Resume Introduction La prise de phenylbutazone expose a des effets indesirables frequents, tres varies et parfois graves. Nous rapportons pour la premiere fois une observation de syndrome de Sweet associe a une sialadenite, induite par la prise de phenylbutazone. Observation Une femme de 54 ans traitee pour des lombalgies, neuf jours auparavant par de la phenylbutazone durant trois jours, voyait apparaitre une parotidite et une sous-maxillite bilaterale, puis une eruption febrile erythemateuse, œdemateuse et pustuleuse, resistant aux antibiotiques. Les examens biologiques montraient un syndrome inflammatoire majeur avec hyperleucocytose a polynucleaires neutrophiles, thrombocytose, anemie et cytolyse hepatique. Le bilan infectieux et auto-immun etait negatif. L’examen histologique d’une biopsie cutanee confirmait le diagnostic de syndrome de Sweet. Une corticotherapie generale entrainait la disparition de la symptomatologie clinique et biologique. Discussion Une sialadenite induite par la phenylbutazone est un effet secondaire rare et meconnu, qui peut s’accompagner d’une reaction systemique. Dans notre observation, le syndrome de Sweet correspond aux criteres bien definis de syndrome de Sweet medicamenteux. Il correspond vraisemblablement a une reaction systemique par hypersensibilite medicamenteuse, au meme titre que la sialadenite.

The analytical approach to the detection of residues of legally used veterinary medicinal products (VMPs) is similar to the approach of forbidden substances. The only difference lies in the quantitative component of the method. Since there is an evolution towards a different strategy in the screening for VMPs in matrices of slaughtered animals, a new approach was developed for determining the residues. The aim of this research was to create an efficient screening approach for determining the identity and/or quantity of legally and illegally used VMPs present at high concentrations in injection sites. The determination of these ‘unknown’ VMPs is combined with a fast report to the customer. Examples are given of the identification of phenylbutazone, penicillin G benzathine and florfenicol. For quantitative purposes, using a mini-validation procedure, concentrations far above the maximum residue limit (MRL) of the identified VMP can be reported. A quantitative validation normally consists of determining the required validation parameters at three levels: 1/2 MRL, MRL, 2 MRL. For highly concentrated injection sites, an alternative approach is proposed. The alternative validation consists of a comparison of the analyte concentration in the sample with the spike at the MRL and 10 times the MRL concentration. © 2002 Elsevier Science B.V. In this article the pharmacokinetic profile of several antibacterial, antiparasitic and antiinflammatory agents in camels has been reviewed. The effect of dehydration on the kinetics of these drugs has also been discussed.

Effects of phenylbutazone treatment on semen and testicular size were evaluated using 6 mature stallions. Three stallions were given 1 g phenylbutazone orally in feed twice daily for 4 weeks, and 3 stallions remained untreated. Semen samples were obtained daily for 7 consecutive days once during a 2-week baseline period (Week 2), once after 4 weeks of treatment (Week 6), and once following 1 complete spermatogenic cycle (60 days) after treatment was discontinued (Week 16). During the remaining weeks (Weeks 1,3,4,5,7,8 and 9), semen was collected twice weekly. Semen samples were evaluated using traditional and computer-assisted techniques at the time of collection and at 24 and 48 hours of storage at 4°C. Testicular volume was estimated using ultrasonography once during each period of the study. Repeated measures analysis of variance revealed no significant measurable effects of phenylbutazone treatment on semen at the time of collection or after storage at 4°C for 24 or 48 hours. Similarly, no effect on testicular volume was found.
