It is unclear whether fluid management goals are best achieved by bolus injection or continuous infusion of loop diuretics. In this study, we compared the effectiveness and safety of a continuous infusion with that of a bolus injection when an increased loop diuretic dosage is required in intensive care unit (ICU) patients.We obtained data from the MIMIC-III database for patients who were first-time ICU admissions and required an increased diuretic dosage. Patients were excluded if they had an estimated glomerular filtration rate15 ml/min/1.73 mThe final dataset included 807 patients (continuous group, n=409; bolus group, n=398). After propensity score matching, there were 253 patients in the bolus group and 231 in the continuous group. The 24h urine output per 40 mg of furosemide was significantly greater in the continuous group than in the bolus group (234.66 ml [95% CI 152.13-317.18, p0.01]). There was no significant between-group difference in the incidence of acute kidney injury (odds ratio 0.96, 95% CI 0.66-1.41, p=0.85).Our results indicate that a continuous infusion of loop diuretics may be more effective than a bolus injection and does not increase the risk of acute kidney injury in patients who need an increased diuretic dosage in the ICU.

Phospholipids are the protective layer of modern cells, but it is challenging for the formation of phospholipids that require a simple abiotic synthesis before the advent of primitive cells. Here, we reported the abiotic synthesis for lysophosphatidic acids (LPAs) with prebiotically plausible reactants in aqueous microdroplets under ambient conditions. The LPAs formation is carried out by fusing two microdroplets streams: one contains glycerol and pyrophosphate in water and the other one contains fatty acids in acetonitrile. Compared with the bulk solution, LPAs were generated in microdroplets without the addition of catalyst and heating. Conditions of reactant concentrations and microdroplet size varied and suggested that LPAs formation occurred near or at the microdroplet surface. The LPAs formation also showed chemoselective toward on chain-length of fatty acids. Finally, the formation of LPAs underwent two-step reactions with glycerol phosphorylation eliminating one water molecule followed by esterification with fatty acids. These results also implicated that pyrophosphate functioned as both catalysts and precursors in prebiotic LPAs synthesis. The approach using fusion aqueous microdroplets has desirable features in studying the substance exchange and interaction in atmosphere.

This study aimed to compare the efficiency of these two combined surgeries [prolapse and hemorrhoids (PPH) combined with the external hemorrhoidectomy and inferior internal hemorrhoid ligation; tissue selecting technique with mega-window stapler (TST-MS) combined with anal canal epithelial preservation operation].This is a single-center, evaluator-blinded randomized controlled trial (RCT). A total of 204 participants were randomly divided into the two groups. The anal function, reoccurrence, intraoperative variables, and operative complications were assessed. The measurement data were compared by paired t test and rank sum test. Chi-squared or Fisher's exact test was used for count data and rank sum test for ranked data. To assess differences within each group in different time points, the repeated-measures analysis of variance was conducted.TST-MS combined with anal canal epithelial preservation operation had lower Wexner score of anal incontinence (Z = 3.062, P = 0.002), higher patients' satisfaction degree (t = 7.32, P 0.001), less residual skin tags (χTST-MS combined with anal canal epithelial preservation operation showed better clinical efficiency in the treatment of prolapsed hemorrhoids.This study has been registered in Chinese Clinical Trial Registry (ChiCTR2000038533).

Radiofrequency ablation (RFA) is one of the effective therapeutic modalities in patients with hepatocellular carcinoma (HCC). However, there is no proper method to evaluate the HCC response to RFA. This study aimed to establish and validate a clinical prediction model based on dual-energy computed tomography (DECT) quantitative-imaging parameters, clinical variables, and CT texture parameters.We enrolled 63 patients with small HCC. Two to four weeks after RFA, we performed DECT scanning to obtain DECT-quantitative parameters and to record the patients' clinical baseline variables. DECT images were manually segmented, and 56 CT texture features were extracted. We used LASSO algorithm for feature selection and data dimensionality reduction; logistic regression analysis was used to build a clinical model with clinical variables and DECT-quantitative parameters; we then added texture features to build a clinical-texture model based on clinical model.A total of six optimal CT texture analysis (CTTA) features were selected, which were statistically different between patients with or without tumor progression (P0.05). When clinical variables and DECT-quantitative parameters were included, the clinical models showed that albumin-bilirubin grade (ALBI) [odds ratio (OR) = 2.77, 95% confidence interval (CI): 1.35-6.65, P = 0.010], λDECT-quantitative parameters, CTTA, and clinical variables were helpful in predicting HCC progression after RFA. The constructed clinical prediction model can provide early warning of potential tumor progression risk for patients after RFA.

Promoter is a key DNA element located near the transcription start site, which regulates gene transcription by binding RNA polymerase. Thus, the identification of promoters is an important research field in synthetic biology. Nannochloropsis is an important unicellular industrial oleaginous microalgae, and at present, some studies have identified some promoters with specific functions by biological methods in Nannochloropsis, whereas few studies used computational methods. Here, we propose a method called DNPPro (DenseNet-Predict-Promoter) based on densely connected convolutional neural networks to predict the promoter of Nannochloropsis. First, we collected promoter sequences from six Nannochloropsis strains and removed 80% similarity using CD-HIT for each strain to yield a reliable set of positive datasets. Then, in order to construct a robust classifier, within-group scrambling method was used to generate negative dataset which overcomes the limitation of randomly selecting a non-promoter region from the same genome as a negative sample. Finally, we constructed a densely connected convolutional neural network, with the sequence one-hot encoding as the input. Compared with commonly used sequence processing methods, DNPPro can extract long sequence features to a greater extent. The cross-strain experiment on independent dataset verifies the generalization of our method. At the same time, T-SNE visualization analysis shows that our method can effectively distinguish promoters from non-promoters.

N6,2'-O-dimethyladenosine (m6Am) is a reversible modification widely occurred on varied RNA molecules. The biological function of m6Am is yet to be known though recent studies have revealed its influences in cellular mRNA fate. Precise identification of m6Am sites on RNA is vital for the understanding of its biological functions. We present here m6AmPred, the first web server for in silico identification of m6Am sites from the primary sequences of RNA. Built upon the eXtreme Gradient Boosting with Dart algorithm (XgbDart) and EIIP-PseEIIP encoding scheme, m6AmPred achieved promising prediction performance with the AUCs greater than 0.954 when tested by 10-fold cross-validation and independent testing datasets. To critically test and validate the performance of m6AmPred, the experimentally verified m6Am sites from two data sources were cross-validated. The m6AmPred web server is freely accessible at: https://www.xjtlu.edu.cn/biologicalsciences/m6am, and it should make a useful tool for the researchers who are interested in N6,2'-O-dimethyladenosine RNA modification.

Chitin, a kind of polysaccharide mainly obtained from food waste, has emerged as an important biodegradable biopolymer in composite materials. The difficulty of aldehyde modification, which greatly limited the application of chitin nanocrystals, was addressed by applying a facile route of partial deacetylation followed by periodate oxidation in this study. Deacetylation occurred on the surface of both crystalline and amorphous regions, which were significantly degraded in the following periodate oxidation due to the inevitable cleavage of chitin chains, leading to an increase in the crystallinity index of obtained di-aldehyde chitin. The degree of deacetylation and periodate addition had limited improvement in the aldehyde content of di-aldehyde chitin with a maximum value of around 0.42 mmol/g. With further 2,2,6,6-tetramethylpiperidine-1-oxyl-mediated oxidation, the carboxyl content of tri-carboxyl chitin was improved to 1.58 mmol/g, which played a critical role in the dispersion efficiency and morphology of chitin nanocrystals. The obtained rod-like chitin nanocrystals with a ζ-potential value of -42 mV and an average size of 97 nm have potential application in dye-adsorption and emulsion stabilizers.

Saline–alkaline (SA) stress is characterized by high salinity and high alkalinity (high pH), which severely inhibit plant growth and cause huge losses in crop yields worldwide. Here we show that a moderate elevation of endogenous abscisic acid (ABA) levels by RNAi-mediated suppression of OsABA8ox1 (OsABA8ox1-kd), a key ABA catabolic gene, significantly increased tolerance to SA stress in rice plants. We produced OsABA8ox1-kd lines in two different japonica cultivars, Dongdao 4 and Nipponbare. Compared with non-transgenic control plants (WT), the OsABA8ox1-kd seedlings accumulated 25.9%–55.7% higher levels of endogenous ABA and exhibited reduced plasmalemma injury, ROS accumulation and Na+/K+ ratio, and higher survival rates, under hydroponic alkaline conditions simulated by 10, 15, and 20 mmol L−1 of Na2CO3. In pot trials using SA field soils of different alkali levels (pH 7.59, 8.86, and 9.29), OsABA8ox1–kd plants showed markedly higher seedling survival rates and more vigorous plant growth, resulting in significantly higher yield components including panicle number (85.7%–128.6%), spikelets per panicle (36.9%–61.9%), branches (153.9%–236.7%), 1000–kernel weight (20.0%–28.6%), and percentage of filled spikelets (96.6%–1340.8%) at harvest time. Under severe SA soil conditions (pH = 9.29, EC = 834.4 μS cm−1), OsABA8ox1-kd lines showed an 194.5%–1090.8% increase in grain yield per plant relative to WT plants. These results suggest that suppression of OsABA8ox1 to increase endogenous ABA levels provides a new molecular approach for improving rice yield in SA paddies.

For this study, NiCrBSi–TiN composite coatings were fabricated on Q235 steel substrates by reactive plasma spraying and the coatings were heat treated at 600 °C, 700 °C, 750 °C and 800 °C for 1 hour. The microstructure evolution of these coatings untreated and treated at different temperatures was analyzed by X-ray diffraction (XRD), scanning electron microscopy (SEM) combined with energy dispersive spectroscopy (EDS) and transmission electron microscopy (TEM). The hardness of the coatings was characterised using the Weibull distribution while the wear and corrosion resistance were studied using a block-ring tribometer and an electrochemical workstation. The phase composition of coatings after heat treatment changed, the compactness of coatings was increased, and the interlayer bonding was significantly improved. Comprehensive experimental results illustrated that the performance of the NiCrBSi–TiN composite coatings was excellent at the heat treatment temperature of 700 °C.

Here, a simple, economical one-step doping strategy is adopted to prepare N, S co-doped graphene hollow spheres (NSGHS). The NSGHS exhibits remarkable oxygen-reduction-reaction (ORR) activity with a high half-wave potential (E1/2) of 0.847 V vs. RHE and considerable long-term stability. When applied as anode materials for sodium-ion batteries (SIBs), it could deliver a high reversible capacity of 385 mAh g−1 at 500 mA g−1, excellent rate performance of 308 mAh g−1 at even 20 A g−1 and superior cycling performance at 10 A g−1 during 5000 cycles with negligible capacity fade. DFT calculation reveals that the doping of N and S endows the surrounding carbon with high positive charge and spin density, respectively, making them be highly active ORR catalytic sites except for S itself. Despite the possible oxidization of C–S–C active sites, the rest C atoms of NSGHS will still hold a considerably high catalytic activity superior to those of mono-doped or undoped cases. In addition, S doping will improve the binding between Na and graphene substrate to get a high capacity and N doping effectively curbs the rising of oxygen levels and oxidization of S in C–S–C active sites, thus the excellent stability for ORR and SIBs. © 2021 Elsevier Ltd

Background Oxacillin-susceptible mecA-positive Staphylococcus aureus (OS-MRSA) represents an important issue, as its oxacillin susceptibility has contributed to misidentification by conventional susceptibility tests and consequently potential therapeutic failure, but limited data on the current status of OS-MRSA infection in Chinese hospitals are available. Methods This multicenter study performed a battery of susceptibility tests and diagnostic tests for 956 S. aureus isolates from 10 hospitals, including automated susceptibility testing on VITEK 2, broth microdilution, disk diffusion, and detection of PBB2a, mecA gene and mecC gene. For all identified OS-MRSA, multi-locus sequence typing (MLST), together with spa typing, SCCmec typing and PVL detecting, was carried out. Results OS-MRSA, most of which were from pediatric inpatients, represented 1.8% (17/956) of total isolates. Of these 17 OS-MRSA, 10 were ST59, followed by ST965 (3/17), and 11 carried SCCmec type IV, while 5 carried SCCmec type V, but only one was Panton–Valentine leucocidin (PVL)-positive, also, 16 had one or two point mutations within mecA promoter. OS-MRSA had inducible oxacillin resistance and significantly lower MDR (Multi-Drug Resistant) rate. We observed that the VITEK 2 system exhibited some deficiency in OS-MRSA detection, whereas cefoxitin disk diffusion was shown to be a reliable and cost-saving alternative and should be supplemented in detecting S. aureus with borderline oxacillin susceptible MICs. Conclusion This study has characterized phenotypically and molecularly OS-MRSA in China, and provided insights into more effective management of OS-MRSA.

Dielectric capacitors have drawn growing attention for their wide application in future high power and/or pulsed power electronic systems. However, the recoverable energy storage density (Wrec) for dielectric ceramics is relatively low up to now, which largely restricts their actual application. Herein, the domain engineering is employed to construct relaxor antiferroelectric NaNbO3-BiFeO3 bulk ceramics, which integrates the merits of antiferroelectrics and relaxors. It is revealed that the antiferroelectric phase transforms from orthorhombic P to R phase, and antiferroelectric domain evolves from micron-sized blocks to nanoscale clusters. Of particular importance is that the 0.90NaNbO3–0.10BiFeO3 ceramic demonstrates ultrahigh Wrec of 18.5 J cm−3 with giant electric breakdown strength of 99.5 kV mm−1, which is superior than state-of-the-art bulk dielectric ceramics. Moreover, the 0.90NaNbO3–0.10BiFeO3 ceramic exhibits superior frequency, cycling and thermal reliability, as well as the substantial current density (2140.6 A cm−2), ultrahigh power density (428.1 MW cm−3) and ultrafast discharge rate (14 ns). These results not only suggest that the NaNbO3-based relaxor antiferroelectric ceramics are promising candidates for advanced energy storage capacitors, but also provide feasible strategy of domain engineering to develop novel lead-free high-performance dielectric materials.

Water distribution Networks (WDNs) are one of the most important infrastructures for modern society. Due to accidental or malicious reasons, water contamination incidents have been repeatedly reported all over the world, which not only disrupt the water supply but also endanger public health. To ensure the safety of WDNs, water quality sensors are deployed across the WDNs for real-time contamination detection and source identification. In the literature, various methods have been employed to improve the performance of contamination source identification (CSI) and recent studies show that there is a great potential to tackle the CSI problem by deep learning models. The success of deep learning based CSI methods often requires a large size of training samples being collected. In real-world situations, the number of contamination events occurring in a single WDN is rather small, especially for a newly built WDN. However, the existing CSI methods in the literature mostly focus on the study of training and applying models on the same WDNs and the knowledge of CSI gained from one WDN cannot be reused by a different WDN. To these ends, based on the application of graph convolutional networks, this paper provides a solution for cross-network CSI that can transfer the CSI knowledge learned from one WDN to a different WDN. Empirically, based on a benchmark WDN in the task of contamination source identification, we show that the proposed cross-network CSI method can achieve comparable accuracy even trained on a different WDN.

Mimicking biological synapses with microelectronic devices is widely considered as the first step in hardware building artificial neuromorphic networks, which is also the basis of brain-inspired neuromorphic computing. Numerous artificial neurons and synapses making up an artificial neuromorphic network have been gained wide attention due to their powerful and efficient data processing capabilities. Recently, artificial synapses, especially memristor-type and transistor-type synapses based on multifarious two-dimensional (2D) materials have been paid much attention. The unique properties of 2D materials make devices perform well in learning ability and power efficiency when mimicking synaptic behaviors, which highlights the feasibility of 2D neuromorphic devices in constructing artificial neuromorphic networks. Herein, the basic structures and principles of biological synapses are introduced, and the definitions of synaptic behaviors in synaptic electronic devices are discussed. Then, the progress of 2D memristor-type and transistor-type neuromorphic devices involving their device architecture, neuromorphic operational mechanism, and promising applications is reviewed. Finally, the future challenges of artificial synaptic devices based on 2D materials are discussed briefly.
