Abstract

Resume Introduction L’infection urinaire (IU) constitue la deuxieme cause d’infection bacterienne communautaire chez le sujet âge. Ce diagnostic est difficile car les symptomes sont souvent atypiques. Nous presentons les caracteristiques des patients ayant une IU parmi une population geriatrique (signes fonctionnels urinaires et/ou syndrome infectieux). Nous avons egalement evalue la sensibilite et la specificite de la bandelette urinaire (BU) pour le diagnostic d’IU dans cette population. Patients et methode Nous avons realise une etude observationnelle, prospective, monocentrique, d’avril 2017 a janvier 2018, incluant tous les patients hospitalises en geriatrie, presentant des signes fonctionnels urinaires et/ou un syndrome infectieux sans point d’appel, pour lesquels un examen cytobacteriologique des urines (ECBU) a ete demande. Une BU etait systematiquement realisee pour tous ces patients. Les caracteristiques cliniques et biologiques des patients ont ete etudiees selon le diagnostic definitif. En outre, les resultats des BU ont ete compares a ceux des ECBU en cas de diagnostic final d’IU. Resultats Au total, 165 patients ont ete inclus. Parmi eux, 67 (40,6 %) avaient une IU et 98 (59,4 %) avaient un autre diagnostic. Il y avait significativement plus de femmes ( p p La BU presentait une sensibilite elevee (92 %) et une VPN elevee (91 %) interessantes dans notre population. Conclusion En cas d’IU chez le sujet âge, peu de criteres sont discriminants avec une autre origine. Par ailleurs, une BU negative suggere l’absence d’IU en raison de sa bonne valeur predictive negative.

Abstract

Resume La prescription d’exercice repose generalement sur la determination des seuils ventilatoires (SV1, SV2) lors d’une epreuve d’effort (CPX). Chez le sujet sain, l’atteinte des seuils ventilatoires est contemporaine de modifications de la valeur RMS de l’activite electromyographique de surface du vastus lateralis (EMGth1, EMGth2). Objectif de l’etude Observer si EMGth1 et EMGth2 apparaissent en concordance avec SV1 et SV2 chez le patient cardiaque. Patients et methode Trente-quatre sujets (62,1 ± 7,3 ans, 172,1 ± 6,3 cm, 81,3 ± 15,3 kg, IMC : 27,3 ± 4,1) ont realise un CPX sur bicyclette a 60 rpm. SV1 correspondait a la cassure dans la relation dioxyde de carbone produit (CO2) – consommation d’oxygene (VO2) ; SV2 a l’intensite pour laquelle l’equivalent respiratoire en CO2 augmentait. EMGth1 et EMGth2 etaient definis comme les premier et second points de rupture dans la relation RMS-intensite. Resultats Les valeurs pics de VO2 (16,3 ± 4,6 mL·min−1·kg−1) et de frequence cardiaque (106,7 ± 13,8 bpm) ont ete atteintes a 112,9 ± 38,5 w (PMT) ; SV1 et SV2 a 71,1 ± 25,9 w (62,5 ± 5,5 % de PMT) et 87,9 ± 28,6 w (78,0 ± 5,1 % de PMT). Chez ces sujets, la relation RMS-intensite presentait deux points de cassure, identifies comme EMGth1 a 68,0 ± 24,7w et EMGth2 a 88,5 ± 30,1w, soit a 60,0 ± 7,6 et 78,6 ± 5,0 % de PMT. Une difference significative a ete observee entre les intensites a EMGth1 et SV1 (p = 0,004), mais pas entre EMGth2 et SV2 (p = 0,13) avec un faible effet de la taille de l’echantillon. Conclusion L’apparition de EMGth1 avant SV1 suppose un lien de causalite entre conditionnement musculaire et reponses ventilatoires a l’effort ; lien a prendre en compte dans la prescription d’activites physiques adaptees du patient cardiaque.

Abstract

The gopher tortoise tick, Amblyomma tuberculatum, has a unique relationship with the gopher tortoise, Gopherus polyphemus, found in sandy habitats across the southeastern United States. We aimed to understand the overall bacterial community associated with A. tuberculatum while also focusing on spotted fever group Rickettsia. These tortoises in the Southern Mississippi region are a federally threatened species; therefore, we have carefully trapped the tortoises and removed the species-specific ticks attached to them. Genomic DNA was extracted from individual ticks and used to explore overall bacterial load using pyrosequencing of bacterial 16S rRNA on 454-sequencing platform. The spotted fever group of Rickettsia was explored by amplifying rickettsial outer membrane protein A (rompA) gene by nested PCR. Sequencing results revealed 330 bacterial operational taxonomic units (OTUs) after all the necessary curation of sequences. Four whole A. tuberculatum ticks showed Proteobacteria, Firmicutes, Actinobacteria and Bacteroidetes as the most dominant phyla with a total of 74 different bacterial genera detected. Together Rickettsiae and Francisella showed >85% abundance, thus dominating the bacterial community structure. Partial sequences obtained from ompA amplicons revealed the presence of an uncharacterized Rickettsia similar to the Rickettsial endosymbiont of A. tuberculatum. This is the first preliminary profile of a complete bacterial community from gopher tortoise ticks and warrants further investigation regarding the functional role of Rickettsial and Francisella-like endosymbionts in tick physiology.

Abstract

Introduction La fasciite necrosante est une infection rare des tissus sous-cutanes atteignant le fascia. Sa mortalite s’eleve a 30 % et sa prise en charge est urgente. L’immunodepression est un facteur de risque reconnu. De causes bacteriennes, elles peuvent etre mono ou polymicrobiennes, a germes aero- ou anaerobies. Les streptocoques du groupe A, enterobacteries, P. aeruginosa et les anaerobies sont le plus souvent impliques. L’origine mycologique est tres rare et exclusivement reservee aux immunodeprimes. Les champignons responsables decrits sont : Mucorales, Aspergillus et Candida. Aucun cas de fasciite necrosante humaine a Nannizziopsis spp. n’etait encore decrit. Observations Un homme africain de 56 ans vivant en France depuis longtemps est greffe renal depuis 2013. Il prend quotidiennement du mycophenolate mofetil, tacrolimus, prednisolone et oracilline. Il n’a pas voyage recemment et n’est pas seropositif VIH. Il est hospitalise en reanimation pour un choc septique sans porte d’entree trouvee avec un traitement probabiliste initial par imipeneme. Quatre jours apres l’admission, il presente une douleur intense de la jambe droite avec apparition de lesions cutanees evocatrices de fasciite necrosante motivant une prise en charge chirurgicale urgente ( Fig. 1 ). Les hemocultures initiales sont positives a S. marcescens, K. pneumonia et P. aeruginosa mais les prelevements peroperatoires ne les retrouveront pas. Ils objectiveront un champignon filamenteux contre lequel on adjoindra a la tazocilline-daptomycine initiee en postoperatoire immediat un traitement par amphotericine B. L’identification de Nannizziopsis spp. et son antifongigramme en centre de reference ont permis le relais par voriconazole. L’ETO n’objective pas d’endocardite, l’IRMc, la PL, l’examen ORL sont normales. La TDM TAP objective des images suspectes hepatiques peut-etre en lien avec une infection invasive a Nannizziopsis spp. L’examen cutane objective des nodules sous-cutanes diffus dont l’anatomopathologie avec coloration de Gomori-Grocott est en attente ( Fig. 2 ). Discussion Nannizziopsis spp. est un champignon keratinophilique connu des veterinaires pour causer des dermatophytoses et granulomes chez les reptiles. Sept cas d’infections invasives humaines a Nannizziopsis ont ete decrites chez les immunodeprimes mais aucune sous forme de fasciite necrosante. Les patients presentaient soit une atteinte pulmonaire, soit des nodules cerebraux, soit des osteites. Ici, comme dans les autres cas decrits, le patient est d’origine africaine mais il n’avait cependant pas voyage recemment. Il n’est pas en contact avec des reptiles. L’imipeneme donne initialement a pu favoriser le developpement fongique intense sur ce terrain. Conclusion Nous rapportons le premier cas de fasciite necrosante a Nannizziopsis spp. chez un patient greffe renal. Nannizziopsis spp. semble etre un champignon emergent en pathologie humaine de l’immunodeprime.

Abstract

SummaryStructure-specific endonucleases resolve DNA secondary structures generated during DNA repair and recombination. The yeast 5′ flap endonuclease Slx1-Slx4 has received particular attention with the finding that Slx4 has Slx1-independent key functions in genome maintenance. Although Slx1 is a highly conserved protein in eukaryotes, no orthologs of Slx4 were reported other than in fungi. Here we report the identification of Slx4 orthologs in metazoa, including fly MUS312, essential for meiotic recombination, and human BTBD12, an ATM/ATR checkpoint kinase substrate. Human SLX1-SLX4 displays robust Holliday junction resolvase activity in addition to 5′ flap endonuclease activity. Depletion of SLX1 and SLX4 results in 53BP1 foci accumulation and H2AX phosphorylation as well as cellular hypersensitivity to MMS. Furthermore, we show that SLX4 binds the XPFERCC4 and MUS81 subunits of the XPF-ERCC1 and MUS81-EME1 endonucleases and is required for DNA interstrand crosslink repair. We propose that SLX4 acts as a docking platform for multiple structure-specific endonucleases.

Abstract

We study the beam-energy and system-size dependence of phi meson production (using the hadronic decay mode phi -> K(+) K(-)) by comparing the new results from Cu + Cu collisions and previously reported Au + Au collisions at root s(NN) = 62.4 and 200 GeV measured in the STAR experiment at RHIC. Data presented in this Letter are from mid-rapidity (vertical bar y vertical bar ) and net strangeness = 2 (Xi). The enhancement for phi mesons is observed to be higher at root s(NN) = 200 GeV compared to 62.4 GeV. These observations for the produced phi(s (s) over bar) mesons clearly suggest that, at these collision energies, the source of enhancement of strange hadrons is related to the formation of a dense partonic medium in high energy nucleus-nucleus collisions and cannot be alone due to canonical suppression of their production in smaller systems. Abstract

We study the energy dependense of the transverse momentum (PT) spectra for charged pions, protons and anti-protons for An + An collisions at root(NN)-N-s = 62.4 and 200 GeV. Data are presented at mid-rapidity (broken vertical bar y broken vertical bar 7 GeV/c) the modification is similar for both energies. The p/pi(+) and (p) over bar/pi(-) ratios for central collisions at root(NN)-N-s = 62.4 GeV peak at PT similar or equal to 2 GeV/c. In the PT range where recombination is expected to dominate, the p/pi(+) ratios at 62.4 GeV are larger than at 200 GeV, while the (p) over bar/pi(-) ratios are smaller. For PT > 2 GeV/c, the (p) over bar/pi(-) ratios at the two beam energies are independent of P-T and centrality indicating that the dependence of the (p) over bar/pi(-) ratio on P-T does not change between 62.4 and 200 GeV. Abstract

The environment of space offers special challenges for the lubrication of components in sliding and rolling mechanisms. Hydrogenated diamond-like carbon (DLC) films are being studied as solid lubricant coatings to simultaneously fulfil specifications regarding wear resistance and low friction behaviour under ambient atmosphere and in vacuum. In this paper, the tribological behaviour of highly hydrogenated DLC coatings ( ∼ 50 at% hydrogen) is assessed. Coating composition was optimised on flat AISI 52100 steel substrates based on ball-on-disc tribotest results in air, vacuum and dry nitrogen environments. The developed DLC coatings can be tailored to yield ultra-low friction values in vacuum ( μ = 0.008 ) . The average friction coefficient range obtained in humid air, dry nitrogen and vacuum for the range of applied loads were, respectively, 0.22 to 0.27, 0.02 to 0.03, and 0.007 to 0.013. New in this work is that optimised DLC coatings were applied to ball bearings for space applications. The torque and life tests of coated pairs of angular contact bearings in air revealed that relatively high bearing torques are generated which increase with time, but the amount of coating wear generated during in-air operation appears relatively light. In vacuum, low torques are generated after a prolonged running-in period. Low-torque life exceeds that observed for MoS 2 by a factor of about two. It is concluded that, in contrast to MoS 2 coated bearings, DLC-coated bearings for space applications might therefore be capable of undergoing in-air ground testing without too much disruption of the subsequent in-space performance.

Abstract

We present the first measurements of charge-dependent correlations on angular difference variables η 1 − η 2 (pseudorapidity) and ϕ 1 − ϕ 2 (azimuth) for primary charged hadrons with transverse momentum 0.15 ⩽ p t ⩽ 2 GeV / c and | η | ⩽ 1.3 from Au–Au collisions at s N N = 130 GeV . We observe correlation structures not predicted by theory but consistent with evolution of hadron emission geometry with increasing centrality from one-dimensional fragmentation of color strings along the beam direction to an at least two-dimensional hadronization geometry along the beam and azimuth directions of a hadron-opaque bulk medium.

Abstract

Identified mid-rapidity particle spectra of pi(+/-), K+/-, and p((p) over bar) from 200 GeV p + p and d + Au collisions are reported. A time-of-flight detector based on multi-gap resistive plate chamber technology is used for particle identification. The particle-species dependence of the Cronin effect is observed to be significantly smaller than that at lower energies. The ratio of the nuclear modification factor (R-dAu) between protons (p + (p) over bar) and charged hadrons (h) in the transverse momentum range 1.2 < p(T) < 3.0 GeV/c is measured to be 1.19 +/- 0.05(stat) +/- 0.03(syst) in minimum-bias collisions and shows little centrality dependence. The yield ratio of (p + (p) over bar)/h in minimum-bias d + Au collisions is found to be a factor of 2 lower than that in Au + Au collisions, indicating that the Cronin effect alone is not enough to account for the relative baryon enhancement observed in heavy ion collisions at RHIC. Abstract

Functional studies strongly suggest that the Mus81-Eme1 complex resolves Holliday junctions (HJs) in fission yeast, but in vitro it preferentially cleaves flexible three-way branched structures that model replication forks or 3' flaps. Here we report that a nicked HJ is the preferred substrate of endogenous and recombinant Mus81-Eme1. Cleavage occurs specifically on the strand that opposes the nick, resulting in resolution of the structure into linear duplex products. Resolving cuts made by the endogenous Mus81-Eme1 complex on an intact HJ are quasi-simultaneous, indicating that Mus81-Eme1 resolves HJs by a nick and counternick mechanism, with a large rate enhancement of the second cut arising from the flexible nature of the nicked HJ intermediate. Recombinant Mus81-Eme1 is ineffective at making the first cut. We also report that HJs accumulate in a DNA polymerase alpha mutant that lacks Mus81, providing further evidence that the Mus81-Eme1 complex targets HJs in vivo.

Abstract

Second harmonic non-linear susceptibilities of Pd(1 1 1) are derived at 800 nm fundamental wavelength from measurements of second harmonic generation (SHG) as a function of azimuth and input and output polarizations. By contrast to other metal surfaces and to other wavelengths, Pd(1 1 1) does not exhibit any anisotropy related to azimuth at 800 nm. SHG is also measured during CO adsorption at 1064 and 800 nm. Opposite trends are observed at the two wavelengths. The results are discussed in the light of previously reported experiments on Pd(1 1 1) at other wavelengths and on other metals. At 800 nm, free electrons and/or the Shockley surface state probably dominate the SHG response of Pd(1 1 1). CO chemisorption probably reduces the density of the d-band states probed at 1064 nm, and on the contrary enhances the density of the states probed at 800 nm.

Abstract

Mus81, a protein with homology to the XPF subunit of the ERCC1-XPF endonuclease, is important for replicational stress tolerance in both budding and fission yeast. Human Mus81 has associated endonuclease activity against structure-specific oligonucleotide substrates, including synthetic Holliday junctions. Mus81-associated endonuclease resolves Holliday junctions into linear duplexes by cutting across the junction exclusively on strands of like polarity. In addition, Mus81 protein abundance increases in cells following exposure to agents that block DNA replication. Taken together, these findings suggest a role for Mus81 in resolving Holliday junctions that arise when DNA replication is blocked by damage or by nucleotide depletion. Mus81 is not related by sequence to previously characterized Holliday junction resolving enzymes, and it has distinct enzymatic properties that suggest it uses a novel enzymatic strategy to cleave Holliday junctions.

Abstract

Mus81, a fission yeast protein related to the XPF subunit of ERCC1-XPF nucleotide excision repair endonuclease, is essential for meiosis and important for coping with stalled replication forks. These processes require resolution of X-shaped DNA structures known as Holliday junctions. We report that Mus81 and an associated protein Eme1 are components of an endonuclease that resolves Holliday junctions into linear duplex products. Mus81 and Eme1 are required during meiosis at a late step of meiotic recombination. The mus81 meiotic defect is rescued by expression of a bacterial Holliday junction resolvase. These findings constitute strong evidence that Mus81 and Eme1 are subunits of a nuclear Holliday junction resolvase.

Abstract

Component PP3 is a phosphoglycoprotein isolated from bovine milk with unknown biological function, which displays in its C-terminal region a basic amphipathic α-helix, a feature often involved in membrane association. According to that, the behaviour of PP3 and of a synthetic peptide from the C-terminal domain (residues 113–135) was investigated in lipid environment. Conductance measurements indicated that the peptide was able to associate and form channels in planar lipid bilayers composed of neutral or charged phospholipids. Electrostatic interactions seemed to promote voltage-dependant channel formation but this was not absolutely required since the pore-forming ability of the 113–135 C-terminal peptide was also detected with the zwitterionic lipid bilayer. Additionally, a spectroscopic study using circular dichroism argues that the peptide adopts an α-helical conformation in interaction with neutral or charged micelles. Thus, the conducting aggregates in bilayers might be composed of a bundle of peptides in helical conformation. Besides, similar conductance measurements performed with the whole PP3 protein did not induce any channel fluctuations. However, with the latter, an early breakdown of the bilayers occurred, a finding that can be tentatively explained by a massive incorporation of PP3. In the light of the present results, it could be inferred that PP3 membrane attachment may be achieved by oligomerization of the C-terminal amphipathic helical region.

Une nouvelle methode de modelisation des potentiels evoques recueillis a partir d'une configuration a plusieurs electrodes est proposee. Le modele multidimensionnel derive dune representation d'etat d'un systeme a une entree et plusieurs sorties selon les principes de la theorie des systemes lineaires. La reponse evoquee est consideree comme etant la reponse impulsionnelle d'un systeme lineaire. Le choix de la methode de Prony, pour l'estimation des parametres du modele, est justifie par la robustesse de cette derniere et sa specificite pour ce genre de modelisation. Une estimation correcte du modele multidimensionnel du potentiel evoque, selon l'approche proposee, ne necessite qu'une moyenne d'au plus 100 reponses elementaires. Enfin, notant que cette methode est bien adaptee pour une application temps reel.

Abstract

Reaction sintering of (3Al 2 O 3 + 2SiO 2 ) powder compacts was studied using either a conventional electric furnace or a 2·45 GHz microwave furnace. Special attention was paid to temperature measurement within the microwave furnace. The reality of a microwave effect that accelerates the kinetics and therefore, decreases the temperatures of mullitization and densification, remains uncertain. In any case, such an effect does not exceed ≈ 50 °C, which is in the order of the temperature gradient between the core and the surface of microwave heated specimens.

Abstract

Etomidate pharmacokinetics were compared in 12 children (P group) (age 7 to 13 years, weight 22 to 48 kg) and in 4 adult women (A group) (age 28 to 52 years, weight 46 to 72 kg), A.S.A. 1, undergoing minor non abdominal surgery. They were unpremedicated and anaesthetized with alfentanil 100 μg · kg−1 and isoflurane 2 vol % in N2O/O2 (1/1). Etomidate was administered as a bolus : 0.3 mg · kg−1 in adults and 0.4 mg · kg−1 in children. Venous plasma samples were frozen until further etomidate assay with a HPLC technique. In all patients but two children, data were fitted to a three rather than a two compartment model. Differences between groups (mean ± SD values) included Vdc (P : 0.66 ± 0.31 l · kg−1 ; A : 0.27 ± 0.15 l · kg−1 ; p < 0.01), t1/2π (P : 5.4 ± 2.9 min ; A : 2.7 ± 5.7 min ; p < 0.05) and plasma clearance (P : 17.2 ± 4.6 ml · kg−1 · min−1 ; A : 10.9 ± 3.3 ml · kg−1 · min−1 ; p < 0.05). No statistical difference was found between A and P groups for the following parameters : t1/2 α (37.1 ± 12.0 min vs 26.8 + 15.1 min), t1/2 β (260 ± 99 min vs 175 ± 99 min), Vdss (2.5 ± 1.1 l · kg−1 vs 2.8 ± 1.6 l · kg−1), Vdβ (4.1 ± 2.4 l · kg−1 vs 4.0 ± 2.2 l · kg−1), and MRT (228 ± 80 min vs 172 ± 101 min). No age-related difference was found inside P group with regard to pharmacokinetic parameters. In conclusion, a 30 % higher etomidate bolus dosage is required in children than in adults to achieve similar plasma concentrations, due to a higher volume of the initial compartment. In comparison to adults the higher clearance suggests higher maintenance dose requirements in children.

Abstract

In the stallion testis, aromatase activity was localized in the microsomal fraction. Androgen aromatization occurred through the loss of 1 beta,2 beta hydrogen atoms and appeared to involve free sulfhydryl groups. A single enzyme system seemed to aromatize androgen and norandrogen at the same rate while having a much lower affinity for norandrogens.
