Herein, the first study is presented using 4D synchrotron X-ray microtomography to capture all stages of crack development in neutron irradiated and radiolytically oxidised nuclear graphite. Employing a novel loading setup, specimens of Gilsocarbon graphite, both unirradiated and irradiated at 301 °C to 19.7 × 1020 neutrons/cm2 (∼2.6 displacements/atom (dpa)), were loaded to generate a crack. All stages of the fracture process were then captured using synchrotron X-ray imaging. Reconstructed tomographic images and 3D segmented crack volumes have been used to observe and analyse the irradiation-induced evolution of the graphite microstructure as well as corresponding changes in the crack initiation, propagation, and arrest behaviour of graphite after neutron irradiation. Close examination of the applied stress-strain curves highlights the suppression of micro-crack-based damage accumulation in irradiated graphite. Moreover, as well as the crack-bridging and deflection mechanisms characteristic of unirradiated graphite, crack arrest in the irradiated graphite is shown to be significantly influenced by crack tip blunting. This change is associated with the growth of the open pore structure of graphite, specifically the enlargement and increased frequency of macro-pores, resulting from the simultaneous radiolytic oxidation of the graphite microstructure during neutron irradiation.

Property measurement of graphite extracted from operational nuclear reactors is vital for forward prediction of the graphite's properties and integrity as part of continued operation and plant life extension assessments. Limited core monitoring material is available, so robust and efficient measurement techniques are required for samples with constrained dimensions. The Young's modulus, Poisson's ratio and flexural strength of 218 beams of radiolytically oxidised nuclear graphite have been determined by combining electronic speckle pattern interferometry, ESPI, and inverse finite element modelling techniques. The graphite was extracted from three operational advanced gas cooled reactor stations where it had been subjected to fast neutron irradiation and radiolytic oxidation resulting in mass loss. The 6 × 6 × 19 mm beam samples had densities ranging from 1063 – 1840 kg.m−3, equivalent to weight losses up to 46 %, with cumulative neutron doses of up to 161 × 1020 n.cm−2 equivalent Dido nickel dose, or 21.1 displacements per atom, significantly extending the datasets available in the open literature to higher levels of both dose and oxidation. In situ ESPI measurements were collected during three-point bend tests, creating full field displacement maps of the evolving deformation to which simulations were matched by chi-squared minimisation of the elastic properties. Potential loading abnormalities were accounted for in the fitting process to ensure the simulations were representative, with total uncertainties in the determined Young's modulus and Poisson's ratio values of ± 4.5 % and ± 0.08, respectively. Consistent trends were observed in the behaviour of the material from the different stations, with strong positive dependences on density for both the flexural strength and Young's modulus. The strength and Young's modulus remained well correlated even to high levels of oxidation mass loss. Conversely, the Poisson's ratio was practically invariant with oxidation with mean values of 0.21 ± 0.03, 0.24 ± 0.05 and 0.25 ± 0.05 for the three stations. The combined ESPI-finite element measurement approach is demonstrated as robust for irradiated small specimens, making it suitable for monitoring graphite in future reactors. The collected data support the safety assessments of the current reactors, enabling predictions of the responses to progressively higher mean core mass loss.

Keyway root cracking and radiolytic weight loss in graphite moderated nuclear reactors may limit future successful operation of the civil nuclear fleet. A novel means of initiating fracture using internally generated stresses has been developed, which uses bromine intercalation to induce fracture in small graphite samples. Successful crack initiation and propagation has been achieved in 1:10 scale reactor components. In situ X-ray Computed Tomography during bromination has enabled observations of failure in real time, allowing the role of microstructure during fracture and weight loss to be determined. Crack paths predicted by models have been assessed.

It is well established that tobacco smokers have reduced levels of monoamine oxidase activities both in the brain and peripheral organs. Furthermore, extensive evidence suggests that smokers are less prone to develop Parkinson's disease. These facts, plus the observation that inhibition of monoamine oxidase B protects against the parkinsonian inducing effects of the nigrostriatal neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine, have prompted studies to identify monoamine oxidase inhibitors in the tobacco plant and tobacco cigarette smoke. Our previous efforts on cured tobacco leaf extracts have led to the characterization of 2,3,6-trimethyl-1,4-naphthoquinone, a non-selective monoamine oxidase inhibitor, and farnesylacetone, a selective monoamine oxidase B inhibitor. We now have extended these studies to tobacco smoke constituents. Fractionation of the smoke extracts has confirmed and extended the qualitative results of an earlier report [J. Korean Soc. Tob. Sci.1997, 19, 136] demonstrating the inhibitory activity of the terpene trans,trans-farnesol on rat brain MAO-B. In the present study, K(i) values for the inhibition of human, baboon, monkey, dog, rat, and mouse liver MAO-B have been determined. Noteworthy is the absence of inhibitory effects on human placental MAO-A and beef liver MAO-B. A limited structure-activity relationship study of analogs of trans,trans-farnesol is reported. Although the health hazards associated with the use of tobacco products preclude any therapeutic opportunities linked to smoking, these results suggest the possibility of identifying novel structures of compounds that could lead to the development of neuroprotective agents.

The purpose of this study was to examine the relationship between high intensity cycle ergometry when resistive forces were optimised and field tests of high intensity performance in an elite sprint trained group (n = 12). Body mass, stature and age of the group were determined prior to testing (66.3 +/- 9.8kg, 177 +/- 7.6cm and 17 +/- 0.74 yrs respectively). Field tests of high intensity performance examined included sprinting. maximal shuttle running and jumping ability (vertical and horizontal). Significant correlations existed between all field tests (P0.01). Only moderate correlations were observed for values generated on the cycle ergometer and performance tests (P0.05). The results of the present study suggest that the optimised cycle ergometer test and the performance tests are unrelated when evaluated statistically. These findings also suggest that the sprinting and jumping tests examined are highly related and may be substituted for each other as possible predictors of high intensity ability.

Traditionally, leg cycle ergometry is used to assess the power output of the lower limbs. However, it is suspected that the upper body makes a significant, albeit as yet unknown, contribution to the measured power output, and as such, the lean mass of the whole body should be considered during ergometric assessment. To test this idea, indices of mechanical power output were obtained from 11 subjects during high intensity leg cycle ergometry tests (20 second duration; 75 grams per kilogram total body mass) using two protocols: one with a standard handle-bar grip (with grip) and one with supinated wrists (without-grip). Peak mechanical power, mean mechanical power, fatigue index and total mechanical work values were calculated for each subject during each test and the sample mean differences associated with the two protocols were compared using paired Student t-tests. The with-grip protocol yielded significantly greater peak mechanical power output than the without-grip protocol (886+/-124 W and 815+/-151 W, respectively), suggesting a significant upper body contribution to the maximum power output measured for the legs. As a first step towards quantifying the upper body involvement during leg cycle ergometry, surface electromyography of the forearm musculature was measured in a twelfth subject whilst performing each of the test protocols. During the with-grip ergometer tests, the intensity of electrical activity in the forearm musculature was similar, if not greater than, the intensity of electrical activity recorded for the forearm musculature during 100% maximum voluntary hand grip-dynamometer contractions, suggesting maximum isometric-type forearm muscle contraction during the with-grip leg ergometry tests. These findings suggest that the performance of traditional-style leg cycle ergometry requires a muscular contribution from the whole body. As such, researchers should be mindful of this, both in terms of the allocation of ergometer loads, and in the analysis of blood-borne metabolites.

The purpose of this study was to compare the maximal exercise performance of 10 men during friction braked cycle ergometry of 20 s duration when resistive forces reflected total body mass (TBM) or fat free mass (FFM). Fat mass was calculated from the sum of skinfold thicknesses. Increases (P0.05) in peak power output (PPO) were found between TBM and FFM (1,015+/-165 W TBM vs 1,099+/-172 W FFM). Decreases (P0.05) were observed for the time taken to reach PPO (3.8+/-1.4 s TBM vs 2.9+/-1 s FFM). Pedal velocity increased (P0.05) during the FFM protocol (129.4+/-8.2 rpm TBM vs 136.3+/-8 rpm FFM). Rating of perceived exertion (RPE) was also (P0.05) greater for FFM (18.4+/-1.6 TBM vs 19.8+/-0.4 FFM). No changes were found for Mean Power Output (MPO), fatigue index (FI) or Work Done (WD) between trials. These findings suggest that high intensity resistive force loading protocols may need to be reconsidered. Results from this study indicate that the active tissue component of body composition needs consideration in resistive force selection when ascertaining maximal cycle ergometer power profiles.

Earthquake sources, as well as contained underground explosions and volcanic explosions, initiate atmospheric waves at the air–ground interface which propagate upward and outward. The propagating atmospheric waves produced are of two types: a high-frequency acoustic wave and a low-frequency gravity wave with horizontal wavelength much longer than its vertical wavelength. Because of the exponential decrease of atmospheric density with height, the acoustic and particularly the gravity waves can grow to significant amplitude in the upper atmosphere, where they can affect the ionosphere causing changes in the distribution of neutral and charged particles. The coherent fluctuations of electron densities and ionization layer boundaries produced by these waves can be detected by electromagnetic sounding methods and hence the occurrence and character of the disturbances can be inferred. A particular application of interest is the detection and discrimination of underground and near surface chemical explosions in a nuclear test monitoring context. Specifically, identification of the different source types is enhanced by combining seismic detection methods with detection of the ionospheric disturbances caused by explosion and earthquake sources. In this study, numerical models of non-linear gravity controlled atmospheric disturbances produced by seismic sources near the surface of the Earth are investigated in order to obtain quantitative predictions that might be used in evaluating detection methods based on gravity wave excitation. Explicit numerical integration of the non-linear finite difference equations is used to simulate the transient flows produced in a three-dimensional ARDC atmosphere. Results from the simulations agree with many results from linear theory approximations and also show non-linear characteristics similar to important gravity wave observations. Electron density changes in the ionosphere are predicted with their spatial and temporal behavior found to be particularly sensitive to the type and magnitude of the dissipative mechanisms that may occur. In the numerical examples studied, the amplitudes of the ionospheric electron density fluctuations due to the gravity waves produced by large explosions and some types of large earthquakes are predicted to be well within the range of detection using E-M ionospheric sounding methods.

Increases in water pressure, if large enough, can be sufficient to open existing fractures in a fractured rock-water system. Such large water pressure increases are expected during earthquake activity and are also produced artificially in high-pressure hydraulic fracturing and slug injection tests. In an in-situ slug injection test, the slug of water is initially released into a packed-off interval of the borehole where the contained fluid is at ambient pressures. We recognize three separate stages in the behaviour of the fracture as the water pressure decreases due to drainage, namely opening, closing and, finally, completely closed. In the first stage, the water will initially drain out along conduits between the faces of the opening fracture. As the fracture opens and grows, it can become long enough to intersect any pre-existing network of open joints and fractures. If this connection occurs, the opened fracture will stop growing and there will be further rapid flow of fluid out along these open conduits. Even if connection to an open conduit does not occur, as the pressure drops the fracture will stop growing and eventually halt. At the pressure where the fracture tip begins to close, drainage into any open conduits will cease and drainage during fracture closing will thus be by flow into the porous walls of the fracture and borehole. This effect of the conduit being shut off may manifest as a kink in the observed height decay of the slug at this pressure due to the effect of the different drainage terms. As the water pressure decreases, the fracture will continue to close until the water pressure drops below the critical pressure and the fracture becomes completely closed. This total closure occurs at the same pressure as that necessary to re-open the fracture, and has been taken equal to the minimum principal stress. This total closure of the fracture also may manifest as a kink in the observed height decay of the slug, as below this closure pressure is the third, and final, stage where drainage is at the usual low pressures controlled by the permeability of the rock just around the packed-off interval of the borehole. We develop the most general mathematical model for this phenomenon using conservation of mass of the water in the borehole-fracture system for each separate pressure stage. This model of fracture behaviour, with separate flow stages and pressure regimes, explains the physical basis for the increasingly used technique in high-pressure injection tests of measuring the so-called re-opening pressure at the sharp change in slope of the rate of change of water height vs. height. Analytic and numerical solutions yield the rate at which the water height drops during the slug test and comparison with high-pressure test data confirms the accuracy of the model. Analysis of data from Yucca Mountain, Nevada, gives values for the fracture opening pressures that indicate the presence of extensional stresses of significant magnitude and of zones of pre-existing open fracture networks in the vicinity of the proposed repository site.

The possibility of utilizing 3H-dopamine as a photo-affinity label for rat brain synaptosomes is investigated. 3H-dopamine interacts covalently with intact and disrupted synaptosomes in the absence of light, but the interaction increases as much as fourfold following flash photolysis with ultraviolet light. The photolytic interaction in intact, but not disrupted, synaptosomes is inhibited by benzotropine and cocaine, but not by haloperidol. It is concluded that a major photolytic reactive site in synaptosomes is that associated with dopamine reuptake.
