18. Therapeutic effect of fastigial nucleus stimulation is mediated by the microRNA-182 & microRNA-382/BDNF signaling pathways in the treatment of post-stroke depression

Abstract

The deregulation of Brain-Derived Neurotrophic Factor (BDNF) was reported to be responsible for the development of post-stroke depression (PSD), while the stimulation of the fastigial nucleus (FN) can be used to treat PDS by down-regulating the expression of miR-182 and miR-382. Therefore, we aim to test the hypothesis that the therapeutic effect of FN stimulation obtained in the treatment of PSD is mediated by the miR-382miR-182/BDNF mRNA signaling pathways. Rat models of PSD were established and divided into sham, stroke, PSD and PSD + FNS groups to receive different treatments. Post-stroke depression-like behaviors were observed after the initiation of the treatments. TUNEL assay, Western Blot, IHC assay, real-time PCR, bioinformatics tools and luciferase assays were performed to examine the effect of FN stimulation on the expression of miR-182, miR-382 and BDNF mRNA/protein, as well as to further clarify the role of miR-382miR-182/BDNF mRNA signaling pathways in FN stimulation. Post-stroke depression-like behaviors were significantly reduced in PSD rats. In contrary, the treatment by FN stimulation alleviated the symptoms of PSD and reduced the apoptosis index in the PSD group. Furthermore, in the PSD group, BDNF mRNA/protein levels were suppressed while the miR-382/miR-182 levels were both significantly up-regulated. After the treatment of FN stimulation, BDNF mRNA/protein levels were partly recovered, while miR-382/miR-182 levels was decreased. Furthermore, BDNF was identified as a virtual target of miR-382 and miR-182. In conclusion, FN stimulation increases the expression of BDNF via down-regulating the expression of miR-382/miR-182, thus exhibiting a positive effect in the management of PSD.

Abstract

Background and aims Repetitive transcranial magnetic stimulation (rTMS) of the left dorsolateral prefrontal cortex (DLPFC) can effectively reduce cravings in methamphetamine use disorder (MUD). However, a considerable group still fails to respond. Cognitive and emotional disturbance, as well as impulsive features, are widespread in patients with MUD and might mediate the treatment response of rTMS. The purpose of this study is to figure out whether these variables can help predicting patients' responses to rTMS treatment. Methods Ninety-seven patients with severe MUD and thirty-one gender- and age-matched healthy subjects were included. Patients were randomized to receive 20 sessions of real or sham rTMS. Intermittent theta burst protocols (iTBS) or sham iTBS were applied every weekday over the DLPFC for 20 daily sessions. Both groups received regular treatment. Craving induced by drug-related cue was measured before and after stimulation. Cognition was evaluated by using the CogState Battery. Baseline characteristics were collected through the Addiction Severity Index, Patient Health Questionnaire-9, General Anxiety Disorder Scale-7, and Barrett Impulsivity Scale-11. Results Results showed that patients with MUD have worse spatial working memory, problem-solving ability, as well as depression and anxiety symptoms compared with healthy controls. Cognition and emotion differed between responders (craving decrease ≥60%) and non-responders in real rTMS group but not in the sham group. Better cognitive and emotional functions means that patients have higher possibility for better response to real rTMS treatment. Conclusions This study suggests that cognitive, emotional and impulsive features could be used to predict the prospective treatment responses of rTMS in patients with MUD.

Abstract

Methamphetamine is the most commonly used illicit drug in China. We previously demonstrated that 10 Hz rTMS over the DLPFC reduces craving in methamphetamine users. Here, we applied intermittent theta burst stimulation (iTBS), a new form of rTMS, in a large sample at four clinical centers. 126 participants (age 31.64 ± 6.33; 106 men) with severe methamphetamine use disorder according to DSM-5 were randomized to receive either iTBS or sham over the DLPFC for four weeks (20 daily sessions, 900 pulses per day). Cue-induced craving and cognitive function were assessed before and after rTMS intervention. Relapse was followed up by urine test after discharge from the rehabilitation center. iTBS significantly reduced craving and improved cognition and sleep quality. This study suggests that rTMS may be a useful and safe treatment option for methamphetamine use disorder, warranting future large scale trials.

Abstract

With the expansion of the scale of the Internet of Things, the transmission distance and speed of wireless networks need to be improved. Due to the limitation of power and energy of sensor nodes, data transmission is basically carried out by multi-hop relay. Collaborative beamforming is a technology that uses multiple sensor nodes to form a virtual antenna array to transmit high-gain and directional beams, which greatly enhances the transmission range. Compared with the muilt-hop network, collaborative beamforming can reduce the numbers of hop or even transmit data to the target node at one time, thus reducing the time required for routing and forwarding. This means that collaborative beamforming can transmit data more quickly and improve data throughput. In this paper, a brief introduction of cooperative beamforming is given. Then we propose an algorithm based on Collaborative beamforming. Simulation results show that the Collaborative beamforming wireless sensor networks can achieve better throughput compared with multi-hop networks.

Abstract

The understanding and prediction of wall-bounded turbulence are of great importance for industrial flows. The stress intensity ratio, Rb, is recognized as one key parameter, and its value was known as being around 0.3 in the wake layer of wall-bounded turbulence and the mixing layer. In this work, Rb is studied using Direct Numerical Simulation data extracted from the canonical flows (channel, pipe, flat plate boundary layer at various Reynolds numbers) and converging-diverging channel flow. It is found that in canonical flows and the converging-diverging channel flow, the stress intensity ratio exhibits a certain pattern if the stress intensity ratio varies with the eddy viscosity ratio r or turbulent Reynolds number Rek. Moreover, the r-Rb distribution shows advanced similarity against the Rek-Rb distribution, which is due to that r is consist of transport variables. The similarity is evidenced by the fact that the r-Rb distribution is insensitive to zero pressure gradient, positive pressure gradient, and geometric effects. The r-Rb distribution seems to be mainly sensitive to the friction Reynolds number. This priori study shows that the r-Rb distribution is of universality in flows of different geometric configurations, and it offers information for the construction and improvement of turbulence models.
