter and exceeded maxima estimated in previous studies by thermal infrared data. Polynya openings developed from a few hours to several days, in the latter case sustained by subsequent katabatic events visible in the IST sequence. Peaks of growing rate above 300 km2/h were observed during intense katabatic events with gusts of 50 m/s. The fluctuations of the open water area resulted well correlated with wind records available from local Automatic Weather Stations within the main katabatic routes, and followed katabatic events at intervals between 0 and 6 h. The analysis of the IST data set derived from the thermal infrared bands of MODIS provides new polynya observations of high horizontal resolution (1 km) and frequent revisit time during the winter season, complementing previously available observations with passive microwave imagery.

We have previously shown that cancer cells can protect themselves from apoptosis induced by type 1 interferons (IFNs) through a ras-->MAPK-mediated pathway. In addition, since IFN-mediated signalling components STATs are controlled by PPAR gamma we studied the pharmacological interaction between recombinant IFN-beta and the PPAR-gamma agonist troglitazone (TGZ). This combination induced a synergistic effect on the growth inhibition of BxPC-3, a pancreatic cancer cell line, through the counteraction of the IFN-beta-induced activation of STAT-3, MAPK and AKT and the increase in the binding of both STAT-1 related complexes and PPAR-gamma with specific DNA responsive elements. The synergism on cell growth inhibition correlated with a cell cycle arrest in G0/G1 phase, secondary to a long-lasting increase of both p21 and p27 expressions. Blockade of MAPK activation and the effect on p21 and p27 expressions, induced by IFN-beta and TGZ combination, were due to the decreased activation of STAT-3 secondary to TGZ. alone also increased p21 and p27 expression through STAT-1 phosphorylation and this effect was attenuated by the concomitant activation of IFNbeta-induced STAT-3-activation. The combination induced also an increase in autophagy and a decrease in anti-autophagic bcl-2/beclin-1 complex formation. This effect was mediated by the inactivation of the AKT-->mTOR-dependent pathway. To the best of our knowledge this is the first evidence that PPAR-gamma activation can counteract STAT-3-dependent escape pathways to IFN-beta-induced growth inhibition through cell cycle perturbation and increased autophagic death in pancreatic cancer cells. Purpose To assess the safety and efficacy of oxaliplatin (OXA) plus dual inhibition of thymidilate synthase during preoperative pelvic radiotherapy (RT) in patients with poor prognosis for rectal carcinoma. Methods and Materials Sixty-three patients with the following characteristics, a clinical (c) stage T4, cN1-2, or cT3N0 of ≤5 cm from the anal verge and/or with a circumferential resection margin (CRM) of ≤5 mm (by magnetic resonance imaging), received three biweekly courses of chemotherapy with OXA, 100 mg/m 2 ; raltitrexed (RTX), 2.5 mg/m 2 on day 1, and 5-fluorouracil (5-FU), 900 mg/m 2 (31 patients) or 800 mg/m 2 (32 patients); levo-folinic acid (LFA), 250 mg/m 2 on day 2, during pelvic RT (45 Gy). Pathologic response was defined as complete pathological response (ypCR), major (tumor regression grade(TRG) 2 to 3, with ypCRM-ve and ypN-ve) or minor or no response (TRG4 to -5, or ypCRM+ve, or ypN+ve). Adjuvant 5-FU/LFA regimen was given in cases of cT4, ypN+ve, or ypCRM+ve. Results Overall, neutropenia (40%) and diarrhea (13%) were the most common grade ≥3 toxicities, and tolerability was better with a 5-FU dose reduction. No significant difference in pathologic response was seen according 5-FU dosage: overall, a ypCR was obtained in 24 (39%) patients, and a major response in 20 (32%) patients. The 5-year probability of freedom from recurrence was 80% (95% confidence interval, 68%–92%); it was 56% for the minor/no response group, while it was around 90% for both the ypCR and the major response group. Conclusions OXA, RTX, and 5-FU/LFA administered during pelvic RT produced promising early and long-term results in rectal carcinoma patients with poor prognosis. The postoperative treatment strategy applied in our study supports the risk-adapted approach in postoperative management.

Data from three Italian CLIMA project cruises between 1997 and 2003 were used to obtain sections of the hydrographic and chemical properties of the main water masses across the shelf break off Cape Adare (western Ross Sea, Antarctica). Dissolved oxygen, nitrate and phosphate data were combined on the basis of the Redfield ratio to obtain the quasi-conservative tracers NO (9[NO3]+[O2]), PO (135[PO4] + [O2]) and phosphate star PO?4 (PO?4 1?4 [PO4] + [O2]/175-1.95). In 1997 and 2003 the presence of the High Salinity Shelf Water at the bottom depth near the sill was traced by both physical and chemical measurements. In 2001 the Modified Shelf Water, characterized by warmer temperature and by a lower dissolved oxygen content than High Salinity Shelf Water, was observed at the shelf edge. The distribution of the chemical tracers together with the hydrographic observations showed recently formed Antarctic Bottom Water on the continental slope during all of the cruises. These observations were confirmed by the extended optimum multiparameter analysis. The calculated thickness of the new Antarctic Bottom Water, as well as the tracer content, were variable in time and in space. The estimated volume of the new Antarctic Bottom Water and the export of dissolved oxygen and nutrient associated with the overflowing water were different over the examined period. In particular, a lower (?55%) export was evidenced in 2001 compared to 1997.

The hydrography of intermediate and deep-water masses in the Northern Ionian Sea (Apulian Plateau) was studied through four quasi-synoptic multidisciplinary surveys carried out in 2004–2006 as an ancillary oceanographic activity in the frame of the APLABES project. This area plays a crucial role for the entire Mediterranean Basin, being influenced by the water outflow of Adriatic origin, which, under severe winter conditions, is a primary source of dense water for the Eastern Mediterranean. At the end of the 1980s such outflow showed a different behavior, and only in the recent years has a gradual re-establishing of the former condition been detected. As such, the Adriatic Sea has regained its role as a main source of the East Mediterranean Deep Water, which was temporarily inhibited during the well-known Eastern Mediterranean Transient which progressively modified the intermediate and deep layers of the Mediterranean Sea. The general structure of water masses was similar through the investigated period, but interesting differences within the bottom layer have been detected. The interaction of the different water types present in the basin is reviewed by means of property–property plots, vertical sections, isopycnal analyses and using an Optimum Multiparameter Analysis (OMP), which is an objective method to measure the mixing of water masses. Due to the lack of any direct information about the dynamics of the water column in the area of the Apulian Plateau during the whole analyzed period, the classical method to infer the baroclinic velocity from the mass field has been applied to hydrographic data. The well-known indeterminacy of this method due to the barotropic component of the velocity field has been resolved using a short time series of current velocities acquired synoptically by a mooring located in the northern part of the studied area. The wavelet transform was adopted for localizing and quantifying the variability of currents simultaneously in both frequency and time domains. The presence of the Adriatic Deep Water close to the bottom was detected on all four surveys, with different signature as underlined by the objectively analysis (with the Optimum Multiparameter Analysis) of the thermohaline field. A core of cold, less-saline and oxygenated water of Adriatic origin coming from the Otranto Channel was identified. This water mass moved in geostrophic balance along the isobaths at 600–1000 m depth at the isopycnal surface of 29.18 kg m−3, not being dense enough to reach the deeper layers of the Ionian Basin, carrying 0.27–0.36 Sv.

The Ross Sea is an important area for the ventilation of the deep layers of the Southern Ocean (e.g. [Jacobs, S.S., Fairbanks, R.G., Horibe, Y., 1985. Origin and evolution of water masses near the Antarctic continental margin: evidence from H218O/H216O ratios in seawater. In: Jacobs, S.S. (Ed.), Oceanology of the Antarctic Continental Shelf. Progress in Oceanography 109, 43–55]). These processes are driven by the atmospheric forcing which, at high latitude, plays a key role in the formation and thickness of sea ice. In order to investigate the effect of the atmospheric forcing variability at different time scales, we analysed the surface heat budget over the Ross Sea continental shelf and in Terra Nova Bay (TNB) polynya, using analyses for the period 1990–2006 provided by European Centre for Medium-range Weather Forecast (ECMWF). This study was also performed using thermohaline data collected within the activities of Climatic Long-term Interaction for the mass-balance in Antarctica project of the Italian National Programme for Antarctic Research for the summer periods from 1994 until 2001. The annual average of the heat budget over the continental shelf of the Ross Sea estimated in the period 1990–2006 shows an interannual variability ranging between −97 and −123 W m−2. Assuming that the heat loss must be compensated by the sensible heat carried by the Circumpolar Deep Water we estimated its transport (3.1 Sv) and its variability (0.2 Sv). Similarly in the TNB polynya the heat loss reaches its maximum in 2003 (−313 W m−2) and its minimum (−58 W m−2) in 1996. The related production of sea ice and the High Salinity Shelf Water (HSSW) were also estimated. The HSSW production switched from the lowest values during the first 10 years of the investigated period (1990–2000) to the highest values for the remaining period (2001–2006). The thermohaline characteristics of the water column in TNB show a general decrease in salinity with a superimposed variability. Comparison between the estimated HSSW production and the salinity observed within the TNB water column show similar tendency in the last years after 2002, while during the period 1995–1998 the behaviour is different. Our hypothesis concern a possible role of the CDW inflow in the TNB area and our results could be explained by a different contribution of CDW transport and HSSW production to the salt content within the water column.

During July and December 2005 two basin-scale hydrographic cruises were performed in the Tyrrhenian basin to monitor hydrographic conditions after the significant changes observed during previous decades. A detailed investigation of the surface layer suggested the presence of a mesoscale structure in the central basin, off Naples. Its principal feature is a subsurface cold lens of Western Intermediate Water, a water mass produced in the Northwestern Mediterranean during the winter season. The lens, which appears as an anticyclonic eddy, was detected for the first time in July. Five months later an eddy signature was detected in the same position with the same hydrographic characteristics. The positive altimetric anomaly in correspondence with the observed structure suggests that the anticyclonic circulation lasted for a long period and its persistence was related to the presence of a seamount in the region. For the first time the importance of isolated topography in influencing the interior Tyrrhenian circulation is highlighted: the Vavilov Seamount, jointly with the weak mean current, seems to be responsible for the persistence of eddy structures in the region. The topographic effect is discussed on the basis of the eddy properties and theoretical findings.
